353 Mechanisms of Luteolin A on Proliferation Invasion and Epithelial Mesenchymal Transition of Hepatocellular Carcinoma Cells through Regulation of MEK/ERK Signaling Pathway

Objective: To investigate the mechanism of luteolin A on proliferation, invasion and epithelial mesenchymal transformation of hepatocellular carcinoma cells by regulating MEK/ERK signaling pathway. Methods: HepG2 cells were divided into control group, OAL group, OAM group, OAH group, PD98059 group and ML-099 group. MTT assay was used to detect cell proliferation; Transwell chamber method was used to detect the invasive ability of cells; Apoptosis was detected by flow cytometry; the expression of MEK.p-MEK, ERK.p-ERK and EMT related proteins were detected by Western blot. Results: The cell proliferation and invasion ability of OAL group, OAM group and OAH group (82.34 ± 2.43), (61.04 ± 1.61) and (32.44 ± 1.04) were significantly lower than those of the control group (110.42 ± 3.86), and the cell apoptosis rate in those group (6.81 ± 0.62), (11.74 ± 0.94) and (18.36 ± 1.05) were significantly higher than those of the control group (5.01 ± 0.53) (Finvasion=1068, Fapoptosis=323.1, P<0.001); the expression of E-cadherin protein (0.42 ± 0.04), (0.58 ± 0.05) and (0.97 ± 0.09) in the cells was higher than that in the control group (0.23 ± 0.03) (F=181.1, P<0.001); N-cadherin (0.63 ± 0.06), (0.51 ± 0.05), (0.38 ± 0.03), Vimentin (1.23 ± 0.11), (0.98 ± 0.09), (0.51 ± 0.05) and p-MEK/MEK (0.72 ± 0.07), (0.51 ± 0.05), (0.32 ± 0.03) and p-ERK/ERK (0.92 ± 0.09), (0.68 ± 0.06). The ratio (0.41 ± 0.04) was lower than that of the control group (FFN-cadherin=39.04, FFVimentin=11.0, FFp-MEK/ME=107.8, FFp-ERK/ERK=82.68, P<0.001). The cell proliferation and invasion ability (30.86 ± 1.01) of PD98059 group was significantly lower than that of the control group (t=48.84, P<0.001), the cell apoptosis ability (17.95 ± 0.98) was significantly higher than that of the control group (t=28.45, P<0.001), the epithelial mesenchymal transformation protein E-cadherin protein (0.92 ± 0.09) was significantly higher than that of the control group (t=17.82, P<0.001), the epithelial mesenchymal transformation protein N-cadherin (0.37 ± 0.04) (t=9.585),vimentin protein (0.41 ± 0.04) (t=19.99) and p-MEK/MEK (0.30 ± 0.03) (t=16.78), p-ERK/ERK (0.39 ± 0.04) (t=14.65) were significantly lower than those in the control group (P<0.001). The cell proliferation and invasion ability (100.86 ± 2.68) of ML-099 group was significantly higher than that of OAH group (t=12.54, P<0.001), and the cell apoptosis ability (5.15 ± 0.86) was significantly lower than that of OAH group (t=23.84, P<0.001); compared with OAH group, E-cadherin protein (0.25 ± 0.03) was significantly decreased in ML-099 group (t=18.59, P<0.001), and N-cadherin protein (0.69 ± 0.07) (t=0.971, P<0.001), Vimentin protein (1.50 ± 0.12) (t=18.65, P<0.0001), p-MEK/MEK (0.94 ± 0.09) (t=16.01, P<0.001), p-ERK/ERK (1.05 ± 0.10) (t=2.367, P=0.0395) were significantly increased in ML-099 group. Conclusion: Luteolin A can inhibit the proliferation, invasion and epithelial mesenchymal transformation of liver cancer cells, and induce apoptosis of liver cancer cells. Its mechanism is related to inhibition of MEK/ERK signaling pathway.

358 circ_0086414 Targeting miR-155-5p Inhibits Hippo/YAP Signaling Pathway to Regulate Proliferation Migration and Invasion of Oral Squamous Cell Carcinoma Cells

Objective: To investigate the effect of circ-0086414 targeting miR-155-5p on the inhibition of Hippo/Yap signaling pathway in regulating the proliferation, migration and invasion of oral squamous cell carcinoma cells. Methods: RT-PCR was used to detect the expression of circ-0086414 in HIOEC, Tca8113, CAL27 and SCC-9 cells.SCC-9 cells which were not transfected with any plasmid were set as control group; SCC-9 cells transfected with pcDNA-circ-0086414, pcDNA-NC, miR-155-5p inhibitor and miR-NC inhibitor plasmids were set as pcDNA-circ-0086414 group, pcDNA-NC group, miR-155-5p group and miR-NC group respectively; the pcDNA-circ-0086414 plasmid was transfected into SCC-9 cells, and 5 μmol/L Verteporfin was co-cultured and set as Verteporfin group. The expression of circ-0086414 and miR-155-5p was detected by RT-PCR; CCK-8 was used to detect cell proliferation; Transwell chamber method was used to detect the invasive ability of cells; the cell migration ability was detected by scratch test; the expression of Lats1, p-Lats1, YAP, p-YAP protein in cells was detected by Western blot; the target relationship between circ-0,086,414 and miR-155-5p was verified by double luciferase reporting system experiment. Results: Compared with human immortalized oral epithelial cells HIOECcirc-0086414 (1.00±0.10) and miR-155-5p (1.00±0.09), the expression of circ-0086414 (0.73±0.07), (0.51±0.05) and (0.36±0.03) in oral squamous cell carcinoma cell lines Tca8113, CAL27 and SCC-9 significantly decreased, and the expression of miR-155-5p (1.36±0.11), (1.57±0.13) and (1.89±0.15) significantly increased (P<0.05); the expression of circ-0086414 and miR-155-5p in SCC-9 cells changed most significantly (P<0.05). Compared with the control group, the expression of circ-0086414 in pcDNA-circ-0086414 group was significantly higher (1.26±0.12), and the expression of miR-155-5p in miR-155-5p group was significantly lower (0.73±0.07) (P<0.001). Compared with the control group, the proliferation, invasion (123.56±5.87) and migration (16.90±1.62) of cells in pcDNA-circ-0086414 group significantly decreased (P<0.05). Compared with pcDNA-circ-0086414 group, the proliferation, invasion and migration of cells in Verteporfin group significantly increased (P<0.05). Compared with the control group, the expression of Last1 (1.12±0.10), p-Last1 (1.49±0.12) and p-YAP (1.4±0.11) protein in pcDNA-circ-0086414 group significantly increased, while the expression of YAP protein (0.45±0.04) significantly decreased (P<0.001). Compared with pcDNA-circ-0086414 group, the expression of Last1 (0.61±0.06), p-Last1 (0.82±0.08) and p-YAP protein (0.58±0.05) in the cells of Verteporfin group significantly decreased, and the expression of YAP protein (0.93±0.09) significantly increased (P<0.001). The luciferase activity of wild-type circ-0086414 (circ-0086414-WT) miR-155-5p group (0.26±0.03) was significantly lower than that of miR-NC group (P<0.05). Conclusion: Overexpression of circ-0086414 can inhibit the proliferation, invasion and migration of oral squamous cell carcinoma SCC-9 cells. Its mechanism is related to inhibition of miR-155-5p expression and activation of Hippo Yap signal pathway.

364 Effects of Downregulation of SIP1 on Apoptosis Invasion and Migration of Endometrial Cancer Cells Based on ROCK-2/Moesin Signaling Pathway

Objective: To investigate the effect of down-regulation of SIP1 on apoptosis, migration and invasion of endometrial cancer cells and its mechanism. Methods: Human endometrial carcinoma cell line HEC-1A was divided into control group, NC group, SIP1 down-regulated group (transfected with si-SIP1), Y27632 group (ROCK inhibitor), SIP1 down-regulated group+Y27632 group (transfected with si-SIP1+ROCK inhibitor). The apoptosis, migration ability and invasion ability of each group were detected by flow cytometry and Transwell assay. The expression levels of apoptosis key protein b-lymphoma-2 gene (Bcl-2), apoptosis gene Bcl-2 related protein X (Bax), caspase family protein 3 (caspase 3), Rho related kinase 2 (ROCK-2) and Moesin were detected by Western blot. Twenty SPF-grade BALB/c mice were tumor-loaded subcutaneously to establish an endometrial cancer tumor model. The control group (n=10) was tumor-loaded with human endometrial cancer HEC-1A cells, and the SIP1 down-regulated group (n=10) was tumor-loaded with human endometrial cancer HEC-1A cells with down-regulated SIP1. Tumor volume was measured with vernier caliper every 3 days, and the tumor growth curve was drawn for 21 days. At the end point, the tumor mass of each mouse was measured and the expression levels of ROCK-2 and Moesin protein in tumor tissues were detected. Results: The results of cell experiment showed that compared with the control group and NC group, the proportion of apoptosis, the number of cell migration and cell invasion in SIP1 down regulation group, Y27632 group and SIP1 down regulation + Y27632 group increased significantly (P<0.01), there was no significant difference between NC group and control group (P>0.05). The results of Western blot showed that the expression levels of ROCK-2 and Moesin in SIP1 down regulated group, Y27632 group and SIP1 down regulated + Y27632 group decreased compared with the control group and NC group (P<0.01). The mice experiment revealed that compared with the control group, the tumor volume, tumor mass and the expression levels of ROCK-2 and Moesin in tumor tissues decreased significantly after down-regulation of SIP1 (P<0.01). Conclusion: Down regulation of SIP1 can promote apoptosis, inhibit migration and invasion of human endometrial cancer cell line HEC-1A, and its may be related to the regulation of ROCK-2/Moesin signal pathway.

