1233 LncRNA-MIAT Promotes Malignant Behavior of Thyroid Cancer by Inhibiting miR-519d-3p to Activate EZH2

Objective: To investigate the regulation and molecular mechanism of long non-coding RNA MIAT (LncRNA-MIAT) on proliferation, apoptosis, migration and invasion of thyroid cancer cells. Methods: The expression of LncRNA-MIAT and histone methyltransferase 2 (EZH2) in thyroid cancer was analyzed by Cancer Genome Atlas (TCGA) and thyroid cancer (THCA) mRNA-seq data. Human normal thyroid cell line Nthy-ori 3-1 and human thyroid cancer cell line FTC-133 were used as target cells. The mRNA expression levels of LncRNA-MIAT, miR-519d-3p and EZH2 were detected by qPCR. RNA fluorescence in situ hybridization (FISH) was used to detect the localization of LncRNA-MIAT in FTC-133 cells, and Western blot was used to detect the expression of EZH2 protein. Luciferase reporter gene was used to detect LncRNA-MIAT and miR-519d-3p and the direct effect of miR-519d-3p on EZH2. FTC-133 cells were divided into 6 groups (control group, Empty vector group, LncRNA-MIAT group (LncRNA-MIAT group), miR-519d-3p inhibitor group (miR-519d-3p group), EZH2 inhibitor EPZ-6438 treatment group (EPZ-6438 group), miR-519d-3p inhibitor combined with overexpression of LncRNA-MIAT group (LncRNA-MIAT+miR-519d-3p inhibitor group)]. Cell proliferation was detected by Edu kit and plate cloning. Apoptosis was detected by flow cytometry. Cell behavior was detected by Transwell cell migration and invasion assay. Results: The expressions of LncRNA-MIAT and EZH2 in thyroid carcinorna tissues and cells were up-regulated, and there was a positive correlation between the two (r=0.466, P<0.05). The expression of miR-519d-3p was down-regulated in FTC-133 cells (P<0.05). Double luciferase reporter assay showed that the target gene of miR-519d-3p was EZH2, and LncRNA-MIAT targeted inhibition of miR-519d-3p to activate EZH2. Compared with Empty vector group, the percentage of Edu positive cells, the number of cell clones, the number of cell invasion and migration in LncRNA-MIAT group and miR-519d-3p inhibitor group were significantly up-regulated (P<0.05), and the cell apoptosis rate was decreased (P<0.05). EZH2 inhibitor EPZ-6438 decreased the percentage of Edu positive cells, the number of cell clones, the number of cell invasion and migration (P<0.05), and increased the rate of apoptosis (P<0.05). Compared with LncRNA-MIAT group or miR-519d-3p inhibitor group, the Edu positive cell percentage, cell clone number, cell invasion and migration number of LncRNA-MIAT+miR-519d-3p inhibitor group were significantly up-regulated (P<0.05), respectively. The apoptosis rate was decreased (P<0.05). Conclusion: LncRNA-MIAT can promote the malignant behavior of thyroid cancer by inhibiting the activation of EZH2 by miR-519d-3p.

1240 The Effect of miR-101 on the Diagnosis of Breast Cancer Patients and Autophagy and Apoptosis of Breast Cancer Cells

Objective: To investigate the expression and clinical characteristics of serum miR-101 in patients with breast cancer and its effects on autophagy and apoptosis of MCF-7 cells. Methods: A total of 96 patients with breast cancer treated from June 2021 to October 2022 were selected as the observation group, 41 patients with benign breast hyperplasia were selected as the benign group, and 35 healthy subjects were selected as the normal control group. The expression level of miR-101 in serum of the 3 groups was detected by qRT-PCR. The expression levels of serum miRNA-101 in different clinicopathological features of breast cancer patients were compared. The diagnostic value of miR-101 was analyzed by ROC curve. Breast cancer cell line (MCF-7) was transfected with miR-101 mimic reagent. The expression of miR-101 in MCF-7 and normal breast cell line (HBL-100) was detected by qPCR. The cell proliferation was detected by CCK-8 method. The expressions of autophagy and apoptosis genes and proteins were detected by qPCR and Western blot. The apoptosis rate of each group was detected by flow cytometry. Results: The relative expression of miRNA-145 in observation group (n= 96,0.27) was significantly lower than that in benign group (n= 41,0.63) and control group (n= 35,1.00), the difference was statistically significant (P<0.01). The expression of serum miRNA-101 was significantly different in lymph node metastasis, differentiation degree, tumor diameter and TNM stage (P<0.01). ROC curve analysis showed that the area under the curve was 0.723(95%CI: 0.813-0.924) and 0.875(95%CI: 0.835-0.981) in the three groups of comparison, respectively. The results of CCK-8, qPCR and Western blot showed that over-expression of miR-101 inhibited the proliferation of MCF-7 cells, significantly increased the gene and protein expression of BAX in MCF-7 cells (P<0.01), and decreased the gene and protein expression of BCL-2, LC3B and Beclin-1 in McF-7 cells (P<0.01). The apoptosis rate of MCF-7 cells was increased. Conclusion: In patients with breast cancer, the expression level of serum miR-101 is significantly decreased, which may be a potential diagnostic indicator of breast cancer. In addition, miR-101 overexpression inhibited the proliferation of MCF-7 breast cancer cells, inhibited autophagy, and promoted apoptosis. These results provide important clues for further research on the mechanism of miR-101 in the occurrence and development of breast cancer, and provide new strategies for the early diagnosis and treatment of breast cancer.

1245 Effect and Mechanism of CircITGB6 on Cisplatin Resistance in Ovarian Cancer Cells by Regulating miR-542-3p/PCNA Axis

Objective: To investigate the influences of circular RNA (Circ) ITGB6 on cisplatin (DDP) resistance of ovarian cancer (OC) cells by regulating miR-542-3p/proliferating cell nuclear antigen (PCNA) axis and its mechanism. Methods: The expression levels of CircITGB6, miR-542-3p, PCNA mRNA in OC tissues, normal ovarian tissues, SKOV3, SKOV3/DDP and normal ovarian epithelial cell line (IOSE-80) were detected by qRT-PCR; SKOV3/DDP was set as control group (no treatment), interference (si CircITGB6) group (transfected with si CircITGB6), negative control (si NC) group (transfected with si NC), si CircITGB6+inhibitor negative control (inhibitor NC) group (co-transfected with si CircITGB6 and inhibitor NC), and si CircITGB6+miR-542-3p inhibitor group (co-transfected with si CircITGB6 and miR-542-3p inhibitor). Apoptosis, proliferation, drug resistance, and the expression levels of CircITGB6, miR-542-3p, PCNA mRNA in SKOV3/DDP cells of each group were detected by flow cytometry, CCK-8 and qRT PCR respectively; double luciferase was used to verify the targeting relationship between miR-542-3p and CircITGB6, PCNA; the PCNA, Bcl-2 associated X protein (Bax) and cell proliferation related nuclear antigen (Ki-67) were detected by Western blot. Results: The expression of CircITGB6 (1.66±0.17 vs 0.95±0.10) and PCNA mRNA (1.59±0.16 vs 0.98±0.10) in OC tissues was higher than that in normal ovarian tissues, and the expression of miR-542-3p (0.64±0.07 vs 1.06±0.11) was lower (all P<0.05); the expression of CircITGB6 and PCNA mRNA in IOSE-80, SKOV3 and SKOV3/DDP increased in turn, the expression of miR-542-3p decreased in turn (P<0.05); compared with the control group and the si NC group, the cell proliferation rate and IC50, CircITGB6 (1.21±0.13 vs 1.88±0.19, 1.84±0.19), Ki-67, PCNA in the si CircITGB6 group were obviously reduced, the apoptosis rate and expression of Bax and miR-542-3p (0.86±0.09 vs 0.51±0.06, 0.55±0.06) were obviously increased (all P<0.05); compared with the si CircITGB6+inhibitor NC group, the cell proliferation rate and IC50, Ki-67, PCNA in the CircITGB6+miR-542-3p inhibitor group were obviously increased, the apoptosis rate and the expression of Bax and miR-542-3p were obviously decreased (all P<0.05). Conclusion: Interfering with CircITGB6 expression can reduce the resistance of SKOV3 to DDP by regulating miR-542-3p/PCNA axis.

