1585 Erianin Inhibits the Malignant Progression of Esophageal Cancer Cells through CCL2-CCR2 Signaling Axis-Mediated Immune Evasion

Objective: To investigate the impact and mechanism of erianin on the malignant progression of esophageal cancer (EC) cells. Methods: The cancer tissues and adjacent tissues 3cm away from the cancer tissues of EC patients (40cases) who underwent radical surgery in our hospital from March 2021 to December 2022 were collected. Real-time fluorescence quantitative PCR (qRT-PCR) and western blot were applied to detect the mRNA and protein expression levels of CCL2 and CCR2 in EC tissues/cells, respectively; ECA109 cells were treated with different concentrations of erianin (0, 10, 20, 40, 60, 80, 160 nmoL/L) for 48 h, and cell proliferation was detected by MTT assay to screen the optimal intervention concentration. ECA109 cells were divided into control group (normal culture), low, medium and high dose erianin groups (20,40,60nmol/L), pcDNA group (transfected with pcDNA3.1), and pcDNA-CCL2 group (transfected with pcDNA3.1-CCL2), MTT assay, plate cloning assay, transwell chamber and flow cytometry were applied to detect cell proliferation, migration, invasion and apoptosis, respectively; after co-culturing ECA109 cells and CD8+T cells, they were grouped into T cell group, co-culture group, erianin group, pcDNA group, and pcDNA-CCL2 group. MTT and flow cytometry were applied to detect CD8+T cell proliferation and apoptosis, enzyme-linked immunosorbent assay (ELISA) was applied to detect the levels of interferon-γ (IFN-γ), interleukin-2 (IL-2), and tumor necrosis factor-α (TNF-α) in the supernatant of CD8+T cells; Western blot was applied to detect the expression of proliferating cell nuclear antigen (PCNA), Bcl-2 associated protein (Bax), programmed death ligand 1 (PD-L1), and CCL2-CCR2 signaling pathway protein in cells. Results: The expression levels of CCL2, CCR2 proteins, and mRNAs increased in EC tissues and EC cells (P<0.05); compared with the control group, the OD490 value, colony formation number, migration and invasion cell numbers, CCL2, CCR2 mRNA, PCNA, PD-L1, CCL2, and CCR2 protein expression levels in ECA109 cells in the low, medium, and high dose erianin groups were obviously reduced, the apoptosis rate and Bax protein expression level were obviously increased (P<0.05); compared with the pcDNA group, the changes in the above indicators of ECA109 cells in the pcDNA-CCL2 group were obviously reversed (P<0.05); compared with the T cell group, the OD490 value, IFN-γ, IL-2, and TNF-α levels of CD8+T cells in the co-culture group decreased, while the cell apoptosis rate increased (P<0.05); compared with the co-culture group, the OD490 value, IFN-γ, IL-2, and TNF-α levels of CD8+T cells in the erianin group increased, while the cell apoptosis rate decreased (P<0.05); compared with the pcDNA group, the changes in the above indicators of CD8+T cells in the pcDNA-CCL2 group were obviously reversed (P<0.05). Conclusion: Erianin may inhibit the malignant progression of EC cells by inhibiting the CCL2-CCR2 signaling pathway.

1593 Effect of Mesenchymal Stem Cell-Derived Exosomes Carrying miR-122a in Diabetic Cardiomyopathy

Objective: To explore the effects of mesenchymal stem cell derived exosomes carrying miR-122a on diabetic cardiomyopathy (DCM) and the related molecular mechanisms. Methods: miR-122a and miR-NC were transfected into isolated mouse bone marrow derived mesenchymal stem cells (BMSC), and BMSC exosomes (BMSC-Exos) were extracted and identified by western blot. Thirty C57/B6 mice were divided into control group, miR-122a group and miR-NC group. In addition to the control group, the miR-122a group and the miR-NC group were treated with high-fat and high-sugar diet combined with STZ to induced DCM model, and then injected with BMSC-Exos-miR-122a and BMSC-Exos-miR-NC through caudal vein, respectively. The control group was intraperitoneally injected with the same amount of sodium citrate buffer. At the end of the experiment, cardiac function indexes of mice in each group were detected. The pathological level of myocardial tissue was detected by HE, the levels of ROS, IL-1β, IL-6 and TNF-α were detected by ELISA, and the protein expression levels of LC3Ⅱ, LC3Ⅰ, Beclin1 and γH2AX in myocardial tissue were detected by western blot. Results: After transfection, compared with BMSC-miR-NC, the expression of miR-122a in BMSC-miR-122a was increased(P<0.05). The extracted BMSC-Exos showed a typical Exo morphology with a particle size of about 100 nm and high expression of exosome marker proteins CD63 and CD81. Compared with the control group, end-diastolic ventricular septal thickness (IVSd), end-diastolic posterior wall thickness (PWd), left ventricular short-axis shortening rate (LVFS), stroke output (SV) and cardiac output (CO) were decreased(P<0.05), while left ventricular end-diastolic diameter (LVDd), left ventricular end-systolic diameter (LVDs) and left ventricular mass (LVM) were increased(P<0.05), the myocardial structure was disturbed and inflammatory cell infiltration increased, and the levels of ROS, IL-1β, IL-6 and TNF-α in myocardial tissue were increased(P<0.05), the expression levels of LC3Ⅱ/LC3Ⅰ and Beclin1 protein were decreased(P<0.05), while the expression levels of γH2AX protein were increased in miR-NC group(P<0.05). Compared with miR-NC group, IVSd, PWd, LVFS, SV and CO were increased(P<0.05), while LVDd, LVDs and LVM were decreased(P<0.05), the myocardial structure recovered and inflammatory cell infiltration decreased, and the levels of ROS, IL-1β, IL-6 and TNF-α were decreased(P<0.05), LC3Ⅱ/LC3Ⅰ and Beclin1 protein expression levels were increased(P<0.05), while γH2AX protein expression levels were decreased in miR-122a group(P<0.05). Conclusion: DCM can significantly reduce the autophagy level of cardiomyocytes. BMSC-Exos carrying miR-122a can improve disease symptoms in DCM by restoring autophagy levels of cardiomyocytes, reduce inflammatory levels and cell damage.

1600 The Impacts and Mechanism of lncRNA LINC01194 on miR-302e in Non-Small Cell Lung Cancer

Objective: To explore the impacts and mechanism of long non-coding RNA (lncRNA LINC01194) on miR-302e in non-small cell lung cancer (NSCLC). Methods: Real-time fluorescence quantitative PCR (qRT-PCR) was applied to detect the expression of lncRNA LINC01194 and miR-302e in NSCLC tissue, paracancerous tissue, human lung normal epithelial cells BEAS-2B, and NSCLC cell lines, respectively, to screen the best intervention cell line; NCI-H1975 cells were divided into: control group (normal culture), si-NC group (transfected with si-NC), si-LINC01194 group (transfected with si-LINC01194), si-LINC01194+anti miR-NC group (co-transfected with si-LINC01194 and anti miR-NC), and si-LINC01194+anti miR-302e group (co-transfected with si-LINC01194 and anti miR-302e); qRT-PCR was applied to detect the expression of lncRNA LINC01194 and miR-302e in cells; MTT assay, plate cloning assay, Transwell chamber flow cytometry were applied to detect cell proliferation, migration, invasion and apoptosis, respectively; Western blot was applied to detect the expression of proliferating cell nuclear antigen (PCNA), matrix metalloproteinase-2 (MMP-2), B-cell lymphoma protein 2 (Bcl-2), and Bcl-2 associated X protein (Bax); and double Luciferase reporter gene experiment was applied to verify the targeting regulation relationship between lncRNA LINC01194 and miR-302e. Results: In NSCLC tissues and cells, the expression level of lncRNA LINC01194 increased, while the expression level of miR-302e decreased (P<0.05); inhibiting the expression of lncRNA LINC01194 was able to obviously inhibit the proliferation, migration, and invasion of NCI-H1975 cells, and promote cell apoptosis (P<0.05); inhibiting miR-302e expression was able to reverse the inhibitory effect of lncRNA LINC01194 expression on NCI-H1975 cell proliferation, migration, invasion, and promote cell apoptosis (P<0.05); double Luciferase reporter gene experiment confirmed that there was a targeted regulatory relationship between lncRNA LINC01194 and miR-302e. Conclusion: lncRNA LINC01194 is up-regulated and miR-302e is down-regulated in NSCLC tissues and cells, inhibiting lncRNA LINC01194 can up-regulate the expression of miR-302e, thereby inhibiting the proliferation, migration, and invasion of NSCLC cells and promoting their apoptosis.