369 Effects of Paeoniflorin on Wound Healing and HIF-1α/VEGF/VEGFR2 Signaling Pathway in Rats with Refractory Ulcers

Objective: To investigate the influence and mechanism of paeoniflorin (PF) on wound healing in rats with refractory ulcer by regulating hypoxia-inducible factor-1α (HIF-1α)/vascular endothelial growth factor (VEGF)/vascular endothelial growth factor receptor 2 (VEGFR2). Methods: Sixty SD rats were randomly grouped into Sham operation (Sham) group, Model group, PF group (30 mg/kg,twice a day), PX-478 group (100 mg/kg HIF-1α/VEGF/VEGFR2 pathway inhibitor PX-478,once a day), and PF+PX-478 group, 12 animals in each group, twice a day, for 2 weeks. The rat model of refractory wound was made by injecting hydrocortisone into the general wound surface. The camera combined with Image J software was used to calculate the wound healing rate of each group of rats; ELISA method was used to detect oxidative stress indicators in rat serum; HE staining was used to detect the pathological changes of rat wounds; TUNEL staining was used to detect apoptosis in wound tissue; RT-qPCR was used to detect the expressions of transforming growth factor β1 (TGF-β1) and platelet-derived factor (PDGF) in rat wound tissue; Western blotting was used to detect the expression of HIF-1α/VEGF/VEGFR2 pathway-related proteins in rat wound tissue. Results: Compared with the Sham group, the wound fibroblasts and capillaries of the rats in the Model group were sparse, the healing rate, levels of SOD, GSH-PX, mRNA levels of TGF-β1, PDGF, and protein levels of HIF-1α, VEGF, VEGFR2 were obviously decreased (P<0.05), the level of MDA and the apoptosis rate of wound tissue were obviously increased (P<0.05); compared with the Model group, the wound fibroblasts and capillaries of the rats in the PF group were denser (P<0.05), the healing rate, levels of SOD, GSH-PX, mRNA levels of TGF-β1, PDGF, and protein levels of HIF-1α, VEGF, VEGFR2 were obviously increased (P<0.05), the level of MDA and the apoptosis rate of wound tissue were obviously decreased (P<0.05), the corresponding indicators in the PX-478 group showed opposite trends (P<0.05); compared with the PF group, the wound fibroblasts and capillaries of the rats in the PF+PX-478 group were sparse (P<0.05), the healing rate, levels of SOD, GSH-PX, mRNA levels of TGF-β1, PDGF, and protein levels of HIF-1α, VEGF, VEGFR2 were obviously decreased (P<0.05), and the level of MDA and the apoptosis rate of wound tissue were obviously increased (P<0.05). Conclusion: PF may improve wound healing in rats with refractory ulcer by activating the HIF-1α/VEGF/VEGFR2 pathway.

375 Influence of D-pinitol Modulation of Notch1/Hes1 Signaling Pathway on Th1/Th2 Balance in Rats with Psoriatic Skin Lesions

Objective: To explore the mechanism of the influence of D-pinitol on Th1/Th2 balance in psoriatic skin lesions in rats. Methods: The imiquimod (IMQ) rat model was constructed and randomly grouped into control group, IMQ group, low-dose D-pinitol group, medium-dose D-pinitol group, high-dose D-pinitol group and positive control group , 10 in each group. After successful modeling, the rats were given intragastric administration of D-pinitol low, medium and high dose groups ( 50 mg / kg, 100 mg / kg, 150 mg / kg ) and positive control group ( 1 mg / kg, methotrexate ). The control group and IMQ group were given the same dose of normal saline once a day for 7 days.On the 8th day of administration, different drug treatments were given to evaluate the psoriasis skin lesion symptoms and index (MPASI) of rats in each group; HE staining was used to observe the pathological changes of psoriatic skin lesions; ELISA was applied to detect serum levels of interferon-γ (IFN-γ), tumor necrosis factor-α (TNF-α), interleukin-4 (IL-4), and interleukin-10 (IL-10); flow cytometry was used to measure the T helper 1 (Th1)/helper 2 (Th2) ratio; Western blot was applied to detect the protein expressions of T box transcription factor (T-bet), GATA-binding protein 3 (GATA-3), Notch 1 and hairy and enhancer of split 1 (Hes1). Results: Compared with the control group, the rats in the IMQ group had psoriasis-like lesions such as erythema, hyperkeratinization, inflammatory cell infiltration, etc. the levels of IFN-γ, TNF-α, Th1 cells, T-bet, Notch1 and Hes1 were obviously increased (P<0.05), the levels of IL-4, IL-10, Th2 cells, and GATA-3 were obviously reduced, the ratio of Th1/Th2 cells was unbalanced (P<0.05); after D-pinitol treatment, the psoriasis-like lesions in rats were alleviated or disappeared, and the skin lesion index decreased, the levels of IFN-γ, TNF-α, Th1 cells, T-bet, Notch1 and Hes1 decreased (P<0.05), the levels of IL-4, IL-10, Th2 cells, GATA-3 increased, Th1/Th2 ratio recovered somewhat (P<0.05). Conclusion: D-pinitol may inhibit Notch1/Hes1 signaling pathway to regulate Th1/Th2 balance, reduce the production of inflammatory factors, and alleviate psoriatic skin lesions in IMQ rats.

380 Therapeutic Effects of Osthol in Modulating Mincle/Syk/NF-κB Signaling Pathway in Rats with Nephrotic Syndrome

Objective: To investigate the therapeutic effect of osthol (OST) on nephrotic syndrome (NS) in rats by regulating macrophage-inducible C-type lectin (Mincle)/spleen tyrosine kinase (Syk)/nuclear factor-κB (NF-κB) signal pathway. Methods: Ten rats were randomly selected and recorded as group N, and other rats were used to construct NS model. The 50 successfully modeled rats were randomly divided into NS group (model group), L-OST group (10 mg/kg OST), M-OST group (20 mg/kg OST), H-OST group (40 mg/kg OST), H-OST+TDB group (40 mg/kg OST+50 μg TDB/week). OST was injected once a day for 4 consecutive weeks. The levels of inflammatory factors (IL-1β, IL-6, TNF-α), oxidative stress indicators (SOD, MDA, LDH) in serum and UP in 24h urine were detected by ELISA; the levels of BUN and Scr was measured by automatic chemical analyzer; HE staining and Masson staining were used to detect the pathological changes of kidney; TUNEL staining was used to detect renal cell apoptosis; Western blot was used to detect the expression of collagen I, Ly6g, Ki-67, and Mincle/Syk/NF-κB pathway proteins. Results: In group N, the renal tissue structure was normal and the staining was clear. In NS group, a large number of interstitial inflammatory cells infiltrated, renal tubules atrophied, and a large number of vacuoles were observed, compared with group N, the contents of 24h UP, Scr, BUN, levels of IL-1β, IL-6, TNF-α, MDA, LDH, the collagen volume fraction, apoptosis rate, the levels of collagen I, Ly6g, Ki-67, Mincle, Syk, p-NF-κB/NF-κB proteins in NS group increased significantly (P<0.05), the level of SOD decreased significantly (P<0.05); compared with NS group, the inflammatory cell infiltration and renal tubule atrophy in the L-OST group, M-OST group and H-OST group were improved, the contents of 24h UP, Scr, BUN, levels of IL-1β, IL-6, TNF-α, MDA, LDH, the collagen volume fraction, apoptosis rate, the levels of collagen I, Ly6g, Ki-67, Mincle, Syk, p-NF-κB/NF-κB proteins decreased significantly (P<0.05), the level of SOD increased significantly (P<0.05), the treatment effect of H-OST group was the best; TDB eliminated the ameliorative effect of OST on renal injury in NS rats. Conclusion: OST may alleviate renal injury in OST rats by inhibiting the Mincle/Syk/NF-κB signal pathway.

