881 The Role and Mechanism of miR-876-5p in the Progression of Gastric Cancer by Regulating Epithelial-Mesenchymal Transition through Negative Feedback

Objective: To explore the effect and mechanism of miR-876-5p regulating epithelial-mesenchymal transition (EMT) on the progression of gastric cancer. Methods: The differences in miR-876-5p expression in gastric cancer tissues and normal tissues adjacent to the cancer were determined by real-time fluorescence quantitative PCR; gastric cancer cells were divided into control group, EMT group, miR-NC+EMT group, miR-876-5p+EMT group, miR-876-5p+EMT+isoproterenol hydrochloride (ISO) group, miR-NC or miR-876-5p were transfected into gastric cancer cells according to the grouping, and 5 ng/mL transforming growth factor-β (TGF-β1) was induced for 24 h to establish an EMT model of gastric cancer cells and 20 μmol/L extracellular. After treatment, the expression of E-cadherin, N-cadherin and vimentin was detected by immunofluorescence staining in each group of cells, and the expression of E-cadherin, Vimentin, N-cadherin, matrix metalloproteinase-2 (MMP-2) and MMP-9 protein was determined by protein immunoblotting (Western blot) in each group of cells. The expression of E-cadherin, Vimentin, N-cadherin, Snail, matrix metalloproteinase-2 (MMP-2) and MMP-9 in each group of cells was determined by western blot. Western blot was performed to determine the expression of ERK signaling pathway-related proteins in each group of cells. Results: The relative expression of miR-876-5p was decreased in gastric cancer tissues compared with normal tissues next to cancer (P<0.05); compared with the EMT group, the fluorescence expression of E-cadherin was enhanced and that of Vimentin and N-cadherin was decreased in the miR-876-5p+EMT group of gastric cancer cells, and the relative expression of E-cadherin protein was upregulated while Vimentin, N-cadherin, Snail, MMP-2, and MMP-9 protein relative expression were all downregulated (P<0.05), cell proliferation activity and EdU positivity were reduced (P<0.05), migration number and invasion number were decreased (P<0.05), and p-ERK1/2 protein relative expression was downregulated (P<0.05); while with miR-876-5p+EMT group, E-cadherin fluorescence expression was diminished, Vimentin and N-cadherin fluorescence expression was enhanced, E-cadherin protein relative expression was down-regulated, and Vimentin, N-cadherin, Snail, MMP- 2, MMP-9 protein relative expression were upregulated (P<0.05), cell proliferation activity and EdU positivity were increased (P<0.05), migration number and invasion number were increased (P<0.05), while p-ERK1/2 protein relative expression was upregulated (P<0.05). Conclusion: miR-876-5p affects tumor progression by regulating the EMT process in gastric cancer cells, and this effect may be related to the inhibition of TGF-β1-mediated activation of the ERK signaling pathway.

887 Effect of Cryptotanshinone on High Glucose-Induced Glomerular Endothelial Cell Injury through the Regulation of Sirt1/FoxO1 Signaling Pathway

Objective: To investigate the effect of cryptotanshinone (CT) on the injury of glomerular endothelial cells (GEC) induced by high glucose by regulating silent information regulator 1 (Sirt1)/forkhead box protein O1 (FoxO1) signal pathway. Methods: GEC was divided into control group (NR group,5mmol/L D-glucose), high glucose group ( HG group, 30mmol/L D-glucose), L-CT group ( GEC treated with 1 μmol/L CT+30 mmol/L D-glucose), M-CT group (GEC treated with 5 μmol/L CT+30 mmol/L D-glucose), H-CT group (GEC treated with 10 μmol/L CT+30 mmol/L D-glucose), and SIRT1-IN-1 group (GEC treated with 10 μmol/L CT+30 mmol/L D-glucose+0.05 mol/L Sirt1/FoxO1 signal pathway inhibitor SIRT1-IN-1); MTT assay was used to detect the effects of CT on the cytotoxicity and viability of GEC cells; the levels of GEC inflammatory factors and oxidative stress indicators were detected by ELISA; the apoptosis of GEC was detected by flow cytometry; Western blot was used to detect the expression of autophagy, apoptosis, fibrosis factors and Sirt1/FoxO1 pathway related proteins in cells. Results: 0-40 μmol/L CT had no obvious toxic effect on GEC. Compared with NR group, OD570 value, content of SOD, the levels of Bcl-2, Beclin1, LC3-Ⅱ/Ⅰ, p-Sirt1/Sirt1, p-FoxO1/FoxO1 proteins in HG group decreased obviously (P<0.05), the apoptosis rate of GEC, the levels of Bax, cleaved Casase-3, Col-I, TGF-1 proteins, contents of IL-6, IL-8, TNF-α, and MDA increased obviously (P<0.05); compared with HG group, the proliferation activity, the levels of autophagic protein and pathway protein in L-CT group, M-CT group and H-CT group increased obviously (P<0.05), the levels of inflammatory reaction, oxidative stress, fibrosis factor protein and apoptosis rate decreased obviously (P<0.05); SIRT1-IN-1 eliminated the beneficial effect of CT on GEC. Conclusion: CT may improve GEC injury induced by high glucose by up-regulating Sirt1/FoxO1 signal pathway.

893 Killing Effect of 5-Fluorouracil Cell Vesicles on Colorectal Cancer Cell Line HCT116

Objective: To explore the killing effect of 5-fluorouracil cell vesicles on colorectal cancer cell line HCT116. Methods: The HCT116 group, cell vesicles group (EVs suspension 5mL, 106 cells/mL), 5-fluorouracil group (5-fluorouracil 5mL, 30μg/mL), and 5-fluorouracil drug-loaded vesicles group (EVs suspension 5mL, 106 cells/mL+5-fluorouracil 5mL, 30μg/mL) were set up, and 6 parallel samples of each well of the above groups were incubated for 72h. After incubation, the levels of cell proliferation, invasion and apoptosis were determined, and the levels of miR-128 and PIK3 were determined by RT-PCR and western blotting. Results: The OD value, survival rate, number of monoclonal formation, number of membrane penetration, migration distance, apoptosis rate, miR-128 level, PI3K mRNA and protein levels in cell vesicles group were not significantly different from those in HCT116 group(P>0.05). OD value, survival rate, number of monoclonal formation, number of membrane penetration, migration distance, PI3KmRNA and protein levels in 5-fluorouracil group and 5-fluorouracil drug-loaded vesicles group were significantly lower than those in HCT116 group and cell vesicles group(P<0.05), and the apoptosis rate and the level of miR-128 were significantly higher than those in HCT116 group and cell vesicle group (P<0.05). OD value, survival rate, number of monoclonal formation, number of membrane penetration, migration distance, PI3K mRNA and protein levels in 5-fluorouracil drug-loaded vesicles group were significantly lower than those in 5-fluorouracil group, and the apoptosis rate and miR-128 level were significantly higher than those in 5-fluorouracil group(P<0.05). Conclusion: Extracellular vesicles loaded with 5-fluorouracil can significantly inhibit the proliferation and invasion level of colorectal cancer cell line HCT116, and enhance its apoptosis level, which may be related to the mechanism that extracellular vesicles loaded with 5-fluorouracil can promote the high expression of miR-128 and low expression of PI3K in colorectal cancer cell line HCT116.