1252 circ_0001658 Regulates Proliferation Cell Cycle and Apoptosis of Breast Cancer Cells by Targeting miR-1179

Objective: To investigate the effects of circ_0001658 on proliferation, cell cycle and apoptosis of breast cancer BT-549 cells and its molecular mechanism. Methods: A total of 30 breast cancer tissues and corresponding paracancer tissues were selected. BT-549 cells were cultured in vitro and divided into si-NC group, si-circ_0001658 group, miR-NC group, miR-1179 group, si-circ_0001658+anti-miR-NC group, si-circ_0001658+anti-miR-1179 group. RT-qPCR was used to detect the expression levels of circ_0001658 and miR-1179. The expression levels of circ_0001658 and miR-1179 were detected by RT-qPCR. Cell proliferation was detected by MTT. Cell cycle was detected by PI single staining. Cell apoptosis was detected by flow cytometry. The expression of related proteins was detected by Western blot. The targeting relationship between circ_0001658 and miR-1179 was detected by dual luciferase reporter assay. Results: circ_0001658 expression was significantly increased (1.00±0.11 vs 4.24±0.53) and miR-1179 expression was significantly decreased (1.00±0.10 vs 0.45±0.05) in breast cancer tissues in comparison with paracancerous tissues (P<0.05). Silencing circ_0001658 decreased the OD value of BT-549 cells (1.45±0.18 vs 0.81±0.07), the proportion of S phase cells (28.19±2.36 vs 15.33±1.16), the protein expression of PCNA (0.66±0.07 vs 0.34±0.04), CyclinD1 (0.69±0.08 vs 0.28±0.03) and Bcl-2 (0.59±0.06 vs 0.21±0.02), and increased the proportion of G0-G1 phase cells (47.35±3.28 vs 62.24±5.14), the apoptosis rate (6.58±0.75 vs 25.34±2.86), and the protein expression of p27 (0.36±0.03 vs 0.79±0.08) and Bax (0.25±0.03 vs 0.65±0.06) (P<0.05). Circ_0001658 regulation of miR-1179 expression (P<0.05), and down-regulated miR-1179 reversed the effects of circ_0001658 silencing (P<0.05). Conclusion: Silencing circ_0001658 inhibited BT-549 cell proliferation and promoted cell apoptosis through targeted up-regulation of miR-1179.

1259 The Mechanism of SFRP1 on the Activity and Angiogenesis of Dental Pulp Stem Cells through MAPK Signaling Pathway

Objective: To explore the molecular regulation mechanism of dental pulp stem cells. Methods: Human dental pulp stem cells (DPSC) were purchased and transfected with si-NC/SFRP1. The mRNA and protein expression level of SFRP1 was detected by qRT-PCR and Western blot. CCK-8 and flow cytometry were used to detect the proliferation activity and apoptosis of cells in each group. Cell wound healing and angiogenesis assays were used to detect the changes of cell migration and angiogenesis ability in each group. Western blot was used to detect the protein changes of p-p38 and p38 in MAPK signaling pathway. Results: After knocking down the expression of SFRP1 in DPSC, the proliferation activity of DPSC decreased and the apoptosis rate increased significantly. Cell migration and angiogenesis ability decreased significantly; the protein content of p-p38 in MAPK signaling pathway was significantly decreased. Conclusion: Knockdown of SFRP1 inhibited the proliferation and angiogenesis of dental pulp stem cells and promoted their apoptosis by inactivating the MAPK signaling pathway.

1263 The Mechanism of NK4 Gene on Proliferation and Apoptosis in Non-Hodgkin Lymphoma Cells Based on Sirt1/p53 Signaling Pathway

Objective: To investigate the effect of NK4 gene on the proliferation and apoptosis of non-Hodgkin’s lymphoma cells. Methods: The expression levels of NK4 mRNA in NHL cell lines (Jurkat and Raji cells) and normal B cells (normal) were detected by qRT-PCR. pcDNA3.1-NK4 was transfected into Raji cells to increase the expression level of NK4. Cell proliferation was detected by CCK-8, apoptosis was detected by flow cytometry, and cell migration and invasion were detected by transwell assay to evaluate cell growth progress. Western blot was used to detect Sirt1/p53 signaling pathway-related proteins. Results: NK4 gene was lowly expressed in NHL cells. After up-regulating NK4 expression in NHL cell line Raji cells, the proliferation, migration and invasion ability of Raji cells decreased significantly, while the apoptosis rate increased. In addition, up-regulation of NK4 expression significantly decreased Sirt1 protein expression and increased p53 level. Addition of Sirt1 activator SRT1720 partially inhibited cell growth. Conclusion: NK4 gene mediated Sirt1/p53 signaling pathway inhibited proliferation and promoted apoptosis of non-Hodgkin lymphoma cells.

1267 Panax Nnotoginseng Powder Ameliorates Gastric Mucosal Injury via Nrf2/Gpx4-Mediated Iron Death in Rats with Cerebral Hemorrhagic Stress Ulcers

Objective: To investigate the protective effect of Panax notoginseng powder on gastric mucosal injury in rats with cerebral hemorrhagic stress ulcers via Nrf2/Gpx4-mediated iron death. Methods: A total of 60 rats were divided into NC group, SGU group, Panax notoginseng powder low dose group (Panax notoginseng powder L group), Panax notoginseng powder medium dose group (Panax notoginseng powder M group), Panax notoginseng powder high dose group (Panax notoginseng powder H group), Panax notoginseng powder high dose + Nrf2 specific inhibitor ML385 group (Panax notoginseng powder H + ML385 group) according to the random number table method, 10 rats in each group. The rats were scored for neurological function; the ulcer index of gastric mucosa was measured; HE staining was performed to detect histopathological changes in gastric tissues; Fe2+, MDA content and SOD activity were detected in gastric tissues; ROS levels in gastric tissues were detected by DHE fluorescence staining; Nrf2, Gpx4 and SLC7A11 protein expression in gastric mucosa tissues were detected by protein blotting. Results: Compared with the NC group, the neurological function score, gastric mucosal ulcer index, Fe2+, MDA content and ROS level increased and SOD activity decreased in the SGU group (P<0.05); compared with the SGU group, the neurological function score, gastric mucosal ulcer index, Fe2+, MDA content and ROS level decreased significantly and SOD activity increased in the Panax ginseng powder L, M and H groups ( P<0.05); compared with the Panax ginseng powder H group, the neurological function score, gastric mucosal ulcer index, Fe2+, MDA content and ROS level of rats in the Panax ginseng powder H+ML385 group were significantly higher and SOD activity was lower (P<0.05). Compared with the NC group, Nrf2, Gpx4 and SLC7A11 protein expression in the gastric mucosa tissues of rats in the SGU group were significantly decreased (P<0.05); compared with the SGU group, Nrf2, Gpx4 and SLC7A11 protein expression in the gastric mucosa tissues of rats in the Panax ginseng powder L, M and H groups were significantly increased in a dose-dependent manner (P<0.05); panax ginseng white and powder H+ Nrf2, Gpx4 and SLC7A11 protein expression in gastric mucosal tissues of rats in ML385 group were significantly lower than those in Panax notoginseng powder H groups (P<0.05). Conclusion: Panax notoginseng powder exerts a protective effect on gastric mucosal injury in rats with cerebral hemorrhagic stress ulcers, and the mechanism is related to the activation of Nrf2/Gpx4 pathway to inhibit iron death.