387 The Expression and Clinical Significance of CD68-TAMs and CD105-MVD in Laryngeal Squamous Cell Carcinoma

Objective: To investigate the expression and clinical significance of CD68-labeled tumor-associated macrophages (CD68-TAMS) and CD105-labeled microvascular density (CD105-MVD) in laryngeal squamous cell carcinoma. Methods: The paraffin specimens of 40 cases of laryngeal squamous cell carcinoma confirmed by surgical resection and pathological examination were retrospectively studied, and 20 cases of adjacent normal tissues (not infiltrated by laryngeal carcinoma cells) were used as control group. Immunohistochemical SP method was used to detect the protein expression of CD68-Tams and CD105 and microvessel density MVD. Kaplan-meier method was used to analyze the relationship between the expression of CD68-Tams and CD105-MVD and the prognosis of patients. Results: The positive expression rate of CD68-TAMS in laryngeal cancer group (77.5%, 31/40) was significantly higher than that in control group (20.00%, 2/20), and the difference was statistically significant (P<0.05). The positive expression rate of CD105-MVD in laryngeal carcinoma group (72.50%, 31/40) was significantly higher than that in control group (30.0%, 6/20) (P<0.05). The expression of CD68-Tams was correlated with lymph node metastasis, tumor differentiation and TNM stage (P<0.05), and the expression of CD105-MVD was correlated with lymph node metastasis and tumor differentiation (P<0.05). The 3-year survival rate of patients with positive CD68-Tams expression was lower than that of patients with low CD68-TAMS expression (P<0.05). The 3-year survival rate of patients with positive CD105-MVD expression was lower than that of patients with low CD105-MVD expression (P<0.05). Cox regression analysis showed that positive CD68-TAMs, positive CD105-MVD, tumor stage and lymph node metastasis were independent influencing factors for poor prognosis of laryngeal cancer (P<0.05). Conclusion: The expression of CD68-TAMs and CD105-MVD in laryngeal cancer tissues indicates that they promote tumor angiogenesis and tumor development, and can be used as markers for the diagnosis of laryngeal squamous cell carcinoma. Positive CD68-TAMs, positive CD105-MVD, tumor staging and lymph node metastasis in laryngeal cancer tissues are closely related to prognosis.It can be used as an independent evaluation indicator of poor prognosis of laryngeal cancer.

393 Correlation between Serum IGF-1 VEGF MMP-9 Levels and Disease Severity in Patients with Combined Pulmonary Fibrosis and Emphysema

Objective: To explore the correlation between serum insulin-like growth factor-1 (IGF-1), vascular endothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9) levels and disease severity in patients with combined pulmonary fibrosis and emphysema (CPFE). Methods: A total of 58 patients with CPFE in the hospital were enrolled as CPFE group, 30 patients with pulmonary fibrosis were enrolled as pulmonary fibrosis group, 30 patients with simple emphysema were enrolled as emphysema group, and 30 healthy controls were enrolled as control group between January 2015 and December 2021. The levels of serum IGF-1, VEGF and MMP-9 were detected, and their differences in different groups were compared. The relationship between serum IGF-1, VEGF, MMP-9 and pulmonary function indexes [forced expiratory volume in 1 second (FEV1), forced vital capacity (FVC), FEV1/FVC], blood gas indexes [partial pressure of arterial blood oxygen (PaO2), partial pressure of carbon dioxide (PaCO2)] was analyzed. Results: The levels of serum IGF-1, VEGF and MMP-9 were the highest in pulmonary fibrosis group, followed by CPFE group, emphysema group and control group (P<0.05). PaO2 level in CPFE group was lower than that in pulmonary fibrosis group and emphysema group (P<0.05), and FEV1/FVC was lower than that in pulmonary fibrosis group and emphysema group (P<0.05). The levels of serum IGF-1, VEGF and MMP-9 was negatively correlated with PaO2 (r=-0.379, -0.357, -0.283, P<0.05), while positively correlated with PaCO2 (r=0.394, 0.302, 0.330, P<0.05). The levels of serum IGF-1, VEGF and MMP-9 was negatively correlated with FEV1 and FEV1/FVC (r=-0.301, -0.246, -0.251, -0.411, -0.373, -0.310, P<0.05), while positively correlated with FVC (r=0.271, 0.253, 0.234, P<0.05). Conclusion: The expression levels of serum IGF-1, VEGF and MMP-9 are high in CPFE patients, which are negatively correlated with lung function, while positively correlated with partial pressure of blood oxygen. They can reflect pulmonary function and limitation degree of ventilation function to a certain extent.

Objective: To explore the expression levels of soluble triggering receptor expressed on myeloid cells-1 (sTREM-1) and pentraxin 3 (PTX3) in the pleural effusion of patients with tuberculous pleurisy(TBP), and analyze their clinical significance. Methods: Ninety-eight patients with TBP (TBP group) admitted in our hospital and 84 non-TBP patients whose general data matched with the TBP group during the same period were selected as the non-TBP group. The patients in the TBP group were divided into a good prognosis group (70 cases) and a poor prognosis group (28 cases) according to the treatment effect. Enzyme-linked immunosorbent assay was used to detect the expression levels of sTREM-1 and PTX3 in pleural effusion, and the differences in sTREM-1 and PTX3 were analyzed between TBP group and non-TBP group, between good prognosis group and poor prognosis group; ROC curve was used to analyze the effects of sTREM-1 and PTX3 expression in pleural effusion on the early diagnosis and prognosis of TBP. Results: The expression levels of sTREM-1 and PTX3 in pleural effusion in TBP group were significantly higher than those in non-TBP group (t=13.516, 12.036, P<0.05); ROC analysis showed that the area under the curve of sTREM-1 combined with PTX3 in diagnosing TBP was 0.962, which was larger than 0.925 and 0.870 for sTREM-1 and PTX3 alone (Z=2.910, 4.233, P<0.05); the expression levels of sTREM-1 and PTX3 in the poor prognosis group were higher than those in the good prognosis group (t=10.051, 11.729, P<0.05); the area under the curve of sTREM-1 combined with PTX3 was 0.971, which was larger than 0.894 and 0.911 for sTREM-1 and PTX3 alone (Z=1.969, 2.356, P<0.05). Conclusion: The expression levels of sTREM-1 and PTX3 in the pleural effusion of patients with TBP increased. The expression levels of sTREM-1 and PTX3 in the pleural effusion of patients with poor prognosis were higher than those of patients with good prognosis. sTREM-1 combined with PTX3 could be used to identify the occurrence of TBP and to diagnose the prognostic effect.

Objective: To investigate the effect of recombinant human brain natriuretic peptide for injection combined with furosemide in the treatment of refractory heart failure, and the influence on cardiac function, blood gas indexes, serum N-terminal pro-brain natriuretic peptide (NT-pro BNP) and procalcitonin (PCT) levels. Methods: The clinical data of 108 cases of refractory heart failure treated in our hospital from January 2018 to December 2021 were retrospectively collected. They were divided into a control group (n=54, furosemide) and an observation group (n=54, furosemide combined with Recombinant Human Brain Natriuretic Peptide for Injection) by different medication methods. Clinical effects, cardiac function, blood gas indexes, serum NT-pro BNP and PCT levels were compared between the two groups. Results: The treatment response rate in the observation group (94.44%) was higher than that in the control group (81.48%) (P<0.05). The values of stroke volume left ventricular ejection fraction, left ventricular end-systolic diameter, and left ventricular end-diastolic diameter in the observation group before and after treatment were higher than those in the control group(P<0.05). The differences in arterial partial pressure of oxygen (PaO2), arterial partial pressure of carbon dioxide (PaCO2), and pH values in the observation group before and after treatment were higher than those in the control group (P<0.05). The differences in serum NT-pro BNP and PCT levels in the observation group before and after treatment were higher than those in the control group(P<0.05). There was no statistically significant difference in the incidence of adverse reactions between the two groups(P>0.05). Conclusion: Recombinant human brain natriuretic peptide for injection combined with furosemide is effective in the treatment of refractory heart failure, which can effectively improve the patient's cardiac function and blood gas indexes, and regulate serum NT-pro BNP and PCT levels.