899 Effect of CircRNA YAP1 on Angiogenesis of Femoral Head Microvascular Endothelial Cells

Objective: To explore the role and mechanism of circular RNA (circRNA) YAP1 in the regulation of microvascular endothelial cell (MVEC) angiogenesis in steroid-induced osteonecrosis of the femoral head (SONFH) rats. Methods: Adult male SD rats were injected with methylprednisolone (MPS, 20 mg/kg/d) for 3 weeks to induce the SONFH model (n=30). The control group (n=30) was injected with an equal amount of saline. Micro-CT scanning and HE staining were performed to evaluate whether the model was successfully established. MVECs were isolated from the femoral heads of the rats using CD31 as a marker and identified by immunofluorescence staining. The MVECs were divided into three groups: the pcDNA-YAP1 group (overexpression of circRNA YAP1), the pcDNA-NC group (negative control), and the pcDNA-YAP1+dickkopf-1 group (treated with Wnt1/β-catenin pathway inhibitor dickkopf-1). The expression of circRNA YAP1 was detected using qPCR, while the expressions of Wnt1, β-catenin, VEGF-A, and vimentin were detected using Western blotting. Cell proliferation was detected using the CCK-8 assay, while cell migration was detected using the Transwell assay. Tube formation assays were performed to detect the angiogenic ability of the cells. Results: The HE staining results showed that the control group was healthy, with no bone necrosis, while the SONFH group showed severe femoral head necrosis and deterioration. The results of micro-CT scanning showed that the control group had dense and regular bone trabeculae, with normal numbers and morphology of bone trabeculae. The SONFH group was accompanied by an empty bone pit, a decrease in the number of bone trabeculae, and shortened bone trabeculae. Compared with the control group (1.00±0.08; 1.00±0.12; 1.00±0.15), the expressions of circRNA YAP1 (0.27±0.04), Wnt1 (0.49±0.03), and β-catenin (0.33±0.03) in the SONFH group were significantly reduced (t=12.158, P=0.009; t=10.596, P=0.012; t=10.080, P=0.014). In addition, MVECs from the femoral heads of the Ctrl and SONFH groups were successfully isolated. Compared with the control group (1.00±0.00; 1.00±0.02; 1.00±0.01), the expressions of circRNA YAP1 (0.15±0.01), Wnt1 (0.28±0.05), and β-catenin (0.31±0.02) in the MVECs from the SONFH group were significantly reduced (t=17.625, P=0.004; t=16.154, P=0.005; t=13.596, P=0.007).In MVECs, the expressions of circRNA YAP1, Wnt1, β-catenin, VEGF-A and vimentin were up-regulated in pcDNA-YAP1 group compared with pcDNA-NC group. The cell proliferation rate, cell migration rate and tubule formation rate were significantly increased (all P<0.05). Compared with pcDNA-YAP1 group, the expression of circRNA YAP1 in pcDNA-YAP1+dickkopf-1 group had no significant change (P>0.05), while the expression of Wnt1, β-catenin, VEGF-A and vimentin were down-regulated. The cell proliferation rate, cell migration rate and tubule formation rate were significantly inhibited (all P<0.05). Conclusion: CircRNA YAP1 promotes the angiogenesis of bone endovascular endothelial cells in SONFH rats by activating Wnt1/β-catenin pathway.

906 Effects of miR-105-5p Regulating the Expression of CDC5L on Proliferation and Migration of Glioma Cells

Objective: To analyze the effects of microRNA-105-5p (miR-105-5p) on the proliferation and migration of glioma cells by regulating cell division cycle 5-like protein (CDC5L). Methods: Samples of 35 surgically treated human glioma tissues and normal brain tissues undergoing decompression due to traumatic brain injury were collected from our hospital. Logarithmic growth phase U251 cells were divided into control (control) group, transfected miR-105-5p negative control (miR-NC) group, transfected miR-105-5p mimics (miR-105-5p mimics) group, transfected silent CDC5L negative control (si-NC) group, transfected silent CDC5L (si-CDC5L) group , co-transfected miR-105-5p mimics and pcDNA (pcDNA) group, and co-transfected miR-105-5p mimics and pcDNA-CDC5L (miR-105-5p mimics+pcDNA-CDC5L) group. qRT-PCR was performed to measure the expression of miR-105-5p and CDC5L mRNA in glioma tissue, normal brain tissue, and U251 cells in each group; CCK-8 method was performed to measure cell proliferation; Transwell was performed to measure cell migration; Western blot method was performed to measure the expression of CDC5L, CyclinDl, matrix metalloproteinase 2 (MMP-2), MMP-9 and cyclin-dependent kinase inhibitor 1A (p21); dual luciferase reporter assay validated the targeting of miR-105-5p to CDC5L Results: Compared with normal brain tissue, the level of miR-105-5p in glioma tissue was significantly decreased,and CDC5L mRNA was significantly increased; miR-105-5p was negatively correlated with CDC5L (r= -0.512,P=0.002). Compared with the control group and si-NC group, the expression of p21 protein was increased in the si-CDC5L group (P<0.05), and the OD values at 24h, 48h and 72h, the number of migrating cells, the expression of CDC5L mRNA and the expression of CDC5L, CyclinDl, MMP-2 and MMP-9 proteins were decreased (P<0.05). Compared with the control group and miR-NC group, the expression of miR-105-5p and p21 protein was increased in the miR-105-5p mimics group (P<0.05), and the OD values at 24h, 48h and 72h, the number of migrating cells, the expression of CDC5L mRNA and the expression of CDC5L, CyclinDl, MMP-2 and MMP-9 proteins were decreased (P<0.05). Overexpression of CDC5L was able to reverse the effects of miR-105-5p on the proliferation and migration of U251 cells and CyclinDl, MMP-2, MMP-9, and p21 proteins (P<0.05); the dual luciferase experiment verified that CDC5L was the downstream target of miR-105-5p. Conclusion: miR-105-5p inhibits the proliferation and migration of glioma cells by down-regulating CDC5L.

912 Effects of XELOX Neoadjuvant Chemotherapy on R0 Resection Rate and Malignant Tumor Marker Levels in Patients with Locally Progressive Low Rectal Cancer Undergoing Radical Resection

Objective: To explore the effects of XELOX neoadjuvant chemotherapy on the R0 resection rate and levels of malignant tumor markers in patients with locally progressive low rectal cancer undergoing radical resection. Methods: Clinical data of 52 patients with locally progressive low rectal cancer who received radical resection in the hospital from August 2019 to August 2022 were retrospectively analyzed and the patients were included in control group. Clinical data of 53 patients with locally progressive low rectal cancer who underwent XELOX neoadjuvant chemotherapy + radical resection in the hospital during the same period were retrospectively analyzed and the patients were enrolled as observation group. The lesion resection status (R0 resection, R1 resection, R2 resection) in the two groups were analyzed. Anorectal dynamics [rectal resting pressure (RRP), rectal maximum tolerance volume (MTV), rectal compliance (RC)], malignant tumor markers [carcinoembryonic antigen (CEA), carbohydrate antigen 199 (CA199), tumor M2 pyruvate kinase (TuM2-PK)] and immune cells [T lymphocytes (CD3+, CD4+, CD4+/CD8+)] were compared between the two groups of patients before treatment and one week after surgery. Results: The R0 resection rate in observation group was higher than that in control group (P<0.05). One week after surgery, RRP in both groups was enhanced compared with that before treatment, but the RRP in observation group was lower than that in control group, and MTV and RC were reduced compared with those before treatment, but the two indicators in observation group were higher than those in control group (all P<0.05). The levels of CEA, CA199 and Tu M2-PK were declined compared to before treatment, and the levels were lower in observation group (all P<0.05). The levels of CD3+, CD4+ and CD4+/CD8+ in observation group were declined compared with those before treatment (all P<0.05), but there were no significant changes in control group (all P>0.05), and the differences between the two groups were statistically significant (all P<0.05). Conclusion: XELOX neoadjuvant chemotherapy can enhance the R0 resection rate, improve the anorectal dynamics and reduce the levels of malignant tumor markers in patients with locally progressive low rectal cancer undergoing radical resection, but it can reduce the immune function of patients.

916 Apriori Algorithm Based Acupuncture for the Treatment of Atopic Dermatitis: A Study on the Regularity of Acupoint Selection

Objective: To analyze the acupoints selected for acupuncture prescriptions and their dispensing patterns in the existing literature reports on acupuncture treatment of atopic dermatitis with Apriori algorithm based on Python, so as to expand the therapeutic ideas for clinical acupuncture treatment of atopic dermatitis. Methods: The contents of the relevant literature in CNKI, Wanfang data, and Wipu repositories (~ February 2021) that met the inclusion criteria of this study were extracted and entered, and the data were read using Python, and the Apriori algorithm was used to analyze the selected acupoints and the rules of combination for the treatment of atopic dermatitis. Results: In this study, 24 acupuncture prescriptions were selected for statistical analysis, involving a total of 38 acupoints, and the total frequency of acupoints applied was 170. Among them, the high frequency acupoints were Quchi, Xuehai, and Zusanli; the high frequency meridians involved were Foot Yang Sun Bladder Meridian, Foot Tai Yin Spleen Meridian, and Hand Yang Ming Large Intestine Meridian; the acupoints were mostly distributed in the extremities and back; Quchi-Tianshu-Zhongbei was the key acupoint for the treatment of atopic dermatitis; the visualization analysis of the combination of acupoints showed that the common combinations of acupoints were: Diaphragmatic Yu, Dazhui, Lung Yu, Heart Yu, Wei Zhong, Gall Bladder Yu, and San Jiao Yu. The cluster analysis of high-frequency acupoints showed that the acupoints could be divided into four effective clusters. Conclusion: The selection pattern of acupuncture points selected from the results of this study can provide reference value for clinical research on the acupuncture treatment of atopic dermatitis.

