1 Influences of Paeoniflorin on LPS-Induced Apoptosis and Osteogenic Differentiation of Periodontal Ligament Stem Cells by Regulating LKB1-AMPK Signaling Pathway

Objective: To investigate the influences of paeoniflorin (PF) on lipopolysaccharide (LPS)-induced apoptosis and osteogenic differentiation of periodontal ligament stem cells (PDLSCs) by regulating liver kinase B1 (LKB1)-adenylate-activated protein kinase (AMPK) signaling pathway. Methods: PDLSCs in logarithmic growth phase were collected and separated into control group, LPS group (10μg/mL), LPS+PF low concentration (PF-L, 10μg/mL LPS+5μmol/L PF) group, LPS+PF medium concentration (PF-M, 10μg/mL LPS+10μmol/L PF) group, LPS+PF high concentration (PF-H, 10μg/mL LPS+20μmol/L PF) group, and LPS+PF-H +LKB1 inhibitor (radicicol, 10μg/mL LPS+20μmol/L PF+10μmol/L radicicol) group. ELISA was used to detect the levels of inflammatory factors [interleukin (IL)-1β, tumor necrosis factor-α (TNF-α), IL-6] in cells; alizarin red staining was used to observe the changes of mineralized nodules; alkaline phosphatase (ALP) was used to detect the cell osteogenic differentiation; cell apoptosis was detected by flow cytometry; the expression levels of osteogenic genes (OPN, RUNX2 and OCN) were detected by qRT-PCR; Western blot was used to detect the expression levels of LKB1-AMPK pathway protein and apoptosis protein (caspase-1). Results: Compared with the control group, the mineralized nodules were reduced in the LPS group, the apoptosis rate, the expression of caspase-1, the contents of TNF-α, IL-1β and IL-6 increased obviously, the level of ALP, the expression of OPN, RUNX2 and OCN, and the phosphorylation levels of LKB1 and AMPK decreased obviously (P<0.05); compared with the LPS group, the mineralized nodules were reduced in the LPS+PF-L group, the LPS+PF-M group, and the LPS+PF-H group, the apoptosis rate, the expression of caspase-1, the contents of TNF-α, IL-1β and IL-6 decreased obviously, the level of ALP, the expression of OPN, RUNX2 and OCN, and the phosphorylation levels of LKB1 and AMPK increased obviously, all were dose-dependent (P<0.05); however, radicicol reversed the effects of PF-H on LPS-induced apoptosis and osteogenic differentiation of PDLSCs (P<0.05). Conclusion: PF inhibits LPS-induced apoptosis of PDLSCs and promotes osteogenic differentiation by up-regulating LKB1-AMPK signaling pathway.

7 Effects of BMP8B on the Differentiation and Proliferation of Human Periodontal Ligament Stem Cells

Objective: To investigate the effects of bone morphogenetic protein 8B (BMP8B) on the proliferation and differentiation of human periodontal ligament stem cells (PDLSCs). Methods: Human PDLSCs were isolated and cultured in vitro, and immunofluorescence staining was used to detect PDLSCs cell protein stromal cell antigen 1 (STRO-1) and CD146 protein. The PDLSCs cells were divided into control group, negative transfection group, BMP8B silencing group and BMP8B overexpression group. Flow cytometry, alizarin red staining and oil red O staining were used to identify PDLSCs. Cell proliferation was detected by CCK-8 method.The PDLSCs was osteogenesis inducted, cell alkaline phosphatase (ALP) activity was detected, mineralized nodules in osteoblasts were detected by alizarin red staining, the expression levels of BMP8B, Osterix, OCN, and Runx2 mRNA in cells were detected by real-time fluorescent quantitative polymerase chain reaction (qRT-PCR), and the cell protein expression was detected by western blot (WB). Results: Under the microscope, PDLSCs cells were mononuclear, polygonal or spindle shaped, with osteogenic potential and lipogenic potential. Compared with control group and negative transfection group, cell proliferation, ALP activity, alizarin red staining, mRNA expression of BMP8B, Osterix, OCN, Runx2 and CyclinD1, CDK2, Osterix, OCN, Runx2 were significantly decreased in BMP8B silencing group (P<0.05). Compared with the control group and the negative transfection group, all indexes of BMP8B overexpression group were significantly increased (P<0.05). There was no significant difference between control group and negative transfection group (P>0.05). Conclusion: BMP8B affects the proliferation and osteogenic differentiation of PDLSCs cells, suggesting that the mechanism of BMP8B in osteogenic differentiation may be a new direction for the treatment of periodontitis.

13 Expression of lncRNA MNX1-AS1 in Non-Small Cell Lung Cancer and the Effect of Regulation of PI3K/AKT Pathway on the Biological Behavior of Cancer Cells

Objective: To investigate the biological function and possible mechanism of long non-coding RNA (lncRNA) MNX1-AS1 (lncRNA MNX1-AS1) in non-small cell lung cancer (NSCLC). Methods: The cancer tissue and adjacent tissue samples of 75 NSCLC patients who underwent surgical treatment in our hospital from June 2017 to March 2019 were taken. RT-qPCR method was performed to detect the expression level of lncRNA MNX1-AS1 in the samples, the correlation between the expression of lncRNA MNX1-AS1 and the clinicopathological characteristics and prognosis of NSCLC was analyzed; the NSCLC cell line PC-9 cells were cultured in vitro and transfected with si-MNX1-AS1 (si-MNX1-AS1 group) and si-NC (si-NC group), respectively. the proliferation of PC-9 cells in each group was detected by MTT assay and EdU assay; Transwell chamber assay was performed to detect cell migration and invasion; Western blot assay was performed to detect the expression of PI3K/AKT pathway-related proteins. Results: The expression level of lncRNA MNX1-AS1 was obviously higher in NSCLC cancer tissues and cell lines (P<0.05). The expression of lncRNA MNX1-AS1 was associated with TNM stage and lymphatic metastasis (P<0.05), Kaplan-Meier survival analysis showed that the 3-year cumulative survival rate of patients in the high expression group of lncRNA MNX1-AS1 was obviously lower than that in the low expression group (42.11% vs 81.08%, χ2=11.132, P=0.001); compared with the si-NC group, the proliferation, migration and invasion abilities of PC-9 cells in the si-MNX1-AS1 group and the protein expression levels of p-PI3K, p-AKT and p-mTOR were obviously decreased (P<0.05). Conclusion: The expression level of lncRNA MNX1-AS1 is increased in NSCLC, which may affect the proliferation, migration and invasion of NSCLC cells by activating the PI3K/AKT pathway, and promote the occurrence and development of NSCLC.