406 Efficacy of Intralesional Injection Combined with Surgical Excision and Laser Therapy in the Treatment of Facial Hyperplastic Scarring and the Influence on VSS Score and Pruritus Score

Objective: To study the effect of intralesional injection combined with surgical excision and laser therapy for treating facial hyperplastic scarring on the Vancouver Scar Scale (VSS) score and pruritus score. Methods: From January 2019 to January 2021, 153 patients with facial hyperplastic scar were treated in the Department of Plastic Surgery, Xijing Hospital. The patients were randomly divided into groups A, B, and C by random number table method, with 51 cases in each group, treated with surgical excision, intralesional injection combined with surgical excision, and intralesional injection combined with surgical excision and laser therapy, respectively. Clinical effects, VSS scores, pruritus and pain scores, and adverse reactions were compared among the groups. Results: There were statistically significant differences in total effective rate among the 3 groups (H=22.236, P≤0.001, χ2=9.590, P=0.008). The clinical recovery rate and total effective rate in group C were significantly higher than those in group A and group B (P<0.05), but there was no statistically significant difference between group A and group B (P>0.05). The scores for scar thickness, color, blood vessel distribution, and flexibility, and the total scores in the 3 groups were similar before treatment (P>0.05). After treatment, the above scores in the 3 groups decreased, and group C had significantly lower scores than group A and group B (P<0.05). The difference values of these scores at T1 and T2 were significantly higher in group C than in group A and group B (P<0.05). The 3 groups had similar NRS scores and VAS scores before treatment (P>0.05). After treatment, the NRS scores and VAS scores in the 3 groups decreased significantly, and group C had significantly lower scores than group A and group B (P<0.05). The difference values of NRS scores and VAS scores at T1 and T2 were significantly higher in group C than in group A and group B (P<0.05). Adverse reactions were observed in all groups but were tolerable. The total incidence of adverse reactions in group C was significantly lower than that in group A or group B (P<0.05). Conclusion: Intralesional injection combined with surgical excision and laser therapy is effective in the treatment of facial hyperplastic scars, and can significantly reduce the VSS score and pruritus score.

412 Influences of LncRNA NEAT1 on Proliferation Apoptosis and Invasion of Human Gastric Cancer Cells by Regulating the miR-506-5p/CFL2 Axis

Objective: To investigate the regulation of long non-coding RNA nuclear-enriched abundant transcript 1 (lncRNA NEAT1) on the miR-506-5p/cofilin-2 (CFL2) axis, and its influences on the biological behaviors of gastric cancer cells such as proliferation, apoptosis, migration, and invasion. Methods: BGC-823 cells were cultured in vitro, and lncRNA NEAT1 low expression vector (si-NEAT1) and miR-506-5p inhibitor (miR-506-5p inhibitor) were transfected into BGC-823 cells, which were divided into the si-NC group, si-NEAT1 group, inhibitor NC group, miR-506-5p inhibitor NC group, si-NEAT1+miR-506-5p inhibitor NC group, and si-NEAT1+miR-506-5p inhibitor group. RT-qPCR was performed to determine the expression of lncRNA NEAT1 and miR-506-5p to determine the transfection efficiency. CCK-8 assay was performed to determine proliferation. Flow cytometry was performed to determine apoptosis; a Transwell assay was performed to determine migration and invasion. Western blot was performed to determine the protein expressions of CFL2 and cyclin-dependent kinase 4 (CDK4). Dual-luciferase validation assay was performed to validate the targeting relationship of lncRNA NEAT1, CFL2, and miR-506-5p; nude mouse tumor-bearing experiments were carried out to further verify the targeted regulation of lncRNA NEAT1, miR-506-5p, and CFL2. Results: Compared with the si-NC group, cell proliferation, invasion ability, and CFL2 expression in the si-NEAT1 group were decreased, while apoptosis rate and miR-506-5p expression were increased (P<0.05). Compared with the inhibitor NC group, cell proliferation, invasion ability, and CFL2 expression were increased in the miR-506-5p inhibitor group, while apoptosis rate and miR-506-5p expression were decreased (P<0.05). LncRNA NEAT1 could target and regulate the expression of miR-506-5p, and CFL2 was the target gene of miR-506-5p. Knockdown of miR-506-5p could weaken the anti-proliferation, anti-invasion, and pro-apoptosis effects of lncRNA NEAT1 low expression (P<0.05). miR-506-5p inhibitor attenuated the anti-graft tumor growth effect of lncRNA NEAT1 low expression (P<0.05). Overexpression of CFL2 attenuated the anti-graft tumor growth effect of overexpression of miR-506-5p (P<0.05). Conclusion: LncRNA NEAT1 can affect the biological behaviors such as proliferation, invasion, and apoptosis of human gastric cancer cells by targeting and regulating the miR-506-5p/CFL2 axis.

418 Short-Term Efficacy of Choledochojejunostomy plus T-tube Drainage in the Treatment of Gallbladder Cancer and Its Effects on Median Survival Time and Postoperative Complications

Objective: To explore the short-term efficacy of cholecystectomy plus bile duct T-tube drainage or choledochojejunostomy (CJ) in the treatment of gallbladder cancer (GC) and its effects on the median survival time and postoperative complications. Methods: The clinical data of 121 patients who underwent laparoscopic cholecystectomy (LC) in department of hepatobiliary surgery of the hospital from January 2018 to December 2018 were retrospectively analyzed. According to the adjuvant treatment methods, the patients were divided into two groups: including control group (n=61, LC+bile duct T-tube drainage) and observation group (n=60, LC+CJ). The perioperative indicators, immune stress response, postoperative complications, short-term efficacy, and median survival time were compared between the two groups of patients. Results: The surgical time, postoperative anal first exhaust time, and intraoperative blood loss in observation group were no different from those in control group (P<0.05), but the hospital stay was shorter than that in control group (P<0.05). The changes of immunoglobulin M (IgM), immunoglobulin G (IgG) and C-reactive protein (CRP) before and after surgery in observation group were lower than those in control group (P<0.05). The total incidence rate of complications in observation group at 14 days after surgery was lower than that in control group (P<0.05). There was no statistical significance in the effective rate of treatment between observation group and control group (P>0.05). There was no statistical difference between the two groups in terms of survival rate (P>0.05). Conclusion: Compared with bile duct T-tube drainage, GC+CJ in the treatment of GC can better shorten the postoperative hospital stay and reduce the postoperative complications and immune stress response, but and the median survival time of the two is similar, which can be promoted in clinical practice.

422 Prediction of Early Neurological Deterioration after Thrombolysis in Acute Cerebral Infarction by Continuous TCD Monitoring of Intracranial Pressure

Objective: To analyze the predictive value of continuous monitoring of intracranial pressure (ICP) by transcranial Doppler (TCD) for early neurological deterioration (END) after thrombolysis in acute cerebral infarction. Methods: The clinical data of 109 patients with acute cerebral infarction treated by thrombolytic therapy in our hospital from January 2019 to January 2022 were retrospectively analyzed. According to the occurrence of END, the patients were divided into END group and non-END group respectively. The clinical data of the two groups and the difference of ICP changes during treatment were compared, and the multivariate logistic regression was used to analyze the influencing factors of END. Results: Among the 109 patients, 28 cases (25.69%) had END. 26 cases (92.86%) in END group had ICP increase during TCD monitoring, which was higher than 43.21% in non-END group, and the difference was statistically significant (P<0.05). Logistic regression analysis showed that increased ICP, BMI≥28.0 kg/m2, baseline NIHSS score ≥10, and large area infarction were all indepENDent risk factors for the end of acute cerebral infarction after thrombolysis (P<0.05). Conclusion: The incidence of END is high after thrombolysis in acute cerebral infarction, and the ICP is generally increased in patients with END during treatment. Combined with the factors of ICP increase, obesity, high NIHSS score at baseline, massive infarction and so on, it is expected to provide a reliable reference for the prediction of END in patients.

427 Effect of Double Pouch and Seromuscular Layer Discontinuous Suture (Xu's Three-Needle Method) Embedding Duodenum on Postoperative Complications of Patients Undergoing Radical Gastrectomy for Gastric Cancer

Objective: To analyze the influence of double pouches and seromuscular layer discontinuous suture (Xu's three-needle method) embedding duodenum on postoperative complications of patients undergoing radical gastrectomy. Methods: A total of 108 patients who underwent radical gastrectomy in the First Affiliated Hospital of Anhui Medical University from January 2018 to March 2022 were selected retrospectively. According to the way of embedding duodenum, those who received intermittent suture of seromuscular layer were included in the control group (n=50), and those who received intermittent suture of two kinds of purse strings and seromuscular layer were included in the observation group (n=58). The operation conditions and postoperative complications of the two groups were compared, and the effects of two duodenal embedding methods on the operation process and safety were analyzed. Results: There was no statistical difference between the observation group and the control group in the total operation time and intraoperative blood loss (P>0.05). The treatment time and hospitalization expenses of duodenal stump in the observation group were lower than those in the control group (P<0.05). The incidence of postoperative complications in the observation group was lower than that in the control group (P<0.05). There was no statistical difference in the incidence of postoperative non-surgical complications between the observation group and the control group (P>0.05). Clavien-Dindo grade of postoperative complications in observation group was lower than that in control group (P<0.05). Conclusion: Compared with simple discontinuous suture of seromuscular layer to embed duodenum, double purse string suture can reduce the incidence of postoperative complications without affecting the surgical process, thus significantly reducing the hospitalization expenses of patients.