922 Expression of sFRP4 and DKK1 in Serum and Clinical Significance in Patients with Primary Osteoporosis

Objective: To analyze the expression and clinical significance of sFRP4 and DKK1 in serum of patients with primary osteoporosis. Methods: Eighty-two patients with primary osteoporosis admitted to our hospital from January 2019 to January 2021 were selected as the study group, in addition, 75 cases from the same period of healthy physical examination were selected as the control group. Serum sFRP4 and DKK1 levels were detected by enzyme-linked immunosorbent assay (ELISA), serum β collagen degradation products (β-CTX), type I collagen amino-terminal elongation peptide (P1NP), osteopontin (OPN) and N-terminal osteocalcin (N-MID) levels were detected by electrochemiluminescence. The BMD value of femur and L1~4BMD value of lumbar spine were measured with bone densitometer. Pearson method was used to analyze the correlation between serum sFRP4, DKK1 levels, bone metabolism indexes, and bone mineral density; the influencing factors of primary osteoporosis were analyzed by logistic regression; the diagnostic value of serum sFRP4 and DKK1 levels in primary osteoporosis was analyzed by ROC curve. Results: The expression levels of serum sFRP4 (62.98±10.79) μg/L and DKK1 (14.28±3.97) μg/mL in the study group were significantly higher than those in the control group (46.87±8.88) μg/L and (11.43±3.04) μg/mL (P<0.05), and the β-CTX (4.52±1.02) ng/mL, P1NP (47.28±4.26) ng/mL and OPN (26.64±5.11) ng/mL in the study group and N-MID (89.87±8.02) ng/mL were significantly higher than those in the control group (3.55±0.92) ng/mL, (30.84±4.53) ng/mL, (19.01±4.12) ng/mL and (81.95±9.50) ng/mL (P<0.05), and the femur BMD (0.58±0.16) g/cm3 and lumbar L1~4BMD (0.64±0.18 g) in the study group. cm3) was significantly lower than that of the control group (0.82±0.19) g/cm3 and (0.83±0.23) g/cm3 (P<0.05). Pearson correlation analysis showed that the expression levels of sFRP 4 and DKK1 were positively correlated (r=0.479, P<0.05), and the expression levels of sFRP 4 and DKK1 were negatively correlated with femoral BMD and lumbar L1~4BMD (P<0.05), and positively correlated with β-CTX, P1NP, OPN and N-MID (P<0.05). Logistic regression analysis showed that the overexpression of sFRP4 and DKK1 was a risk factor for primary osteoporosis (P<0.05). The AUC of serum sFRP4 in diagnosis of primary osteoporosis was 0.874, and that of DKK1 in diagnosis of primary osteoporosis was 0.809, the AUC of the combination of the two methods in the diagnosis of primary osteoporosis was 0.921, which was superior to the diagnosis of serum sFRP4 and DKK1 separately (Z combination vs sFRP4=2.851, Z combination vs DKK1=4.365, P<0.05). Conclusion: Serum sFRP4 and DKK1 are significantly elevated in patients with primary osteoporosis, which is closely related to bone metabolism and BMD, and the combination of the two can better diagnose primary osteoporosis.

928 Expression of hTERC RNA Fraction in Cervical Squamous Intraepithelial Neoplasia by RNAscope Technique and Its Application Value

Objective: To investigate the diagnostic significance of hTERC RNA fractions expression detected by RNA scope technique in cervical squamous intraepithelial neoplasia. Methods: A total of 80 cervical biopsy specimens diagnosed with HR-HPV infection in the Department of Pathology, Affiliated Hospital of North China University of Science and Technology from September 2021 to February 2022 were screened in 3 groups: inflammation group (14 cases), low-grade group (34 cases), and high-grade group (32 cases). The amplification status of hTERC gene and the expression of its RNA components in paraffin-embedded tissues were detected by fluorescence in situ hybridization and RNAscope techniques, respectively. Results: FISH detection of hTERC gene amplification was different among the three groups (P<0.05). Detection rate by RNAscope was statistically different among the three groups (P<0.05). In the histological diagnostic subgroups, the degree of positive expression of hTERC RNA fraction was (-/++) in all 14 cases in the inflammation group, the hTERC RNA fraction (++) was predominant in the low-grade group (19/34), and the hTERC RNA fraction (++++/++++) was predominant in the high-grade group (27/32), and the expression of the above staining patterns in the histological graded diagnosis had statistical differences (P<0.05). Conclusion: The RNAscope technique to detect the expression of hTERC RNA fraction in cervical intraepithelial neoplasia tissue helps in the histological diagnosis and progression prediction of cervical lesions.

933 Correlation Study of the Changes in Expression Levels between TNF-α IL-6 and VEGF in Invasive Pituitary Adenoma

Objective: To investigate the correlation between the changes in the expression levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) and vascular endothelial growth factor (VEGF) in invasive pituitary adenoma tissues and their significance. Methods: Fifty-six patients with pituitary adenomas surgically resected in the neurosurgery department of Chengde Central Hospital were selected and divided into patients with non-functional invasive pituitary adenomas (observation group, 38 patients) and non-functional non-pituitary adenomas (control group, 18 patients) according to the invasiveness of the tumor. The expression levels of TNF-α,, IL-6 and VEGF in the observation and control groups were measured by real-time quantitative PCR and immunohistochemistry to understand the role of the three in the aggressive growth of pituitary adenomas. Results: The expression levels of TNF-α, IL-6 and VEGF mRNA were higher in the observation group than in the control group (all P<0.05), and the results of immunohistochemical staining also showed that the expression of TNF-α, IL-6 and VEGF were higher in the observation group than in the control group, and the differences were of high research value and statistical significance (all P<0.05) . Conclusion: High expression of TNF-α, IL-6 and VEGF in pituitary adenoma tissues may predispose pituitary adenomas to aggressive growth, and low or no expression may lead to even less aggressive or non-invasive growth. TNF-α, IL-6 and VEGF may contribute to aggressive pituitary adenoma growth.

937 Changes in Expression Levels of MAGE-As Protein and mRNA in Lung Cancer Patients and Their Clinical Significance

Objective: To investigate the expression and clinical significance of melanoma associated antigen (MAGE)-As protein and mRNA in non-small cell lung cancer. Methods: A total of 88 patients with non-small cell lung cancer treated in our hospital from January 2015 to January 2020 were selected, and the adjacent tissues were selected as the control, the expression of MAGE-As protein in lung cancer and adjacent tissues was detected by Western blot, and the expression of MAGE-As mRNA in lung cancer and adjacent tissues was detected by RT-PCR. Results: The relative expression levels of MAGE-As protein and mRNA in NSCLC tissues were (0.343 ± 0.101) and (0.728 ± 0.112), which were significantly higher than those in adjacent tissues (P<0.05); the relative expression of MAGE-As protein in stage Ⅲ ~Ⅳ lung cancer was (0.385 ± 0.102), which was significantly higher than that in stage Ⅰ~ Ⅱ lung cancer (P<0.05); there were no significant difference in the relative expression of MAGE-As protein in different gender, age, tumor diameter, pathological type and differentiation degree (P>0.05); the relative expression of MAGE-As mRNA in clinical stage Ⅲ~ Ⅳ and moderately and moderately differentiated lung cancer tissues were (0.762 ± 0.112) and (0.742 ± 0.107), which were significantly higher than those in stage Ⅰ~Ⅱ and well differentiated lung cancer tissues (P<0.05); there were no significant difference in the relative expression of MAGE-As mRNA in different gender, age, tumor diameter and pathological type (P>0.05). The median overall survival of patients with high and low MAGE-As mRNA expression in lung cancer tissues was 33 and 44 months, with a difference in survival between patients with high and low MAGE-As mRNA expression (P<0.05). The difference in median overall survival between patients with high and low MAGE-As protein expression was not statistically significant (P>0.05). Conclusion: The expression of MAGE as protein and mRNA are up-regulated in non-small cell lung cancer, is related to the severity of the disease, and the prognosis of patients with high expression of MAGE As mRNA is poor.

941 The Clinical Significance of Microinflammatory Status and Serum PTH (1-84) and BALP Levels in Maintenance Hemodialysis Renal Bone Disease