18 The Mechanism of LncRNA HCP5 Regulating miR-27b-3p/H2AFZ Axis on the Proliferation Invasion and Migration of Oral Squamous Cell Carcinoma

Objective: To investigate the influences of long non-coding RNA (LncRNA) human leukocyte antigen complex P5 (HCP5) proliferation, invasion and migration of oral squamous cell carcinoma (OSCC) by regulating the microRNA (miR)-27b-3p/H2A histone family member Z (H2AFZ) axis. Methods: The cancer tissues and adjacent paracancerous tissues, OSCC cell lines (Tca-811, SCC-9, SCC-25, HN4) and human normal oral keratin-forming cells (hNOK) were collected from 48 OSCC patients treated surgically at our hospital from 2021 to 2022, and the LncRNA HCP5, miR-27b-3p, H2AFZ mRNA expression levels; logarithmic growth phase Tca-811 cells were divided into control group, small interfering RNA negative control (si NC) group, interfering HCP5 expression (si HCP5) group, si HCP5+ inhibitor negative control (inhibitor NC) group, si HCP5+ miR-27b-3p inhibitor (miR-27b-3p) group. Double luciferase was used to verify the targeting relationship between LncRNA HCP5 and miR-27b-3p, miR-27b-3p and H2AFZ; qRT-PCR to detect the expression levels of LncRNA HCP5, miR-27b-3p and H2AFZ mRNA; flow cytometry and MTT to detect apoptosis and proliferation changes of Tca-811 cells. Transwell assay was performed to detect changes in cell invasion and migration; Western blot to detect expression of cyclinD1, cleaved caspase-3, matrix metalloproteinase 2 and H2AFZ protein. Results: Compared with hNOK cells and adjacent tissues, the expressions of LncRNA HCP5 and H2AFZ mRNA in OSCC cells and tissues were increased, and the expression of miR-27b-3p was decreased (P<0.05). There was a targeting relationship between miR-27b-3p with LncRNA HCP5, H2AFZ. Compared with the si NC group and the control group, the apoptosis rate, cleaved caspase-3 and miR-27b-3p expressions in the si HCP5 group were higher than those in the si NC group and the control group (P<0.05), the OD570 (24 h, 48 h), the number of invasive and migrating cells and the expressions of CyclinD1, MMP-2, HCP5, H2AFZ were lower (P<0.05); inhibiting the expression of miR-27b-3p reversed the inhibitory effect of interfering with HCP5 on the malignant behavior of Tca-811 cells (P<0.05). Conclusion: Interfering with LncRNA HCP5 can inhibit the proliferation, invasion and migration of OSCC by targeting up-regulation of miR-27b-3p and inhibiting the expression of H2AFZ.

25 Influences of Safflower Yellow on Cardiomyocyte Pyroptosis and Inflammatory Injury in Rats with Myocardial Infarction by Regulating NF-κB/NLRP3 Signaling Pathway

Objective: To explore the influences of Carthamin yellow (CY) on the cardiomyocyte pyroptosis and inflammatory injury in myocardial infarction (MI) rats by regulating nuclear factor-κB (NF-κB)/NOD-like receptor protein 3 (NLRP3) signaling pathway. Methods: Rat MI model was established by ligation of left anterior descending coronary artery. The 72 SPF male rats were separated into: control group, sham group, model group, low-dose CY group (CY-L group, 20 mg/kg), high-dose CY group (CY-H group, 40 mg/kg), and high-dose CY+NF-κB activator group (CY-H+LPS group, CY 40 mg/kg+LPS 10mg/kg), 12 cases per group. After modeling, intraperitoneal injection was administered, once a day, for a total of 28 days. After the administration, the rats in each group were examined by echocardiography to detect the changes of cardiac function. 2,3,5-Triphenyltetrazolium chloride (TTC)-Evans blue (EB) method and immunofluorescence method were applied to detect myocardial infarction area and cardiomyocyte pyroptosis ratio in each group. Histopathological changes of myocardial tissue were observed by hematoxylin-eosin (HE) and Masson staining. The kits were used to measure the levels of interleukin (IL)-1β, IL-18 and lactate dehydrogenase (LDH) in rat myocardium. Western Blot was performed to detect the protein expression of NF-κB, NLRP3, cleaved-caspase1 and gastrin D-N (GSDMD-N) in myocardial tissue. Results: Compared with the sham group, the model group had severe myocardial tissue pathological damage and decreased cardiac function, the myocardial infarction area, the ratio of cardiomyocyte pyroptosis, the levels of IL-1β, IL-18, LDH, and the protein expression levels of NF-κB, NLRP3, GSDMD-N and cleaved-caspase1 in myocardial tissue were obviously increased (P<0.05). Compared with the model group, the cardiac function of the rats in the CY-L group and the CY-H group was obviously improved, the myocardial tissue damage was alleviated, and the cell pyroptosis was reduced, the levels of inflammatory factors IL-1β, IL-18 and LDH were obviously decreased, and the expression of NF-κB/NLRP3 signaling pathway-related proteins was obviously decreased (P<0.05). LPS attenuated the inhibitory effects of high-dose CY on cardiomyocyte pyroptosis and inflammatory injury in MI rats (P<0.05). Conclusion: CY may reduce the pyroptosis and inflammatory injury of cardiomyocytes in MI rats by inhibiting the NF-κB/NLRP3 signaling pathway.

31 Experimental Study on Reducing High Glucose-Induced Injury in H9c2 Cardiomyocytes by Modulating PI3K/Akt/mTOR Signaling Pathway with Zicui Tongmai Decoction

Objective: To investigate the possible signaling pathways (PI3K-Akt-mTOR) involved in autophagy and apoptosis of H9c2 cardiomyocytes induced by high glucose, and to investigate the mechanism of action of Zicui Tongmai Decoction in inhibiting cardiomyocyte apoptosis. Methods: SD rats were randomly divided into blank plasma group and Zicui Tongmai Decoction group, with 10 rats in each group. The corresponding medicated plasma was prepared by gavage for 7 days,the drug concentration of Zicui Tongmai Decoction group was 4.4g/mL and the gavage volume was 10mL·kg-1·d-1,the blank plasma group was gavaged with the same dose of sterilized ultrapure water (the administration volume was 10mL/kg). H9c2 myocardial cells were divided into normal group, blank plasma group and drug containing plasma group, fetal bovine serum, blank plasma and drug containing plasma were added respectively. A concentration gradient of 5%, 10% and 15% was set for each group. The proliferation of H9c2 cardiomyocytes in each group was compared by CCK-8 method, the cell survival rate was calculated, and the drug containing plasma was screened. H9c2 cardiomyocytes induced by high glucose were randomly divided into three groups: model group, Zicui Tongmai Decoction medicated plasma group and engliejing group, and additionally there was normal group. Expression of LC3B in H9c2 cardiomyocytes was detected 24h after administration of IF, and the expression of PI3K Akt mTOR signal pathway protein and apoptosis related protein caspase-3 was detected by Western blot. Results: Drug containing plasma and blank plasma have no significant effect on cell proliferation at the concentration level of 10%, which is used as the intervention concentration of drug containing plasma experiment. The expression of LC3B Ⅱ/LC3BⅠ decreased significantly compared with the normal group(P<0.05),but the protein expression of PI3K, P-Akt, mTOR and Caspase-3 increased compared with the normal group(P<0.05). The expression of LC3B Ⅱ/LC3BⅠin Zicui Tongmai Decoction medicated plasma group and engliejing group increased significantly compared with the model group(P<0.05),but the protein expression of PI3K,P-Akt,mTOR and Caspase-3 in Zicui Tongmai Decoction medicated plasma group and engliejing group decreased compared with the model group(P<0.05). There was no significant difference in expression of LC3BⅡ/LC3BⅠ and protein expression of PI3K,P-Akt,mTOR and Caspase-3 between Zicui Tongmai Decoction medicated plasma group and engliejing group (P>0.05). Conclusion: Zicui Tongmai Decoction can activate autophagy and inhibit apoptosis of cardiomyocytes, which may be related to the inhibition of PI3K/p-Akt / mTOR signaling pathway.