1606 Mechanisms of SIRT1 in M/CMCS for Alveolar Bone Loss and TLR4/NF-κB Signaling in Chronic Periodontitis

Objective: To investigate the mechanism of SIRT1 in the treatment of alveolar bone loss and TLR4/NF-κB signaling in rats with chronic periodontitis by metronidazole carboxymethyl chitosan gel. Methods: Fifty rats were randomly divided into five groups: NC group, Model group, M/CMCS group, MNZ group and M/CMCS+EX527 group, with 10 rats in each group. Clinical indicators of periodontal tissues (BI, PD) were measured in the rats. ELISA was used to measure the levels of IL-6, IL-1βand TNF-α in serum. Micro-CT was used to measure the bone mineral density (BMD), bone volume/total volume (BV/TV), and the distance between the cemento-enamel junction (CEJ) and alveolar bone crest (ABC). HE staining was performed to examine the histopathological changes in periodontal tissues. Protein immunoblotting was used to detect the expression of TLR4, NF-κB p65, and SIRT1 proteins in periodontal tissues. Results: Compared to the NC group, the rats in the model group showed significant increases in PD (2.28±0.21), BI (3.76±0.30), levels of TNF-α (234.05±11.62), IL-1β (203.62±15.31), and IL-6 (181.35±11.27) in serum, CEJ-ABC distance (615.34±51.45), and protein expression of TLR4 (0.53±0.06) and NF-κB p65 (0.75±0.08) in periodontal tissues, as well as decreases in BMD (0.32±0.03), BV/TV (31.02±1.57), and protein expression of SIRT1 (0.21±0.03) in periodontal tissues (P<0.0001). Compared to the model group, the M/CMCS group and MNZ group showed significant decreases in PD (0.81±0.08), (1.23±0.13), BI (1.24±0.12), (1.89±0.15), levels of TNF-α (75.81±7.54), (115.36±8.23), IL-1β (93.21±8.15), (122.48±9.02), and IL-6 (76.81±6.32), (105.04±7.11) in serum, CEJ-ABC distance (276.17±27.28), (361.35±30.94), and protein expression of TLR4 (0.21±0.03), (0.28±0.03) and NF-κB p65 (0.36±0.04), (0.42±0.05) in periodontal tissues, as well as increases in BMD (0.54±0.05), (0.42±0.05), BV/TV (45.91±2.06), (39.87±2.41), and protein expression of SIRT1 (0.72±0.08), (0.25±0.06) in periodontal tissues, with the M/CMCS group showing the most significant changes (P<0.0001). Compared to the M/CMCS group, the rats in the M/CMCS+EX527 group showed significant increases in BI (3.18±0.27), PD (2.10±0.18), levels of TNF-α (208.54±10.81), IL-1β (191.25±12.37), and IL-6 (173.26±10.04) in serum, CEJ-ABC distance (601.82±50.29), and protein expression of TLR4 (0.50±0.05) and NF-κB p65 (0.71±0.07) in periodontal tissues, as well as decreases in BMD (0.35±0.04), BV/TV (35.62±1.64), and protein expression of SIRT1 (0.25±0.03) in periodontal tissues (P<0.0001). Conclusion: Metronidazole carboxymethyl chitosan gel can promote alveolar bone reconstruction and repair periodontal tissues in rats with chronic periodontitis and inhibit TLR4/NF-κB signaling pathway by a mechanism related to the activation of SIRT1 expression.

1612 Significance of Methylation of Dwarf Homology Box Gene 2 and Ras-Related Structural Domain Protein Family 1A Genes in Non-Small Cell Lung Cancer

Objective: To explore the relationship between the gene methylation of dwarf stature homeobox-containing gene (SHOX2) and Ras-related domain protein family 1A (RASSF1A) and the invasion and metastasis of non-small cell lung cancer (NSCLC). Methods: Methylation-specific PCR (MSP) was used to analyze the methylation status of SHOX2 and RASSF1A gene promoter regions in NSCLC and its corresponding adjacent tissues, and the relationship between methylation status and clinical pathological features of patients was analyzed. Results: The methylation rates of SHOX2 and RASSF1A gene promoter regions in NSCLC cancer tissues were 68.97% (80/116) and 33.62% (39/116) respectively, which were significantly higher than those in normal adjacent tissues with 12.93% (15/116) and 10.34% (12/116) (P<0.05). SHOX2 and RASSF1A gene methylation distribution had no relationship with gender, age, smoking history, tumor diameter, and differentiation degree of NSCLC patients (P>0.05), but had something to do with TNM stage, pathological classification, lymph node metastasis, and survival time (P<0.05). The methylation rate of SHOX2 gene in patients with TNM stage IIIa +IIIb was significantly higher than that in patients with stage I + II (P<0.05), and the gene methylation rate of patients with squamous carcinoma was significantly higher than that of patients with adenocarcinoma (P<0.05), and the gene methylation rate in patients with lymph node metastasis was significantly higher than that in patients without lymph node metastasis (P<0.05), and the gene methylation rate in patients with survival time <2 years was significantly higher than that in patients with survival time ≥ 2 years (P<0.05). The RASSF1A gene methylation rate in patients with TNM stage IIIa + IIIb was significantly higher than that in patients with stage I + II (P<0.05), and the gene methylation rate of patients with adenocarcinoma was significantly higher than that in patients with squamous carcinoma (P<0.05), and the gene methylation rate in patients with lymph node metastasis was significantly higher than that in patients without lymph node metastasis (P<0.05), and the gene methylation rate in patients with survival time< 2 years was significantly higher than that in patients with survival time≥ 2 years (P<0.05). Conclusion: The methylation of SHOX2 and RASSF1A gene promoter regions in NSCLC tissues may be one of the reasons for the invasion and metastasis of NSCLC. Detection of SHOX2 and RASSF1A gene promoter methylation can provide reference value for diagnosis of NSCLC and evaluation of prognosis.

1617 Analysis of the Therapeutic Effect and Mechanism of Bovine Basic Fibroblast Growth Factor on Burn Scarring

Objective: To investigate the effect and mechanism of dot-matrix laser combined with bovine basic fibroblast growth factor in the treatment of burn scarring. Methods: A total of 158 cases of post-burn scarring patients treated in our hospital from January 2021 to July 2022 were selected, and the patients were randomly divided into 79 cases each in the combined group and the control group using a simple random grouping method (randomized numerical table method). In terms of basic treatment measures, the patients in the control group received fractional laser treatment at the same time, and the combined group received bovine basic fibroblast growth factor (BBGF) treatment for 4 consecutive weeks on top of the control group treatment. The combination group received bovine basic fibroblast growth factor (BFGF) on top of the treatment in the control group for 4 consecutive weeks, and compared the levels of transforming growth factor β1 (TGF-β1) and bone morphogenetic protein 7 (BMP-7) in the scar tissues of the two groups before and after treatment, as well as the scores of the Vancouver Scar Scale (VSS), the Scar Evaluation Scale (PSAS), and the differences in the clinical efficacy and the incidence of adverse reactions. Results: Before treatment, there was no statistically significant difference in the levels of TGF-β1, BMP-7, PSAS score, scar color, blood vessel distribution, scar thickness, and softness score of scar in the scar tissues of patients in the combined group and the control group (P>0.05); after treatment, the levels of TGF-β1 in the scar tissues of the patients in the two groups were reduced, and the levels of BMP-7 increased, and the difference was statistically significant ( P<0.05); comparing the difference between the two groups before and after treatment, the decrease of TGF-β1 and the increase of BMP-7 in the combined group were greater than that in the control group, and the difference was statistically significant (P<0.05); after treatment, the scar color, vascular distribution, scar thickness, scar softness score, and PSAS score of the patients in the two groups were reduced, and the decrease of the combined group was greater than that of the control group, and the differences between the above indexes were statistically significant (P<0.05). The differences between the groups were statistically significant (P<0.05); after treatment, the overall distribution of efficacy of the patients in the combined group was better than that of the control group, and the difference was statistically significant (P<0.05); in the course of treatment, the complication rate of the combined group was 7.59%, and that of the control group was 20.25%, and the difference between the two groups was statistically significant (P<0.05). Conclusion: For patients with burn scar formation, the use of fractional laser + bovine basic fibroblast growth factor treatment has significant and reliable clinical efficacy, and can significantly promote and improve the state of scar tissue proliferation and the aesthetic degree of the patient's appearance, which is worthy of clinical promotion.

1623 Expression and Clinical Significance of miR-29b-3p and FOXO3A in Colon Cancer Tissues

Objective: To discuss the clinical significance of microRNA-29b-3p (miR-29b-3p) and forkhead box O3A (FOXO3A) in colon cancer tissues by detecting their expression level. Methods: A total of 82 patients with colon cancer treated in our hospital from January 2015 to January 2017 were selected. Colon cancer tissues and adjacent normal tissues ≥5cm from the tumor margin were collected during the operation. Real-time fluorescence quantification (qRT-PCR) was used to detect the expression levels of miR-29b-3p and FOXO3A mRNA in various tissues, at the same time, Western blot was used to detect the expression level of FOXO3A protein in each tissue. Pearson method was used to analyze the correlation between miR-29b-3p and FOXO3A mRNA expression in colon cancer; Kaplan-Meier method was used to analyze the relationship between the expression of miR-29b-3p, FOXO3A mRNA and the survival rate of patients; the factors affecting the 5-year mortality of patients with colon cancer were analyzed by COX regression. Results: The expression level of miR-29b-3p in colon cancer tissues (1.55±0.40) was higher than that in adjacent normal tissues (1.01±0.23). The expression levels of FOXO3A mRNA and protein in colon cancer tissues [(0.48±0.13), (0.72±0.19)] were lower than those in adjacent normal tissues [(0.99±0.25), (1.43±0.37)]. The difference was statistically significant (t=23.87, 35.501, 33.318, P<0.05). The expressions of miR-29b-3p and FOXO3A mRNA in colon cancer were correlated with TNM stage and lymph node metastasis of colon cancer patient (P<0.05). There was a negative correlation between the expression of miR-29b-3p and FOXO3A mRNA in colon cancer (r=-0.479, P<0.05). The 5-year overall survival rate of patients with low expression of miR-29b-3p (83.33%) was higher than that of patients with high expression (55.00%), and the difference was statistically significant (χ2=6.820, P<0.05). The 5-year overall survival rate of patients in the FOXO3A mRNA high-expression group (88.64%) was higher than that of patients in the low-expression group (47.37%), and the difference was statistically significant (χ2=16.005, P<0.05). The high expression of miR-29b-3p and low expression of FOXO3A mRNA in TNM Ⅲ stage and colon cancer tissues were risk factors for 5-year death in colon cancer patients (P<0.05). Conclusion: The expression of miR-29b-3p is high, and the expression of FOXO3A mRNA and its protein is low in colon cancer tissues. miR-29b-3p and FOXO3A mRNA are correlated with TNM stage, lymph node metastasis, and prognosis.