1274 Correlation of HPV Infection with Immunoregulatory Imbalance and Abnormal Cell Proliferation in Patients with Cervical Lesions

Objective: To investigate the relationship between human papillomavirus (HPV) infection and immune dysregulation and abnormal cell proliferation in cervical lesions. Methods: In order to investigate this relationship, a study was conducted from January 2020 to April 2022, in which 56 cases of high-risk HPV-infected cervical lesion patients (high-risk HPV group), 51 cases of low-risk HPV-infected cervical lesion patients (low-risk HPV group), 57 cases of HPV-negative cervical lesion patients (HPV-negative group), and 60 healthy women (normal group) were selected. The cellular and humoral immune indicators, cytokines, p16 protein, Ki67 protein, Prdx4 mRNA, and STAT3 mRNA were compared among the groups. Results: The CD4+ cells, immunoglobulin G (IgG), and immunoglobulin A (IgA) levels were significantly lower in the high-risk HPV group and low-risk HPV group than in the HPV-negative group and normal group (P<0.05). Furthermore, the levels of CD4+ cells, IgG and IgA were significantly lower in the high-risk HPV group than in the low-risk HPV group (P<0.05). The levels of interleukin-2 (IL-2) were significantly lower in the high-risk HPV group and low-risk HPV group than in the HPV-negative group and normal group (P<0.05), with the high-risk HPV group showing significantly lower IL-2 levels than the low-risk HPV group (P<0.05). The levels of interleukin-4 (IL-4), interleukin-17 (IL-17), and transforming growth factor-β (TGF-β) were significantly higher in the high-risk HPV group and low-risk HPV group than in the HPV-negative group and normal group (P<0.05), with the high-risk HPV group showing significantly higher levels of IL-4, IL-17, and TGF-β than the low-risk HPV group (P<0.05).The positive expression rates of p16 and Ki67 proteins were significantly higher in the high-risk HPV group and low-risk HPV group than in the HPV-negative group and normal group (P<0.05). The relative expression levels of Prdx4 and STAT3 mRNA were significantly higher in the high-risk HPV group and low-risk HPV group than in the HPV-negative group and normal group (P<0.05), with the high-risk HPV group showing significantly higher levels of Prdx4 and STAT3 mRNA than the low-risk HPV group (P<0.05). Conclusion: HPV infection exacerbates immune dysregulation and promotes abnormal cell proliferation in cervical lesion patients, particularly in high-risk HPV infections.

1279 Correlation Analysis of Serum IL-32 sRAGE and NLR with Multiple Myeloma Complicated with Renal Injury

Objective: To investigate the correlation between serum interleukin-32(IL-32), soluble advanced glycation end-product receptor (sRAGE) and neutrophil/lymphocyte ratio (NLR) and multiple myeloma complicated with kidney injury. Methods: A total of 82 MM patients diagnosed and treated in Rugao People's Hospital from June 2020 to June 2022 were selected as the observation group, and were divided into renal injury group (n=27) and non-renal injury group (n=55) according to renal function, and 41 healthy physical examination patients were selected as the healthy control group during the same period. IL-32, sRAGE and NLR levels were compared. Binary Logistic regression was used to analyze the effects of IL-32, sRAGE and NLR on kidney injury in MM patients. ROC curve model was used to analyze the AUC value, sensitivity and specificity of IL-32, sRAGE and NLR in predicting kidney injury in MM patients. Results: IL-32 and NLR in observation group were higher than those in control group, while sRAGE was lower than those in control group (P<0.05). IL-32 and NLR in renal injury group were higher than those in control group, while sRAGE was lower (P<0.05). Logistic regression model analysis showed that increased IL-32, increased NLR and decreased sRAGE levels were risk factors for kidney injury in multiple myeloma patients (P<0.05). ROC curve analysis showed that the AUC values of serum IL-32, sRAGE and NLR in predicting kidney injury in multiple myeloma patients were 0.761, 0.765 and 0.802, P<0.05. Conclusion: Serum IL-32, sRAGE, and NLR expression levels increased with kidney injury in MM patients.

1283 Correlation between Levels of Serum β2 Microglobulin Cystatin C and Serum α1 Microglobulin and Prognosis in Patients with Acute Renal Failure

Objective: To analyze the correlation between levels of serum β2 microglobulin (β2-MG), cystatin C (Cys C) and serum α1 microglobulin (α1-MG) and prognosis in patients with acute renal failure. Methods: The clinical data of 105 patients with acute renal failure who were treated between August 2020 and August 2021 were retrospectively analyzed. According to the severity of disease at admission, the patients were divided into stage I patients (n=41), stage II patients (n=40), and stage III patients (n=24). The differences in levels of serum β2-MG, Cys C and α1-MG at admission were compared among patients at different stages. According to the prognosis, the patients were classified into recovery group (patients with renal function recovery, n=67) and injury group (patients with permanent renal function injury, n=38). The differences in serum β2-MG, Cys C and α1-MG at admission among patients with different prognosis status were compared, and the evaluated efficiency of serum β2-MG, Cys C and α1-MG on the prognosis in patients with acute renal failure was explored. Results: The levels of serum β2-MG, Cys C and α1-MG in stage III patients were higher than those in stage II patients and stage I patients (P<0.05). The level of serum β2-MG and Cys C in stage II patients was higher than that in stage I patients (P<0.05). The levels of β2-MG, Cys C and α1-MG in recovery group were lower than those in injury group (P<0.05). Serum β2-MG, Cys C and α1-MG curves AUC were significantly higher than those of the reference lines (AUC>0.5), and their cut off values were 3.59mg/L, 3.21mg/L and 33.36mg/L respectively. Conclusion: Serum β2-MG, Cys C and α1-MG levels were correlated with the disease condition and the prognosis in patients with acute renal failure. It is necessary to take effective treatment measures before serum β2-MG level >3.59mg/L, Cys C level >3.21mg/L and α1-MG level >33.36mg/L in the clinical treatment of acute renal failure so as to improve the prognosis of patients.

1287 Changes of Peripheral Blood Absolute Lymphocyte Count Serum CRP and PCT in Patients with COPD and Their Relationship with Disease Severity

Objective: To analyze the changes of peripheral blood absolute lymphocyte count (ALC), serum C-reactive protein (CRP) and procalcitonin (PCT), and their relationship with disease severity in patients with chronic obstructive pulmonary disease (COPD). Methods: A total of 102 patients with COPD treated in the hospital were enrolled between January 2019 and February 2022. According to disease progression and the severity of pulmonary function, they were divided into stable COPD group and acute exacerbation of COPD (AECOPD) group, and grades I-IV groups. A total of 100 healthy controls were enrolled as control group. The differences in levels of ALC, CRP and PCT in different groups were detected and compared, and the correlation between the above indexes and pulmonary function index [forced expiratory volume in 1 second (FEV1) as a percentage of predicted value (FEV1%)] was analyzed. The changes of ALC, CRP and PCT before and after treatment were observed. Results: The peripheral blood ALC was significantly lower in COPD group than control group, while levels of CRP and PCT were significantly higher in COPD group (P<0.05). The peripheral blood ALC was significantly lower in AECOPD group than stable COPD group, while levels of CRP and PCT were significantly higher in AECOPD group (P<0.05). The differences in levels of ALC, CRP and PCT among patients with different severity of pulmonary function were statistically significant (P<0.05). Further pairwise comparison showed that ALC level in patients at grade IV was lower than that at grades I and II, while CRP and PCT levels were the highest in patients at grade IV, followed by grade III, grade II and grade I (P<0.05). The level of peripheral blood ALC was positively correlated with FEV1%, while CRP and PCT were negatively correlated with it (P<0.05). After treatment, ALC was significantly increased, while levels of CRP and PCT were significantly decreased (P<0.05). Conclusion: The levels of peripheral blood ALC, serum CRP and PCT are closely related to disease severity in COPD patients, which can also guide the treatment of AECOPD.