37 The Mechanism of Vascular Secretory Factor Rspondin3 Regulating Wntβ-catenin Signaling Pathway to Promote Tissue Repair in Acute Lung Injury

Objective: To investigate the expression of vascular secretory factor Rspondin3 through Wnt/ β- catenin signaling pathway in the molecular mechanism of acute lung injury rat model. Methods: The acute lung injury model of SD rats was established. The rats were divided into control group (n=10), model group (n=10), NC group (n=10) and Rspondin3 high group (n=10). The LPS induced acute lung injury model of rats was established in the model group, NC group and Rspondin3 group. After the NC group and Rspondin3 were established, NC plasmid and Rspondin3 over-expression plasmid were injected through tail vein respectively. The control group and model group were given the same amount of normal saline. After 2 weeks, HE staining was used to detect the level of pathological changes in the lung tissues of rats in each group. ELISA was used to detect the changes of interleukin-1 (interleukin-1, IL-1), interleukin-2 (interleukin-4, IL-2) and interleukin-6 (interleukin-8, IL-6) levels in the lung tissues of rats in each group. Western blot was used to detect Wnt-3a and β- catenin protein expression level. The ultrastructural changes of lung tissue in each group were detected by transmission electron microscopy. Results: Compared with the control group, the model group and NC group showed obvious lung tissue injury, and the degree of lung tissue pathological injury in Rspondin3 group was relatively reduced. ELISA results showed that compared with the control group, the expression levels of inflammatory factors IL-1, IL-2 and IL-6 in the lung tissue of model group, NC group and Rspondin3 group were increased, and compared with the model group and NC group, the expression levels of inflammatory factors IL-1, IL-2 and IL-6 in the lung tissue of Rspondin3 group were decreased. The difference was statistically significant (P<0.01). Western blot results showed that Wnt-3a and β-catenin protein expression levels were reduced in the model group and NC group compared to the control group, and Wnt-3a and β-catenin protein expression was increased and reduced in the lung tissue of the Rspondin3 group compared to the model group and NC group, with statistically significant differences (P<0.01). Transmission electron microscopy showed that compared with the control group, the lung tissue structure of the model group and the NC group was loose, the nucleus disintegrated, cell was incomplete, and the cells were vacuolated, while the ultrastructure of the lung group of the Rspondin3 group showed that the cell structure damage was reduced, the nucleus disintegrated and the cell was relatively complete. Conclusion: Overexpression of Rspondin3 has a protective effect on the lung tissue of rats with acute lung injury, and the mechanism may be related to the activation of the activation of the Wnt-3a/ β-catenin signaling pathway.

42 Therapeutic Effect of Theaflavins Modulation of AMPK/SIRT1/NF-κB Signaling Pathway in Rats with Knee Osteoarthritis

Objective: To investigate the therapeutic effect of theaflavins on knee osteoarthritis (KOA) rats by regulating AMP-activated protein kinase (AMPK)/silent information regulator 1 (SIRT1)/nuclear factor-κB (NF- κB) signaling pathway. Methods: 50μL (0.01mg/μL) sodium iodoacetate (MIA) was injected into the right knee joint of 63 SD rats, another 13 rats were only injected with normal saline into the knee joint as blank group, two weeks later, 3 rats were randomly selected from KOA rats and blank group, and HE staining, Mankin and lequesne MG scores were performed to identify whether the KOA model was successfully prepared; the remaining successful modeling rats were randomly separated into model group, low-dose theaflavin group (20mg/kg theaflavin), middle-dose theaflavin group (40mg/kg theaflavin), and high-dose theaflavin group (80mg/kg theaflavin), celecoxib group (18mg/kg positive drug celecoxib), and inhibitor group (80mg/kg theaflavins + 0.2mg/kg AMPK inhibitor Compound C). Low-dose, medium-dose and high-dose theaflavin groups and celecoxib group were given theaflavin and celecoxib by intragastric administration; the blank group and model group were intragastrically administered with the same volume of distilled water, the inhibitor group was administered the corresponding dose of Compound C into the tail vein based on the corresponding dose of theaflavin, once a day, for 4 consecutive weeks. On the 15th day (one day after modeling), the 29th day (the 14th day of administration) and the 43rd day (the 28th day of administration), the rat mechanical pain threshold and rat joint width were detected; the behavioral changes of the rats were observed, and the lequesne MG score was performed; the pathological changes of cartilage tissue were observed by HE staining and Mankin score was performed; the kits were used to measure serum inflammatory factors (IL-1β, TNF-α and COMP) and oxidative stress markers (SOD and MDA) levels; Western Blot was used to detect the AMPK/SIRT1/NF-κB signaling pathway-related protein expression. Results: Compared with the blank group, the cell arrangement of the KOA rats cell arrangement was disordered, the surface was rough, the fissures were uniform, the arrangement was uneven, the number of chondrocytes was reduced, the moisture marks were not obvious, and the Mankin, lequesne MG scores were obviously increased. Compared with the blank group, on the 15th day, the pain threshold of rats in the model group, theaflavin low, medium and high dose groups, the celecoxib group and the inhibitor group was obviously decreased, and the joint width was obviously increased (P<0.05); compared with the blank group, the cartilage layer of the rats in the model group was thinner, the soles of the rats were swollen, the HE staining was uneven, the cell arrangement was disordered, the surface was rough, the fissures were uniform, the arrangement was uneven, the number of chondrocytes was reduced, and the moisture marks were not obvious, the Mankin and lequesne MG scores, serum IL-1β, TNF-α, COMP and MDA levels, and cartilage tissue p-NF-κB p65/NF-κB p65 level were obviously increased (P<0.05), the serum SOD level and cartilage tissue p-AMPKα/AMPKα, SIRT1 expression levels were obviously decreased (P<0.05); compared with the model group, the pain threshold of the theaflavin low, medium and high dose groups and the celecoxib group was obviously increased (P<0.05), the width of the joint was reduced, the swelling of the soles of the rats' feet was reduced, the cartilage surface was slightly damaged, and the staining was more uniform, chondrocytes were visible, the Mankin and lequesne MG scores, serum IL-1β, TNF-α, COMP and MDA levels, and cartilage tissue p-NF-κB p65/NF-κB p65 level were obviously decreased, the serum SOD level and cartilage tissue p-AMPKα/AMPKα, SIRT1 expression levels were obviously increased (P<0.05); AMPK inhibitors attenuate the therapeutic effect of theaflavins in KOA rats. Conclusion: Theaflavins have therapeutic effects on KOA rats by regulating AMPK/SIRT1/NF-κB signaling pathway.

49 Effect of Erythropoietin on Proliferation and Apoptosis of Neural Stem Cells Induced by Hypoxia

Objective: To investigate the influences of erythropoietin (EPO) on the proliferation and apoptosis of neural stem cells induced by hypoxia by regulating hypoxia-inducible factor-1α (HIF-1α)/extracellular signal-regulated kinase (ERK) signaling pathway. Methods: Neural stem cells were divided into NC group, Model group, low-dose EPO group (EPO-L group, 5 U/mL), and high-dose EPO group (EPO-H group, 25 U/mL) , YC-1 group, and EPO-H+YC-1 (HIF-1α inhibitor) group (25 U/mL +10μmol/L). Western blot was used to detect the protein expression of Cyclin D1, Bcl-2/adenovirus interfering protein 3 (BNIP3), Cleaved Caspase-3, HIF-1α, and p-ERK1/2. Results: Compared with NC group, the proportion of Brd U positive cells, and the protein expression of Cyclin D1, HIF-1α and p-ERK1/2 in Model group decreased, and the apoptosis rate, the protein expression of BNIP3 and Cleaved Caspase-3 increased (P<0.05); compared with the Model group, the proportion of Brd U positive cells, and the protein expression of Cyclin D1, HIF-1α and p-ERK1/2 in EPO-L group and EPO-H group increased, and the apoptosis rate, the protein expression of BNIP3 and Cleaved Caspase-3 decreased (P<0.05), the corresponding indexes in YC-1 group were opposite to those above (P<0.05); YC-1 weakened the promotion of high dose EPO on the proliferation of neural stem cells induced by hypoxia and the inhibition of apoptosis. Conclusion: EPO may promote hypoxia-induced neural stem cell proliferation and inhibit cell apoptosis by activating the HIF-1α/ERK pathway.