1629 Relationship between Changes in Peripheral Blood TLR4 MyD88 NF-κB and PG Levels and Hp Infection Lesion Progression and Inflammatory Response in Patients with Atrophic Gastritis

Objective: To investigate the changes in the expression of serum Toll-like receptor 4 (TLR4), myeloid differentiation factor-88 (MyD88), nuclear factor κB (NF-κB), and pepsinogen (PG Ⅰ, PG Ⅱ) in patients with atrophic gastritis, and their clinical significance. Methods: A total of 98 patients with atrophic gastritis confirmed by gastroscopy taking pathological tissue examination during the period from December 2020 to March 2023 in the First Hospital of Yulin City were selected as Group A, 100 patients with non-atrophic gastritis confirmed by examination during the same period were selected as Group B, and 100 volunteers for health physical examination were selected as Group C. The serum of the study subjects of the three groups were examined and compared with those of the three groups in terms of the expression changes of serum TLR4, MyD88, NF- κB, PGⅠ, PGⅡ, interleukin-6 (IL-6), IL-1β, and tumor necrosis factor-α (TNF-α) levels of the three study groups, and stratified according to whether the patients were co-infected with Hp infection or not, and endoscopic grading criteria of atrophic gastritis for comparing the differences of the above indexes. Results: The TLR4, MyD88, NF-κB, PGII, IL-6, IL-1β, and TNF-α of patients in Group A were significantly higher than those in Groups B and C, and the PGⅠ assay was significantly lower than those in Groups B and C, with a statistically significant difference (P<0.05); the TLR4, MyD88, NF- κB, PGII, IL-6, IL-1β, and TNF-α were significantly higher than those in group B, and the values of PGⅠ assay were significantly lower than those in group C, and the difference was statistically significant (P<0.05); in the Hp-positive group, the values of TLR4, MyD88, PGII, IL-6, IL-1β, and TNF-α were significantly higher than those in the Hp-negative group, and the values of PGⅠ assay were significantly lower than those in the Hp-negative group, and the difference was statistically significant (P<0.05); TLR4, MyD88, NF-κB , PGⅡ, IL-6, IL-1β, and TNF-α of patients with grade III lesions were significantly higher than those of patients with grades II and I, and the values of PGⅠ assay were significantly lower than those of patients with grades II and I, and the difference was statistically significant (P<0.05); TLR4 of patients with grade II TLR4, MyD88, NF-κB , PGII, IL-6, IL-1β, TNF-α were significantly higher in patients with grade II lesions than in patients with grade I. The PGⅠ assay was significantly lower than that in patients with grade I, and the difference was statistically significant (P<0.05); TLR4, MyD88, NF-κB with IL-6, IL-1β, and TNF-α were all significantly positively correlated (P<0.05), and there was no significant correlation between PGII and PGⅠ and IL-6, IL-1β, and TNF-α (P>0.05). Conclusion: The TLR4/MyD88/NF-κB pathway is related to the degree of lesions and the occurrence of Hp infection in patients with atrophic gastritis, and regulates the severity of inflammatory response in patients to a certain extent.

1635 Relationship of LGR5 Her-2 and Ki-67 with Clinicopathological Parameters and Prognosis in Colorectal Cancer Tissues

Objective: To investigate the relationship of leucine-rich G-protein-coupled receptor 5(LGR5), human epidermal growth factor receptor 2 (Her-2), Ki-67 and clinicopathologic parameters and prognosis in colorectal cancer (CRC). Methods: Clinical data of 132 CRC patients admitted to the First People's Hospital of Fangchenggang City from January 2020 to July 2021 were retrospectively analyzed. The expressions of LGR5, Her-2 and Ki-67 in CRC tissues were detected by immunohistochemical method, and the relationship between them and clinicopathological parameters was analyzed. After 12 months of follow-up, patients were divided into good prognosis group (114 cases) and poor prognosis group (18 cases) according to the prognosis. Receiver operating characteristic curve (ROC curve) was used to analyze the prognostic values of LGR5 and Her-2 in CRC patients. Results: The positive expression rates of LGR5, Her-2, and Ki-67 in CRC tissues were significantly higher than those in adjacent normal tissues (P<0.05). The protein expressions of LGR5, Her-2, and Ki-67 were not significantly correlated with gender, age, tumor site, and differentiation degree of CRC patients (P>0.05). The expressions of LGR5 and Ki-67 were correlated with the degree of invasion, TNM stage, lymph node metastasis and distant metastasis (P<0.05). The expression of Her-2 protein was not correlated with the degree of invasion (P>0.05), but with TNM staging, lymph node metastasis and distant metastasis (P<0.05). The positive expression rates of LGR5, Her-2, and Ki-67 protein in poor prognosis group were higher than those in good prognosis group (P<0.05). ROC curve showed that the combination of LGR5 and Her-2 protein (AUC=0.804, P<0.01) had higher predictive value. Conclusion: LGR5, Her-2, and Ki-67 are highly expressed in CRC tissues, and their expression levels are closely related to the clinicopathological parameters of patients. The combined detection of LGR5 and Her-2 has high predictive value for the prognosis of patients.

1640 Clinical Efficacy of Hernia Crochet Assisted Laparoscopic Single Site Transumbilical Treatment for Children with Inguinal Incarcerated Oblique Hernia

Objective: To study the clinical effect of hernia crochet assisted laparoscopic single site transumbilical treatment of inguinal incarcerated oblique hernia in children. Methods: A retrospective analysis was performed on 100 children with inguinal incarcerated oblique hernia admitted from January 2021 to January 2023. The subjects were divided into study group (50 cases) and control group (50 cases) according to the surgical methods. The study group received hernia crocheted assisted laparoscopic single site transumbilical treatment, while the control group received open surgery. The levels of inflammatory factors [interleukin-6 (IL-6), C-reactive protein (CRP)] before and after surgery were compared with intraoperative indicators (incision length, intraoperative blood loss, and operation time), postoperative indicators (first ventilation time, first time of getting out of bed, and length of hospital stay) and the incidence of complications (scrotal redness, chronic pain, and incision infection) between the two groups. Results: There were no significant differences in IL-6 and CRP levels between the two groups before surgery (P>0.05). Postoperative IL-6 (32.91±6.06) mg/L and CRP (14.53±2.91) ng/L in the study group were significantly lower than those in the control group (42.58±7.84) mg/L and 18.14±3.08 ng/L, and the difference was statistically significant(P<0.05). Incision length (0.39±0.14) cm and intraoperative blood loss (3.41±0.78) ml in the study group were significantly lower than those in the control group (3.37±1.13) cm and (19.42±3.96) ml. The time of operation (32.41±6.73) min, the time of primary ventilation (8.87±1.77) h, the time of first getting out of bed (13.58±2.93) h and the time of hospitalization (2.81±0.69) d were significantly shorter than those of the control group (56.66±10.14) min and the time of primary ventilation (17.18±3). 56) h, the first time out of bed (22.53±3.15)h, the length of hospital stay (4.97±1.85)d, the difference was statistically significant (P<0.05). The incidence of scrotal swelling, chronic pain and incision infection in the study group was significantly lower than that in the control group, the difference was statistically significant (P<0.05). Conclusion: Hernia crochet assisted laparoscopic single site transumbilical treatment of inguinal incarcerated oblique hernia in children with significant clinical efficacy, can reduce inflammatory response in children, can also shorten the length of incision and operation, primary ventilation, initial ambulation, and hospital stay in children, reduce intraoperative blood loss and complications.

