WEILIJIANG·Saimaiti, MA Jun, WANG Yujie
DOI: 10.3969/j.issn.1006-6233.2023.04.01
Objective: To investigate the effects of microRNA (miR)-30A-3p on migration and angiogenic phenotype of renal cell carcinoma (RCC) cells. Methods: The expression characteristics of miR-30a-3p in RCC cells were analyzed based on the expression data of miR-30a-3p in the Cancer Genome Atlas (TCGA) database. Predict the binding of miR-30a-3p to the 3'untranslated region (3' -UTR) of B cell activicting factor (BAFF) by bioinformatics websites. RCC cell lines ACHN and 786O were divided into LV-miR-30a-3p group [infected with lentiviral vector overexpressing miR-30a-3p], LV-NC group [infected with lentiviral vector negative control], LV-miR-30a-3p+pcDNA-BAFF group [co-infected with LV-miR-30a-3p and LV-Mir-30a-3p Overexpressing BAFF vector (pcDNA-BAFF)] and LV-miR-30a-3p+pcDNA-EV group [co-infected with LV-miR-30a-3p and empty vector (pcDNA-EV)]. Transwell migration chamber assay was used to detect cell migration ability, and tubule formation assay was used to detect cell angiogenesis ability. The expression of miR-30a-3p was detected by qPCR. The protein expressions of BAFF and vascular endothelial growth factor (VEGFA) were detected by Western blot. Targeting regulation of miR-30a-3p against BAFF in ACHN cells was detected by dual luciferase gene reporting assay. Results: Analysis of the data in the TCGA database showed that miR-30a-3p expression was significantly lower in the Cancer group compared to the Normal group (P<0.05). Compared with HK-2 cells, the expression of miR-30a-3p in ACHN and 786O cell lines was significantly decreased (both P<0.05). Compared with the LV-NC group, the expression of miR-30a-3p was increased in the LV-miR-30a-3p group (all P<0.05), while the cell mobility, tubule formation rate, and the expressions of BAFF and VEGFA were down-regulated in the LV-miR-30a-3p group (all P<0.05). According to bioinformatics prediction and double luciferase reporter assay, BAFF was found to be the target gene of miR-30a-3p. Compared with the LV-miR-30a-3p+pcDNA-EV group, cell mobility, tubule formation rate and BAFF expression in the LV-miR-30a-3p+pcDNA-BAFF group were up-regulated (all P<0.05). Conclusion: miR-30a-3p inhibits the migration and angiogenesis of RCC cells by targeting inhibits BAFF.

DOI: 10.3969/j.issn.1006-6233.2023.04.03
Objective: To explore the expression of Centromeric protein F(CENPF)and its biological function in LUAD. Methods: CENPF mRNA expression in LUAD was analyzed by searching the Oncomine database, CENPF protein expression levels were determined by immunohistochemical (IHC) staining using the HPA database, and IHC images were obtained from the HPA database. In addition, subgroups and prognosis of LUAD were analyzed by using the Ualcan and GEPIA database tools, respectively. To further explore the CENPF sequence mutations in LUAD patients, the c-Bioportal website was applied. Finally, the LinkedOmics database was applied to analyze the co-expressed genes of CENPF in the LUAD cohort for functional and pathway analysis by the LinkInterpreter module. Results: CENPF was highly expressed in LUAD (P<0.05), which could lead to poor prognosis by Kaplan-Meier analysis (P<0.05). And CENPF mRNA expression was related to smoking status, TP53 mutation and tumor stage in LUAD patients (P<0.05). In addition, CENPF was mutated in LUAD, but it had no effect on LUAD prognosis (P=0.698). Co-expression analysis showed that CENPF co-expressed genes were closely related to cell cycle and cell division process. Conclusion: CENPF can be involved in the development of LUAD as a pro-oncogene and has potential applications as a diagnostic, prognostic indicator and targeted therapy for LUAD.