53 Panax Notoginseng Saponins Inhibit the Proliferation and Promote Apoptosis of Human Rheumatoid Arthritis Synovial Fibroblast Cell Line

Objective: To investigate the effect and possible mechanism of Panax notoginseng saponins on the proliferation, apoptosis, migration and invasion of human rheumatoid arthritis synovial fibroblasts. Methods: MH7A cells were intervened with different doses (0.5, 1.0, 2.0 mg/mL) of Panax notoginseng saponins for 24h, and then cell proliferation, apoptosis, migration and invasion were detected by CCK-8, flow cytometry, scratch assay and Transwell, respectively. The expression of proteins (Ki-67, PCNA, E-cadherin and N-cadherin) were detected by Western blotting, and the expression of miR-335-5p was detected by qRT-PCR. The miR-335-5p mimic was transfected into MH7A cells, or the miR-335-5p inhibitor was transfected into MH7A cells and then intervened with 2.0mg/mL Panax notoginseng saponins for 24h, and then the same methods as above were used to detect cell proliferation, migration and invasion. Results: Compared with the control group, different doses of Panax notoginseng saponins decreased the A value, scratch healing rate, invasion number and the expression of Ki-67, PCNA and N-cadherin protein in MH7A cells (P<0.05), but increased the apoptosis rate and E-cadherin protein expression(P<0.05), and promoted the expression of miR-335-5p in cells (P<0.05). After up-regulating miR-335-5p, the A value, wound healing rate, invasion number and the expression of Ki-67, PCNA and N-cadherin protein in MH7A cells were all decreased (P<0.05), but the apoptosis rate and E-cadherin protein expression was decreased (P<0.05). Down-regulation of miR-335-5p reversed the effects of Panax notoginseng saponins on the proliferation, apoptosis, migration and invasion of MH7A cells. Conclusion: Panax notoginseng saponins may inhibit the proliferation, migration and invasion of rheumatoid arthritis synovial fibroblasts MH7A and promote cell apoptosis by up-regulating miR-335-5p.

59 Experimental Study on the Effect of miR-24-3p Targeting BMP8B on the Function of Osteoblasts

Objective: To investigate the targeting relationship between miR-24-3p and bone morphogenetic protein 8B (BMP8B), and to analyze their effects on osteoblast function. Methods: Human periodontal ligament stem cells (hPDLSCs) were isolated and cultured in vitro and induced to differentiate into osteoblasts. The osteoblasts were identified by alkaline phosphatase (ALP) staining and alizarin red staining; the osteoblasts were divided into blank control group, negative control group (inhibitor-NC group), silencing miR-24-3p expression group (miR-24-3p-inhibitor group), and co-transfection group (miR-24-3p-inhibitor +BMP8B-siRNA group). Real-time fluorescent quantitative PCR method was used to detect the levels of miR-24-3p and BMP8B mRNA; CCK-8 method and flow cytometer were used to detect the proliferation and apoptosis of bone cells in each component; ALP activity detection kit was used to detect ALP activity; Western blotting was used to detect the levels of BMP8B protein and functional activity-related proteins (PICP, BGP, OPN) in each component of bone cells. The TargetScan database and the dual luciferase reporter gene experiment were used to predict and verify the targeting relationship between miR-24-3p and BMP8B respectively. Results: Compared with the blank control group and inhibitor-NC group, the osteoblast proliferation inhibition rate and apoptosis rate in the miR-24-3p-inhibitor group were decreased, ALP activity and expression of PICP, BGP, OPN were increased (P<0.05); compared with miR-24-3p-inhibitor group, the osteoblast proliferation inhibition rate and apoptosis rate in the miR-24-3p-inhibitor+BMP8B-siRNA group were increased, ALP activity and expression of PICP, BGP, OPN were decreased (P<0.05). The dual luciferase reporter gene experiment confirmed that miR-24-3p targeted regulation of BMP8B. Conclusion: Inhibition of miR-24-3p may promote the proliferation and differentiation of osteoblasts and inhibit cell apoptosis by targeting up-regulation of BMP8B expression.

65 Probing the Effect of Syringin on High Glucose-Induced MPC5 Damage in Mouse Podocyte Based on the mTOR/p70S6K Signaling Pathway

Objective: To investigate the effects of syringin on the injury of mouse podocyte MPC5 induced by high glucose and mammalian target of rapamycin (mTOR)/p70 ribosomal protein S6 kinase (p70S6K) signal pathway. Methods: MPC5 of mouse podocytes were divided into control group (5.5 mmol/L glucose), high glucose induced group (30mmol/L glucose), syringin low concentration group (2.5μmol/L syringin+30mmol/L glucose), syringin medium concentration group (5μmol/L syringin+30mmol/L glucose), syringin high concentration group (10μmol/L syringin+30mmol/L glucose). Each group added the corresponding reagent or drug of the above concentration to RPMI 1640 medium and cultured for 72h. The absorbance of MPC5 cells in each group was measured by microplate reader, and the cell viability was calculated; the apoptosis of MPC5 cells in each group was determined by flow cytometry; the cytoskeleton morphology of MPC5 cells in each group was observed by phalloidin staining; the expression of mTOR, p70S6K messenger RNA (mRNA) and protein in MPC5 cells of each group were determined by fluorescence quantitative PCR and Western blot. Results: The cytoskeleton morphology of MPC5 cells in the control group was normal, the long axis of the cells was linearly distributed, and the fluorescence staining was obvious; compared with the control group, the cytoskeleton of MPC5 cells in the high glucose induced group was broken, the cells were atrophied, the fluorescence staining was dim, the absorbance value and cell viability were significantly decreased, and the apoptosis rate, mTOR, p70S6K mRNA and protein expression levels were significantly increased (P<0.05); compared with the high glucose induced group, the cytoskeleton structure of MPC5 cells in the syringin low, medium and high concentration groups recovered clearly, the fluorescence intensity gradually became bright, the peduncle tended to be obvious, the absorbance value and cell viability increased in turn, and the apoptosis rate, mTOR, p70S6K mRNA and protein expression levels were decreased in turn (P<0.05). Conclusion: Syringin has a protective effect on MPC5 cells damage induced by high glucose, which can significantly promote MPC5 cells proliferation and inhibit its apoptosis. Its mechanism may be related to the inhibition of syringin on the activation of MPC5 cells mTORC1/p70S6K pathway under high glucose state.