1644 The Prognostic Value of Combined Detection of NT-proBNP Creatinine and Hemoglobin in Patients with Chronic Heart Failure

Objective: To analyze the prognostic value of NT-proBNP, creatinine and hemoglobin in patients with chronic heart failure. Methods: The clinical data of 220 patients with CHF from March 2020 to March 2021 were retrospectively analyzed. The patients were followed up for 1 year and divided into good prognosis group (n=141) and poor prognosis group (n=79) according to whether the patients had concurrent adverse events. The levels of NT-proBNP, creatinine and hemoglobin in the two groups were compared, and the prognostic risk factors of CHF patients were analyzed by multivariate Logistic regression. Pearson method was used to analyze the correlation of serum NT-proBNP, creatinine and hemoglobin with left ventricular ejection fraction (LVEF) and left ventricular end-diastolic diameter (LVEDD) in the case group. The area under receiver operating characteristic (ROC) curve (AUC) evaluated the value of NT-proBNP, creatinine and hemoglobin in predicting the prognosis of CHF. Results: The levels of NT-proBNP and creatinine in poor prognosis group were higher than those in good prognosis group (P<0.05), and the levels of hemoglobin in poor prognosis group were lower than those in good prognosis group (P<0.05); Multifactor stepwise binary Logistic regression analysis showed that the levels of NT-proBNP, creatinine and hemoglobin were independent risk factors affecting the prognosis of CHF (P<0.05); Pearson correlation analysis showed that NT-proBNP, creatinine and hemoglobin were significantly correlated with LEVF and LVEDD in CHF patients (P<0.05); with the increase of NYHA grade, the levels of NT-proBNP and creatinine were increased (P<0.05), while the hemoglobin level was decreased (P<0.05); ROC curve showed that the AUC of NT-proBNP, creatinine, hemoglobin, and their combination in predicting the prognosis of CHF was 0.765,0.656,0.720,0.866, and the sensitivity was 60.8%,46.8%,53.2% and 86.1%, respectively,the specificity was 83.0%,91.5%,83.7% and 73.8%. Conclusion: NT-proBNP, creatinine and hemoglobin are correlated with the severity of heart failure in CHF patients, and the combined detection has better efficacy in predicting the prognosis of CHF patients, and the combined detection has higher clinical value than the single factor detection.

1649 Correlation between Serum HMGB1 and Inflammatory Indicators and Lung Function Parameters in Patients with Acute Exacerbation of COPD

Objective: To analyze the correlation of serum high mobility group protein 1 (HMGB1) with inflammatory indicators and pulmonary function parameters in patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD). Methods: A total of 104 patients with AECOPD who were treated in the hospital from January 2020 to June 2022 were selected and included in AECOPD group, and another 104 patients with stable chronic obstructive pulmonary disease (COPD) were enrolled as COPD group. 104 healthy subjects with physical examination during the same time period were included in healthy control group. The levels of peripheral blood related indicators [HMGB1, hypoxia-inducible factor-1α (HIF-1α), white blood cell count (WBC)] and serum inflammatory factors [interleukin-6 (IL-6), C-reactive protein (CRP), procalcitonin (PCT)] and pulmonary function parameters [6-minute walk distance (6MWD), pulmonary artery pressure, partial pressure of carbon dioxide (PaCO2), partial pressure of oxygen (PaO2)] were compared among the groups at admission. The relationship between serum HMGB1 and the above inflammatory indicators and pulmonary function parameters was analyzed. Results: The levels of serum HMGB1, HIF-1α, IL-6, CRP, PCT and WBC in the three groups at admission were manifested as AECOPD group>COPD group> healthy control group (P<0.05). The 6MWD and PaO2 at admission were shown as AECOPD group<COPD group< healthy control group (P<0.05). The pulmonary artery pressure and PaCO2 of the three groups at admission showed AECOPD group>COPD group>healthy control group (P<0.05). Pearson correlation analysis showed that serum HMGB1 level at admission was positively correlated with IL-6, CRP and pulmonary artery pressure in patients with AECOPD (r=0.566, 0.287, 0.230, all P<0.05), and was negatively correlated with PaO2 (r=-0.212, P<0.05). Conclusion: Compared with patients with stable COPD and healthy subjects, serum HMGB1 level in patients with AECOPD is abnormally increased, and serum HMGB1 is closely related to IL-6, CRP, pulmonary artery pressure and PaO2.

1654 Diagnostic Accuracy of CT-Guided Percutaneous Transthoracic Needle Biopsy for Non-Small Cell Lung Cancer and Its Influencing Factors

Objective: To explore the diagnostic accuracy of CT-guided percutaneous transthoracic needle biopsy (PTNB) for non-small cell lung cancer (NSCLC) and its influencing factors. Methods: A total of 98 patients with NSCLC treated in the hospital were enrolled as the research subjects between May 2020 and May 2022. All underwent PTNB. Taking the results of postoperative pathological examination as the golden standard, diagnostic results of PTNB for NSCLC were statistically analyzed, and the influencing factors of diagnostic accuracy were analyzed. Results: The diagnostic accuracy of PTNB for NSCLC was 87.76% (86/98), and the accuracy rates of adenocarcinoma, squamous cell carcinoma, large cell carcinoma and adenosquamous carcinoma were 91.53% (54/59), 93.94% (31/33), 0.00% (0/2) and 25.00% (1/4), respectively. The results of univariate analysis showed that there was no significant difference in gender, age, calcification, size of biopsy tissues or biopsy frequency between accuracy group and non-accuracy group (P>0.05), but there were significant differences in lesion sites, lesion size, distance between lesion and chest walls, necrosis and puncture depth (P<0.05). The results of multivariate analysis showed that lesions at inferior lobe, lesion <2cm, the distance between lesions and chest walls <2cm and necrosis were independent influencing factors of diagnostic accuracy rate. Conclusion: PTNB has high diagnostic value for adenocarcinoma and squamous cell carcinoma, but has low diagnostic value for large cell carcinoma and adenosquamous carcinoma. The independent influencing factors of diagnostic accuracy rate include lesions at inferior lobe, lesions <2cm, the distance between lesions and chest walls <2cm and necrosis. Targeted measures is beneficial to increase diagnostic accuracy rate.

1659 Changes and Significance of T Lymphocyte Subsets, B Cells and NK Cells in Infertility Patients with Cervical High-Risk HPV Infection

Objective: To investigate the changes and significance of T lymphocyte subsets, B cells, and natural killer (NK) cells in infertility patients with high-risk cervical human papilloma (HPV) infection. Methods: The clinical data of 1,150 infertility patients with cervical high-risk HPV infection (study group) and 580 HPV-negative infertility patients (control group) who were treated in the gynecology department at the hospital between November 2019 and October 2020 were collected retrospectively. The changes in T lymphocyte subsets, B cells, and NK cells were compared between the two groups. The relationship of the above-mentioned indicators with subtypes of HPV infection and grades of cervical lesions was analyzed. Results: There were no significant differences in the proportions of CD3+ and B cells between the two groups (P>0.05). The proportions of CD4+, CD4+/CD8+, and NK cells in the study group were lower than those in the control group, and the proportion of CD8+ was higher than that in the control group (P<0.05). In the study group, the proportion of HPV 16/18 infected patients was higher than patients infected with other subtypes, and the proportion of HPV 16/18 was significantly different in patients with different grades of HPV infection (P<0.05). The proportions of CD4+, CD4+/CD8+, and NK cells in HPV 16/18 infected patients were lower than those in patients infected with other subtypes, and the proportion of CD8+ was higher than that in patients infected with other subtypes (P<0.05). The proportions of CD3+ and B cells showed no significant differences between patients infected with different subtypes of HPV (P>0.05). With the progression of cervical lesions, the proportions of CD4+, CD4+/CD8+, and NK cells decreased, while the proportion of CD8+ increased (P<0.05), but no significant changes in the proportions of CD3+ and B cells were observed (P>0.05). Conclusion: High-risk HPV-infected infertility patients are accompanied by a certain degree of immune dysfunction. Clinically, changes in the proportions of T lymphocyte subsets, B cells, and NK cells can be detected to provide an objective basis for clinical assessment of HPV subtypes and the progression of cervical lesions.

1664 Analysis of Diagnostic Value of BIRADS Combined with Elastography for Breast Cancer Diagnosis

Objective: To explore the diagnostic value of BIRADS grade combined with elastography for breast cancer. Methods: A total of 105 patients with highly suspected breast cancer admitted to our hospital from January 2020 to December 2021 were selected as the study subjects, and all patients underwent BIRADS grade and elastography examination, taking the pathological test results of patients as the gold standard to explore the diagnostic value of BIRADS, elastography and the combined application. Results: This study included a total of 105 patients and 156 lesions. Pathological examination results showed that 49 lesions were benign, including 19 cases of proliferative adenoma, 14 cases of intraductal papilloma, 11 cases of fibroadenoma, and 5 cases of mastitis. The remaining 107 lesions were malignant, including 41 cases of infiltrating carcinoma, 37 cases of infiltrating ductal carcinoma, 13 cases of ductal carcinoma in situ, 8 cases of lobular carcinoma, 4 cases of mucinous carcinoma, 2 cases of papillary carcinoma, 1 case of signet ring cell carcinoma, and 1 case of sarcomatoid carcinoma. The accuracy rate of BIRADS diagnosis for breast malignancy was 81.41%, with a sensitivity of 77.57%, a specificity of 89.80%, a positive predictive value of 94.32%, and a negative predictive value of 64.71%. The accuracy rate of elasticity imaging diagnosis for breast malignancy was 85.26%, with a sensitivity of 87.85%, a specificity of 79.59%, a positive predictive value of 90.38%, and a negative predictive value of 75.00%. The accuracy rate of combined BIRADS and elasticity imaging diagnosis for breast malignancy was 91.67%, with a sensitivity of 90.65%, a specificity of 93.88%, a positive predictive value of 97.00%, and a negative predictive value of 82.14%. The accuracy of combined diagnosis was higher (91.67%) than BIRADS (81.41%) and elastography (85.26%), with statistically significant (P<0.05). Conclusion: BIRADS and elastography combined diagnosis can improve the diagnostic accuracy, specificity and sensitivity of malignant breast lesions, and have high diagnostic value.