FAN Jing, et al
DOI: 10.3969/j.issn.1006-6233.2023.04.04
Objective: To explore whether miR-153-3p can improve the inflammatory response of human periodontal ligament stem cells (hPDLSCs) induced by lipopolysaccharide (LPS) by targeting regulates chitinase 3-like protein 1 (CHI3L1). Methods: The hPDLSCs were isolated and cultured, and inflammatory cell models were established using LPS treatment and transfected with miR-NC, miR-153-3p mimics, si-NC, si-CHI3L1, and miR-153-3p mimics+pc-NC, miR-153-3p mimics+pcDNA-CHI3L1. 48 h after transfection, the expressions of miR-153-3p and CHI3L1 mRNA in the cells of each group were detected by qRT-PCR, the levels of IL-6, IL-1β and TNF-α in the cell supernatant were detected by ELISA, cell apoptosis was detected by flow cytometry, the expression of CHI3L1 protein in cells was detected by Western Blot, the targeting relationship between miR-153-3p and CHI3L1 was detected by dual-luciferase reporter gene assay. Results: The expression of miR-153-3p was down-regulated in hPDLSCs after LPS treatment (P<0.05), while the expressions of CHI3L1 mRNA and protein were significantly up-regulated (P<0.05); over-expression of miR-153-3p or inhibition of CHI3L1 expression could reduce the levels of IL-6, IL-1β and TNF-α in LPS-induced hPDLSCs, down-regulate the expressions of CHI3L1 mRNA and protein, and reduce the apoptosis rate, and the differences were statistically significant (P<0.05). The dual-luciferase reporter gene assay confirmed that miR-153-3p could targeting regulates the expression of CHI3L1 (P<0.05). In addition, the over-expression of CHI3L1 could significantly reverse the inhibitory effects of the over-expression of miR-153-3p on LPS-induced apoptosis and inflammatory response of hPDLSCs (P<0.05). Conclusion: MiR-153-3p can reduce the LPS-induced inflammatory response and apoptosis of hPDLSCs by targeting regulates CHI3L1 expression.

Postoperative clinical characteristics of the two groups were recorded, and the influencing factors of SA were screened by logistic regression analysis. Results: There were statistically significant differences in general data [age, Karnofsky Performance Scale (KPS) score, body mass index (BMI), and serum prealbumin (PA)] between the two groups (P<0.05), but there was no statistically significant difference in the other clinical indexes (P>0.05). Logistic regression analysis showed that age, KPS score and BMI were influencing factors of SA. Conclusion: The occurrence of SA in elderly patients after esophageal squamous carcinoma surgery is related to multiple factors. Clinically, attention should be paid to the related factors of SA, timely and targeted intervention measures should be taken to reduce the incidence of postoperative SA.

DOI: 10.3969/j.issn.1006-6233.2023.04.014
Objective: To investigate the effect of enteric probiotics supplementation in children with bronchial asthma and its mechanism of action. Methods: A total of 105 children with bronchial asthma admitted to our hospital from January 2019 to January 2022 were randomly divided into routine group (52 cases) and probiotic group (53 cases). The routine group was given routine treatment, and the probiotic group was given intestinal probiotics adjuvant treatment on the basis of routine treatment. The two groups were detected the differences of intestinal flora, immune function and sputum eosinophil percentage (EOS%), integrin α4β1 and αvβ6 before and after treatment, and the adverse effects and the number of acute attacks within 6 months were counted. Results: Before treatment, there was no significant difference in intestinal flora between the two groups (P>0.05). After treatment, bifidobacterium and lactobacillus in probiotics group were higher than before, while Escherichia coli was lower. There was no statistical significance in intestinal flora in conventional group compared with before treatment (P>0.05). The difference values of bifidobacterium, lactobacillus and Escherichia coli before and after treatment in probiotic group were larger than those in conventional group (P<0.05). Before treatment, there were no significant differences in sputum EOS%, integrin α4β1, αvβ6 and immune function between the two groups (P>0.05). After treatment, CD8 Anhui+, EOS% of sputum, integrin α4β1, αvβ6 were decreased, and CD4 Anhui+, CD4 Anhui+/CD8 Anhui+ were increased in both groups (P<0.05). The difference before and after treatment in probiotic group was larger than that in conventional group (P<0.05). There were 3 cases of adverse reaction in probiotics group, the incidence was 5.66%, and the adverse reaction rate was not statistically significant compared with 11.54% in conventional group (P>0.05). During the 6-month follow-up, the number of acute attacks in the probiotic group (0.41±0.16) was significantly lower than that in the conventional group (0.87±0.23), and the difference was statistically significant (P<0.05). Conclusion: Intestinal probiotics in the adjuvant treatment of bronchial asthma can improve the immune function of children and reduce the number of acute attacks. The possible mechanism is related to improving the structure of intestinal flora and reducing the expression of EOS%, integrin α4β1 and αvβ6 in sputum.