431 Correlation of EGR1 Expression in Triple-Negative Breast Cancer Tissues with Clinicopathological Features and Prognosis

Objective: To analyze the correlation of early growth response gene 1 (EGR1) expression and clinicopathological features and prognosis in triple negative breast cancer (TNBC). Methods: Fifty patients with TNBC and 53 patients with non-TNBC breast cancer treated in our hospital from January 2016 to March 2018 were enrolled for the research. The expression levels of EGR1 of TNBC cancer tissue, non-TNBC cancer tissue and adjacent normal tissue were compared, and the correlation between EGR1 expression in TNBC cancer tissue and clinicopathological features and prognosis was analyzed. Results: The incidence of EGR1 high expression of TNBC cancer tissue was significantly lower than that of non-TNBC cancer tissue and adjacent normal tissue, and the incidence of EGR1 high expression of non-TNBC cancer tissue was significantly lower than that of adjacent normal tissue (P<0.05). The incidence of EGR1 high expression in cancer tissue of TNBC patients with tumor diameter ≥ 3cm, histological grade G3, metastasis of lymph node, TNM stage Ⅲ and Ki-67 index ≥ 50% was significantly lower than those patients with tumor diameter < 3cm, G2 and G1, without metastasis of lymph node, stage I and < 50%, and the incidence of EGR1 high expression of TNBC cancer tissue with G2 and stage II was significantly lower than those with G1 and stage I (P<0.05). The median survival time of TNBC patients with high EGR1 expression was significantly lower than those with low EGR1 expression (P<0.05).Results: of multivariate Cox regression analysis showed that lymph node metastasis was an independent risk factor for poor prognosis of TNBC patients, and the high expression of EGR1 in cancer tissue was an independent protective factor of poor prognosis (P<0.05). Conclusion: The expression of EGR1 in TNBC cancer tissue was low, and the high expression of EGR1 in cancer tissue was an independent protective factor for the death of patients.

436 The Value of OASIS Score in the Evaluation and Prognosis of Elderly Patients in ICU

Objective: To explore the use of the Oxford Acute Disease Severity score (OASIS) in the outcome prediction of elderly patients in the intensive care unit (ICU). Methods: In this study, 101 elderly critically ill patients treated in the ICU of our hospital from July 2020 to July 2022 were selected, and the patients were divided into 64 cases in the survival group and 37 cases in the death group according to their prognostic survival at 28 days of hospitalization, and the general clinical data, comorbid diseases, relevant laboratory indices, OASIS score, combined comorbidity score (CCS), Charlson Comorbidity Index (CCI), Acute Physiological and Chronic Health Score (APACHE Ⅱ), logistic regression model was used to analyze the relationship between OASIS score and poor prognostic outcome of patients, and ROC curve was used to analyze the value of the above four scoring methods in predicting unfavourable prognostic outcome of the elderly in ICU. Results: The age of patients in the death group was significantly higher than that in the survival group (P<0.05); there was no significant difference in BMI, sex, smoking, alcohol consumption and surgical operation between the death group and the survival group (P>0.05); the ALT, AST, WBC, Hb, NLR, BUN, Scr, blood sugar, PaO2 and PaCO2 of the dead group were higher than those of the survival group, while the ALB and pH values of the dead group were lower than those of the survival group, the difference was statistically significant (P<0.05); there was no significant difference in the blood sodium level between the death group and the survival group (P>0.05); the OASIS score, CCS score, CCI score and APACHE Ⅱ score of patients in the death group were higher than those in the survival group, and the difference was statistically significant (P<0.05); logistic regression model showed that age, OASIS score, CCS score, CCI score, APACHE II score, NLR, Scr, pH, PaO2, PaCO2 and ALB were independent risk factors for death of elderly ICU patients (P<0.05); the AUC values of the area under the curve of OASIS score, CCS score, CCI score and APACHE Ⅱ score predicting the death of elderly ICU patients were 0.870, 0.764, 0.747 and 0.745, respectively. Conclusion: OASIS score is closely related to the death of elderly patients in ICU after treatment, and has high clinical value in the judgment of patients' condition and prognosis.

441 Effects of Sextant Minimally Invasive Percutaneous Pedicle Screw Internal Fixation and Open Pedicle Screw Internal Fixation for Spinal Fractures on Imaging Indicators and Recurrent Fractures

Objective: To analyze the effects of Sextant minimally invasive percutaneous pedicle screw internal fixation and open pedicle screw internal fixation on imaging indicators and recurrent fractures in the treatment of spinal fractures. Methods: A total of 80 patients with spinal fractures treated in our hospital from March 2020 to May 2021 were randomly divided into an observation group (Sextant internal fixation) and a control group (open internal fixation) by simple random number table method, 40 cases in each. The indexes during operation, imaging indexes, VAS, ODI, JOA scores before operation and 3 months after operation, and the incidence of complications were compared between the two groups. Results: During the operation, the incision length, intraoperative blood loss, postoperative drainage volume and operation time of the observation group were significantly lower than those of the control group (all P<0.05). Before surgery, there were no statistical differences in imaging indicators, VAS, ODI and JOA scores between the two groups (all P>0.05). Three months after operation, the imaging indexes (SI, COBB, anterior edge height of injured vertebrae) in both groups showed significant changes compared with those before surgery (all P<0.05),the increased value of SI, decreased value of COBB and increased height of injured vertebral front in the observation group were greater than those in the control group (all P<0.05); VAS, JOA, and ODI in both groups showed significant changes compared with those before surgery (all P<0.05),the decrease of VAS, increase of JOA and decrease of ODI in the observation group were all greater than those in the control group (all P<0.05); Compared with 25.00% of the total incidence of postoperative complications in the control group, the observation group was significantly lower at 7.50% (P<0.05). Conclusion: Compared with open pedicle screw internal fixation, Sextant minimally invasive percutaneous pedicle screw internal fixation in the treatment of spinal fractures can better protect the vertebral body stability and spinal nerve function, and reduce postoperative complications.

Objective: To compare the dosimetry differences between volumetric modulated arc therapy (VMAT) and intensity modulated radiation therapy (IMRT) in the treatment of nasopharyngeal carcinoma. Methods: A total of 114 patients with nasopharyngeal carcinoma undergoing radiotherapy who were treated in the Second Affiliated Hospital of Hainan Medical University from January 2019 to December 2021 were selected as the research subjects, and they were divided into group A and group B according to the random number table method, with 57 cases in each group. Group A received VMAT radiotherapy while group B was given radiotherapy with IMRT. The efficacy, target dose, organ at risk (OAR) dose, treatment efficiency (the number of accelerator monitor units, treatment time), and adverse reactions were compared between the two groups of patients. Results: After treatment, there were no statistical differences in the total treatment remission rate and target dose between the two groups (all P>0.05). The doses of optic nerve, lens, parotid gland and mandible of group A were less than those of group B (all P<0.05). The number of accelerator monitor units and treatment time in group A were less or shorter compared with those in group B (all P<0.05). The differences in the incidence rates of adverse reactions were not statistically significant between the two groups during treatment (P>0.05). Conclusion: VMAT and IMRT have similar effects in the treatment of patients with nasopharyngeal carcinoma, but the application of VMAT can reduce the doses of normal organs around the lesions, and it is beneficial to improving the treatment efficiency and safety.

451 Application of Continuous Full-Thickness Reinforcement Suture Duodenal Stump Protocol in Radical Gastric Cancer Surgery and Analysis of Survival Quality

Objective: To investigate the surgical effect of continuous full-thickness reinforcement and suture of duodenal stump in Bi-type radical gastrectomy for gastric cancer and its effect on postoperative quality of life. Methods: A retrospective research plan was adopted, and 106 gastric cancer patients who were surgically treated in our hospital from March 2017 to June 2021 were selected for study. According to whether the patients adopted continuous full-thickness reinforcement and suture of duodenal stump during the operation, they were divided into studies. 60 cases in the group (Bi II radical operation for gastric cancer + continuous full-thickness reinforcement and suture of duodenal stump), 46 cases in the control group (Bi II radical operation for gastric cancer + conventional purse-string reinforcement and suture of duodenal stump or no reinforcement) suture), and the differences in surgical process indicators, gastrointestinal Visick classification, postoperative recovery, patient quality of life scores and surgical complications were compared between the two groups. Results: The data of operation time, digestive tract reconstruction time, operation bleeding, first postoperative anal exhaust time, first postoperative feeding time, and hospitalization time were statistically analyzed and compared. There was no significant difference between the study group and the control group (P>0.05). There was no significant difference in the distribution of Visick grading between the study group and the control group (P>0.05). The incidence of surgical complications in the study group was 8.33% lower than that in the control group (28.26%), and the difference was statistically significant (P<0.05). The score of nausea and vomiting in the study group was lower than that in the control group, and the difference was statistically significant (P<0.05). Conclusion: The use of continuous full-thickness reinforcement and suture of duodenal stump in Bi-type radical gastrectomy for gastric cancer is beneficial to reduce the surgical complication rate and improve the survival quality of patients after surgery to a certain extent.