70 Effects of Fluvastatin Sodium on the Proliferation and Apoptosis of Colorectal Cancer SW620 Cells by Down-Regulating the Expression of LncRNA PTPRG-AS1

Objective: To explore the effect and possible mechanism of fluvastatin sodium on the proliferation and apoptosis of colorectal cancer SW620 cells. Methods: SW620 cells and normal colorectal epithelial FHC cells were cultured in vitro, and the expression of lncRNA PTPRG-AS1 in the cells was detected by qRT-PCR. SW620 cells were divided into control group, fluvastatin sodium-low, medium and high groups, si-PTPRG-AS1 group, si-NC group, fluvastatin sodium+pcDNA-PTPRG-AS1 group, fluvastatin sodium+pcDNA group. CCK-8 method, clone formation assay and flow cytometry were used to detect cell viability, clone formation number, apoptosis rate, and Western blot was used to detect the expression of apoptosis-related proteins (cleaved-caspase3, cleaved-caspase9). Results: The expression of lncRNA PTPRG-AS1 in colorectal cancer SW620 cells was higher than that in FHC cells (4.38±0.31 vs 1.00±0.00, P<0.05). The cell viability and the number of colonies formed in the fluvastatin sodium-low, medium and high groups were lower than those in the control group(P<0.05), but the apoptosis rate and protein (cleaved-caspase3, cleaved-caspase9) expression were higher than those in the control group(P<0.05), and the expression of lncRNA PTPRG-AS1 was lower than that of the control group(P<0.05). The cell viability and the number of colonies formed in the si-PTPRG-AS1 group were lower than those in the si-NC group(P<0.05), but the apoptosis rate and protein (cleaved-caspase3, cleaved-caspase9) expression were higher than those in the si-NC group(P<0.05). The cell viability and the number of colonies formed in the fluvastatin sodium+pcDNA-PTPRG-AS1 group were higher than those in the fluvastatin sodium+pcDNA group(P<0.05), but the apoptosis rate and protein (cleaved-caspase3, cleaved-caspase9) expression were lower than those in the fluvastatin sodium+pcDNA group(P<0.05). Conclusion: Fluvastatin sodium may inhibit the proliferation and promote apoptosis of colorectal cancer SW620 cells by down-regulating the expression of lncRNA PTPRG-AS1.

76 Ameliorative Effects and Mechanism of Action of Atropine in Rats with Form Deprivation Myopia Model

Objective: To investigate the improvement effect of atropine (ATR) on form deprivation myopia model rats by regulating Wnt/β-catenin (β-catenin) signaling pathway and related mechanisms. Methods: Sixty SD rats were randomly grouped into control (NC) group, form deprivation myopia model (Model) group, ATR group (3mg/kg), TAK-715 group (10mg/kg Wnt/β- catenin pathway activator TAK-715), and ATR+TAK-715 group (3mg/kg ATR+10mg/kg TAK-715), with 12 per group. The right eye of NC group and Model group was subconjunctively injected with equal amount of normal saline. After 4 weeks, the axial length and diopter of the right eye of the rats in each group were recorded; the morphology of sclera was observed by HE staining; ELISA kits were used to detect the levels of serum interleukin (IL)-6, IL-1β and tumor necrosis factor-α (TNF-α); RT-qPCR was used to detect the mRNA expression of scleral tissue transforming growth factor β1 (TGF-β1), matrix metalloproteinase 2 (MMP-2), and matrix metalloproteinase inhibitor-2 (TIMP-2); Western blot was used to detect the expression of Wnt/β-catenin pathway proteins in sclera tissue. Results: The sclera of the rats in the NC group was normal; the sclera in the Model group became thinner, and the collagen fibers were sparsely arranged and disordered; the thickness of sclera and the diameter of collagen fibers increased in ATR group; the sclera thickness of the rats in the ATR+TAK-715 group decreased, and the fibers were arranged disorderly. Compared with the normal sclera in the NC group, the axial length, refraction, the levels of IL-1β, IL-6 and TNF-α, the mRNA level of MMP-2, and the protein levels of Wnt and MMP-2 in the Model group were obviously increased (P<0.05), the mRNA levels of TGF-β1 and TIMP-2, and the protein levels of TGF-β1, TIMP-2, and p-β-catenin/β-catenin were obviously decreased (P<0.05); compared with the Model group, the axial length, refraction, the levels of IL-1β, IL-6 and TNF-α, the mRNA level of MMP-2, and the protein levels of Wnt and MMP-2 in the ATR group were obviously decreased (P<0.05), the mRNA levels of TGF-β1 and TIMP-2, and the protein levels of TGF-β1, TIMP-2, and p-β-catenin/β-catenin were obviously increased (P<0.05), the corresponding index of TAK-715 group showed the opposite trends (P<0.05); compared with the ATR group, the axial length, refraction, the levels of IL-1β, IL-6 and TNF-α, the mRNA level of MMP-2, and the protein levels of Wnt and MMP-2 in the ATR+TAK-715 group were obviously increased (P<0.05), the mRNA levels of TGF-β1 and TIMP-2, and the protein levels of TGF-β1, TIMP-2, and p-β-catenin/β-catenin were obviously decreased (P<0.05). Conclusion: ATR may reduce inflammatory response and inhibit extracellular matrix remodeling by regulating the Wnt/β-catenin pathway, thereby improving form deprivation myopia in rats.

81 Study on the Mechanism of Effect of Protocatechuic Acid on Inflammatory Response and Angiogenesis in Rats with Endometriosis

Objective: To observe the effect and mechanism of protocatechuic acid (PA) on angiogenesis in rat endometriosis (EMS). Methods: Fifty-four female SD rats in estrus were selected to establish the EMS disease model by endometrial autograft and the rats were randomly divided into blank group, model group, PA (high, medium and low) dose group and gestrinone group. PA and gestrinone groups were administered gavage once a day. The model group was given the same volume of normal saline for 28 days. The rats in the blank group were fed normally. The ectopic tissue volume of rats was measured, and the tumor necrosis factor in the peritoneal fluid of rats in each group was detected by enzyme-linked immunosorbent assay (ELISA) α (TNF-α), Interleukin 6 (IL-6) and interleukin 1 β (IL-1 β). The levels of angiopoietin (ANG)-1 and Ang-2, the contents of vascular endothelial growth factor (VEGF), matrix metalloproteinase 2 (MMP-2) and matrix metalloproteinase 9 (MMP-9) in serum and tissues, and the protein expression of total extracellular signal regulated kinase 1/2(t-ERK1/2) and phosphorylated total extracellular signal regulated kinase 1/2(p-ERK1/2) in tissues were detected by Western blot. Results: Compared with the blank group, the lesion volume increased at 14d and 42d, and the inflammatory factor TNF-α, IL-6, IL-1 β and Ang-1 and Ang-2, VEGF, MMP-2 and MMP-9, p-ERK1/2, VEGF, MMP-2 and MMP-9 in endometriosis were all significantly increased in the model group rats (P<0.05). The changes of the above indexes were negatively regulated in a concentration dependent manner in the low-dose group, the middle dose group and the high-dose group of protocatechuic acid (P<0.05). Conclusion: Protocatechuic acid can reduce wound injury and inhibit inflammatory reaction and angiogenesis in rats with endometriosis. The underlying mechanism may be related to inactivation of ERK / VEGF / MMP pathway.

Objective: To study the effect of microRNA-122-5p on the expression, invasion and migration of MMP-9 in human colorectal cancer SW480 cells induced by TPA. Methods: Bioinformatics was used to screen colorectal cancer differential genes and predict their target miRNA. SW480 cells were treated with TPA (100 nM), and gene and protein expression levels were detected by RT-qPCR and Western blot; gelatin zymography was used to detect MMP-9 cell exocytosis; Transwell was used to detect cell invasion and migration ability. Results: MMP-9 was highly expressed in colon cancer tissues, and the upstream target gene miR-122-5P was screened; TPA induced a time-dependent upregulation of MMP-9 mRNA expression (P<0.01) and a downregulation of miR-122-5p expression (P<0.01); overexpression of miR-122-5p significantly downregulated TPA-induced MMP-9 mRNA and protein expression (P<0.05), the extracellular secretion of MMP-9 was reduced, and the invasion and migration ability of cells were significantly decreased (P<0.01). Conclusion: miR-122-5p may induce MMP-9 expression by regulating TPA and thus inhibit the invasive and migratory ability of colorectal cancer SW480 cells, providing a new idea to overcome the metastasis study of colorectal cancer.