Objective: To analyze the clinical significance of micro-inflammatory state and serum levels of intact parathyroid hormone [PTH(1-84)] and bone-specific alkaline phosphatase (BALP) on patients with renal osteodystrophy undergoing maintenance hemodialysis (MHD). Methods: A total of 104 patients with MHD admitted to department of nephrology of the hospital from January 2019 to July 2022 were selected as the research subjects. According to the presence or absence of renal osteodystrophy, the patients were divided into renal osteodystrophy group (n=36) and non-renal osteodystrophy group (n=68). Another 50 healthy volunteers were enrolled as control group. The levels of micro-inflammatory state [C-reactive protein (CRP), interleukin-6 (IL-6), β2-microglobulin (β2-MG)] and bone turnover markers [PTH(1-84), BALP, osteoprotegerin (OPG)] were compared among the three groups at enrollment. Pearson correlation analysis was used to analyze the relationship between micro-inflammatory state and bone turnover markers in patients with renal osteodystrophy. Receiver operating characteristic (ROC) curve was applied to analyze the predictive value of the above micro-inflammatory indicators and bone turnover markers on MHD with renal osteodystrophy. Results: The levels of serum CRP, IL-6 and β2-MG in the three groups at enrollment were as follows: renal osteodystrophy group>non-renal osteodystrophy group>control group, with statistically significant differences (P<0.05). The levels of serum PTH(1-84), BALP and OPG in the three groups were shown as renal osteodystrophy group>non-renal osteodystrophy group>control group (P<0.05). Pearson correlation analysis showed that the level of serum IL-6 at enrollment in renal osteodystrophy group was positively correlated with PTH(1-84) (P<0.05), and β2-MG was positively correlated with BALP (P<0.05). ROC curve revealed that serum CRP, IL-6, β2-MG, PTH (1-84), BALP and OPG had good predictive efficiency in the diagnosis of MHD with renal osteodystrophy (with AUC, sensitivity and specificity of 0.769, 77.80% and 77.90% when CRP cutoff value was 18.41 mg/L, 0.791, 83.30% and 69.10% when IL-6 cutoff value was 243.53 pg/mL, 0.731, 73.90% and 70.28% when β2-MG cutoff value was 5.605 mg/L, 0.839, 77.80% and 85.30% when PTH (1-84) cutoff value was 173.235 ng/L, 0.810, 94.40% and 64.70% when BALP cutoff value was 23.575 U/L and 0.826, 83.30% and 76.50% when OPG cutoff value was 365.93 pg/mL respectively). Conclusion: The levels of bone turnover markers [PTH(1-84), BALP, OPG] and micro-inflammatory indicators (CRP, IL-6, β2-MG) are abnormally increased in patients with MHD, and they have good predictive efficiency in the diagnosis of MHD with renal osteodystrophy.

947 Expression Levels and Significance of CD4+CD25+T Regulatory Cell and Related Cytokines in Patients with COPD Complicated with Bacterial Infection

Objective: To study the expression levels and clinical significance of CD4+CD25+T regulatory cell and related cytokines of interleukin-4 (IL-4), IL-12 and interferon-γ (IFN-γ) in patients with chronic obstructive pulmonary disease (COPD) complicated with bacterial infection. Methods: A total of 108 patients with COPD admitted between August 2020 and August 2022 were included as the research subjects, and they were divided into infected group (n=53) and control group (n=55) according to whether the patients were complicated with bacterial infection. The expression levels of CD4+CD25+ regulatory T cell and IL-4, IL-12 and IFN-γ were detected in the peripheral blood samples, and the differences between the two groups were compared, and the relationship between levels of related indicators and COPD with infection was analyzed. The 28-d prognosis of patients was statistically analyzed, and the predictive value of the above indicators on prognosis was analyzed. Results: The levels of serum IL-4 and IL-12 of COPD patients in infected group were higher than those in control group while the IFN-γ/IL-4 was lower than that in control group (P<0.05). Follow-up results showed that 15 patients died within 28 d, with a mortality rate of 13.89%. The level of serum IL-4 of COPD patients in death group was higher than that in control group while the levels of CD4+CD25+, IL-12, IFN-γ and IFN-γ/IL-4 were lower than those in control group (P<0.05). Logistics regression analysis showed that serum levels of IL-4 and IL-12 were associated with COPD with pulmonary infection (P<0.05), and CD4+CD25+, IL-12 and IFN-γ/IL-4 were associated with the prognosis of COPD patients (P<0.05). ROC curve showed that the AUC values of CD4+CD25+, IL-12, IFN-γ/IL-4 and the combination of the three were 0.747, 0.827, 0.709, and 0.924 respectively in judging prognosis of COPD patients. Conclusion: CD4+CD25+ Treg cell and related cytokines are related to COPD infection and disease progression. Detection of CD4+CD25+ Treg distribution as well as IL-4, IL-12 and IFN-γ can provide reference information for diagnosis of pulmonary infection and evaluation of prognosis in patients with COPD.

952 Application Value of Low-Dose CT Three-Dimensional Reconstruction in Rhinoplasty after Nasal Bone Fracture

Objective: To explore the application value of low-dose CT three-dimensional reconstruction in rhinoplasty after nasal bone fracture. Methods: 80 patients with nasal bone fracture were randomly divided into two groups according to the random number table, 40 cases in each group. The experimental group 1 was scanned with low dose (50mAs) CT, and the control group 1 was scanned with standard dose (250mAs) CT. The quality of the two groups of volume rendering (VR) images was scored, and the CT dose length product (DLP) values of the two groups were recorded, and the relevant data were statistically analyzed. There were 19 cases of rhinoplasty in the above 80 patients. They were divided into two groups according to the random number table method. All the 19 patients underwent osteotomy + nasal septum correction + prosthetic ear cartilage rhinoplasty. Four weeks after operation, 10 cases in the experimental group 2 were scanned with low dose CT, and 9 cases in the control group 2 were scanned with standard dose CT. The bony nasal root point and the corresponding skin surface point on the median sagittal plane were directly marked in the software, and the distance between the two was measured. The change of distance before and after rhinoplasty was recorded. The angles of nasal frontal angle, nasal tip angle and nasal lip angle of the two groups were marked, and the angle changes were recorded. The CT dose length product (DLP) value and effective dose (ED) value of the two groups were recorded and analyzed statistically. Results: There was no significant difference in the image quality score of CT scan between the two groups in the diagnosis of simple nasal bone fracture and nasal bone fracture with maxillary frontal process fracture (P>0.05), but there was significant difference in the diagnosis of nasal bone fracture with skull base fracture (P<0.05). The radiation dose of the experimental group 1 was significantly lower than that of the control group 1 (P<0.05). There was no significant difference between the experimental group 2 and the control group 2 in the changes of the line distance of the landmark points and the angle of the nasal frontal angle, nasal tip angle and nasal lip angle before and after operation (P>0.05). The DLP and ED values of the two groups before and after operation were compared, and the difference was statistically significant (P<0.05). Conclusion: Low-dose CT scan can meet the needs of accurate diagnosis of nasal bone fracture and nasal bone fracture with maxillary frontal process fracture and preoperative and postoperative evaluation of patients with rhinoplasty after nasal bone fracture. At the same time, it can significantly reduce the radiation dose and improve the safety of scanning, which is worthy of popularization and application.

957 Interactive Effects of HPV Infection and Key Factors of the ERK Signaling Pathway in Cervical Cancer and Clinical Implications

Objective: To investigate the interactive effects and significance of HPV infection and key factors of ERK signaling pathway in cervical cancer. Methods: Ninety patients with cervical cancer admitted to our hospital from October 2020 to October 2022 were selected as the study group, and another 90 patients with cervical intraepithelial neoplasia were included as the control group. The HPV infection status, ERK signaling pathway key factors [nuclear inhomogeneous ribonucleoprotein E1 (HnRNP E1), phosphorylated extracellular signal-regulated kinase 1/2 (p-ERK1/2), and phosphorylated non-receptor tyrosine kinase (p-Src)] expression were counted in both groups, and the expression of ERK signaling pathway key factors in patients with different clinicopathological characteristics in the study group was compared. The correlation between the two was analyzed with Spearman analysis and Logistic regression equation analysis was conducted to determine the interaction effect between HPV infection and key factors of the ERK signaling pathway in cervical cancer. Results: The HPV infection rate and the positive rates of HnRNP E1, p-ERK1/2 and p-Src protein in the study group were higher than those in the control group for CIN grade Ⅰ~Ⅱ and Ⅲ (P<0.05); the FIGO stage, HPV infection and differentiation degree of cervical cancer patients were positively correlated with HnRNP E1, p-ERK1/2 and p-Src protein (P<0.05); HnRNP E1, p-ERK1/2, p-Src protein and the interaction of the three were high-risk factors for HPV infection in cervical cancer patients (P<0.05). Conclusion: Both HPV infection and positive expression of key factors of ERK signaling pathway can increase the risk of cervical cancer, and there is a positive interaction between the two, which can provide favorable reference for the identification and treatment of cervical cancer.