1668 Analysis of the Predictive Value of Peripheral Blood PT PTA and PLT Levels for Progression of Primary Liver Cancer in Patients with Chronic Hepatitis B

Objective: To analyze the predictive value of platelet count (PLT), prothrombin time (PT) and prothrombin activity (PTA) in hepatocellular carcinoma after hepatitis B. Methods: A total of 161 patients with chronic hepatitis B who were hospitalized from January 2017 to December 2019 were selected. They were divided into two groups according to whether there was abnormal coagulation (n=76) and normal coagulation (n=85). At the same time, the demography data and clinical data of patients were recorded. The observation results were observed for three consecutive years. A comparative analysis was carried out to find 46 patients with primary liver cancer. Single factor and multi factor analysis were carried out for patients with liver cancer. The predictive value of PLT, PT, and PTA for primary liver cancer was evaluated using the receiver operating characteristic (ROC) curve. Results: The differences in ALT, AST, and AFP levels between the two groups were statistically significant (P<0.05). The levels of PLT, PTA and PT in chronic hepatitis B group and primary liver cancer group were significantly different (P<0.05). Using statistically significant variables in univariate analysis as independent variables and the occurrence of primary liver cancer as dependent variables for multivariate logistic regression analysis, the results showed that PLT (OR=3.525), PTA (OR=215), and PT (OR=2.312) were the influencing factors for the occurrence of primary liver cancer in patients (P<0.05). The area under the receiver operating characteristic of PLT, PT and PTA was 0.790, 0.744 and 0.682, respectively, which had statistical significance in predicting the development of hepatocellular carcinoma in patients with hepatitis B (P<0.001). Conclusion: PLT, PTA, and PT have a good predictive value in the progression of hepatitis B virus related hepatocellular carcinoma, and the ability of PTA and PT is better than PLT.

1672 Preoperative Three-Dimensional Reconstruction of the Liver Applied to Precision Resection of Primary Hepatocellular Carcinoma

Objective: To investigate the impact of preoperative liver reconstruction in accurate resection for primary liver cancer. Methods: From January 2019 to January 2022, a total of 107 patients were admitted to our hospital. Patients were grouped by computer random numbers: Excel was used to generate 107 random numbers, each corresponding to a patient, and the first 53 numbers were assigned to the control group, and the remaining 54 numbers to the research group according to the order of the generated random numbers from smallest to largest. In the control group (n=53), conventional preoperative evaluation was performed based on conventional two-dimensional imaging of the liver, and in the study group (n=54), preoperative evaluation was performed by applying three-dimensional reconstruction of the liver, and the clinical indexes, hepatic resection volume, liver function indexes, and the levels of serum glycoantigen 19-9 (CA19-9), insulin-like growth factor 1 (IGF-1), carcinoembryonic antigen (CEA), and alpha-fetoprotein (AFP) of the patients in the two groups were compared. ) and alpha-fetoprotein (AFP) levels, and postoperative complications were recorded. The patients were followed up for 1 year and their recurrence metastasis and death were recorded. Results: There was no significant difference in operation time and blood transfusion rate between the two groups (P>0.05). Compared with the control group, the amount of blood loss during operation in the study group was reduced, and the length of hospital stay and anal exhaust time were shortened (P<0.05). There was no significant difference in the actual resected liver volume between the two groups (P>0.05). The preoperative measurement value of the study group was lower than that of the control group, and the actual resected liver volume ratio was higher than that of the control group (P<0.05). There was no significant difference in liver function indexes between the two groups before surgery (P>0.05). There were statistically significant differences in postoperative liver function indexes between the two groups, with AST, ALT, TBIL and AFP levels decreased and ALB levels increased, and the increase and decrease of patients in the study group was greater than that in the control group (P<0.05). There was no significant difference in serum levels of lGF-1, CEA, CA19-9 and AFP between 2 groups before surgery (P>0.05). After treatment, the serum levels of lGF-1, CEA, CA19-9 and AFP were all decreased, and the increase and decrease of patients in the study group were greater than those in the control group (P<0.05). In the control group, there were 1 case of biliary leakage, 4 cases of wound infection, 3 cases of pulmonary infection, and 3 cases of abdominal effusion. The total complication rate was 20.75% (11 cases /53 cases), and there were 8 cases of recurrence and metastasis (15.09%). In the study group, there were 2 cases of pulmonary infection, 1 case of wound infection and 1 case of abdominal effusion, the total complication rate was 7.41% (4 cases /54 cases), and there were 2 cases of recurrence and metastasis (3.70%). The incidence of postoperative complications (7.41%) and recurrence and metastasis (3.70%) in the study group were lower than those in the control group (20.75%, 15.09%), and the differences were statistically significant (P<0.05). Conclusion: Three-dimensional preoperative liver reconstruction technology can improve the accuracy and safety of primary liver cancer resection, reduce the risk of complications after surgery, reduce the damage to their liver function, and has a broad application prospect and high clinical application value.

1677 Clinical Efficacy and 5-Year Follow-up Analysis of Breast Conserving Surgery for TNBC Patients

Objective: To analyze the effect of breast conserving surgery in triple negative breast cancer (TNBC). Methods: A total of 124 cases of early TNBC patients admitted to our hospital from February 2015 to December 2017 were selected, and the surgical plan was chosen according to the wishes of patients and their families, their own conditions, surgical indications and contraindications, and divided into observation and control groups, 60 cases in the control group were treated with modified radical surgery, and 64 cases in the observation group were treated with breast-conserving surgery. Comparison of intraoperative condition, hospitalization time, recent efficacy after treatment, postoperative shoulder mobility, postoperative complications, psychological function, somatic function, social function scores, and 5-year postoperative follow-up between the two groups. Results: The hospitalization time, operation duration, objective remission rate, disease control rate, postoperative rate of local recurrence rate, and distant metastasis rate of the two groups were compared, and there was no difference (P>0.05); intraoperative bleeding, postoperative drainage, and the total incidence of complications in the observation group were lower than those in the control group (P<0.05); and the postoperative flexion, abduction, posterior extension, internal rotation, external rotation mobility, psychological function, somatic function and social function scores were elevated more than those of patients in the control group (P<0.05). Conclusion: Breast-conserving surgery for the treatment of early TNBC has no significant difference in recent efficacy, 5-year postoperative local recurrence rate, and distant metastasis rate compared with modified radical surgery. Breast-conserving surgery has less intraoperative bleeding and postoperative drainage, greater shoulder mobility, fewer complications, and higher postoperative quality-of-life scores, and is worthwhile to popularize its use in the clinic.

1683 Efficacy of Maintenance Therapy with Decitabine-Containing Chemotherapy in Patients with Relapsed AML after Allogeneic Hematopoietic Stem Cell Transplantation

Objective: To explore the therapeutic effects of decitabine combined with FLAG chemotherapy regimen on patients with recurrent acute myeloid leukemia (AML) after allogeneic hematopoietic stem cell transplantation (allo-HSCT). Methods: The clinical data of 95 patients with recurrent AML after allo-HSCT treatment between January 2014 and June 2020 were retrospectively analyzed. According to different treatment regimens, 95 patients were divided into combined group (51 cases) and FLAG group (44 cases). Combined group was given decitabine (5~7mg/m2/d, subcutaneous injection, d1~d5) combined with FLAG regimen, and FLAG group was treated with FLAG regimen, and the two groups had the same symptomatic and supportive treatment. The efficacy was reviewed after 4 weeks of treatment, and the toxic and side effects of chemotherapy and status of symptomatic and supportive treatment were recorded in the two groups. June 31, 2022 was the deadline for follow-up, and Kaplan-meier survival analysis was used to record the overall survival time and 2-year survival rate in the two groups. Results: After treatment, the complete remission rate (CR) and overall response rate (ORR) with 50.98% and 80.39% in combined group were higher than those in FLAG group with 34.09% and 56.82%. There was a statistically significant difference in the ORR (P<0.05). During chemotherapy, there were no statistically differences between the two groups in terms of adverse reactions such as bone marrow suppression (Z=0.18, P=0.855), gastrointestinal reactions (Z=0.63, P=0.531), liver injury (Z=1.06, P=0.288), bleeding (Z=0.61, P=0.540), and pulmonary infection (Z=0.33, P=0.739) (P>0.05). There was no statistically significant difference in the comparison of the rates of deleterious erythrocyte suspension, platelets, and antibiotic application between the 2 groups (P>0.05). As of June 31, 2022, the median overall survival time and 2-year overall survival rate with 14.5 (1.2-34.8) months and 41.18% in combined group were higher than those in FLAG group with 11.4 (0.9~26.9) months and 22.73%. However, Log-rank test showed that there was no significant difference in survival rate between the two groups (Log-rank χ2=3.758, P=0.053). Conclusion: On the basis of FLAG regimen, combined with decitabine in the treatment of patients with recurrent AML after allo-HSCT can achieve better short-term efficacy without increasing the adverse reactions of chemotherapy.