1292 Predictive Value of Serum miR-499 Combined with PLTP Levels for Atrial Fibrillation after Off-Pump Isolated Coronary Artery Bypass Grafting

Objective: To investigate the predictive value of serum microRNA-499 (miR-499) combined with phospholipid transfer protein (PLTP) levels on atrial fibrillation (AF) after off-pump isolated coronary artery bypass grafting (OPCABG). Methods: A total of 237 patients with coronary artery disease who underwent OPCABG admitted to our hospital from January 2018 to June 2022 were selected and divided into 69 cases in the AF group and 168 cases in the non-AF group according to whether AF occurred after OPCABG.Serum miR-499 and PLTP levels were measured by qPCR and ELISA.Spearman correlation was used to analyze the correlation between serum miR-499 and PLTP levels in patients with AF after OPCABG,multi-factor logistic regression was used to analyze the factors influencing AF after OPCABG,and ROC curve was used to analyze the predictive value of serum miR-499 and PLTP levels on AF after OPCABG. Results: Serum miR-499 levels were higher in the AF group (1.53±0.29) than in the non-AF group (1.22±0.33),and PLTP levels[1.36 (1.00,2.01)] were lower than in the non-AF group[2.33 (1.39,3.53)],with statistically significant differences (t/Z=6.731,5.879,all P<0.001).Spearman correlation analysis showed that serum miR-499 was negatively correlated with PLTP levels in AF patients after OPCABG (rs=-0.789,P<0.001).Multifactorial logistic regression analysis showed that age (OR=1.085,95% CI:1.036 to 1.136),diabetes (OR=3.399,95% CI:1.472 to 7.853),left atrial diameter (OR=1.194,95% CI:1.063 to 1.341) and miR-499 (OR=2.961,95% CI:1.944 to 4.508) were independent risk factors for AF after OPCABG and PLTP (OR=0.319,95% CI:0.195 to 0.522) was an independent protective factor (P<0.05).ROC curve analysis showed that serum miR-499 combined with PLTP levels (AUC=0.842,95% CI:0.789 to 0.886) predicted a greater AUC for AF after OPCABG than miR-499 (AUC=0.759,95% CI:0.699 to 0.812),PLTP (AUC=0.743,95% CI:0.683 to 0.798) predicted individually (Z=3.154,3.804,all P<0.05). Conclusion: Increased serum miR-499 levels and decreased PLTP levels were independently associated with post-OPCABG AF,and serum miR-499 combined with PLTP levels were of high value in predicting post-OPCABG AF.

1298 Comparison of the Value of US-FNA and US-CNB in the Diagnosis of Thyroid Nodules of Different Sizes

Objective: To explore the value comparison of fine needle biopsy (US-FNA) and crude needle biopsy (US-CNB) in the diagnosis of different size thyroid nodules. Methods: A total of 106 patients with thyroid nodules admitted to our hospital from March 2020 to March 2022 were selected and grouped according to the maximum diameter of the nodule, Patients with a nodule diameter less than 10mm were included in group A and the maximum diameter more then 10mm was included in Group B. All patients were examined for US-FNA and US-CNB, and postoperative pathological diagnosis was performed. Postoperative pathological diagnosis was taken as the gold standard to explore the value of US-FNA and US-CNB in the diagnosis of thyroid nodules of different sizes. Results: There were 37 malignant nodules (2 follicular carcinomas, 28 papillary carcinomas, 3 thyroid lymphomas, and 4 thyroid sarcomas) and 13 benign nodules (2 nodular goiters with follicular adenomatous nodules, and 11 nodular goiters) in patients in group A. There were 39 malignant nodules (3 follicular carcinoma, 31 papillary carcinoma, 2 thyroid lymphoma, 3 thyroid sarcoma) and 17 benign nodules (4 nodular goiter with follicular adenoma nodules, 13 nodular goiter) in patients in group B. The accuracy of diagnosis of thyroid nodules by US-CNB and US-FNA in patients in group A was 80.00% and 82.00%, sensitivity was 86.49% and 83.78%, specificity was 61.54% and 76.92%, positive predictive value was 86.49% and 91.18%, negative predictive value was 61.54% and 62.50%, and kappa Consistency test analyzed kappa values were 0.480 (P=0.001), 0.565 (P<0.001) respectively. The accuracy of diagnosis of thyroid nodules by US-FNA and US-CNB in patients in group B was 73.21% and 78.57%, the sensitivity was 76.92% and 79.49%, the specificity was 64.71% and 76.47%, the positive predictive value was 83.33% and 88.57%, and the negative predictive value was 55.00% and 61.90%, respectively, and the kappa Consistency test was analyzed and the kappa values were 0.525 (P<0.001), 0.397 (p=0.003), respectively. Conclusion: When the patient's thyroid nodule is less then10 mm, US-FNA is chosen for diagnosis with high accuracy; when it is more 10 mm, US-CNB should be chosen for diagnosis.

1302 Predictive Value of IL-6 Combination with RDW and Extravascular Lung Water Index in Severe Acute Pancreatitis Complicated by ARDS

Objective: To investigate the predictive value of serum interleukin-6 (IL-6) combined with red blood cell distribution width (RDW) and extravascular lung water index (ELWI) in severe acute pancreatitis (SAP) complicated by acute respiratory distress syndrome (ARDS). Methods: A total of 134 patients with SAP admitted to our hospital from January 2017 to July 2022 were selected and divided into 48 cases in the ARDS group and 86 cases in the non-ARDS group according to whether they had concomitant ARDS. Multifactorial logistic regression was used to analyze the influencing factors of SAP-complicated ARDS, and ROC curves were used to analyze the predictive value of serum IL-6, RDW levels, and ELWI for SAP-complicated ARDS. Results: Serum IL-6, RDW levels and ELWI were elevated in the ARDS group compared to the non-ARDS group (P<0.05). Multifactorial logistic regression analysis showed that increased respiratory rate (OR=1.042, 95% CI: 1.002 to 1.083) and IL-6 (OR=1.012, 95% CI: 1.004 to 1.019), RDW (OR=2.225, 95% CI: 1.339 to 3.696), ELWI (OR= 1.482, 95% CI: 1.193 to 1.840) were elevated as independent risk factors for SAP complicated by ARDS, and elevated oxygenation index (OR=0.990, 95% CI: 0.983 to 0.996) was an independent protective factor (P<0.05). ROC curve analysis showed that serum IL-6 and RDW combined with ELWI (AUC=0.889, 95% CI: 0.824 to 0.937) predicted a greater area under the curve (AUC) for SAP complicated by ARDS than IL-6 (AUC=0.755, 95% CI: 0.673 to 0.825), RDW (AUC=0.743, 95% CI: 0.660 to 0.815) and ELWI (AUC=0.766, 95% CI: 0.685 to 0.835) predicted individually (P<0.05). Conclusion: Elevated serum IL-6, RDW levels and ELWI were independently associated with SAP complicated by ARDS, and serum IL-6 and RDW combined with ELWI were of high value in predicting SAP complicated by ARDS.

1308 Therapeutic Effect of Dagliflozin Combined with Glucagon-Like Peptide-1 Analog in Diabetes Mellitus with Chronic Heart Failure

Objective: To investigate the effects of dagliprazin combined with glucagon-like peptide-1 (GLP-1) analogs on blood glucose fluctuation and ventricular remodeling in patients with diabetes mellitus and chronic heart failure. Methods: Ninety diabetic patients with chronic heart failure were randomly divided into two groups. The patients in the control group were treated with GLP-1 analogues, and the patients in the study group were treated with dapagliflozin combined with GLP-1 analogues. The efficacy, blood glucose fluctuation and ventricular remodeling were compared between the two groups. Results: Compared with the control group, the overall response rate was higher in the study group (P<0.05); before and after treatment, glycosylated hemoglobin (HbA1c), fasting plasma glucose (FPG), 2h postprandial plasma glucose (2hPG) levels, amplitude of blood glucose excursion (LAGE), mean absolute difference of daytime blood glucose (MODD), amplitude of postprandial blood glucose excursion (PPGE), standard deviation of blood glucose level (SDBG), left ventricular remodeling index (LVRI), left ventricular mass index (LVMI), left ventricular end diastolic diameter (LVEDD) and left ventricular ejection fraction (LVEF) differences were greater in the study group than in the control group (P<0.05). Conclusion: Dapagliflozin combined with GLP-1 analogues can reduce blood glucose fluctuation and improve ventricular remodeling in diabetic patients with chronic heart failure.