963 Detection and Correlation Analysis of Gal-3 IL-33 Expression, MP Load and Neutrophil Ratio in Alveolar Lavage Fluid of Children with MPP

Objective: To investigate the levels of galectin-3 (Gal-3), interleukin-33 (IL-33), Mycoplasma pneumoniae (MP) -DNA, neutrophil proportion and clinical significance in alveolar lavage fluid of children with mycoplasma pneumoniae pneumonia (MPP). Methods: A total of 102 children with MPP who were treated in our hospital from January 2020 to March 2022 were selected as the observation group, including 59 children with mild disease and 43 children with severe disease, while 60 children with bronchial foreign bodies were selected as the control group, and the differences in Gal-3, IL-33, MP-DNA load, neutrophils, etc. in alveolar lavage fluid in each group were compared, the changes of Gal-3, IL-33, MP-DNA load and neutrophil in alveolar lavage fluid in the observation group before and after treatment were analyzed. Results: Gal-3, IL-33, MP-DNA load, neutrophils and macrophages in alveolar lavage fluid of the observation group were (18.82 ± 3.10) pg/ml, (20.01 ± 3.43) pg/ml, (5.94 ± 1.01) log copy/ml, (18.21 ± 3.11)% and (88.14 ± 9.92)%, respectively, which were significantly higher than those in the control group (P<0.05). Gal-3, IL-33, MP-DNA load and neutrophil in alveolar lavage fluid of severe children in the observation group were (20.32 ± 3.02) pg/ml, (22.03 ± 3.05) pg/ml, (7.07 ± 1.70) log copy/ml and (20.31 ± 2.88)% respectively, which were significantly higher than those of mild children (P<0.05). Gal-3, IL-33, MP-DNA load, neutrophils and macrophages in alveolar lavage fluid of mild and severe children in the observation group decreased after treatment compared with those before treatment (P<0.05). Gal-3, IL-33, neutrophils in alveolar lavage fluid were positively correlated with MP-DNA load (P<0.05), while no significant correlation was found in the rest. Conclusion: Gal-3, IL-33, MP-DNA load and the proportion of neutrophils in the alveolar lavage fluid of children with MPP are significantly increased, which is related to the severity of the disease, and Gal-3, IL-33, neutrophils in the alveolar lavage fluid are positively related to MP-DNA load.

967 Expression and Clinical Significance of the Inflammation-Related Factors TLR4 and NF-κB in Renal Tissue of Idiopathic Membranous Nephropathy

Objective: To investigate the expression levels of inflammation-related factors toll-like receptor 4 (TLR4) and nuclear transcription factor-kappa B (NF-κB) in renal tissues of patients with idiopathic membranous nephropathy (IMN) and to explore the clinical significance of the inflammatory factors TLR4 and NF-κB in the pathogenesis of IMN. Methods: In this study, 45 patients who were initially diagnosed with IMN by renal biopsy in the Department of Nephrology of the First Affiliated Hospital of Bengbu Medical College were included in the group of idiopathic membranous nephropathy (IMN group), and 20 patients who had nephrectomy for renal contusion, renal tumor or ureteral tumor and left healthy kidney tissues in the same period were selected as the normal control group (NC group). The relevant clinical indexes were collected and analyzed to compare the differences between the two groups. The expressions of inflammatory factors TLR4 and NF-κB in the kidney tissues of the two groups were detected by immunohistochemistry, and the mean optical density values were analyzed to compare the differences in inflammatory factor expressions between the two groups. The IMN group was divided into 29 cases in the fibrosis group and 16 cases in the no-fibrosis group, and the differences in inflammatory factor expression between the two groups were analyzed and compared to understand the role of inflammation in renal interstitial fibrosis. Pearson correlation analysis was applied between the two groups of inflammatory factors and their correlation with clinical indices. Results: The renal tissue expression levels of TLR4 and NF-κB were significantly higher in the IMN group than in the NC group (P<0.01). The expression of TLR4 and NF-κB was significantly stronger in the IMN fibrosis group than in the group without fibrosis (P<0.05). Correlation analysis revealed a close correlation between TLR4 and NF-κB (P<0.01). The TLR4 was negatively associated with serum albumin and high density lipoprotein (P<0.01) and positively associated with urinary protein quantification, serum cholesterol, and low density lipoprotein (P<0.05, P<0.01). The NF-κB was negatively associated with serum albumin and high density lipoprotein (P<0.05) and positively associated with 24-hour urinary protein quantification (P<0.05). Conclusion: The expression level of inflammatory factors TLR4 and NF-κB are upregulated in IMN kidney tissue, and the interaction causes cell damage in renal tissue, leading to renal interstitial fibrosis, which is involved in the occurrence and development of IMN.

972 The Value of Changes in Serum CRP, AMY and MPV in Diagnosis and Disease Assessment of Patients with Biliary Pancreatitis

Objective: To investigate the changes and clinical significance of serum C-reactive protein (CRP), amylase (AMY), and mean platelet volume (MPV) in patients with biliary pancreatitis. Methods: Ninety eight patients with acute biliary pancreatitis (biliary source group) and 100 patients with acute non biliary pancreatitis (control group) diagnosed at the same time in Huangshan People's Hospital from June 2019 to June 2022 were selected. The levels of CRP, AMY, MPV, liver function indicators, blood routine biochemical indicators, acute physiological function and chronic health (APACHE II) scores, and CT severity index scores (CTSI) of the two groups were compared. Simple linear correlation was used to analyze the correlation between CRP, AMY, MPV levels, APACHE Ⅱ score and CTSI. Results: There was no statistically significant difference between the two groups in serum CRP and AMY measured values (P>0.05); the measured value of MPV in peripheral blood of the biliary group was higher than that of the control group, and the difference between the above indexes was statistically significant (P<0.05); the serum ALT, AST, TBIL and DBIL of patients in the biliary group were higher than those in the control group, and the difference between the above indexes was statistically significant (P<0.05); there was no statistically significant difference in urea nitrogen, WBC, blood sugar, blood calcium, lactate dehydrogenase, hemoglobin, hematocrit, platelet count, APACHE Ⅱ score and CTSI score between the biliary group and the control group (P>0.05); the correlation analysis of CRP, AMY, MPV, APACHE Ⅱ score and CTSI score of patients in biliary group showed that there was significant positive correlation (P<0.05); the level of MPV in peripheral blood has high value in differential diagnosis of patients with acute biliary pancreatitis or acute non biliary pancreatitis, with sensitivity of 70.63%, specificity of 64.75%, AUC value of 0.746 (95% CI: 0.679-0.813), and cutoff value of ≥ 9.20 fL. Conclusion: The levels of CRP and AMY in patients with acute biliary pancreatitis or acute non-biliary pancreatitis are not significantly different, MPV is higher than that in patients with acute non-biliary pancreatitis, CRP, AMY, MPV and acute biliary pancreatitis patients Depending on the severity, MPV has a certain value in the differential diagnosis of acute biliary pancreatitis or acute non-biliary pancreatitis.

977 The Relationship between Lipid Metabolism IGF-1 and HbA1c Levels and the Nature of T2DM Combined with Thyroid Nodules

Objective: To investigate the relationship between lipid metabolism, serum levels of insulin-like growth factor-1 (IGF-1) and glycosylated hemoglobin (HbA1c) and the nature of thyroid nodules in patients with type 2 diabetes mellitus (T2DM). Methods: 137 patients with T2DM combined with thyroid nodules treated in our hospital from May 2020 to May 2022 were selected as the observation group, while 140 patients with T2DM alone were selected as the control group, the differences in lipid metabolism, IGF-1, HbA1c, etc. between the two groups were compared, and the differences in lipid metabolism, IGF-1, HbA1c, etc. between patients with benign and malignant nodules in the observation group were analyzed. Results: The course of T2DM, fasting blood glucose (FBG), fasting insulin (FINS), glycosylated hemoglobin (HbA1c), insulin resistance index (HOMA-IR), triglyceride (TG), TG/HDL-C and IGF-1 in the observation group were (10.50 ± 2.43) years, (11.40 ± 2.11) mmol/L, (24.49 ± 3.32) mIU/L, (9.60 ± 1.83)%, (12.45 ± 2.11), (4.14 ± 1.02) mmol/L, (3.05 ± 0.92) and (220.10 ± 70.20) ng/ml, respectively, which was significantly higher than that in the control group (P<0.05). There was no significant difference between the observation group and the control group in blood glucose at 2h after meal (2hPG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), TC/HDL-C and LDL-C/HDL-C (P>0.05). HbA1c, TG, TG/HDL-C and IGF-1 in patients with malignant nodules of observation group were (10.21 ± 1.60)%, (4.76 ± 1.56) mmol/L, (3.50 ± 0.90) and (243.32 ± 51.26) ng/ml, respectively, higher than those in patients with benign nodules (P<0.05). TG, TG/HDL-C were positively correlated with HbA1c (P<0.05). IGF-1 had no significant correlation with TG, TG/HDL-C and HbA1c (P>0.05). The area under ROC curve predicted by TG, TG/HDL-C, IGF-1, HbA1c to predict malignant nodules were 0.621, 0.705, 0.727, 0.687 respectively (P<0.05). Conclusion: In T2DM patients with thyroid nodules, lipid metabolism is significantly abnormal, IGF-1 and HbAIc are significantly increased, and TG, TG/HDL-C, IGF-1 and HbA1c has certain application value in predicting benign and malignant thyroid nodules.