91 Systematic Review of Fecal Microbiota Transplantation for Functional Constipation Based on Randomized Controlled Trials

Objective: To evaluate the clinical efficacy of FMT combined with drugs in the treatment of functional constipation and to provide evidence-based medical basis for clinical treatment. Methods: A systematic literature search of six domestic and international databases was conducted to screen randomised controlled trials (RCTs) or observational trials of FMT for functional constipation, with screening, data extraction and bias assessment carried out independently by two researchers. The Cochrane Risk of Bias Assessment Tool and StataSE 15.0 software were used for quality assessment and statistical analysis. Results: Eleven RCT studies with a total of 958 patients were eventually included.Meta-analysis showed that FMT combined with pharmacological treatment significantly improved clinical outcomes compared with pharmacological treatment alone for functional constipation [RR=1.39,95%CI(1.23,1.58),P<0.001]; significantly improved stool properties (BSFS score) [WMD=0.81,95%CI(0.53,1.08),P<0.001]; reduced quality of life in constipation (PAC-QOL score) [WMD=-13.47,95%CI(-15.24,-11.7),P<0.001]; reduced Wexner constipation score WMD=-2.78,95%CI(-3.02,-2.53),P<0.001]; while significantly improving gastrointestinal function [WMD=-30.99,95%CI(-38.26,-23.72),P<0.001]; and regulating the balance of serum gastrointestinal hormone levels [WMD=3.78,95%CI(0.30,7.26), P<0.001]. The difference in the regulation of intestinal flora was statistically significant in reducing the level of pathogenic bacteria as well as increasing beneficial bacteria; the difference in the occurrence of adverse events [RR=0.49, 95% CI (0.20,1.25), P<0.001] was not statistically significant. Conclusion: FMT combined with medication has better efficacy in the treatment of functional constipation than conventional treatment alone.

97 Analysis of the Relationship between CYP2C19 Gene Polymorphism and Valproic Acid Metabolism in Han Chinese Patients with Mental Illness in Hebei Province

Objective: To analyze the CYP2C19 gene polymorphism and explore the relationship between CYP2C19 gene polymorphism and valproic acid metabolism in Han Chinese patients with mental illness in Hebei Province. Methods: The frequencies of alleles, genotypes and metabolic types of CYP2C19 in 464 Han Chinese patients with mental illness in Hebei Province were analysed, and the differences in men and women were compared separately. Fifty-two patients with schizophrenia treated with olanzapine and valproic acid were selected from the above cases.The blood concentration of valproic acid was determined by high performance liquid chromatography tandem mass spectrometry.One-way ANOVA was used to analyze the differences in valproic acid blood concentrations and blood concentrations per unit dose of valproic acid between different CYP2C19 metabolic types, and multi-factor linear regression was applied to analyze the factors influencing valproic acid blood concentrations. Results: The frequencies of CYP2C19* 1, CYP2C19* 2, CYP2C19* 3 and CYP2C19* 17 in 464 patients with mental illness were 66.60%, 27.26%, 4.74% and 1.40% respectively. A total of 9 genotypes were detected, in descending order of frequency: CYP2C19*1/*1, CYP2C19*1/*2, (CYP2C19* 2/* 2, CYP2C19*1/*3), CYP2C19*1/*17, CYP2C19*2/*3, CYP2C19*3/*3, (CYP2C19*2/*17, CYP2C19*3/*17); the frequencies of distribution of superfast, fast, medium and slow metabolic phenotypes were 2.37%, 42.46%, 46.33% and 8.84%.There was no statistically significant difference in the frequency of distribution of alleles, genotypes and metabolic phenotypes between male and female (χ2=1.668, P=0.644; χ2=13.487, P=0.096; χ2=6.113, P=0.106). Differences in valproic acid blood concentrations between metabolic forms were not statistically significant (F=0.146, P=0.864; F=0.006, P=0.994). The linear regression equation was VPA=43.62 + 38.03 dose - 0.428 age ( F=7.689, P<0.001). Conclusion: The CYP2C19 gene is polymorphic in Han Chinese psychiatric patients in Hebei Province, and the differences in the distribution of alleles, genotypes and metabolic phenotypes between male and female were not statistically significant. Gene polymorphisms of CYP2C19 did not significantly affect the metabolism of valproic acid.

103 Predictive Value of Multiple Inflammatory Indices for Cardiovascular Complications in Children with Kawasaki Disease

Objective: To explore the predictive value of multiple inflammatory indices for cardiovascular complications in children with Kawasaki disease. Methods: A total of 127 children with KD admitted to our hospital from January 2020 to May 2022 were continuously collected as the research subjects, and they were divided into high MII group and low MII group according to the median of MII. Demographic data such as gender and age were collected using self-made general information questionnaire, and clinical data such as fever time, conjunctival hyperemia, and lymphadenopathy were obtained through electronic medical record system. Children were followed up for 12 months with cardiovascular complications as the outcome. Results: There was no significant difference in gender, fever time, and lymph node enlargement rate between the high MII group and the low MII group (P>0.05). The age and conjunctival hyperemia rate in the high MII group were higher than those in the low MII group, and the differences were statistically significant (P<0.05). The results of the K-M survival curve analysis showed that the median cardiovascular complication rates were 87.50% and 90.63% in the high MII-1 and high MII-2 groups, respectively, and 76.19% and 74.60% in the low MII-1 and low MII-2 groups, respectively, with statistically significant differences between the two groups (χ2=10.680, 10.860 , P=0.001, 0.001). Multivariate analysis showed that MII-1 (OR=2.247, P<0.001) and MII-2 (OR=2.134, P<0.001) were independent influencing factors of cardiovascular complications in children with KD. The results of ROC analysis showed that when the values of MII-1 and MII-2 were 784.64 and 21876.76, respectively, the maximum predictive performance of cardiovascular complications in children with KD after 12 months of follow-up was achieved, and the areas under the curve were 0.680 and 0.726, respectively. Conclusion: Multiple inflammatory indices have certain clinical value in predicting cardiovascular complications in children with Kawasaki disease, and are worthy of clinical application

Objective: To analyze the coagulation function, levels of plasma hemoglobin (Hb) and D-dimer (D-D) in patients with diabetic nephropathy (DN) and their relationship with thrombosis of arteriovenous fistula (AVF). Methods: A total of 99 patients with DN confirmed in the hospital were enrolled between February 2020 and February 2022. According to presence or absence of AVF thrombosis, they were divided into thrombosis group (n=34) and simple DN group (n=65). The clinical data in both groups were recorded. The prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time (TT) and fibrinogen (FIB) were detected by freezing method. The level of antithrombase-III (AT-III) and D-D were detected by chromogenic substrates assay. The international normalized ratio (INR) was the ISI power of PT to normal control PT ratio. The level of D-D was detected by immunoturbidimetry. The level of Hb was detected by colorimetry. The predictive value of the above indexes for AVF thrombosis was analyzed by ROC curves. Results: The difference in age, gender, body index (BMI), disease course and blood urea nitrogen between thrombosis group and simple DN group was not statistically significant (P>0.05). The dialysis time and usage time of AVF in simple DN group were significantly shorter than those in thrombosis group (P<0.05), and levels of AT-III, PT, APTT, TT, FIB, INR, Hb and D-D were significantly lower than those in thrombosis group (P<0.05). AUC values of AT-III, PT, APTT, TT, INR, FIB, Hb and D-D for predicting AVF thrombosis were 0.771, 0.788, 0.748, 0.778, 0.673, 0.778, 0.771 and 0.745, respectively. Combined test showed an AUC of 0.963, a sensitivity of 82.35% and a specificity of 81.54%. Conclusion: Coagulation function indexes, Hb and D-D can predict AVF thrombosis in hemodialysis patients.