1313 Analysis of Serological Risk Indexes for Continuous Peritoneal Dialysis-Associated Peritonitis

Objective: To investigate continuous peritoneal dialysis-associated peritonitis (peritonitis), and to analyze the related serological risk indexes. Methods: A total of 416 patients with uremia undergoing peritoneal dialysis in the hospital were retrospectively enrolled between March 2021 and July 2022. The clinical data of patients were collected, including general data and laboratory indexes. The peritonitis infection and distribution of pathogens were recorded. According to presence or absence of peritonitis, they were divided into infection group and non-infection group, and differences in clinical data were compared between the two groups. The influencing factors of infection were analyzed. The risk prediction model based on laboratory indexes was constructed by binary Logistic regression model, and its predictive value was analyzed by ROC curves. Results: Among the 416 patients, there were 102 cases (24.52%) with peritonitis. There were 102 strains of pathogens, mainly on Gram-positive bacteria [62.75% (64/102), including Staphylococcus epidermidis 32.81% (21/64) and Staphylococcus aureus 26.56% (17/64)] and Gram-negative bacteria [34.31% (35/102)], including Escherichia coli [45.71% (16/35)]. The proportion of cases with education level of primary school and below in infection group was higher than that in non-infection group, dialysis age was longer than that in non-infection group, and residual renal function (RRF) was lower than that in non-infection group (P<0.05). The levels of hemoglobin (Hb) and albumin (ALB) in infection group were lower than those in non-infection group, while levels of serum C-reactive protein (CRP), white blood cell count (WBC) and ferritin (SF) were higher than those in non-infection group (P<0.05). The results of Logistic regression analysis showed that CRP, WBC, SF and ALB were risk factors of peritonitis. ROC curve analysis showed that the risk prediction model based on the above 4 indexes was of good predictive value (P<0.05). Conclusion: The main pathogen is Gram-positive bacteria in uremia combined with peritonitis. Low education level, long dialysis age and low RRF may increase the risk of peritonitis infection. CRP, WBC, SF and ALB are serological risk indexes to predict peritonitis, which have good predictive value for infection.

1318 Value of Concentrated Growth Factors Combined with Biooss Bone Substitute Materials for the Treatment of Bone Loss in Mandibular Posterior Region with Immediate Implantation

Objective: To study the application value of concentrated growth factor (CGF) and Biooss bone substitute materials in mandibular implants. Methods: A total of 101 patients with immediate implant bone loss in mandibular posterior area in our hospital from June 2019 to June 2022 were retrospectively selected, and the patients were grouped according to differences in treatment regimen: 51 in observation group and 50 in control group, The control group was treated with the Biooss bone substitute material, the observation group was given concentrated growth factors on the basis of the control group.The patients were followed up by telephone and outpatient clinic 6 months after the treatment to compare the horizontal and vertical bone grafting, bone thickness and bone regeneration effects between the two groups; the clinical efficacy of the patients was evaluated according to the changes in the postoperative stability coefficients (ISQ), and the levels of bone thickness, bone density, and bone width were compared between the two groups at the immediate postoperative stage, at 3 months after the surgery and at 6 months after the surgery; and the patients were compared with respect to the degree of recovery of the postoperative masticatory function, the height of gingival papilla attachment and overall dental esthetic degree index measurements. The measured values of the patients' postoperative masticatory function recovery, gingival papilla attachment height, and overall dental esthetics were compared. At the end of the follow-up period, patients' satisfaction with the treatment was collected and the differences between groups were compared. Results: The effect of bone regeneration in the horizontal and vertical directions (94.00%,92.00%) was better than that of the control group (90.19%,86.27%). The ISQ mean of patients in control group decreased first and then increased, with the lowest ISQ score of the patients, and the ISQ mean of observation group showed an increasing trend. The ISQ mean was different between both groups at 1 week and 4 weeks after the operation, and the mean of observation group was higher than that of control group (P<0.05), but the difference between the immediate postoperative and 8 weeks was not significant (P>0.05). The immediate bone thickness, bone width and bone density values in the two groups were comparable (P>0.05); compared 3 and 6 months after surgery, and observation group was higher than serums (P<0.05). Compared with serums, the recovery degree of masticatory function, the height of the whole gingival nipple increased, with a statistically significant difference (P<0.05). The treatment satisfaction of observers was (96.00%)was higher than that of serums (84.31%), (P<0.05). Conclusion: Concentrate growth factor (CGF) and Biooss bone replacement material can improve patient satisfaction and bone regeneration effect, maintain the stability of postoperative implant, and have good clinical application effect.

1324 Efficacy of Laparoscopic Radical Prostatectomy with Different Bladder Approaches and Its Effect on Perioperative Parameters Postoperative Urinary Control Function, and Erectile Function

Objective: To investigate the curative effect of laparoscopic radical resection of prostate cancer with different bladder approaches. Methods: A total of 106 patients with prostate cancer admitted to our hospital from January 2020 to January 2022 were selected as research subjects, and were divided into observation group (53 cases, treated with laparoscopic anterior bladder approach) and control group (53 cases, treated with laparoscopic posterior bladder approach) according to simple randomization method. The clinical effect, perioperative indexes, urinary control function, erectile function, and complications of the two groups were observed. Results: After treatment, the total clinical effective rate of observation group was 98.11%, which was higher than 86.79% (P<0.05). All perioperative indexes in observation group were lower (P<0.05). The residual urine volume of observation group was lower, and the maximum urine flow rate was higher (P<0.05). The erectile function scores of sexual intercourse, sexual activity, ejaculation and sexual stimulation in both groups were higher (P<0.05). The above scores in the observation group were higher (P<0.05). The total complication rate of observation group was 1.89%, which was lower 5.66% (P>0.05). Conclusion: The anterior approach can improve the clinical efficacy, promote the recovery of patients, improve the urinary control function and erectile function after surgery, without obvious complications, and is worthy of clinical promotion.

1329 Prognostic Value of D-dimer Irisin and Caprini Thrombosis Risk Assessment Scale in Chronic Obstructive Pulmonary Disease Combined with Pulmonary Embolism

Objective: To explore the prognostic value of D-dimer (D-D), Irisin and Caprini thrombus analysis in chronic obstructive pulmonary disease patients with pulmonary embolism. Methods: A total of 151 patients with chronic obstructive pulmonary disease complicated with pulmonary embolism treated in our hospital from January 2019 to August 2022 were selected as the observation group, while 151 chronic obstructive pulmonary disease patients without complicated with pulmonary embolism were selected as the control group. The D-dimer, Irisin and Caprini thrombus Risk Assessment Scale were compared between the two groups. At the same time, the differences of D-dimer, Irisin and Caprini thrombus Risk Assessment Scale in patients with different clinical characteristics and prognosis in the observation group were analyzed. Results: The D-dimer and Caprini thrombus risk assessment scale in the observation group were (1.50±0.34) mg/L and (5.40±0.98) points respectively, which were significantly higher than those in the control group (P<0.05), while Irisin was (22.54±4.03) ng/ml, which was significantly lower than that in the control group (P<0.05). The D-dimer and Caprini thrombus risk assessment scale of extremely high-risk patients in the observation group were (1.58±0.36) mg/L and (7.89±0.91) points respectively, which were significantly higher than other patients (P<0.05), while Irisin was (19.98 ± 4.03) ng/ml, which was significantly lower than other patients (P<0.05). There was a positive correlation between D-dimer and Caprini thrombus risk assessment scale score in the observation group (r=0.465, P<0.05). Irisin in the observation group was negatively correlated with the score of the thrombus risk assessment scale (r=-0.511, P<0.05). The D-dimer and Caprini thrombus risk assessment scale of the dead patients in the observation group were (1.72±0.30) mg/L and (6.72±0.91) points respectively, which were significantly higher than those of the surviving patients (P<0.05), while Irisin was (18.89±4.12) ng/ml, which was significantly lower than those of the surviving patients (P<0.05). The area under the ROC curve of D-dimer, Irisin, Caprini thrombus risk assessment scale prediction of death in patients with chronic obstructive pulmonary disease and pulmonary embolism were 0.811, 0.715, 0.890, respectively, P<0.05. Conclusion: The D-dimer, and the Caprini Thrombus Risk Assessment Scale score of COPD patients with pulmonary embolism increase, but the level of Irisin decrease, the Caprini Thrombosis Risk Assessment Scale has a high value in evaluating the prognosis of COPD with pulmonary embolism.