983 Effectiveness and Safety of Delta Large Channel Endoscopic Spinal Decompression in the Treatment of Lumbar Spinal Stenosis

Objective: To investigate the clinical effectiveness of Delta channel endoscopic spinal decompression in the treatment of lumbar spinal stenosis. Methods: A total of 95 patients with lumbar spinal stenosis admitted to our hospital from January 2021 to January 2022 were selected in this study. Among them, 48 patients were treated with Delta channel spinal endoscopic surgical decompression (observation group), and the other 47 patients were treated with unilateral double-channel spinal endoscopic treatment (control group). The differences of surgery-related clinical indicators, pain degree before and after surgery, Oswestry disability index (ODI), CT imaging parameters of spinal stenosis, and clinical efficacy were compared between the two groups. Results: The operation time of the patients in the observation group was shorter than that in the control group, and the operation bleeding volume was less than that in the control group, the difference was statistically significant (P<0.05). There was no statistically significant difference between the two groups in the postoperative time to the ground and hospital stay (P>0.05). Before operation, there was no significant difference in VAS score of waist pain and VAS score of leg pain between the observation group and the control group (P>0.05). After operation, the pain degree of waist and leg in the two groups was significantly reduced, and the VAS score was significantly lower than that before operation (P<0.05). Before operation, there was no statistically significant difference in the measured values of sagittal diameter of spinal canal (APDC), spinal canal area (CAC), area of vertebral canal (ICA), dural sac area (CAD) and epidural space (LAC) between the observation group and the control group (P>0.05); three months after operation, CT reexamination of patients in both groups showed that the stenosis of the spinal canal of the patients was improved very well, and the measured values of APDC, CAC, ICA, CAD and LAC in the lumbar intervertebral space were significantly higher than those in the group before operation (P<0.05); before operation, the lumbar function of the patients was evaluated by ODI. There was no significant difference in the ODI value between the observation group and the control group (P>0.05); the lumbar function of patients in the two groups was significantly improved after operation, and the measured value of ODI was significantly lower than that before operation (P<0.05). After operation, the postoperative complications were 8.33% in the observation group and 4.26% in the control group, with no significant difference (P>0.05). Conclusion: Both Delta channel endoscopic spinal decompression and unilateral double-channel endoscopic spinal decompression can achieve good clinical results in the treatment of lumbar spinal stenosis, but the former has the advantages of simple operation, less operation time, and less surgical blood loss.

990 A Comparative Study on the Application Value of PRF and Bio-Oss Bone Powder in Extra-Maxillary Sinus Lift Surgery

Objective: To investigate the clinical value of platelet-rich fibrin (PRF) and Bio-Oss bone meal in the clinical value of extra-maxillary sinus lift surgery. Methods: A total of 120 patients who were treated in our hospital from July 2020 to January 2021 and received extra-maxillary sinus lift surgery were selected and simply randomly divided into 58 patients in PRF group and 62 patients in Bio-Oss group by random number table method, with platelet-rich fibrin (PRF) implantation intervention procedure in PRF group and Bio-Oss bone powder implantation intervention procedure in Bio-Oss group. Pain, mucosa at the surgical site, postoperative bone defect regeneration and surrounding soft tissue health were observed in the two groups, bone density changes, platelet-derived growth factor (PDGF), vascular endothelial growth factor (VEGF), transforming growth factor-β(TGF-β) index levels before and after treatment in both groups, Pearman analysis of bone density and PDGF, VEGF, TGF-β correlation was conducted. Results: Pain performance was significantly lower in the PRF group compared with the Bio-Oss group, the proportion of patients with normal mucosal color was significantly higher in the PRF group compared with the Bio-Oss group, and the degree of mucosal swelling was significantly lower compared with the Bio-Oss group; the levels of bleeding index, probing depth, and attachment loss index were significantly lower in the PRF group compared with the Bio-Oss group, and the bone graft height and osteogenic thickness index were significantly higher in the PRF group compared with the Bio-Oss group. Oss group (all P<0.05); BMD in both groups decreased significantly compared to the preoperative level 1 month after surgery, but the PRF group had a higher BMD level than the Bio-Oss group (P<0.05); BMD values in the Bio-Oss group increased significantly at 3 months after surgery compared to 1 month after surgery, but were significantly lower than the preoperative level and significantly lower than the PRF group (all P<0.05); . The levels of each index in the PRF group were significantly higher than those in the Bio-Oss group (all P<0.05); Pearson correlation analysis of BMD and the levels of growth factors PDGF, VEGF and TGF-β indexes showed positive correlation (all P<0.05). Conclusion: PRF implantation in patients undergoing extra-maxillary sinus lift surgery can promote the early formation of new bone in the extra-maxillary sinus and achieve good clinical results.

995 Multifactorial Analysis of Postpartum Hemorrhage in Patients with Gestational Diabetes Mellitus

Objective: To investigate the influencing factors that may lead to postpartum hemorrhage in patients with gestational diabetes mellitus, in order to ensure maternal and infant safety and to provide interventional guidance for promoting good pregnancy outcomes. Methods: The baseline data of 152 patients with gestational diabetes admitted to our hospital between January 2021 and December 2022 were retrospectively analysed. The patients were divided into a haemorrhage group (42 patients with bleeding ≥500 mL 24 h after delivery in natural delivery and ≥1000 mL within 24 h in caesarean section) and a non-haemorrhage group (110 patients) according to the presence or absence of postpartum haemorrhage. The baseline data of the two groups were compared to explore the factors that may have contributed to the patients' postpartum haemorrhage. Results: The 152 patients with gestational diabetes mellitus enrolled had 42 cases of postpartum haemorrhage (27.63% (42/152), with a mean postpartum haemorrhage volume of (920.38±40.56) mL, and they were included in the haemorrhage group, and 110 cases of postpartum haemorrhage did not occur (72.37% (110/152), with a mean postpartum haemorrhage volume of (280.25±20.45) mL, and they were included in the non-hemorrhage group; compared with the non-hemorrhage group, patients in the hemorrhage group had a higher proportion of pregnancy (≥3), combined gynaecological inflammation (yes), excess amniotic fluid (yes), combined pre-eclampsia (yes), low expression of serum FIB and AT-III, and high expression of HbA1c, all with statistically significant differences (P<0.05); the differences in age, pre-pregnancy body mass index, week of delivery, number of deliveries, mean systolic blood pressure, mean diastolic blood pressure, history of uterine surgery, history of induced abortion and history of spontaneous abortion between the two groups were not statistically significant (P>0.05); the results of logistic regression analysis showed that the number of pregnancies (≥3), combined gynaecological inflammation (yes), excess amniotic fluid (yes), combined pre-eclampsia (yes), serum FIB, AT-III and HbA1c expression were all influential factors in the development of postpartum haemorrhage in patients with gestational diabetes (OR > 1, P<0.05). Conclusion: Postpartum haemorrhage in patients with gestational diabetes is associated with the number of pregnancies, combined gynaecological inflammation, excess amniotic fluid, combined pre-eclampsia, serum FIB, AT-III and HbA1c expression. For the high-risk groups with ≥3 pregnancies, combined gynaecological inflammation, excess amniotic fluid, combined pre-eclampsia, low serum FIB, AT-III expression and high HbA1c expression, clinical interventions should be tailored to the specific conditions of the patients, with the aim of reducing the risk of postpartum haemorrhage in patients with gestational diabetes and ensuring maternal and infant safety.

1000 Efficacy and Safety of Ultrasound-Guided Subcutaneous Methylene Blue Injection Combined with Acupotomy in the Treatment of Postherpetic Neuralgia

Objective: To explore the clinical efficacy and safety of ultrasound-guided subcutaneous methylene blue injection combined with acupotomy in the treatment of postherpetic neuralgia (PHN). Methods: The clinical data of patients with PHN who were treated with ultrasound-guided subcutaneous methylene blue injection in the hospital from March 2020 to May 2022 were retrospectively analyzed, and 40 cases were randomly selected as control group. The clinical data of patients with PHN who received ultrasound-guided subcutaneous methylene blue injection combined with acupotomy in the hospital during the same period were retrospectively analyzed, and 50 cases were randomly enrolled as study group. The clinical efficacy of the two groups of patients after 10 days of treatment was counted. The pain status [Zoster Brief Pain Inventory (ZBPI), Houston Pain Outcome Instrument (HPOI)], sleep quality [Athens Insomnia Scale (AIS), Pittsburgh Sleep Quality Index (PSQI)] and quality of life [Quality of Life Index Questionnaire (QL-index), Brief Pain Inventory (BPI)] were compared between the two groups of patients before treatment and after 10 days of treatment, and the occurrence of adverse reactions during treatment in the two groups were recorded. Results: At weeks after the end of treatment, the total effective rate of clinical treatment in study group (88.0%) was significantly higher than that in control group (70.0%) (P<0.05). After 10 days of treatment, the scores of ZBPI, PSQI, AIS and BPI in the two groups were significantly lower than those before treatment, and the scores in study group were significantly lower compared with those in control group (P<0.05). The HPOI score and QL-index score in the two groups after 10 days of treatment were significantly higher than those before treatment, and the scores were significantly higher in study group than those in control group (P<0.05). During treatment, there was no statistically significant difference in the total incidence rate of adverse reactions between study group (14.0%) and control group (7.5%) (P>0.05). Conclusion: Ultrasound-guided subcutaneous methylene blue injection combined with acupotomy can effectively enhance the clinical therapeutic effectiveness and reduce the pain degree of patients with PHN, and it has a good improvement effect on sleep and quality of life and has good safety.