457 Analysis of the Relationship between Neutrophil/Lymphocyte Ratio and CAD in Postmenopausal Women

Objective: To investigate the relationship between neutrophil/lymphocyte ratio (NLR) in blood and CAD in postmenopausal women, and to provide a strong basis for clinical treatment of coronary artery lesions in postmenopausal women with CAD. Methods: A total of 260 postmenopausal female CAD patients diagnosed by coronary angiography (CAG) in our hospital from June 2020 to June 2021 were selected as the case group. At the same time, 260 women with suspected CAD during the same period, but whose CAD was ruled out by clinicians and ancillary tests, were selected as a control group. A retrospective analysis was conducted. Results: There was no significant difference in BMI, age at menopause and family history of CAD between the two groups of patients (P>0.05). Patients in the study group with older age, longer menopause, and combined hypertension and diabetes mellitus were statistically analyzed (P<0.05). It can be seen from the laboratory indicators that the white blood cells, NLR, TG and aspartate aminotransferase of the case group were significantly higher than those of the control group, with statistical differences (P<0.05); PCT, HDL, fibrin, and ApoA were significantly lower than those of the control group, there was a statistical difference (P<0.05). There were 119 cases in the severe group, 72 cases in the moderate group, and 69 cases in the mild group of CAD patients. Statistical multi-group comparison was performed by ANOVA, F=3.538, P<0.05, the difference was statistically significant. The difference was not statistically significant (P>0.05) when comparing the mild group with the moderate group by the LSD method between two groups, and the difference was statistically significant (P<0.05) when comparing the mild group with the severe group and the moderate group with the severe group. Logistic regression equation showed that age, hypertension, NLR, TG and fibrin were independent risk factors for coronary heart disease (CAD) in postmenopausal women. Conclusion: The NLR is a convenient and inexpensive blood test that allows clinicians to identify high-risk patients early and can effectively assess and predict the severity of CAD coronary arteries in postmenopausal women.

461 Effect of Continuous Bedside Hemofiltration Combined with Desmophosphate and VitB6 on Patients with Acute Organophosphorus Pesticide Poisoning and the Effect on Myocardial Injury and Inflammatory Symptoms

Objective: To explore the clinical effect of continuous bedside hemofiltration combined with antiphosphoridine and vitamin B6 (VitB6) in the treatment of acute organophosphorus pesticide poisoning. Methods: 92 patients with acute organophosphorus pesticide poisoning treated in our hospital from March 2018 to April 2022 were randomly divided into the observation group (n = 46) and the control group (n = 46), the control group was given continuous bedside hemofiltration treatment, and the observation group was treated with jiefosidine and VitB6 on the basis of the control group. The differences in atropine dosage, mechanical ventilation time and hospitalization time between the two groups were observed, and the differences of myocardial enzyme spectrum and so on before and after treatment were analyzed. Results: The atropine dosage,and hospitalization time in the observation group were significantly less than those in the control group (P<0.05). The difference before and after treatment of cholinesterase (ChE), superoxide dismutase (SOD) and malondialdehyde (MDA) in the observation group were significantly higher than that in the control group (P<0.05). The difference before and after treatment of creatine kinase isoenzyme MB (CK-MB), creatine kinase (CK), troponin (cTnI), aspartate aminotransferase (AST), brain natriuretic peptide (BNP) and myoglobin (MYO) in the observation group were significantly higher than those in the control group (P<0.05). The difference before and after treatment of C-reactive protein (CRP), tumor necrosis factor (TNF-α) and interleukin-8 (IL-8) in the observation group were significantly higher than those in the control group (P<0.05). Conclusion: Continuous bedside hemofiltration combined with desmophosphate and VitB6 has a good clinical effect in the treatment of acute organophosphorus pesticide poisoning, has the advantages of promoting the recovery of myocardial injury and reducing the level of inflammation.

467 Effects of Vitamin D Supplementation Lactoalbumin Supplementation and Resistance Exercise on Nutritional Status and Health Level in Elderly Patients with Sarcopenia Syndrome

Objective: To investigate the effects of vitamin D and lactoalbumin supplementations and resistance exercise on nutritional status and health level of elderly sarcopenia syndrome. Methods: 150 elderly patients with sarcopenia syndrome who were treated in the hospital from January 2020 to January 2022 were selected as the study subjects. According to the random number table method, the study subjects were divided into control group (75 cases) and observation group (75 cases) by the simple randomization. The control group received routine health education and ordinary diet intervention while the observation group was given vitamin D supplementation, lactoalbumin supplementation and resistance exercise intervention, and they were treated for 6 months. The nutritional indicators, upper and lower limb muscle strengths, muscle mass, muscle function, limb motor function and health level were observed before and after intervention in the two groups. Results: After 6 months of intervention, the levels of serum total protein, prealbumin and albumin in both groups were higher than those before intervention, and the differences of the above indicators before and after intervention in observation group were greater than those in control group (P<0.05). The knee extension muscle strength, flexion muscle strength and muscle mass in the two groups of patients were higher than those before intervention, and the differences of the above three indicators before and after intervention were higher in observation group than those in control group (all P<0.05). The muscle function grades in observation group (excellent in 49 cases, medium in 24 cases, poor in 2 cases) were better than those in control group (excellent in 31 cases, medium in 35 cases, poor in 9 cases) ( P<0.05). The scores of simple test for evaluating hand function (STEF) and Berg balance scale (BBS) in both groups were higher than those before intervention, and the differences of STEF score and BBS score before and after intervention in observation group were higher than those in control group (all P<0.05). The scores of physical health, psychological health and social health in the two groups were higher than those before intervention, and the differences of the above scores before and after intervention were higher in observation group compared to control group (all P<0.05). Conclusion: Supplementations of vitamin D and lactoalbumin and resistance exercise can effectively improve the nutritional status, enhance the muscle strength, improve the muscle mass and muscle function, promote the recovery of limb motor function, and improve the overall health status of elderly patients with sarcopenia syndrome.

472 Predictive Value of Coagulation Function and Nutritional Index on Prognosis in Patients with Esophageal Varices Bleeding Undergoing Endoscopic Variceal Ligation

Objective: To analyze the predictive value of coagulation function and nutritional index on prognosis in patients with esophageal varices bleeding undergoing endoscopic variceal ligation(EVL). Methods: One hundred and fifty-two patients with esophageal variceal bleeding who underwent EVL in our department from December 2019 to December 2021 were divided into rebleeding group and non-rebleeding group according to the prognosis. The clinical data were compared between the two groups, and multivariate Logistic regression analysis was used to explore the independent predictors of early rebleeding. The area under the curve(AUC)of receiver operating curve(ROC) was performed to evaluate the predictive value of index for early postoperative rebleeding. Results: All patients successfully completed EVL, and the success rate of short-term hemostasis was 100%. The follow-up result showed that 15 patients(9.9%) had rebleed within 6 weeks after EVL, which occurred 15.3 days(4~36 days) after EVL. The incidences of Child-Pugh grade C, portal vein thrombosis, severe esophageal varices, the total number of ligation rings, prothrombin time(PT), activated partial thromboplastin time and total bilirubin level in rebleeding group were significantly higher than those in non-rebleeding group, and the levels of hemoglobin, platelet, albumin and prognostic nutritional index(PNI)score were significantly lower than that of non-rebleeding group(P<0.05). Multivariate logistic regression analysis showed that portal vein thrombosis, PT and PNI score were the independent predictors of early rebleeding after EVL (P<0.05). The result of ROC curve showed that the preoperative PT and PNI score of all patients had a high predictive value for early postoperative rebleeding, and their AUC (95% CI) were 0.751 (0.569 ~ 0.932) and 0.731 (0.466 ~ 0.974) respectively. Conclusion: PT and PNI score of patients with esophageal variceal bleeding before EVL were the independent predictors of postoperative early rebleeding, and they had a high predictive value for surgical prognosis.