1335 Effect of Less Invasive Surfactant Administration Combined with Caffeine Citrate on Neonatal Respiratory Distress Syndrome and Its Influence on Blood Gas Indicators and Ventilation Time

Objective: To explore the effect of less invasive surfactant administration (LISA) combined with caffeine citrate in the treatment of neonatal respiratory distress syndrome (NRDS) and its influence on blood gas indicators and ventilation time. Methods: A total of 143 children with NRDS admitted to the hospital from September 2020 to September 2022 were selected and divided into combination group (n=72, LISA combined with caffeine citrate treatment) and conventional group (n=71, LISA treatment) according to the two-color sphere random grouping method. The clinical efficacy, respiratory mechanics indicators [endogenous positive end-expiratory pressure, airway resistance, static compliance], blood gas indicators [partial pressure of carbon dioxide (PaCO2), partial pressure of oxygen (PaO2), blood power of hydrogen (pH) value], mechanical ventilation time, oxygen therapy time, apnea and treatment complications were compared between the two groups. Results: There was no statistical difference in the overall effective rate between the two groups (P>0.05), but the markedly effective rate in combination group was higher than that in conventional group (P<0.05). After treatment, the endogenous positive end expiratory pressure and airway resistance in both groups were lower than before treatment, while static compliance was higher than before treatment (P<0.05). Moreover, the amplitude of changes in endogenous positive end expiratory pressure, airway resistance, and static compliance in the combination group before and after treatment was greater than that in the conventional group (P<0.05). After treatment, the PaCO2 levels in both groups were lower than before treatment, while the PaO2 and pH values were higher than before treatment (P<0.05). Moreover, the changes in PaCO2 and PaO2 levels in the combination group before and after treatment were greater than those in the conventional group (P<0.05). The mechanical ventilation time, oxygen therapy time, and apnea frequency in combination group were shorter or less than those in conventional group (P<0.05). The incidence rates of bronchopulmonary dysplasia and ventilator-associated pneumonia in combination group were lower than those in conventional group (P<0.05). Conclusion: LISA combined with caffeine citrate has a significant effect in the treatment of NRDS, and it can obviously improve the blood gas analysis indicators and shorten the ventilation time, and it has good safety and is worthy of clinical promotion and application.

Objective: To investigate the clinical value of carotid artery ultrasound parameters combined with plasma small and dense low-density lipoprotein (sd-LDL) in evaluating atherosclerotic cardiovascular disease (ASCVD) in diabetic patients. Methods: The clinical data of 104 diabetic patients admitted to the hospital from July to December 2022 were analyzed retrospectively. Patients with simple diabetes were defined as the control group (n=51) and those with ASCVD were defined as the observation group (n=53). Carotid artery ultrasound parameters and sd-LDL were compared between groups. Logistic regression analysis was performed to screen the influencing factors of ASCVD in diabetic patients. The clinical value of carotid artery ultrasound parameters and sd-LDL in evaluating ASCVD in diabetic patients was discussed. Results: Carotid artery ultrasound parameters (carotid intima-media thickness (IMT) and plaque score) and sd-LDL level in the observation group were larger/higher than the control group (P<0.05). The numbers of smokers in the two groups were close (P<0.05). Logistic regression analysis found that IMT, plaque score and sd-LDL level were influencing factors of ASCVD in diabetic patients (P<0.05). Conclusion: Carotid artery ultrasound parameters and sd-LDL are much helpful for evaluating ASCVD in diabetic patients. Clinically, combination of IMT and plaque score and sd-LDL level can be adopted to provide a basis for clinical diagnosis and treatment of ASCVD.

Objective: To analyze the relationship between the changes of coagulation parameters and the severity of the disease and adverse pregnancy outcomes in patients with pregnancy induced hypertension disease (PIH). Methods: A total of 199 hospitalized patients with pregnancy induced hypertension in late pregnancy from May 2019 to May 2021 were retrospectively selected as the study group: 55 cases of pregnancy induced hypertension, 75 cases of pre eclampsia, 69 cases of severe pre eclampsia, and 59 healthy pregnant women of the same age and week as the control group. The anticoagulation related parameters of each group and the adverse pregnancy outcome rate of the study group were statistically analyzed. Results: There were significant difference in 9 parameters between the hypertensive group during pregnancy and the control group (P<0.05,P<0.01), and there were significant difference in activated partial thromboplastin time (APTT), fibrinogen (FIB), thrombin time (TT), platelet count (PLT), mean platelet volume (MPV), and platelet hematocrit (PCT) between preeclampsia and severe preeclampsia and the control group (P<0.05,P<0.01). Spearman showed that FIB was positively correlated with them (P<0.01), MPV, APTT, TT, PLT, PCT were negatively correlated with them (P<0.01). With the aggravation of the disease, the adverse pregnancy outcome rate increased, P<0.01, and the difference was statistically significant. Conclusion: FIB, MPV, APTT, TT and PLT have potential predictive value for disease severity and adverse outcome.

1348 Effect of Q Self-Traction Method on Early Esophageal Cancer and Its Influence on the Complication Rate

Objective: To explore the effect of Q self-traction method in the treatment of early esophageal cancer and its influence on the incidence rates of complications. Methods: Retrospective analysis was performed on the clinical data of 184 patients with early esophageal cancer admitted to the hospital from June 2019 to June 2022. According to different surgical methods, the patients were divided into observation group (89 cases, Q self-traction method) and control group (95 cases, standard endoscopic submucosal dissection). Perioperative indicators, therapeutic effect, occurrence of complications, and recurrence and scores of Generic Quality of Life Inventory-74 (GQOLI-74) at 6 months after surgery were compared between the two groups. Results: The intra-operative blood loss, dissection time, surgical time and hospital stay in observation group were shorter than those in control group (P<0.05). There were no statistically significant differences in the en bloc resection rate and curative resection rate between the two groups (P>0.05). There were no statistical differences in the incidence rates of various complications between the two groups (P>0.05), but the total incidence rate of complications in observation group was lower than that in control group (P<0.05). At 6 months of follow-up after surgery, there was 1 case of recurrence (1.12%) in observation group and 3 cases of recurrence (3.16%) in control group (P>0.05). The scores of items of GQOLI-74 showed no statistical differences between the two groups before surgery (P>0.05), and the GQOLI-74 scores at 6 months after surgery were significantly increased (P<0.05), and the increase of scores in observation group were higher compared with those in control group (P<0.05). Conclusion: Q self-traction method can optimize ESD operation for early esophageal cancer, and its therapeutic effect is comparable to that of traditional ESD, but the incidence rates of complications are low and the quality of life of patients is significantly improved.

1353 Clinical Analysis of Deep Lymph Node Dissection of Right Recurrent Laryngeal Nerve in Patients with Papillary Thyroid Carcinoma Complicated with Hyperthyroidism

Objective: To analyze the clinical value of right recurrent laryngeal nerve deep lymph node dissection (LN-prRLN) in patients with papillary thyroid carcinoma (PTC) complicated with hyperthyroidism. Methods: From March 2019 to March 2021, 115 patients with right or bilateral PTC complicated with hyperthyroidism in our hospital were selected. All patients underwent total thyroidectomy, central lymph node dissection, and LN-prRLN dissection. The lymph node metastasis of patients was analyzed, and the clinical data of patients with LN-prRLN metastasis and those without LN-prRLN metastasis were compared. Logistic regression analysis was used to summarize the influencing factors of LN-prRLN metastasis. Results: The total metastasis rates of clinical lymph node negative patients in sub-region ⅳ a+cervical region, sub-region ⅳ b+cervical region and right cervical lymph nodes were lower than those in clinical lymph node positive patients, with statistical significance (P<0.05). The total metastatic rate of LN-prRLN in 15 patients was 26.96%(31/115). Logistic regression analysis showed that with LN-prRLN metastasis as the dependent variable, the influencing factors of tumor diameter ≥1cm, multiple lesions, invasion of extraglandular lobe, metastasis in subregion Ⅳa and right cervical lymph node metastasis were statistically significant (P<0.05). Conclusion: The LN-prRLN metastasis rate is high in PTC patients with hyperthyroidism. For patients with LN-prRLN metastasis risk factors, it is recommended to actively carry out LN-prRLN cleaning.