1006 Application of Transvaginal and Transabdominal Volor Doppler Ultrasound in the Diagnosis of Early Ectopic Pregnancy

Objective: To compare the application value of transvaginal and transabdominal color Doppler ultrasound in the diagnosis of early ectopic pregnancy. Methods: The clinical data of 123 pregnant women with ectopic pregnancy who were admitted to our hospital from September 2019 to July 2022 were retrospectively analyzed, including the characteristics of transvaginal color Doppler ultrasound and abdominal color Doppler ultrasound. Taking the postoperative pathological results as the gold standard, the efficacy of the two color Doppler ultrasound in diagnosing early ectopic pregnancy was analyzed. Results: Postoperative pathological examination revealed 86 cases of ectopic pregnancy, accounting for 69.92%. Among them, there were 71 cases of mass type, 6 cases of rupture type, 9 cases of gestational sac survival type. The sensitivity, accuracy and negative predictive value of transvaginal color Doppler ultrasound in diagnosing early ectopic pregnancy are higher than those of abdominal color Doppler ultrasound (P<0.05). Kappa value of early ectopic pregnancy diagnosed by transvaginal color Doppler ultrasound is 0.825, which is higher than 0.452 of abdominal color Doppler ultrasound. In early ectopic pregnancy, the visible detection rates of the original heart tube beat, uterus rectal fossa effusion, intrauterine pregnancy sac, adnexal mass were higher than those of abdominal ultrasound (P<0.05). Conclusion: Both transvaginal color Doppler ultrasound and abdominal color Doppler ultrasound can provide some reference for the diagnosis of early ectopic pregnancy, and the diagnostic efficiency and reference value of transvaginal color Doppler ultrasound are higher.

1010 Analysis of the Characteristics and Influencing Factors of Subdural Effusion Complications after Neurosurgical Treatment of Severe Craniocerebral Injury

Objective: To explore the characteristics and risk factors of complications of subdural effusion after neurosurgical treatment of severe craniocerebral injury. Methods: One hundred and thirty-two patients with severe cranial brain injury (sTBI) treated at our hospital from January 2020 to January 2022, all of whom underwent surgery, were selected to analyse the differences in clinical data, serum matrix metalloproteinase-9 (MMP-9) and matrix metalloproteinase-2 (MMP-2) between patients who developed and did not develop subdural effusions after surgery. Results: Subdural effusion occurred in 41 cases after surgery, with an incidence of 31.06%. The distribution of subdural effusion was mainly frontal and temporal. The proportions of patients with postoperative subdural effusion aged ≥60 years, with ventricular haemorrhage, cortical incision, midline shift distance >5 cm and bone flap edge distance >2 cm from midline were 65.85%, 51.22%, 51.22%, 68.29% and 41.46% respectively, which were higher than those without subdural effusion (P<0.05). Serum MMP-9 and MMP-2 were (110.20 ± 23.32) ng/ml and (63.30 ± 8.87) ng/ml, respectively, higher in patients who developed subdural effusion after surgery than in patients who did not develop subdural effusion (P<0.05). logistic regression analysis showed that cortical incision, midline shift distance and bone flap edge distance from the midline were The results of the logistic regression analysis showed that cortical incision, midline shift distance and distance of the bone flap edge from the midline were factors influencing the occurrence of subdural effusion in patients (P<0.05). 1015 Influence of HoLEP and TURP on Urinary Flow Rate Residual Urine Volume and Sexual Function in Patients with Benign Prostatic Hyperplasia

Objective: To investigate the influence of HoLEP and TURP on urinary flow rate, residual urine volume and sexual function in patients with BPH. Methods: This study retrospectively reviewed 256 patients with BPH admitted to the hospital between May 2020 and April 2022. Patients treated with HoLEP and TURP were defined as HoLEP group (n=126) and TURP group (n=130). Comparison was made between groups in terms of urinary flow rate, residual urine volume, and sexual function. Results: After operation, the maximum flow rate and international erectile function index (IIEF-5) of both groups were higher than before operation, while residual urine volume, international prostate symptom score (IPSS), and quality of life score (QOL) were lower than before operation (P<0.05). The difference in maximum flow rate, residual urine volume, IIEF-5 score, IPSS score, and QOL score before and after operation in the HoLEP group was higher than that in the TURP group (P<0.05). The incidence of retrograde ejaculation after operation in the HoLEP group was lower than that in the TURP group (P<0.05). Conclusion: Compared with TURP, HoLEP can improve urine flow rate and reduce residual urine volume, reduce erectile dysfunction and retrograde ejaculation, and improve sexual function in patients with BPH. It is minimally invasive, with high security.

1020 Predictive Value of CMR-FT Imaging for Quantitative Assessment of Prognosis in Dilated Cardiomyopathy

Objective: To investigate the predictive value of cardiac magnetic resonance signature tracking (CMR-FT) imaging for quantitative assessment of myocardial strain before and after treatment for dilated cardiomyopathy (DCM) on prognosis. Methods: Ninety-four patients with DCM admitted to our hospital from April 2018 to April 2021 were selected and given standard treatment according to guideline recommendations and followed up for 6 months, and divided into poor prognosis group (MACE occurred, 23 cases) and good prognosis group (no MACE occurred, 71 cases) according to whether major adverse cardiovascular events (MACE) occurred, and clinical data and cardiac function indexes before and after treatment were compared between the two groups [left ventricular ejection fraction (LVEF), amino-terminal brain natriuretic peptide precursor (NT-proBNP)], myocardial strain parameters [total strain (εs), passive strain (εe), active strain (εa), peak positive left ventricular systolic strain rate (SRs), peak negative left atrial strain rate in early left ventricular diastole (SRe), peak negative left atrial strain rate in late left ventricular diastole ( SRa)], to analyze the correlation between each myocardial strain parameter and cardiac function indexes, and to evaluate the prognostic impact and predictive value of the difference of each myocardial strain parameter before and after treatment. Results: LVEF before and after treatment was lower in the poor prognosis group than in the good prognosis group [(45.27±3.62)% versus (49.82±4.30)%, t=4.572, P<0.05, (48.96±4.15)% versus (56.41±5.03)%, t=6.423, P<0.05], and NT-proBNP was higher in the good prognosis group [( 1275.84±326.55) pg/mL versus (1014.79±259.87) pg/mL, t=3.924, P<0.05 and (1001.35±241.98) pg/mL versus (665.21±202.43) pg/mL, t=6.591, P<0.05]. The pre- and post-treatment εs, εe, εa, SRs, SRe, SRa and differences were smaller in the poor prognosis group than in the good prognosis group (all P<0.05); myocardial strain parameters were positively correlated with LVEF and negatively correlated with NT-proBNP before and after treatment (all P<0.05); logistic regression analysis showed that myocardial strain parameter differences all had a significant effect on prognosis (all P< Logistic regression analysis showed that the difference in myocardial strain parameters had a significant effect on prognosis (all P<0.05). 1026 Effect of Different Surgical Approaches on the Treatment Outcome and Prognosis of Patients with Multi-Segmental Spinal Cord Cervical Spondylosis

Objective: To analyze the effects of anterior-posterior approach and simple posterior approach on the clinical curative effect, quality of life and postoperative complications in patients with multi-level cervical spondylotic myelopathy and optimize the surgical treatment. Methods: One hundred and twenty patients with multi-segmental spinal cord type cervical spondylosis were collected from our hospital between January 2016 and December 2021, and were divided into the posterior-only group (n=83) and the combined anterior-posterior group (n=37) according to the different surgical approaches. The duration of surgery, bleeding, length of hospital stay and hospital costs were recorded for both groups; the recovery of neurological function and quality of life at 1, 3 and 12 months after surgery were assessed by JOA, VRS and NRS scores and PF, RP and GH rating scales; patient satisfaction was recorded by telephone follow-up. Results: Compared with the posterior group, there were no statistical differences in JOA scores, JOA recovery rates, PF, RP, GH scores and satisfaction among patients with combined anterior and posterior surgery at each postoperative follow-up time point (P>0.05); NDI scores were significantly higher in the combined anterior and posterior group at 3 and 12 months postoperatively, with a statistical difference (P<0.05); VRS and NRS scores were statistically higher in the combined anterior-posterior group than in the posterior-only group (P<0.05); the incidence of postoperative axial symptoms was significantly higher in the combined anterior-posterior group (P<0.05); and the duration of surgery, intraoperative blood loss, postoperative hospital stay and hospital costs were significantly higher in the combined anterior-posterior group (P<0.01). NDI scores were statistically higher in the combined anterior-posterior group of K-line negative patients at 1 and 12 months postoperatively compared to the posterior-only group of K-line negative patients (p<0.05). Conclusion: Combined anterior and posterior surgery does not offer significant advantages in terms of neurological recovery, improvement in quality of life and patient satisfaction, but rather entails greater surgical trauma, cost and complications.