477 Experience of Radical Surgical Thyroid Cancer Treatment by Lumpectomy with Scarless Transaxillary Approach to the Neck

Objective: To explore the safety and efficacy of scarless transaxillary approach to the neck for radical lumpectomy of thyroid cancer. Methods: The medical records of 224 patients with unilateral micropapillary thyroid cancer treated by transaxillary approach lumpectomy and conventional neck incision from December 2020 to May 2022 were collected from the Department of Thyroid Surgery, Affiliated Hospital of Chengde Medical University. They were divided into two groups according to the mode of surgery: the axillary group of 100 cases, 13 males and 87 females, and the neck group of 124 cases, 19 males and 105 females.Comparative analyses were made in terms of operative time, surgical approach, drainage, surgical bleeding, number of lymph nodes dissected in the central region, the average length of stay, postoperative complications, tumor recurrence or metastasis, and cosmetic outcome. Results: The axillary group was younger than the neck group (P<0.01); the operation took longer than the neck group (P<0.01); and there was more postoperative drainage than the neck group (P<0.01). There was no statistically significant difference between the two groups in terms of mean length of stay, the number of lymph nodes dissected in the central region, and mean bleeding (P>0.05). In terms of postoperative complications, there were no statistically significant differences in postoperative hematoma, temporary damage to the recurrent laryngeal nerve, postoperative incision infection, lymphatic leakage, postoperative limitation of neck movement, and tumour recurrence (P>0.05). Statistically significant (P<0.01) in terms of postoperative pain and swallowing discomfort, postoperative numbness in the supraclavicular region, and cosmetic satisfaction. Conclusion: Compared to conventional open neck surgery, axillary approach lumpectomy is a safe and feasible procedure for patients with microscopic papillary thyroid cancer, as it can achieve radical cure of the tumor while preserving the function of the anterior neck area and taking into account the cosmetic effect.

481 Relationship between Different Types of Dyslipidemia and Bone Mineral Density in Physical Examination Population in Changsha

Objective: To explore the relationship between different types of dyslipidemia and bone mineral density in physical examination population in Changsha. Methods: A retrospective analysis was performed on the related data of 527 objects undergoing physical examination in the hospital between May 2019 and May 2022. The basic data (blood lipid, bone mineral density) were collected. The objects with normal blood lipid, high triacylglycerol (TG), high cholesterol (TC) and mixed hyperlipidemia were included in normal group (n=140), high TG group (n=70), high TC group (n=159) and mixed group (n=158). The levels of blood lipid and bone mineral density among the four groups were compared. The correlation between blood lipid and bone mineral density, and the effects of blood lipid on bone mineral density were analyzed. Results: LDL-C level was the highest in high TC group, and low in normal group and high TG group. Except for normal group and high TG group, differences in LDL-C level among the other groups were statistically significant (P<0.05). TC in high TC group and mixed group was significantly higher than that in normal group and high TG group (P<0.05). HDL-C level was the highest in high TG group, and the lowest in mixed group (P<0.05). TG level was the highest in mixed group, and the lowest in normal group and high TC group. Except for normal group and high TC group, differences in TG level among the other groups were statistically significant (P<0.05). The ultrasonic velocity of right calcaneus in high TC group and mixed group were significantly lower than those in normal group and high TG group (P<0.05). The ultrasonic velocity of right calcaneus was negatively correlated with LDL-C, TC, HDL-C and TG (P<0.05). Multivariate linear regression analysis showed that influencing factors of bone mineral density were LDL-C, TC, TG and HDL-C. Conclusion: In physical examination population in Changsha, bone mineral density is lower in those with dyslipidemia. Especially, patients with TC and mixed dyslipidemia need regular screening of bone mineral density in order to detect bone mass changes early and conduct targeted preventive treatment.

486 Efficacy of Recombinant Human Erythropoietin Combined with Levocarnitine in the Treatment of Renal Anemia in Patients with Maintenance Hemodialysis and Its Influence on Inflammatory Factors and Iron Metabolism

Objective: To observe the therapeutic effect of taking combination of recombinant human erythropoietin (rHuEPO) and levocarnitine as treatment regimen on renal anemia caused by maintenance hemodialysis, and to record the influence on serum inflammatory factors and iron metabolism. Methods: A total of 113 patients with renal anemia who underwent hemodialysis in the hospital from January 2019 to January 2021 were selected as the study subjects. 57 cases were included in observation group and 56 cases in control group by adopting non-randomized controlled trial design. Both groups were treated with rHuEPO, folic acid and other drugs, and the observation group was added with levocarnitine after dialysis. After 3 months of treatment, the therapeutic effect of renal anemia of both groups was recorded. Serum inflammatory factors [interleukin-6 (IL-6), C-reactive protein (hs-CRP), tumor necrosis factor α (TNF-α)], serum iron metabolism parameters [serum transferrin receptor (sTfR), serum ferritin (SF), transferrin saturation (TAST)] and oxidative stress indicators [malondialdehyde (MDA), superoxide dismutase (SOD)] were compared before treatment and 3 months after treatment. The adverse reactions (gastrointestinal reactions, abnormal liver function, elevated blood pressure, fever) were recorded within 3 months of treatment. Results: The total effective rate of renal anemia treatment of 82.46% in the observation group after 3 months of treatment was higher than 66.07% in the control group (P<0.05). Compared with before treatment, serum inflammatory factors (IL-6, hs-CRP, TNF-α), some iron metabolism parameter (sTfR) and some oxidative stress indicator (MDA) were reduced after 3 months of treatment in both groups, and the reductions were greater in the observation group compared with those in the control group (P<0.05). After 3 months of treatment, the levels of some serum iron metabolism parameters (SF, TAST) and some oxidative stress indicator (SOD) were increased in the two group compared to before treatment, and the increases in the observation group were greater in comparison with the control group (P<0.05). Within 3 months of treatment, the total incidence rates of adverse reactions in the two groups were 14.04% and 26.79% respectively (P>0.05). Conclusion: The use of rHuEPO and other drugs combined with levocarnitine for maintenance hemodialysis-induced renal anemia not only has considerable clinical therapeutic effect, but also reduces inflammatory response, regulate iron metabolism and control oxidative response. In addition, it has safety and deserves clinical promotion.

491 Analysis of the Effect of Calcium Hydroxide Combined with Chlorhexidine on the Pain Level and Disinfection Effect of Root Canal Disinfection in Patients

Objective: To compare the influence of calcium hydroxide combined with chlorhexidine and calcium hydroxide alone on the disinfection effect and postoperative pain degree in patients with root canal disinfection, and to explore the clinical application effects of different drugs. Methods: A total of 102 patients with root canal disinfection who were treated in the hospital from Jun. 2019 Dec. 2022 were selected as the research subjects, and were simply randomized into combined group (n=51) and conventional group (n=51) by using the random number table method. The conventional group adopted calcium hydroxide alone for root canal disinfection, while the combined group received calcium hydroxide combined with chlorhexidine for root canal disinfection. At 7 days after medicine sealing, the disinfection effect, Mohdsulong pain grading, and incidence rates of complications were compared between the two groups. The colony counts of aerobic bacteria and anaerobic bacteria in root canal before and after root canal preparation and at 7 days after medicine sealing and the sealing time of the two groups of patients were compared. Results: The effective rate of disinfection of the combined group was 98.04%, which was higher than 80.39% of the conventional group (P<0.05). There were no statistical differences in the counts of aerobic and anaerobic bacteria between the two groups before and after root canal preparation (P>0.05). At 7 days after medicine sealing, the counts of aerobic bacteria and anaerobic bacteria in the root canal of the combined group were less than those of the conventional group (P<0.05). The pain grading of the combined group was better compared with that of the conventional group (P<0.05), and the sealing time was shorter than that in the conventional group (P<0.05). The incidence rate of gingival swelling and total incidence rate of complications were lower in the combined group than those in the conventional group (P<0.05). Conclusion: Compared with calcium hydroxide alone, the application of calcium hydroxide combined with chlorhexidine for root canal disinfection can achieve better disinfection effect, and can more effectively inhibit the bacterial reproduction, relieve the pain of root canal treatment, and shorten the root canal treatment time, with higher safety.

496 Exploring the Curative Efficacy and Mechanism of Thalidomide Combined with GemOx Regimen in Treating B-NHL Patients

Objective: To explore the efficacy and mechanism of thalidomide combined with GemOx regimen (oxaliplatin + gemcitabine) in patients with relapsed and refractory B cell non-Hodgkin's lymphoma (B-NHL). Methods: Forty patients with relapsed refractory B-NHL were randomly divided into two groups. 20 patients in the treatment group were treated with thalidomide plus GemOx regimen, and 20 patients in the control group were treated with GemOx regimen. In addition, 20 healthy subjects in the same period were selected as the normal group. The levels of TNF-α and IL-6 in peripheral blood were detected by Elisa, and TH17 and NK cell ratio in peripheral blood were detected by Flow cytometry. Results: Compared with the control group, the total effective rate of the treatment group was higher, and the incidence of thrombocytopenia, leucopenia, anemia, gastrointestinal reaction, liver and kidney dysfunction was lower (P<0.05). The levels of TNF-α, IL-6, TH17 and NK cells were significantly higher in treatment group and control group than those in control group (P<0.05). Compared with before treatment, the levels of TNF-α and IL-6 in treatment group and control group were significantly lower, the ratio of Th17 cells was higher (P<0.05) , the ratio of NK cells was lower in control group, and the ratio of NK cells was higher in treatment group (P<0.05). Compared with the control group, the levels of TNF-α, IL-6, Th17 and NK cells in the treatment group were lower than those in the control group (P<0.05). Before treatment, there was no difference in TNF-α, IL-6, Th17 and NK cell ratio between treatment group and control group (P>0.05). Conclusion: Thalidomide combined with GemOx regimen is expected to improve the efficacy of treatment in B-NHL patients, decrease serum levels of TNF-α and IL-6, and increase Th17 and NK cell ratio.