1688 Correlation between Noninvasive Cardiac Function Assessment Indexes and NYHA Classification in Patients with Acute Exacerbation of Chronic Heart Failure

Objective: To investigate the correlation between noninvasive cardiac output (CO), cardiac index (CI), stroke volume (SV), left ventricular work (LCW), and NYHA cardiac function classification in patients with acute exacerbation of chronic heart failure (Chronicheartfailure, CHF). Methods: One hundred and forty-four patients with acute exacerbation of CHF admitted to the Department of Cardiology of Chengde City Central Hospital from November 2021 to October 2022 were selected and categorized into class IV (50 patients), class III (48 patients), and class II (46 patients) according to the cardiac function grading criteria of NYHA (American College of Cardiology). All patients underwent noninvasive hemodynamic cardiac function index testing and B-type natriuretic peptide (BNP) and cardiac ejection fraction (EF) testing, comparing the levels of CO, CI, SV, LCW, BNP, and EF in each group, and analyzing the correlation between CO, CI, SV, and LCW and the NYHA cardiac function classification of patients with acute exacerbation of CHF. Results: The values of CO, CI, SV, LCW, and EF in class Ⅱgroup were higher than those in class Ⅳgroup (P<0.05). The values of CO, CI, SV, LCW, and EF in class Ⅲ group were higher than those in class Ⅳ group (P<0.05). The values of CO, CI, SV, LCW, and EF in class Ⅱgroup were higher than those in the class Ⅲgroup. The BNP of the cardiac function class Ⅲ and Ⅳ groups was higher than that of the cardiac function class Ⅱgroup, but there was no significant difference in BNP between the cardiac function grade Ⅲ and Ⅳ groups (P>0.05). Spearman correlation analysis showed that CO, CI, SV, and LCW were negatively correlated with NYHA cardiac function grade in patients with acute exacerbation of CHF (r=-0.426, -0.368, -0.419, -0.348, P<0.05). Conclusion: As NYHA cardiac function classification worsened, CO, CI, SV, LCW, and EF values decreased, and noninvasive hemodynamic cardiac function assessment indexes were all negatively correlated with NYHA classification, and noninvasive hemodynamic cardiac function assessment indexes were helpful in guiding the assessment of cardiac function classification.

1692 Effectiveness of Balloon Dilatation Angioplasty in the Treatment of Atherosclerotic Intracranial Artery Stenosis Combined with Acute Cerebral Infarction and Its Effect on Quality of Life

Objective: To study the surgical effect and quality of life of balloon dilated angioplasty in the treatment of atherosclerotic intracranial artery stenosis combined with acute cerebral infarction. Methods: A total of 35 patients with atherosclerotic intracranial artery stenosis complicated with acute cerebral infarction who were treated with balloon dilatation angioplasty in our hospital from October 2021 to December 2022 were selected as the balloon dilatation group, another 35 patients with atherosclerotic intracranial artery stenosis complicated with acute cerebral infarction who were treated with intravenous thrombolytic therapy were randomly selected as the intravenous thrombolytic group, ang then 35 patients with atherosclerotic intracranial artery stenosis complicated with acute cerebral infarction who were treated with internal medicine conservative treatment were selected as conservative treatment group. The therapeutic effect, National Institutes of Health Stroke Scale (NIHSS) score, modified Rankin Scale (MRS) score, cerebral blood perfusion index, and quality of life of the three groups were compared. Results: The total effective rate of balloon expansion group was higher than that of intravenous thrombolysis group and conservative treatment group (P<0.05), and the total effective rate of intravenous thrombolysis group was higher than that of conservative treatment group (P<0.05). There was no significant difference in NIHSS score and MRS Score between the three groups before treatment (P>0.05). After treatment, NIHSS score and MRS Score of the three groups were lower than those before treatment, and the reduction rate of balloon dilation group was greater than that of intravenous thrombolysis group and conservative treatment group (P<0.05). The reduction rate of intravenous thrombolysis group was greater than that of conservative treatment group (P<0.05). Comparison of regional cerebral blood flow velocity (rCBF), mean transit time (MTT) and regional cerebral blood volume (rCBV) among the three groups before treatment showed no statistical significance (P>0.05), while rCBF and rCBV were higher in the balloon expansion group than in the intravenous thrombolysis group and the conservative treatment group after treatment (P>0.05). The rCBF and rCBV of the three groups after treatment were higher than those before treatment, and the magnitude of increase in the balloon dilatation group was greater than that of the intravenous thrombolysis group and the conservative treatment group (P<0.05), and the magnitude of increase in the intravenous thrombolysis group was greater than that of the conservative treatment group (P<0.05), the MTT of the three groups after treatment was lower than that of the preoperative group, and the magnitude of decrease in the balloon dilatation group was greater than that of the intravenous thrombolysis group and the conservative treatment group (P<0.05). group (P<0.05), and the decrease in the intravenous thrombolysis group was greater than the MTT in the conservative treatment group (P<0.05). After treatment, the scores of physiology, psychology, social relations, subjective quality of life, and subjective health status in balloon dilation group were higher than those in intravenous thrombolytic group and conservative treatment group (P<0.05), the scores of intravenous thrombolytic group were higher than those in conservative treatment group (P<0.05), and the scores of environmental field in balloon dilation group were higher than those in intravenous thrombolytic group and conservative treatment group (P<0.05). There was no significant difference in environmental field scores between the intravenous thrombolytic group and the conservative treatment group (P>0.05). Conclusion: Balloon dilated angioplasty for atherosclerotic intracranial artery stenosis combined with acute cerebral infarction can significantly improve the neurological function and cerebral blood perfusion index, with higher quality of life.

1698 Diagnostic Value of Circulating miRNA-208a in Heart Failure with Reduced Ejection Graction

Objective: To investigate the clinical value of circulating miRNA-208a expression levels in the diagnosis of heart failure with reduced ejection fraction (HFrEF). Methods: From January 2021 to December 2022, 120 patients with heart failure (HF) admitted to the Department of Cardiovascular Medicine of our hospital were selected. Among them, 56 patients with HFrEF were included in the HFrEF group, 64 patients with non-HFrEF were included in the non-HFrEF group, and 60 healthy people in the same period were selected as the control group. Left ventricular ejection fraction (LVEF), left ventricular shortening fraction (LVFS), left atrial diameter (LAD), and left ventricular mass index (LVMI) were measured by echocardiography, Detection of miRNA-208a β- Myosin heavy chain( β- MHC) mRNA expression, and the serum level of N-terminal pro-brain natriuretic peptide precursor (NT-proBNP) was measured by electrochemiluminescence method. Results: LVEF and LVFS in HFrEF group and non-HFrEF group were significantly lower than those in the control group, while LAD and LVMI were significantly higher than those in the control group (P<0.05); LVEF and LVFS in HFrEF group were significantly lower than those in non-HFrEF group, and LAD and LVMI were significantly higher than those in non-HFrEF group (P<0.05). Serum NT-proBNP and miRNA-208a in HFrEF group and non-HFrEF group β- The expression of MHC mRNA was significantly higher than that of the control group. The serum NT-proBNP and miRNA-208a β- The expression of MHC mRNA was significantly higher than that of non-HFrEF group (P<0.05). The expression of miRNA-208a in three groups was negatively corre- lated with LVEF and LVFS, and with LAD, LVMI, NT-proBNP and β- MHC mRNA was positive- ly correlated and the difference was statistically significant (P<0.05). The area under curve (AUC) of miRNA-208a in diagnosis of HFrEF is 0.757, the best cut-off value is 5.19, the Jordan index is 0.417, the sensitivity is 89.29, and the specificity is 52.42%. The AUC of the combined diagnosis ofSerum NT-proBNP, miRNA-208a and β- MHC mRNA is 0.996, the best cut-off value is 0.592, the Jordan index is 0.956, the sensitivity is 96.43%, and the specificity is 99.19%. Conclusion: The expression of miRNA-208a in the circulating blood of patients with HFrEF is significantly incre- ased, which can be used as a biomarker for diagnosis of HFrEF, and can be compared with NT-proBNP β- The combined application of MHC mRNA has better diagnostic efficacy.