DOI: 10.3969/j.issn.1006-6233.2023.04.016
Objective: To analyze the expression of microRNA (miR)-155 and miR-143 in patients with premature ovarian insufficiency (POF), and to explore their predictive effect on successful pregnancy. Methods: A total of 172 patients with POF in our hospital from June 2020 to December 2021 were selected as the research objects, and 86 healthy people without POF during the same period were selected as the control. The expressions of miR-155 and miR-143 were compared between POF patients and non-POF healthy subjects, and the pregnancy was counted. Logistic regression was used to analyze the influencing factors of successful pregnancy in POF patients. The value of successful pregnancy in patients was analyzed with POF assessed by miR-155 and miR-143 by subject operating characteristic curve (ROC), decision curves were plotted to analyze the benefit of successful pregnancy in patients with POF assessed by miR-155 and miR-143, and the rate of successful pregnancy in patients with POF was compared with different expression of miR-155 and miR-143. Results: Patients with POF had lower miR-155 expression levels and higher miR-143 expression levels than healthy subjects with non-POF (P<0.05). There were 79 successful pregnancies and 93 non-pregnant cases in 172 POF patients; compared with non-pregnant POF patients, those with successful pregnancies had higher basal follicle counts, shorter duration of infertility, younger female age, younger male age, higher expression levels of anti-müllerian hormone and miR-155, and lower expression levels of miR-143 (P<0.05). Basal follicle count of 2-4 and high miR-155 expression were independent protective factors for pregnancy failure in POF patients, and low miR-143 expression was an independent risk factor for pregnancy failure in POF patients (P<0.05). The AUC value of the combined miR-155 and miR-143 assay to assess successful pregnancy in patients with POF was 0.812 (95% CI: 0.746 to 0.867), which was higher than the AUC values of miR-155 (χ2=2.625, P=0.009) and miR-143 (χ2=2.973, P=0.003) alone. Decision curve analysis showed that the net benefit of the combined miR-155 and miR-143 test in assessing successful pregnancy in POF patients was higher than that of the test alone when the risk threshold was 0.10 to 0.63. Decision curve analysis showed that the net benefit of the combined miR-155 and miR-143 test in assessing successful pregnancy in POF patients was higher than that of the test alone when the risk threshold was 0.10 to 0.63. miR-155+/miR-143- had a significantly higher rate of successful pregnancy than miR-155+/miR-143+, miR 155-/miR-143- and miR-155-/miR-143+ (P<0.05). Conclusion: POF patients with low miR-155 expression and high miR-143 expression, both of which have a direct impact on the success of pregnancy in POF patients. miR-155 high expression and miR-143 low expression have a higher success rate of pregnancy in POF patients, and the detection of miR-155 and miR-143 can predict pregnancy, which has certain clinical application value.

CHAI Jing, WANG Doudou, WANG Jiao, et al
DOI: 10.3969/j.issn.1006-6233.2023.04.018
Objective: To investigate the distribution of pathogenic bacteria, drug resistance and influencing factors of postoperative pseudojoint infection in patients with knee osteoarthritis in Shijiazhuang Third Hospital from 2016 to 2020. Methods: Totally 1509 patients with knee osteoarthritis who underwent knee surgery in Shijiazhuang Third Hospital from January 2016 to January 2020 were selected and divided into infection group (38 cases) and non-infection group (1471 cases) according to postoperative infection. The knee joint effusion of the infected patients was collected and cultured to detect the types of pathogenic bacteria and drug resistance. Clinical data were collected and multivariate Logistic regression was used to analyze the factors influencing postoperative infection in patients with knee osteoarthritis. Results: A total of 46 strains of pathogenic bacteria were detected, of which 86.96% were Gram-positive bacteria, Gram-positive bacteria were resistant to β-lactam, quinolones and other antibiotics.Duration of operation ≥3h, type 2 diabetes, rheumatoid arthritis, smoking, postoperative urinary tract infection and prevention without antibiotics were risk factors for postoperative periprosthetic infection in patients with knee osteoarthritis (P<0.05). Conclusion: Gram-positive bacteria are the main pathogenic bacteria of pseudojoint infection after knee osteoarthritis, and have high drug resistance rate. Long operation time, type 2 diabetes mellitus, rheumatoid arthritis, smoking, postoperative urinary tract infection and failure to use antibiotics to prevent infection are the main factors leading to infection around pseudarthrosis. Infection monitoring of high-risk patients should be strengthened and antibiotics should be rationally used to reduce the risk of infection.