112 Diagnostic Value of HRCT Signs of Pure Ground Glass Nodules in Non-Lepidic Invasive Lung Adenocarcinoma

Objective: To evaluate the diagnostic value of HRCT signs of pure ground glass nodules (pGGN) in non-lepidic invasive lung adenocarcinoma (IAC), and to find the optimal imaging indicator. Methods: The clinical data of 85 patients with IAC confirmed by surgical pathology and HRCT imaging data of 86 pGGNs were retrospectively analyzed. According to the pathology, they were divided into lepidic dominant group (48 cases, 47 patients) and non-lepidic dominant group (38 cases, 38 patients). The HRCT signs such as mean CT value, maximum diameter, burrs sign, abnormal vascular sign and abnormal bronchial sign were compared between the two groups. The ROC curve was analyzed with non-lepidic dominant adenocarcinoma as the dependent variable, and the area under the curve (AUC) was used to evaluate the diagnositic efficacy of each imaging indicator. Results: The mean CT value and maximum diameter of non-lepidic dominant IAC were higher than lepidic dominant IAC. The incidence of abnormal vascular sign, abnormal bronchial sign and burrs sign of non-lepidic dominant IAC was higher than lepidic dominant IAC (P<0.05). ROC curve analysis showed that the area under the curve of non-lepidic dominant IAC was 0.877,the efficiency was 0.895,the sensitivity was 0.729, and its diagnostic efficiency was higher than other indicators. Conclusion: The mean CT value has a high diagnositic value for non-lepidic dominant IAC with pGGN and is the optimal imaging indicator.

115 Relationship of Serum miR-181a-5p Expression with Th17/Treg Immune Homeostasis in Patients with Acute Pancreatitis

Objective: To investigate the expression of serum microRNA-181a-5p (miR-181a-5p) in patients with acute pancreatitis (AP) and the relationship with T helper cell 17/T regulatory cell (Th17/Treg) immune homeostasis. Methods: 110 patients with AP admitted to our hospital from January 2020 to December 2021 (AP group) were selected and divided into the mild AP group (n=41), the moderately severe AP group (n=37)and the severe AP group (n=32) according to the severity of their conditions, and 58 healthy people with physical examination (control group) were also selected for the same period. Peripheral blood interleukin-6 (IL-6), IL-10, IL-17 and transforming growth factor-β1 (TGF-β1) levels were measured by enzyme-linked immunosorbent assay, peripheral blood Th17 and Treg percentages were measured by flow cytometry, and serum miR-181a-5p expression was measured by qPCR. Correlation of serum miR-181a-5p expression with Th17/Treg in AP patients was analyzed using Pearson/Spearman correlation. Results: Compared with the control group, serum miR-181a-5p expression and peripheral blood Treg percentage decreased, and Th17 percentage and Th17/Treg increased in AP group (all P<0.001). Compared with the control group, the peripheral blood levels of IL-6 and IL-17 in the AP group were increased, and the levels of IL-10 and TGF-β1 were reduced (all P<0.001). Serum miR-181a-5p expression and peripheral blood IL-10, TGF-β1, and Treg percentage were sequentially lower and peripheral blood IL-6, IL-17, Th17 percentage, and Th17/Treg were sequentially higher in the mild, moderately severe, and severe AP groups (all P<0.001). Pearson/Spearman correlation analysis showed that serum miR-181a-5p expression in AP patients was negatively correlated with IL-6, IL-17, Th17, and Th17/Treg (r/rs=-0.688, -0.652, -0.782, -0.825, all P<0.001) and positively correlated with IL-10, TGF-β1, and Treg were positively correlated (r/rs=0.743, 0.667, 0.739, all P<0.001). Conclusion: Serum miR-181a-5p is lowly expressed in AP patients with Th17/Treg immune imbalance. miR-181a-5p may be involved in the development of AP through Th17/Treg immune imbalance.

120 The Value of Serum Apelin-13 Combined with Rotterdam-CT Score in Assessing the Condition and Prognosis of Patients with Traumatic Brain Injury

Objective: To investigate the value of serum Angiotensin II receptor-like 1 endogenous ligand 13 (Apelin-13) combined with Rotterdam-CT score in assessing the condition and prognosis of patients with traumatic brain injury (TBI). Methods: 153 patients with TBI admitted to our hospital from March 2020 to March 2022 (TBI group) were selected and divided into 62 cases in the mild TBI group, 52 cases in the moderate TBI group and 39 cases in the severe TBI group according to the Glasgow Coma Scale (GCS), and 42 cases in the poor prognosis group and 111 cases in the good prognosis group according to the Glasgow Prognosis Scale (GOS), and another 49 healthy individuals undergoing physical examination (control group) were selected during the same period. Rotterdam-CT scores were calculated from cranial CT examinations in patients with TBI, and serum Apelin-13 levels were measured by enzyme-linked immunosorbent assay. Spearman correlation was used to analyze the correlation between serum Apelin-13 levels and Rotterdam-CT scores and GCS scores in patients with TBI, multi-factor logistic regression was used to analyze the factors influencing poor prognosis in patients with TBI, and ROC curves were used to analyze the value of serum Apelin-13 levels and Rotterdam-CT scores in assessing poor prognosis in patients with TBI. Results: The serum level of Apelin-13 in the TBI group was lower than that in the control group, and the difference was statistically significant (P<0.05). The serum Apelin-13 level in the mild TBI group, moderate TBI group and severe TBI group decreased sequentially, and the Rotterdam-CT score increased sequentially, and the difference was statistically significant (P<0.05). GCS scores in patients with TBI were positively correlated with serum Apelin-13 levels and negatively correlated with Rotterdam-CT scores (rs=0.654, -0.675, all P<0.001). Basal pool abnormalities, midline shift ≥5 mm, and increased Rotterdam-CT score were independent risk factors for poor prognosis in patients with TBI, and increased GCS score and elevated Apelin-13 were independent protective factors [3.460 (1.166 to 10.262), 3.993 (1.569 to 10.163), 2.416 (1.169~4.992), 0.681 (0.536 to 0.864), 0.871 (0.805 to 0.941)]. ROC curve analysis showed that the area under the curve for serum Apelin-13 levels and Rotterdam-CT scores alone and in combination to assess poor prognosis in patients with TBI were 0.797, 0.808 and 0.896, respectively, with sensitivities of 59.52%, 95.24% and 92.86% and specificities of 59.52%, 95.24% and 92.86%. Conclusion: Decreased serum Apelin-13 levels and increased Rotterdam-CT scores are strongly associated with exacerbation and poor prognosis in patients with TBI, and serum Apelin-13 combined with Rotterdam-CT scores are of high value in assessing poor prognosis in patients with TBI.