1704 Effectiveness of iRoot SP a Novel Bioceramic Material in Minimally Invasive Root Canal Therapy and Its Impact on Patients' Periodontal Health

Objective: To observe the recent application effect of a new bioceramics material iRoot SP in minimally invasive root canal therapy and the long-term periodontal pain and periodontal health status. Methods: A total of 116 patients undergoing minimally invasive root canal treatment admitted to Chengde Stomatological Hospital from January 2020 to February 2022 were divided into AH-PLUS group and iRoot SP group by random number table method, with 58 patients in each group. The root canal filling in the AH-PLUS group was filled with AH-PLUS paste, while the root canal filling in the iRoot SP group was filled with a new bioceramical material iRoot SP. Both groups were observed 7 days after treatment and followed up for 6 months. The short-term curative effect 7 days after surgery and the success rate of treatment 6 months after surgery were analyzed. Periodontal probing depth (PD), gingival crevicular bleeding index (BI), gingival index (GI), and plaque index (PLI) were used to evaluate the patients' periodontal health before and 7 days after surgery. Visual simulation scores were used to evaluate the patients' postoperative pain at 1, 3, 5, and 7 days after surgery. According to the Mohd Sulong criteria, the patients' emergency response was evaluated during the postoperative observation period, and the oral medicine treatment satisfaction Evaluation scale was used to evaluate the patients' treatment satisfaction at 7 days after the operation. Results: Compared with the AH-PLUS group, the total effective rate at 7 days after surgery and the success rate at 6 months after surgery in the iRoot SP group were significantly increased, with statistical significance (94.83%vs77.59%, χ2=6.202, P=0.013, 94.83%vs75.86%, χ2=8.340, P=0.013). Compared with before operation, PD, scores of BI, GI and PLI of the two groups were significantly decreased after treatment, and the differences in PD, scores of BI, GI and PLI between the two groups were statistically significant (P<0.05). On 1, 3, 5 and 7 days after surgery, the score of pain of the two groups showed a gradually decreasing trend, and the score of pain of the iRoot SP group were lower than those of the AH-PLUS group at different time points (P<0.05). Compared with the H-PLUS group, the emergency response rate of the iRoot SP group was significantly decreased during postoperative observation, with statistical significance (5.17%vs17.24%, χ2=4.245, P=0.013). Compared with the H-PLUS group, the total satisfaction rate of the iRoot SP group was significantly increased at 7 days after operation, with statistical significance (93.10%vs74.14%, χ2=74.14, P=0.013). Conclusion: Compared with AH-PLUS paste, iRoot SP, a novel bioceramical material, had a better effect on root canal filling in patients with minimally invasive root canal treatment, and was worthy of clinical promotion.

1709 Effect of Bovine Pulmonary Surfactant Combined with Caffeine on Blood Gas Markers, Inflammatory Indexes and Pulmonary Circulation Function in Children with Neonatal Pulmonary Hyaline Membrane Disease

Objective: To explore the effect of pulmonary surfactant combined with caffeine on blood gas markers, inflammatory indexes, and pulmonary circulation function in children with neonatal pulmonary membrane disease. Methods: A total of 145 newborns with pulmonary dialysis who received treatment in our hospital from January 2020 to December 2022 were included as the study subjects. The children were randomly divided into two groups using simple random grouping of children using computer randomization method. The control group (n=72) received caffeine citrate treatment, while the observation group (n=73) received bovine lung surfactant combined with caffeine treatment. The clinical data of the two groups of children were collected to compare the blood gas markers, inflammatory factor levels and pulmonary circulation function before and after treatment, and to compare the differences in treatment effects between the groups. Results: The response rate of the observation group was 86.30%, higher than 73.61% in the control group (P<0.05). After treatment, the blood gas indexes of the children improved, the pO2, BE, and pH values increased significantly, but the pCO2 level of the children was significantly reduced, and the blood gas index of the observation group improved more than that of the control group (P<0.05). IL-6 and LDH in the two groups were at the same level (P>0.05); after treatment, IL-6 levels in the children increased first and decreased, LDH levels decreased over time, and the IL-6 and LDH levels in the observation group were lower than the corresponding time node in the control group (P<0.05). There was no significant difference in the pulmonary circulation function (P>0.05). After treatment, the EVLW, PVR and MPAP all decreased, and the RVSWI level increased, and the increase and decrease of the observation group were higher than those of the control group, with a significant difference (P<0.05). Conclusion: In the treatment of neonatal pulmonary transparent membrane disease, the treatment of cow pulmonary surfactant combined with caffeine can effectively improve the blood gas and inflammation index of children, improve the pulmonary circulation function of children, and effectively improve the treatment effect.

1714 Effectiveness of Isotretinoin Combined with M22-ResurFx Non-Ablative Fractional Laser in the Treatment of Severe Acne

Objective: To explore the effects of isotretinoin combined with M22-ResurFx non-ablative fractional laser in the treatment of severe acne. Methods: A total of 104 patients with severe acne were selected and randomly divided into control group and observation group, with 52 cases in each group. The control group was given M22-ResurFx non-ablative fractional laser treatment, and the observation group was given isotretinoin on this basis. The clinical efficacy of the two groups was evaluated after 5 months of treatment. The skin physiological indexes (VISIA digital skin analysis indexes) and wound basal tissue biochemical indexes [vascular endothelial growth factor (VEGF), transforming growth factor-β1 (TGF-β1), transforming growth factor-β activated kinase (TAK)] were compared between the two groups before treatment and after 5 months of treatment. The occurrence of adverse reactions and recurrence rate within 3 months of follow-up were recorded in both groups. Results: The total effective rate in observation group was higher than that in control group after 5 months of treatment (96.15% vs 80.77%, P<0.05). Before treatment, there were no statistical differences in the score of lactic acid sting test and scores of items of skin VISIA between the two groups (P>0.05). After 5 months of treatment, the score of lactic acid sting test in both groups was higher than that before treatment (P<0.05) while the five VISIA scores of redness zone, sclererythrin, spot, texture, and pore were lower compared with those before treatment (P<0.05), and the differences of the above indicators in observation group were greater than those in control group (P<0.05). There were no statistically significant differences in the levels of VEGF, TGF-β1 and TAK in wound basal tissue between both groups before treatment (P>0.05), and the above levels in the two groups after treatment were significantly decreased (P<0.05), and the differences in observation group were greater compared to control group (P<0.05). There was no statistical significance in the total incidence rate of adverse reactions between the two groups (15.38% vs 17.31%, P>0.05). The recurrence rate in observation group was significantly lower than that in control group (1.92% vs 13.46%, P<0.05). Conclusion: Isotretinoin combined with M22-ResurFx non-ablative fractional laser has significant efficacy in the treatment of severe acne, and it can significantly improve the acne severity and skin physiological indexes, and can inhibit the levels of VEGF, TGF-β1 and TAK in wound basal tissue to reduce the acne scar regeneration, and it does not increase adverse reactions, and it has low recurrence rate and good safety.

1720 Effect of Negative Pressure Therapy Combined with Jade Ointment on Serum Th1/Th2 Cells and miR-155 Expression in DFU Patients

Objective: To explore the effect of negative pressure therapy combined with jade ointment on serum expression of Th 1, Th 2 cells and miR-155 in patients with diabetic foot (DFU). Methods: A total of 146 DFU patients admitted to our hospital from June 2021 to January 2022 were selected and grouped by Excel computerized randomized simply random method into a combination group and a control group, with 73 patients in each group. The patients were given conventional debridement and basic treatment, with negative pressure wound therapy, the control group was treated with calcium alginate dressing, and the combined group was treated with reduced muscle Yuhong ointment for treatment. The total course of treatment was 8 weeks. The efficacy of the two groups, the relative expression of miR-155 and serum Th1-related cytokines Th2-related cytokines in DFU granulation tissues before treatment and at 1, 2, 4, and 8 weeks of treatment were compared, and blood and liver and renal function indexes were recorded before and after treatment. Results: The observation group had a higher response rate (91.78%, 67 / 73) than the control group (79.45%, 58 / 73), with differences between groups (P<0.05). The effect of treatment, time and interaction on the relative expression of miR-155 was significant (P<0.05). The relative expression of miR-155 decreased in both groups over time and showed a stronger trend in the observation group compared to the control group. There was no statistically significant difference in the patients' Th1- and Th2-related factors before treatment by intergroup t-test (P>0.05), and after treatment, the patients' levels of the factors improved significantly after 8 weeks, with a decrease in TNF-α, IL-2, and IFN-γ, and an increase in IL-4, IL-5, and IL-10, and the difference between pre- and post-treatment was greater in the Observation Group compared with the Control Group (P<0.05). There was no statistical difference in the comparison of WBC, RBC and PLT levels between the two groups before treatment (P>0.05); after treatment, the patients' WBC levels were significantly reduced, and the difference between the observation group before and after treatment was greater than that of the control group, with statistically significant differences (P<0.05), but there was no statistically significant difference in the comparison of RBC and PLT levels between the two groups before and after treatment (P>0.05). There was no statistically significant difference in the comparison of liver and kidney function indexes between the two groups before and after treatment (P>0.05). Conclusion: Patients with diabetic foot can significantly improve the serum Th1Th2 cells and miR-155 expression level, and improve the therapeutic effect.

1725 Short-Term Efficacy and Safety of Levosimendan in the Treatment of Patients with Acute Decompensated Heart Failure

Objective: To analyze and study the results and clinical safety of a treatment regimen of conventional drugs combined with levosimendan in hospitalized patients with acute decompensated heart failure (ADHF) over a short course of treatment. Methods: A total of 160 patients with ADHF admitted to the Second Affiliated Hospital of Anhui Medical University from July 2018 to July 2022 were selected and divided into control group with conventional medication and experimental group with levosimendan combined with conventional medication, 80 cases in each group. Left ventricular ejection fraction (LVEF), left ventricular end diastolic diameter (LVEDD) , N-terminal pro-B-type natriuretic peptide(NT-proBNP) , cardiac troponin I (cTnI), and cardiac functional grading were collected at admission and one week after treatment. At the same time, the therapeutic effect was analyzed and evaluated. Results: After treatment, LVEF levels of all subjects increased( both P<0.05 ). There were a significant decrease in the level of LVEDD、NT-proBNP、cTnI for both groups compared with those before treatment( both P<0.05 ). After the treatment of levosimendan, the magnitude of changes in LVEF, LVEDD, NT-proBNP and cTnI of the patients in the experimental group was better than that in the control group, and the difference between the two groups was significant (all P<0.05). Conclusion: Levosimendan in combination with conventional drugs improves myocardial function and slows down clinical symptoms in hospitalized patients with acute decompensated heart failure, and is safe and effective.