XIE Xiaoli, et al
DOI: 10.3969/j.issn.1006-6233.2023.04.025
Objective: To investigate the effects of domestic rapamycin drug-eluting stent and paclitaxel drug-eluting balloon on heart rate variability, levels of serum Kallikrein 1 (KLK1) and tissue inhibitor of matrix metalloproteinase-1 (TIMP-1) and short-term prognosis in patients with coronary heart disease. Methods: The data of 105 patients with coronary artery disease who received percutaneous coronary intervention in the hospital from November 2019 to May 2021 were reviewed, and they were divided into the observation group (56 cases, using domestic rapamycin drug-eluting stent) and the control group (49 cases, treated with domestic paclitaxel drug-eluting balloon) according to different stents used. The heart rate variability-related indicators [standard deviation of normal sinus RR interval within 5 min (SDNN), standard deviation of the 5-minute average RR intervals (SDANN), percent of differences between adjacent NN intervals greater than 50 ms (pNN50)] before surgery, at 1 week after surgery and at 1 year follow-up, levels of KLK1, TIMP-1, endothelin-1 (ET-1) and nitric oxide (NO) before surgery and at 1 week after surgery as well as short-term prognosis [vascular re-stenosis, vascular re-thrombosis, major adverse cardiovascular events (MACE)] at 1 year of follow-up were compared between the two groups. Results: There were statistical differences in the SDNN, SDANN and pNN50 in repeated measures analysis of variance between the two groups form the aspect of time-point effect (P<0.017), and the above three indicators showed upward trends with the prolongation of postoperative time. The absolute values of differences of serum KLK1, TIMP-1, NO and plasma ET-1 before and after surgery revealed statistically significant differences between the two groups (P<0.05), and the changes in the observation group were greater than those in the control group. At 1 year of follow-up, the incidence rate of vascular re-stenosis in the observation group was lower than that in the control group (P<0.05), but there were no statistically significant differences in the incidence rates of vascular re-thrombosis and MACE between the two groups (P>0.05). Conclusion: Domestic rapamycin drug-eluting stent can effectively regulate heart rate variability in patients with coronary artery disease, and it has a positive effect on improving serum KLK1 and TIMP-1 levels, and can help protect and repair the endothelial function, reduce the risk of stent stenosis, and achieve the good short-term prognosis.

DOI: 10.3969/j.issn.1006-6233.2023.04.026
Objective: To explore the correlation of the mean platelet volume to lymphocyte ratio (MPVLR), Glasgow prognosis score (Glasgow) and the prognosis of patients with acute coronary syndrome after percutaneous coronary intervention (PCI). Methods: A total of 98 patients with acute coronary syndrome treated in our hospital from January 2019 to July 2021 were selected, according to whether major adverse cardiovascular events (MACE) such as cardiac or all-cause death and acute heart failure occurred one year after operation, they were divided into poor prognosis group and good prognosis group, with 30 cases and 68 cases respectively. The differences of clinical data such as GRACE score, Glasgow score and MPVLR of patients with poor prognosis and those with good prognosis were observed and compared. Results: The admission GRACE score of patients with poor prognosis was (145.50 ± 19.29), which was significantly higher than that of patients with good prognosis (P<0.05). The proportion of Glasgow score 1-2 in patients with poor prognosis was 80.00%, which was significantly higher than that in patients with good prognosis (P<0.05). The peak values of MPVLR and cardiac troponin T (cTnT) in patients with poor prognosis were (6.90 ± 1.01) and (6.69 ± 1.32) ng / ml, respectively, which were significantly higher than those in patients with good prognosis (P<0.05). The left ventricular ejection fraction (LVEF) of patients with poor prognosis was (46.60 ± 7.82)%, which was significantly lower than that of patients with good prognosis (P<0.05); Logistic regression analysis showed that Glasgow score, MPVLR and LVEF were the influencing factors of poor prognosis after PCI in patients with acute coronary syndrome (P<0.05). Conclusion: MPVLR and Glasgow scores at admission are related to poor prognosis of patients with acute coronary syndrome after PCI, which is worthy of further study.

DOI: 10.3969/j.issn.1006-6233.2023.04.028
Objective: To analyze the relationship between 25 hydroxyvitamin D, microinflammation and fall in patients treated with hemodialysis. Methods: A total of 80 patients receiving maintenance hemodialysis (MHD) treatment in Chengde Central Hospital from January 2020 to June 2020 were divided into 25(OH)D low group and normal group according to their 25(OH)D level. Gender, age, dialysis age, height, weight, primary disease ,blood purification method of the patients and the occurrence of falls in the past one year were recorded. Blood procalcitonin (PCT), haemoglobin (Hb), blood albumin, blood calcium, blood phosphorus, parathyroid hormone, triglyceride (TG), total cholesterol (TC), low density lipoprotein cholesterol and other indicators were tested. The relationship between 25(OH)D, PCT and fall was analyzed by Pearson correlation analysis, multiple linear regression and binary Logistic analysis. Results: The level of PCT, TG and TC in 25 (OH) D normal group were lower than those in 25 (OH) D low group, and the differences were statistically significant (P<0.05). The level of Hb in the 25 (OH) D normal group was higher than that in the 25(OH)D low group, and the difference was statistically significant (P<0.05). Pearson correlation analysis showed that there was a negative correlation between PCT,TC and 25(OH)D level. Multivariate regression analysis showed that PCT and TC were independent influencing factors of 25(OH)D level, and 25(OH)D level was an protective factor affecting fall in MHD patients. Conclusion: PCT plays an important role in the 25(OH)D level of MHD patients, it is an independent risk factor affecting 25(OH)D level, clinical attention should be paid to the correction of PCT level in MHD patients. Active vitamin D supplementation is also needed to improve 25(OH)D levels, reduce the incidence of falls, and continuously improve the quality of survival.