1032 Relationship between Serum HMGB1 and Cav-1 Levels and Delayed Healing after Intramedullary Nail Fixation of Femoral Fractures

Objective: To investigate the relationship between serum high mobility group protein B1 (HMGB1) and caveolin-1 (Cav-1) levels and delayed healing after intramedullary nail fixation of femoral fractures. Methods: A total of 70 patients admitted to our hospital from January 2018 to July 2022 with delayed healing after intramedullary nail fixation of femur fractures were selected as the delayed healing group, and 70 patients with normal healing after intramedullary nail fixation of femur fractures were selected in a 1:1 ratio as the normal healing group. Clinical data of the patients were collected and serum HMGB1 and Cav-1 levels were measured by ELISA. The factors influencing delayed healing after intramedullary nail fixation of femoral fractures were analyzed by multi-factor logistic regression, and the ROC curve was used to analyze the predictive value of serum HMGB1 and Cav-1 levels on delayed healing after intramedullary nail fixation of femoral fractures. Results: Serum HMGB1 (2.71±0.42 ng/ml vs 3.68±1.04 ng/ml) and Cav-1 (10.85±1.89 ng/ml vs 13.03 ± 2.07 ng/ml) levels were reduced in the delayed healing group compared to the normal healing group (t=7.204, 6.497, all P<0.001). Multi-factor logistic regression analysis showed that increasing age (OR=1.095, 95% CI: 1.036 to 1.157) was an independent risk factor for delayed healing after intramedullary nail fixation of femoral fractures, and HMGB1 (OR=0.936, 95% CI: 0.904 to 0.969) and Cav-1 (OR=0.599, 95% CI: 0.470 to 0.763) were elevated as independent protective factors (all P<0.05). ROC curve analysis showed that serum HMGB1, Cav-1 levels combined predicted a greater area under the curve (AUC) for delayed healing after intramedullary nail fixation of femoral fractures than HMGB1, Cav-1 predicted individually (Z=2.352, 2.621, all P<0.05). Conclusion: Decreased serum HMGB1 and Cav-1 levels are closely associated with delayed healing after intramedullary nail fixation of femoral fractures and may be an auxiliary predictor of delayed healing after surgery.

1037 Correlation between Postpartum Hemorrhage and Antenatal FIB:A Systematic Review

Objective: To investigate the correlation between postpartum haemorrhage and antenatal FIB through systematic review and analysis. Methods: Studies published in the last decade (June 2012-June 2022) on the relationship between postpartum haemorrhage and antenatal FIB levels were collected through a comprehensive search of domestic and international databases. Data were extracted independently by two researchers, and the quality of the extracted literature was evaluated using the evaluation criteria of the NOS literature quality evaluation form. Those with a score ≥6 (high quality) were included in the Meta-analysis, and Meta-analysis was performed using RevMan 5.3 software to evaluate the predictive value of antenatal FIB levels on postpartum haemorrhage. The included literature was analysed by subgroup according to different modes of delivery, degree of bleeding and sample size to evaluate the predictive value of antenatal FIB levels for postpartum haemorrhage; funnel plots were produced using stata software to evaluate the publication bias of the included literature. Results: A total of 10 papers were included in this Meta-analysis. Meta-analysis showed that a reduced antenatal FIB level was predictive of postpartum haemorrhage, mean (MD)=-0.30, 95% confidence interval (95% CI) -0.51 to -0.08, P=0.007; subgroup analysis showed that antenatal FIB level was predictive of postpartum haemorrhage in patients with vaginal delivery. The prenatal FIB level was predictive of the occurrence of severe postpartum haemorrhage. The prenatal FIB level was predictive of the occurrence of postpartum haemorrhage in studies with sample size >1000 and sample size <1000. Conclusion: Prenatal FIB levels can be used as a predictor of postpartum haemorrhage.

Methods: A random number table was used to divide 126 newly diagnosed PCOS patients admitted to the People's Hospital of Xuancheng City, Anhui Province from January 2021 to June 2022 into a control group (treated with CC alone, n=63) and an observation group (treated with LE combined with CC, n=63). Glucose and lipid metabolism-related indicators, sex hormone levels, ovulation status and adverse effects was made between the control group and the observation group before and after treatment. Results: The FPG, FINS, HOMA-IR, TG, TC, FSH, LH, E2 and T of the patients in the observation group were all significantly lower than those of the control group after treatment, and the levels of changes in these indicators before and after treatment were all significantly higher than those of the control group, and the differences were all statistically significant (P<0.05). The number of mature follicles and the diameter of mature follicles in the observation group were significantly higher than those in the control group, and the difference was statistically significant (P<0.05). There was no statistically significant difference in the incidence of adverse reactions between the observation group and the control group (P>0.05). Conclusion: LE combined with CC significantly improves glucolipid metabolism and promotes ovarian ovulation in patients with primary PCOS.

1047 Effect of Different Doses of Remazolam on Cardiovascular Responses during Induction of General Anesthesia in Patients Undergoing Laparoscopic Cholecystectomy

Objective: To observe the effects of different doses of Remazolam on cardiovascular responses in patients undergoing laparoscopic cholecystectomy under general anesthesia. Methods: A total of 122 patients who underwent laparoscopic cholecystectomy in our hospital from January 2021 to February 2022 were prospectively selected and divided into two groups according to the random number table generated by statistical software using a simple random design scheme. The control group was anesthetised with 0.1 mg/kg remazolam, and the observation group was anesthetised with 0.2 mg/kg Remazolam. The differences of hemodynamics between the two groups were detected, and anesthesia and adverse reactions were compared between the two groups. Results: There was no significant difference in blood glucose level between the two groups at T0 (P>0.05). When the blood glucose level of the two groups was higher than T0 at 1 min (T1) and 5 min (T2) after anesthesia intubation, the blood glucose level of the observation group at T1 and T2 was lower than that of the control group (P<0.05). At T0, there was no significant difference in hemodynamics between the two groups (P>0.05). The heart rate (HR) and mean arterial pressure (MAP) levels of the control group were higher at T1 and T2 than at T0 (P<0.05). The levels of HR and MAP in the observation group at T1 were higher than those at T0, but there was no statistical difference between the levels of HR and MAP at T2 and those at T0 (P>0.05). The levels of HR and MAP in the observation group at T1 and T2 were lower than those in the control group (P<0.05). The onset time of anesthesia and the duration of PACU in the observation group were shorter than those in the control group, while the recovery time and extubation time had no statistical difference (P>0.05). There was no significant difference in the incidence of adverse reactions between the two groups (P>0.05). Conclusion: In laparoscopic cholecystectomy, 0.2mg/kg Remazolam for anesthesia induction can stabilize blood flow.

1051 Efficacy of Fuzheng Yiai Decoction Supplemented with Laparoscopic Radical Resection of Rectal Cancer in the Treatment of Early Rectal Cancer

Objective: To explore the efficacy of Fuzheng Yiai decoction supplemented with laparoscopic radical resection of rectal cancer in the treatment of early rectal cancer. Methods: A total of 103 patients with early rectal cancer who underwent radical resection of rectal cancer in the hospital between February 2018 and September 2021 were selected as the research subjects, and they were divided into the treatment group and the control group according to the simple randomization method. The control group (n=51) were given mosapride citrate tablets and FOLFOX4 chemotherapy after surgery, and the treatment group (n=52) was additionally treated with Fuzheng Yiai decoction on the basis of the control group. The scores of TCM syndromes, intestinal microecology, immune function, serum tumor markers [serum carcinoembryonic antigen (CEA), alpha-fetoprotein (AFP), Eotaxin-2, cyclooxygenase-2 (COX-2), carbohydrate antigen 199 (CA199)] and adverse drug reactions were compared between the two groups. Results: After 2 months of treatment, the scores of TCM syndromes in the two groups were significantly decreased (P<0.05), the densities of Escherichia coli and Enterococcus were significantly declined (P<0.05) while the densities of Lactobacillus and Bifidobacterium were significantly increased (P<0.05), the proportions of CD3+, CD4+ and NK cells in both groups were significantly enhanced (P<0.05) while the proportion of CD8+ cells was significantly reduced (P<0.05), and the levels of serum CEA, AFP, CA199, COX-2 and Eotaxin-2 were significantly lowered (P<0.05), and the improvement degrees of the above indicators in the treatment group were significantly higher than those in the control group (P<0.05). Conclusion: Laparoscopic radical resection supplemented with Fuzheng Yiai decoction for the treatment of patients with early rectal cancer has significant clinical efficacy and obvious improvements on intestinal microecology and body' immunity.