1730 Analysis of the Efficacy of Cyclosporin A-based Compound Medication Regimen in the Treatment of Non-Severe Aplastic Anemia

Objective: To study the efficacy of compound Zaofan pills combined with cyclosporine A and testosterone undecanoate in the treatment of non-severe aplastic anemia (NSAA). Methods: A retrospective analysis was performed on 80 patients with NSAA admitted to the hospital from January 2020 to January 2023. According to the treatment methods, they were divided into study group (40 cases) and control group (40 cases). The study group was treated with compound Zaofan pills combined with cyclosporine A and testosterone undecanoate, and the control group was treated with cyclosporine A and stanazolol. The lymphocyte subsets (CD4+, CD8+), white blood cell count, platelet count, hemoglobin level before and after treatment, liver function indexes [serum alkaline phosphatase (ALP), glutamyltransferase (GGT), aspartate aminotransferase (AST), alanine aminotransferase (ALT)], total effective rate, side effects (toxic side effects, liver function) were compared between the two groups. Results: There were no significant differences in CD4+, CD8+, white blood cell count, platelet count and hemoglobin level between the two groups before treatment (P>0.05). Compared with before treatment, CD4+, white blood cell count, platelet count and hemoglobin level were higher in the two groups after treatment, and CD8+ was lower, the difference was statistically significant (P<0.05). Compared with the control group, the CD4+, white blood cell count, platelet count, hemoglobin level and total effective rate of the study group were higher, and the CD8+, ALP, GGT, AST, ALT levels and the incidence of toxic side effects and abnormal liver function were lower, the differences were statistically significant (P<0.05). Conclusion: Compound Zaofan pill combined with cyclosporine A and testosterone undecanoate is effective in the treatment of patients with NSAA, which can enhance the cellular immune function, improve white blood cell count, platelet count and hemoglobin level, and reduce the degree of liver damage, with few side effects and high safety.

The conventional group was given salvianolate combined with metformin and other conventional DKD treatments, and the Bailing capsule group was combined with Bailing capsule on the basis of the regular group. The blood glucose indicators, renal function indicators, serum-related factors, and vascular endothelial function indicators were compared between the two groups of patients before treatment and at 16 weeks of treatment. The clinical efficacy of the two groups of patients was compared at 16 weeks of treatment. Results: There were no significant differences in the levels of blood glucose indicators (FPG, 2hPG, HbA1c, HOMA-IR), renal function indicators (Scr, BUN, 24h UAER) and serum-related factors (ACA-IgA, ACA-IgM, ACA-IgG, NGAL) between the two groups of patients before treatment (all P>0.05), and at 16 weeks of treatment, the above indicators of the two groups were significantly decreased compared with those before treatment, and the decreases were greater in Bailing capsule group (all P<0.05). The level of ET-1 of Bailing capsule group at 16 weeks of treatment was significantly lower than that of conventional group, and the decline was greater (all P<0.05). The NO, FMD and NMD of Bailing capsule group were significantly higher than those of conventional group at 16 weeks of treatment, and the increases were greater (all P<0.05), and its clinical efficacy was significantly improved compared to conventional group (Z=2.123, P=0.034). Conclusion: On the basis of conventional treatment of patients with DKD, combination of Bailing capsule can effectively regulate the glucose metabolism level, relieve the inflammatory response, improve the renal function, and enhance the therapeutic effect.

1740 Analysis of the Efficacy and Impact of Renin-Angiotensin System Inhibitors Combined with Hydroxychloroquine in the Treatment of IgA Nephropathy and Nephroprotective Effects

Objective: To investigate the efficacy and renal protective effect of renin angiotensin system inhibitor (RASI) combined with hydroxychloroquine in the treatment of IgA nephropathy. Methods: The study subjects were 84 patients with IgA nephropathy admitted from January 2018 to December 2022. According to the random number table method, the study subjects were divided into the control group (42 cases) and the observation group (42 cases). The patients in the control group took RASI, and the observation group took hydroxychloroquine sulfate tablets on the basis of the control group for 3 months. The clinical effects of the two groups were compared. The renal function indicators and treatment safety of the two groups of patients before and after treatment were compared. Results: The difference in serum 24-UPro content between the observation group after treatment and before treatment was significantly greater than that of the control group (P<0.05). After treatment, the levels of 24-USPro in both groups were lower than before treatment (P<0.05), and the levels of Scr were higher than before treatment (P<0.05).There was no statistically significant difference in serum Scr level and UA level between the observation group after treatment and before treatment (P>0.05); there was no statistically significant difference in the incidence of adverse reactions between the two groups of patients (P>0.05). Conclusion: The combination of renin-angiotensin system inhibitor and hydroxychloroquine has a good effect on patients with IgA nephropathy, helping to control proteinuria, protect the kidney and maintain the stability of renal function.

1743 Effects of Camrelizumab Combined with TP Regimen on Efficacy Immune Function and Peripheral Blood VEGF bFGF CA125 and CEA in Patients with Intermediate to Advanced NSCLC

Objective: To observe the effects of camrelizumab combined with TP chemotherapy regimen (paclitaxel + cisplatin) on efficacy, immune function, peripheral blood vascular endothelial growth factor (VEGF), carbohydrate antigen 125 (CA125), carcinoembryonic antigen (CEA) and basic fibroblast growth factor (bFGF) in patients with advanced non-small cell lung cancer (NSCLC). Methods: A total of 102 patients with advanced NSCLC admitted to our hospital from January 2020 to January 2023 were selected and divided into two groups by simple random method. The control group was treated with TP regimen, and the observation group was additionally treated with camrelizumab. The effects, immune function, and peripheral blood factors of the two groups were compared, and the toxic and side effects of the two groups were counted. Results: The objective remission rate (ORR) and disease control rate (DCR) in the observation group were 56.86% (29/51) and 80.39% (41/51), which were higher than those in the control group (33.33% (17/51) and 58.82% (30/51), and the difference was statistically significant (P<0.05). Before treatment, there was no significant difference in VEGF, bFGF, CA125, CEA and immune function between the two groups (P>0.05). Compared with before treatment, VEGF, bFGF, CA125, CEA, and CD8+ decreased after treatment, while CD4+ and CD4+/CD8+ increased after treatment. The difference before and after treatment in the observation group was greater than that in the control group (P<0.05). There was no significant difference in the incidence of stomatitis, peripheral nerve abnormality, alopecia, bone marrow suppression, gastrointestinal reaction, abnormal liver or kidney function, reactive cutaneous capillary hyperplasia grade Ⅲ to grade Ⅳ between 2 groups (P>0.05). Conclusion: Camrelizumab combined with TP regimen in the treatment of advanced NSCLC can effectively improve ORR and DCR, improve immune function, inhibit the expression of VEGF, bFGF, CA125 and CEA in peripheral blood, while not increasing toxic and side effects.

1748 Efficacy and Safety of Immunotherapy Combined with Targeted Therapy for Metastatic Colorectal Cancer

Objective: To evaluate the efficacy and safety of carrellizumab combined with anti-angiogenic agents in the treatment of metastatic MSS colorectal cancer. Methods: A total of 40 colorectal patients with metastatic MSS were enrolled in the radiotherapy Department of Tangshan People's Hospital from June 2021 to June 2022, who received third-line treatment with carrilizumab combined with anti-angiogenic agents (fuquitinib/regorafenib) or single-agent anti-angiogenic agents (fuquitinib/regorafenib) (20 in the combination group and 20 in the monotherapy group). Overall survival (OS), disease control rate (DCR) and objective response rate (ORR) of the two groups were evaluated, and the incidence of adverse events in the two groups were statistically analyzed. Results: Compared with the monotherapy group, there was no significant difference in the general clinical data of the combined group. The median OS in the combination and monotherapy groups was 11 and 7.5 months, DCR was 85% and 60%, and ORR was 30% and 10%, respectively. The incidence of adverse events in the combination group and the monotherapy group was 95% and 90%, respectively. Most of the adverse events were grade 1-2, and the incidence of serious adverse events (≥ grade 3) was 15% and 10%, respectively, without grade 4 adverse events. The incidence of capillaries in combination group was higher than that in monotherapy group. Conclusion: The combination group has improved benefit for patients with OS, but the difference is not statistically significant compared with the monotherapy group. Immunotherapy combined with anti-angiogenic drugs is a promising strategy, and the combination of the two can improve the survival benefits of patients, showing better anti-tumor activity and high safety in the treatment of advanced metastatic colorectal cancer, providing an important basis for the development of immunotargeted therapy for CRC.