1358 Effect of Vitrectomy Combined with Panretinal Photocoagulation on Refractive Status and Tear Film Function in Patients with Diabetes Retinopathy

Objective: To investigate the effect of vitrectomy combined with panretinal photocoagulation on refractive status and tear film function in patients with diabetes retinopathy. Methods: A total of 68 patients with diabetes retinopathy were selected. The standard three channel 23G vitrectomy combined with panretinal photocoagulation was used in the vitrectomy combined group, and the fundus laser therapeutic apparatus was used in the panretinal photocoagulation group. The efficacy was compared. Results: Before treatment, there was no significant difference in the best corrected visual acuity, tear film rupture time, and tear secretion test between the whole retina photocoagulation group and the glass cutting combined group (P>0.05). After 6 months of treatment, the glass cutting combined group had higher best corrected visual acuity, tear film rupture time, and tear secretion test than the whole retina photocoagulation group (P<0.05). The difference in tear film rupture time before and after treatment and the difference in tear secretion test before and after treatment in the combination group was higher than that in the whole retina photocoagulation group (P<0.05). Before treatment, there was no significant difference in refractive status and axial length between the whole retina photocoagulation group and the glass cutting combined group (P>0.05). The refractive status and axial length of the glass cutting combined group were higher than those of the whole retina photocoagulation group (P<0.05). Conclusion: Vitrectomy combined with panretinal photocoagulation is helpful to reduce the changes of refractive status and stabilize tear film function in patients with diabetes retinopathy.

1362 Clinical Study on the Combination of Yaotongning Capsule and Spinal Micro-Adjustment Manipulation on Improving Pain and Lumbar Dysfunction in Patients with Discogenic Low Back Pain

Objective: To study the clinical efficacy and safety of spinal micro-adjustment manipulation combined with drug Yaotongning capsule in treating discogenic low back pain (DLBP) with cold-dampness obstruction syndrome. Methods: Seventy patients with DLBP were randomly divided into a treatment group and control group,35 cases in each group. Yaotongning capsules and spinal micro-adjustment manipulation were used to treat the patients in the treatment group, whereas Diclofenac sodium sustained-release tablets and spinal micro-adjustment manipulation were used to treat the patients in the control group.The Oswestry disability index (ODI), Japanese orthopaedic association scores (JOA), visual analogue scale (VAS) and safety indicators of the two groups were compared and evaluated before treatment and 1,3 and 6 weeks after treatment. The satisfaction rate with the evaluation of lumbar function improvement is 97%, the improvement rate of lumbar function is 86%, and the overall excellent and good rate is 97%.. Results: After 1 week,3 weeks and 6 weeks of treatment, patients in the two groups had improvements in their VAS, ODI, and JOA scores when compared to their pre-treatment scores (P<0.05). The treatment group's effectiveness was inferior to the control group's before three post-operative weeks, but it was comparable to that of the control group by six weeks. However, the treatment group experienced fewer adverse responses than the control group did at that time. Conclusion: Spinal micro-adjustment manipulation combined with Chinese patent medicine Yaotongning capsule could significantly improve the clinical symptom of DLBP with cold-dampness obstruction syndrome, and the effect was more obvious after 6 weeks of treatment.

1367 Efficacy and Postoperative Complications of PVP and PKP for Osteoporotic Thoracolumbar Compression Fractures under CT Localization

Objective: To investigate the efficacy and postoperative complications of percutaneous vertebroplasty (PVP) and percutaneous kyphoplasty (PVP) under the localization of computed tomography (CT) in the treatment of osteoporotic thoracolumbar compression fractures. Methods: The clinical data of 98 patients with osteoporotic thoracolumbar compression fracture admitted to our hospital from February 2020 to June 2021 were retrospectively analyzed. According to different surgical methods, they were divided into PVP group (47 cases) and PKP group (51 cases). The general operation conditions of the two groups, the vertebral height, Cobb angle, visual analog scale (VAS) and Barthel index evaluation scale of the two groups were compared before and 3 days after operation, and the complications of the two groups were observed. Result: The operative time, hospital stay and bone cement injection amount in PVP group were lower than those in PKP group, P<0.05; 3 days after operation, the vertebral body height of both groups increased and PKP group was higher than PVP group, Cobb Angle of both groups decreased and PKP group was lower than PVP group; preoperative and postoperative 3d vertebral height and Cobb Angle difference in PKP group were higher than those in PVP group (P<0.05). 3 days after surgery, VAS scores of both groups decreased and PKP group was lower than PVP group, Barthel index scores of both groups increased and PKP group was higher than PVP group; the difference values of 3dVAS score and Barthel index score in PKP group before and after surgery were higher than those in PVP group, P<0.05; the total complication rate of PKP group was 7.84% lower than that of PVP group 17.02%, P>0.05. Conclusion: Both CT-guided PKP and PVP have ideal effects and high safety in the treatment of osteoporotic thoracolumbar compression fractures. However, compared with CT-guided PVP, CT-guided PKP can improve the patient's vertebral body condition and reduce pain, which is worthy of clinical reference.

1372 Analysis of the Predictive Value of the Geriatric Nutritional Risk Index for Osteoporosis in Elderly Patients with Type 2 Diabetes Mellitus

Objective: To investigate the relationship between the Geriatric Nutrition Risk Index (GNRI) and bone mineral density in elderly patients with T2DM and the predictive value of GNRI for osteoporosis in elderly patients with type 2 diabetes. Methods: A total of 445 elderly patients with T2DM attending the First Affiliated Hospital of Chengdu Medical College from January 2019 to June 2022 were retrospectively selected, and the general data, bone mineral density data and relevant blood biochemical indexes of the study subjects were collected to calculate the geriatric nutritional risk index (GNRI) score. According to GNRI, they were divided into non-nutritional risk group (GNRI>98, n=283), low nutritional risk group (92≤GNRI≤98, n=95), and medium-high nutritional risk group (GNRI<92, n=67). Spearman correlation was used to analyze the correlation between BMD and the above indexes, and dichotomous logistic regression was used to analyze the relationship between GNRI and osteoporosis in elderly T2DM patients. The predictive value of GNRI for osteoporosis was analyzed using subject work characteristic (ROC) curves. Results: The age and prevalence of osteoporosis gradually increased in the non-nutrition risk group, low-nutrition risk group, and medium-high nutrition risk group, with statistically significant differences between the groups (P<0.001). Total protein (TP), serum albumin (ALB), serum calcium level, femoral neck BMD, lumbar spine BMD, and total hip BMD decreased sequentially in the non-nutrition risk group, low-nutrition risk group, and medium-high nutrition risk group, and the difference between the groups was statistically significant (P<0.05). spearman correlation results showed that GNRI was positively correlated with BMD at all sites (r = 0.211, 0.183, 0.180, P<0.001). The results of dichotomous logistic regression analysis showed that GNRI was an independent protective factor for the development of osteoporosis [OR=-0.155, 95% CI (0.753, 0.974), P<0.05]. The results of ROC curve analysis showed that GNRI had 43.43% sensitivity, 74.32% specificity and 17.76% Yordon index for the diagnosis of osteoporosis. The best cut-off value was 98.282. Conclusion: GNRI levels in elderly T2DM patients were positively correlated with BMD at all sites; GNRI is an independent protective factor for the development of osteoporosis and may also be a valid predictor of the development of osteoporosis in elderly T2DM patients.

1377 Effect of Modified Anterolateral Approach Combined with Modified Posteromedial Approach on Stability and Functional Recovery of Ankle Joint in the Treatment of Trimalleolar Fracture

Objective: To explore the impact of improved anterolateral approach combined with posterior medial approach on ankle stability and functional recovery in patients with trimalleolar fractures. Methods: A retrospective analysis was conducted on 102 patients with trimalleolar fractures treated in our hospital from January 2017 to December 2021. According to different surgical methods, they were divided into a modified group of 49 cases (modified anterolateral approach combined with posterior medial approach) and a traditional group of 53 cases (traditional posterior lateral approach combined with posterior medial approach). The perioperative related indicators, clinical treatment efficacy, incidence of incision complications, ankle stability, and recovery of ankle function were compared between the two groups. Results: The intraoperative bleeding volume, surgical time, time from surgery to bed activity, and fracture healing time in the improved group were significantly lower than those in the traditional group, with statistical significance (P<0.05). The excellent and good treatment rate of the improved group was significantly higher than that of the traditional group, and the difference was statistically significant (P<0.05); the incidence of incision complications in the improved group was significantly lower than that in the traditional group (P<0.05). The comparison of Baird Jackson scores between the two groups showed a statistically significant difference (P<0.05) at 6 months after surgery>2 months after surgery>1 month after surgery>before surgery, and the postoperative improvement group had a significantly higher score, with a statistically significant difference (P<0.05); the comparison of AOFAS scores between the two groups showed a statistically significant difference (P<0.05) at 6 months after surgery>2 months after surgery>1 month after surgery>before surgery, and the postoperative improvement group was significantly higher, with a statistically significant difference (P<0.05). Conclusion: The combination of modified anterior lateral approach and modified posterior medial approach for the treatment of trimalleolar fractures can more effectively increase ankle stability, promote ankle function recovery, and achieve better treatment efficacy with fewer complications.