500 Effects of Remifentanil Compounded with Propofol on Cerebral Protection and Inflammatory Factors in Patients Undergoing Aneurysm Clipping

Objective: To study the effect of remifentanil compounded with propofol on brain protection and inflammatory factors in patients undergoing aneurysm clipping. Methods: A total of 105 patients who came to our hospital for cerebral aneurysm clipping from January 2019 to December 2021 were selected as the research subjects, and were retrospectively analyzed. According to the different anesthesia induction methods,the observation group (remifentanil complex propofol, 55 cases) and the control group (conventional fentanyl anesthesia, 50 cases) were compared between the two groups of patients before anesthesia induction (T0), 1 h after induction (T1), after surgery (T2), and 6 h after surgery (T3), and cerebral oxygen metabolism and inflammatory factors at 5 time points 12 h (T4) after operation. The recovery and cognitive status of the two groups of patients were compared. Results: The results of repeated measures ANOVA showed that the anesthesia induction method, time, and the interaction of both affected CERO2, CjvO2, and Da-jvO2 (P<0.05). CERO2, CjvO2, and Da-jvO2 will change significantly with the trend of the time point. CERO2 and CjvO2 overall show a trend of rising first and then decreasing. CERO2 gradually decreases from the time point T1, and Da-jvO2 mainly shows the time point trend of decreasing, and gradually decreases from the time point T0. The effects of the anesthesia induction method, time, and the interaction effect on S100 β and NSE were all statistically significant (P<0.05). S100 β and NSE increased with time, and the trend was more obvious than the control group (P<0.05); the effect of time on IL-6 was statistically significant (P<0.05), but slightly increased over time; and the interaction of anesthesia induction method was not statistically significant on IL-6 (P>0.05). The observed force recovery time, eye opening time and extubation time were lower than that of the control group, statistically significant (P<0.05), but there was no difference in respiratory recovery time between the two groups (P>0.05). The effect of anesthesia induction method and time on MMSE and OAAS score were statistically significant (P<0.05). The interaction between method and time had little effect on MMSE scores (P>0.05) and a statistically significant effect on OAAS scores (P<0.05), and MMSE and OAAS scores in both groups, and the observation group was significant compared with the control group (P<0.05). Conclusion: Remifentanil compounded with propofol can significantly protect the brain of patients undergoing aneurysm clipping, and can increase the inflammatory response, but the recovery time, consciousness and cognitive function recovery are more superior.

507 Effects of Rituximab on Visual Acuity and Intestinal Flora in Patients with Neuromyelitis Optica

Objective: To investigate the effect of rituximab on vision and intestinal flora in patients with neuromyelitis optica. Methods: A total of 127 patients with neuromyelitis optica admitted to our hospital from March 2018 to March 2021 were selected for research, and they were divided into a study group (n=64) and a control group (n=63) by a simple random method using a random number table. Both groups were given hormone pulse therapy in the acute phase, the study group was given rituximab therapy in the remission phase, and the control group was given cyclophosphamide intravenous infusion therapy in the remission phase. The Disability Status Scale (EDSS) was used to evaluate the treatment effect. The visual acuity and intestinal flora distribution before and after 8 weeks of treatment were compared between the two groups, and the incidence of adverse reactions during the treatment was compared. The study was followed up for 1 year to compare the recurrence of the two groups. Results: After treatment, the function of EDSS subitems in the two groups recovered significantly compared with that before treatment (P<0.05). The differences of cone system function, brain stem function, vesicorectal function and visual function scores in the study group before and after treatment were higher than those in the control group (P<0.05). The worst eye, the best eye and the average logMAR corrected visual acuity after treatment in the two groups. The difference before and after treatment in the study group was higher than that in the control group (P<0.05), and the recurrence rate was lower than that in the control group (P<0.05). After treatment, the Chaol index and Shannon index were (233.15±15.26) and (3.36±0.68), respectively, which were higher than those of the control group (P<0.05), and there was no significant difference in adverse reactions between the two groups (P>0.05). Conclusion: Rituximab has obvious effects on improving vision and intestinal flora structure in patients with neuromyelitis optica, and has the value of popularization and application.

511 Effectiveness of Domestic Sodium Hyaluronate Combined with Polyethylene Glycol Eye Drops in the Treatment of Xerophthalmia after Strabismus Surgery

Objective: To explore the effectiveness of the combination of two eye drops (domestic sodium hyaluronate plus polyethylene glycol) on xerophthalmia after strabismus surgery. Methods: 89 patients with xerophthalmia after strabismus surgery (89 affected eyes) in the hospital were recruited between January 2021 and February 2022, and they were randomly assigned to regular group (receiving domestic sodium hyaluronate eye drops, n=44, 44 eyes) and combined group (given eye drops of domestic sodium hyaluronate combined with polyethylene glycol, n=45, 45 eyes). The xerophthalmia symptom score, tear film function parameters, clinical efficacy and changes in interleukin-8 (IL-8), tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) were compared before and after 1 month of treatment. Dry Eye-related Quality-of-life Score (DEQS) was used for the evaluation of quality of life. Results: After treatment, symptom scores and corneal fluorescence staining (Fluorescein, FL) were significantly lower in both groups, while basal tear secretion test (Schirmer I test, SIt) and tear breakup time (BUT) were elevated, and the difference between symptom scores, FL, SIt and BUT before and after treatment in the combined group were ( (4.52±0.35), (2.60±0.93), (6.02±1.55) mm, (5.64±1.73) s, respectively, which were higher than those of the regular group (2.20±0.35), (2.02±1.03), (4.81±1.36) mm, (3.82±1.60) s (P<0.05); 95.56% of the total clinical efficiency and efficacy of the combined group were higher than those of the regular group. The total clinical efficiency and efficacy of 95.56% in the combined group was significantly better than that of 81.82% in the regular group (P<0.05); the levels of IL-8, IL-1β and TNF-α decreased in both groups after treatment, and the differences of IL-8, IL-1β and TNF-α levels in the combined group before and after treatment were (3.06±0.60) ng/L, (20.60±7.66) ng/L and (2.29±0.86) μg/L, respectively. (7.66) ng/L, (2.29±0.86) μg/L, respectively, which were higher than (2.11±0.65) ng/L, (11.06±8.31) ng/L, (1.38±0.93) μg/L in the regular group (all P<0.05); the DEQS scores in the combined group after 2 weeks and 1 month of treatment were (40.17±1.71), ( 32.44±1.16) points, both lower than (44.38±1.89) and (37.56±1.45) points in the regular group (both P<0.05), and the DEQS scores in both groups showed a downward trend after treatment, but the downward trend in the combined group was more obvious than that in the regular group (P<0.05). Conclusion: Domestic sodium hyaluronate combined with polyethylene glycol eye drops can improve xerophthalmia symptom and tear film function after strabismus surgery, with significant efficacy, and can relieve postoperative inflammatory response and improve quality of life.

516 Effects of Selective α-2 Agonists on Patients Undergoing Carotid Endarterectomy and Their Cerebral Protective Effects

Objective: To investigate the effects of selective α-2 receptor agonists dexmedetomidine on inflammation, oxidative stress and brain functions in patients undergoing carotid endarterectomy. Methods: A total of 60 patients who underwent carotid endarterectomy in our hospital from January 2018 to December 2021 were randomly divided into control group and observation group. The observation group was given dexmedetomidine anesthesia, and the control group was given routine anesthesia. The neurological function of the two groups were compared. Serum inflammatory factor TNFα, ICAM-1 and oxidative stress factor MDA, IL-6, S100β, neural function markers BDNF, TGF-β1,NSE, were detected. Results: The decreases of TNFα, IL-6, ICAM-1, MDA, S100β and NSE in the observation group was significantly higher than the control group(P<0.05). MMSE score, BDNF, and TGF-β1 in the observation group were significantly higher than in the control group(P<0.05). Conclusion: The selective α-2 receptor agonists dexmedetomidine could inhibit inflammation and oxidative stress, and protect brain functions.