1382 Observation of the Analgesic Effect of Esketamine after Hysteromyomectomy under Gynecological Laparoscopy

Objective: To investigate the effect of low-dose Esketamine combined with sufentanil on postoperative intravenous analgesia after gynecological laparoscopic surgery. Methods: A total of 80 cases of elective gynecological laparoscopic myomectomy were randomly divided into S group and KS group, with 40 cases in each group. Both groups of patients were given general anesthesia and PCIA analgesia after operation. Group S: sufentanil 50 μ g+ondansetron 8 mg, group KS: sufentanil 25 μ g+esketamine 25 mg+ondansetron 8 mg, and 0.9% sodium chloride was added to 50ml. To evaluate the postoperative analgesia effect of the two groups, the main monitoring indicators were VAS at 2h (T1), 6h (T2) and 24h (T3) after operation, and the secondary indicators were MAP, HR and PCIA press times, adverse reactions within 24h, preoperative and postoperative anxiety and depression (HADS scale), and postoperative analgesia satisfaction. Results: VAS of S group at T2 and T3 was higher than that of KS group (P<0.05), and there was no significant difference between the two groups at T1 (P>0.05). The number of PCIA single press in group S was higher than that in group KS (P<0.01). In T1, T2 and T3, there was no significant difference in MAP and HR between the two groups (P>0.05). The score of HADS-A in group S was significantly higher than that in group KS (P<0.05). The number of cases of adverse reactions such as drowsiness and skin itching in group S was higher than that in group KS (P<0.05), while the number of cases of nausea, vomiting and dizziness was not statistically significant (P>0.05). The percentage of pain relief and satisfaction of postoperative analgesia in group S were lower than those in group KS. Conclusion: Low-dose Esketamine combined with sufentanil for intravenous analgesia after gynecological endoscopic surgery has a good analgesic effect, reduces adverse reactions, and improves patients' satisfaction with postoperative analgesia.

1388 Effectiveness of Low-Dose Sacubitril Valsartan versus Perindopril in the Treatment of Elderly Patients with HFrEF and Its Effect on Hemodynamics and Cardiac Remodeling

Objective: To explore the effect of low-dose sacubitril/valsartan and perindopril in the treatment of elderly patients with heart failure with reduced ejection fraction (HFrEF) and the influence on hemodynamics and cardiac remodeling. Methods: The clinical data of 133 elderly patients with HFrEF admitted to the hospital from January 2020 to October 2022 were retrospectively analyzed, and the patients were divided into observation group (conventional treatment+low-dose sacubitril/valsartan, n=69) and control group (conventional treatment+perindopril, n=64) according to the treatment methods. Cardiac function grading, cardiac remodeling indicators, hemodynamic indicators and serum cardiac function indicators were compared between the two groups before treatment and at 3 months of treatment. Adverse drug reactions were recorded in the two groups. Results: The NYHA cardiac function grading in observation group was significantly better than that in control group at 3 months of treatment (Z=2.112, P=0.035). At 3 months of treatment, the LVEF in the two groups was higher than that before treatment (P<0.05) while the LVEDd and LVESD were lower than those before treatment (P<0.05), and the differences before and after treatment in the two groups showed statistical significance (P<0.05). The HR in observation group at 3 months of treatment was lower than that before treatment (P<0.05) while the CO and CI were higher than those before treatment (P<0.05), and the differences before and after treatment revealed statistical significance in the two groups (P<0.05). The levels of serum NT-proBNP, hs-CRP and cTnT in observation group at 3 months of treatment were lower than those before treatment (P<0.05), and the differences before and after treatment in the two groups revealed statistical significance (P<0.05). There were no statistically significant differences in the incidence rates of adverse reactions such as hypotension, dry cough and dizziness between the two groups (P>0.05). Conclusion: Low-dose sacubitril/valsartan and perindopril are safe and effective in the treatment of HFrEF. The former one is more effective in improving the cardiac function and cardiac remodeling and stabilizing the hemodynamics.

1393 Clinical Efficacy Analysis of Duloxetine Combined with Pregabalin in the Treatment of Fibromyalgia Syndrome

Objective: To study the clinical effect of duloxetine combined with pregabalin in the treatment of fibromyalgia syndrome on improving pain symptoms and psychological disorders of patients. Methods: In this prospective study, 120 patients with fibromyalgia syndrome who were treated in our hospital from June 2020 to June 2022 were selected as research subjects. All patients were treated with pregabalin, and patients in the observation group were treated with duloxetine on this basis, and both groups were treated for 6 months. Differences in treatment outcomes, pain, negative emotions, sleep, fatigue, and safety were compared between the two groups. Results: The comparison of VAS scores between the control group and the observation group before treatment was statistically insignificant (P>0.05), and significantly decreased after treatment in both groups, and VAS scores in the observation group were significantly decreased at each time point after treatment compared with the control group (P<0.05). Statistically, there was no significant difference in HAMA and HAMD scores between the control group and the observation group before treatment (P>0.05). After treatment, the HAMA and HAMD scores in the observation group were significantly lower than those in the control group. The total effective rate of observation group (95.00%) was significantly higher than that of control group (83.33%). The comparison of PSQI score and FSS score between the control group and the observation group before treatment was statistically insignificant (P>0.05), and the two groups were significantly reduced after treatment. The change degree of PSQI score and FSS score in the observation group was significantly higher than that in the control group. There was no significant difference in the incidence of adverse reactions between the two groups during treatment(P>0.05). Conclusion: When duloxetine combined with pregabalin was used to treat fibromyalgia syndrome, the pain, sleep quality and psychological status of the patients were significantly improved, and the therapeutic effect was good.

1398 Effects of Oxycodone and Remifentanil Combined with Propofol on Hemodynamics, Neurological Function and Prognosis of Patients Operated for Craniocerebral Injury

Objective: To explore the effects of oxycodone and remifentanil combined with propofol on hemodynamics, neurological function and prognosis of patients with craniocerebral injury. Methods: A total of 150 patients with craniocerebral injury admitted from September 2019 to September 2021 were selected as the study subjects and randomly divided into study group and control group, with 75 patients in each group. The study group was given oxycodone combined with propofol, and the control group was given remifentanil combined with propofol. The levels of hemodynamics [Oxygen saturation (SpO2), mean arterial pressure (MAP), heart rate (HR)] before operation (T0), 30 minutes after anesthesia (T1), immediately after operation (T2), and 24 hours after operation (T3) were compared between the two groups, and the levels of nerve function [Tau protein (Tau), neuron-specific enolase (NSE), glial fibrillary acidic protein (GFAP)] at T0 and T3 were compared. The prognosis of the two groups was compared after 3 months follow-up. Results: The differences of SpO2, MAP and HR between the two groups and at different time points and the interaction between time and groups were statistically significant (P<0.05). At T0, there were no differences in serum Tau, NSE and GFAP levels between two groups (P>0.05). At T3, the levels of serum Tau, NSE and GFAP in both groups were lower than those at T0, and the reduction degree in the study group was greater than that in the control group (P<0.05). There were no deaths in the two groups, and the prognosis of the study group was better than that of the control group (P<0.05). Conclusion: Compared with remifentanil combined with propofol, oxycodone combined with propofol can obtain a more stable hemodynamic level in patients with craniocerebral injury, and can significantly improve the neurological function and prognosis of patients, which is more worthy of clinical application.