126 Expression of CircKIF4A and miR-942-5p in Ovarian Cancer Tissues and Their Relationship with Clinicopathological Characteristics and Prognosis of Patients

Objective: To investigate the expressions of miR-942-5p and circKIF4A in ovarian cancer tissues and their relationship with the prognosis of patients. Methods: A total of 154 ovarian cancer patients who came to our hospital for surgical treatment from January 2015 to January 2017 were collected as the research subjects. The cancer tissues and adjacent tissues of the surgical patients were collected, and the relationship between the expressions of miR-942-5p and circKIF4A with clinicopathological characteristics and prognosis was retrospectively analyzed. The levels of Mir-942-5p and circKIF4A were detected by real-time fluorescence quantitative PCR (QRT-PCR); Kaplan-Meier method was used to analyze the relationship between the expression of different miR-942-5p and circKIF4A and the survival time of patients; multivariate Cox regression risk model was used to analyze the prognostic factors. Results: The expression of circKIF4A was higher in cancer tissues of ovarian cancer patients (4.27±0.33) compared with paraneoplastic tissues (1.00±0.05), and miR-942-5p expression was lower (0.63±0.12) compared with paraneoplastic tissues (1.01±0.19), with a negative correlation between the two expressions (r=-0.312, P<0.05); CircKIF4A and miR-942-5p expression in cancer tissues of ovarian cancer patients was not associated with age, pathological type, WHO histological grade, tumor diameter and degree of infiltration, but with degree of differentiation, lymph node metastasis, TNM stage, tumor grade and presence of ascites (P<0.05); Kaplan-Meier analysis showed that the 5-year cumulative survival rate of patients in the circKIF4A high expression group was significantly lower at 26.92% (21/78) than that of patients in the low expression group at 47.37% (36/76), with a statistically significant difference (Log rank χ2=6.651, P=0.010); The 5-year cumulative survival rate of patients in the miR-942-5p high expression group was significantly higher at 46.25% (37/80) than that of patients in the low expression group at 27.03% (20/74), with a statistically significant difference (Log rank χ2=9.978, P=0.000); Cox results showed that lymph node metastasis, TNM stage, degree of differentiation, high expression of circKIF4A, and miR-942-5p low expression were all risk factors affecting the survival status of ovarian cancer patients. Conclusion: The expression of miR-942-5p was low and circKIF4A was high in cancer tissues of ovarian cancer patients, and the expression of both was negatively correlated with the disease development and prognosis of ovarian cancer patients.

131 Effects of Combined Orthodontic Periodontal Treatment on the Expression of Periodontal Pathogens and Inflammatory Factors in Periodontitis Patients with Malocclusion

Objective: To investigate the expression of periodontal pathogens and inflammatory factors in patients with periodontitis associated with malocclusion by orthodontic combined periodontal treatment. Methods: All the enrolled patients were 84 patients with malocclusion periodontitis who were admitted to our hospital from January 2017 to January 2021. According to random number table method, they were divided into control group (n=42, orthodontic treatment alone) and combined group (n=42, orthodontic periodontal treatment combined). The total effective rate was compared between the two groups. Compared the two groups before treatment, after treatment of periodontal related index [plaque index (PLI), periodontal pocket probing depth (PD), attachment level (AL), gingival bleeding index (SBI)], periodontal pathogens [gum porphyrin (Pg) bacterium, fossett steiner bacteria (Tf), teeth tartar treponema (Td), intermediate platts bacteria (Pi)], periodontal tissue inflammation Cytokines [tumor necrosis factor -α (TNF-α), interleukin-6 (IL-6), interleukin-10 (IL-10)] levels. Results: The total effective rate in combined group was 92.86%, which was higher than 76.19% in control group (P<0.05). Before treatment, there were no significant differences in periodontal related indicators, pathogenic bacteria and periodontitis factors in gingival crevicular fluid between 2 groups (P>0.05). After treatment, PLI, PD, SBI, AL, Pg, Tf, Td, Pi, TNF-α and IL-6 in both groups were decreased (P<0.05), while IL-10 was increased (P<0.05), and the difference between before and after treatment in combined group was greater than that in control group (P<0.05). Conclusion: Orthodontic periodontitis combined with malocclusion can improve the periodontal state of patients, reduce the level of pathogenic bacteria and reduce the inflammatory state.

136 Effects of Deep Muscle Stimulation Combined with Flexi-Bar Therapy on Psoas Endurance, Pain and Prognosis in Patients with Nonspecific Low Back Pain

Objective: To investigate the effect of deep muscle stimulation combined with Flexi-bar therapy on psoas endurance, pain and prognosis in patients with nonspecific low back pain (NLBP). Methods: A total of 100 patients with NLBP in our hospital from June 2019 to June 2021 were selected and randomly divided into groups, 50 cases in each group. The control group was given routine rehabilitation training, and the observation group was given Flexi-bar therapy combined with deep muscle stimulation on top of the control group for 2 months of continuous intervention. Statistical comparison of visual analogue scoring (VAS), Oswestry dysfunction index (ODI), static and dynamic lumbar muscle endurance, lumbar EMG signals, pain mediators [β-endorphin (β-EP), 5-hydroxytryptamine (5-HT)], prognosis was conducted between the 2 groups. Results: After 1 month and 2 months of intervention, the VAS and ODI scores of the observation group were lower than those of the control group, and the serum β-EP and 5-HT levels were higher than those of the control group (P<0.05). The psoas endurance and RMS of the observation group were higher than those of the control group, and the MF was lower than that of the control group (P<0.05); there was no significant difference in the SF-36 scores between the two groups before the intervention, 1 month after the intervention, and 2 months after the intervention (P>0.05). Conclusion: Deep muscle stimulation combined with Flexi-bar therapy can effectively relieve pain, improve psoas muscle endurance, restore lumbar spine function, and improve prognosis in patients with NLBP.

Objective: To investigate the microbiological characteristics and the predictive value of Clinical Pulmonary Infection Score (CPIS) and Sequential Organ Failure Score (SOFA) in patients with chronic obstructive pulmonary disease (COPD) complicated by ventilator-associated pneumonia (VAP). Methods: A total of 136 COPD patients admitted to our hospital from November 2019 to November 2021 were selected and divided into study group (n=60) and reference group (n=76) according to whether the patients had VAP or not, and 50 healthy subjects during the same period were selected as the control group. The baseline characteristics and CPIS and SOFA scores of the three groups of patients with different prognosis were compared, and the multi-factors associated with VAP-related pathogen resistance, COPD combined with VAP and poor prognosis were analyzed; the predictive value of CPIS, SOFA and combined scores on the prognosis of COPD combined with VAP was analyzed. Results: Diabetes, pulmonary tuberculosis, multiple hospitalizations due to COPD, and antibiotic use within 3 months in the study group were higher than those in the reference group, and the CPIS and SOFA scores were higher than those in the reference group, which were risk factors for COPD complicated with VAP (P<0.05); the resistance rate of Klebsiella pneumoniae, Pseudomonas aeruginosa, and Acinetobacter baumannii to imipenem and imipenem was less than 10%, and the drug resistance rate was low; the CPIS and SOFA scores in the poor prognosis group were significantly higher than those in the good prognosis group (P<0.05); CPIS and SOFA scores were risk factors for poor prognosis of COPD combined with VAP (P<0.05); the AUC of CPIS combined with SOFA score in predicting the prognosis of COPD combined with VAP was higher than that of CPIS and SOFA score (P<0.05). Conclusion: Elevated CPIS and SOFA scores are risk factors and prognostic risk factors for COPD complicated with VAP. VAP antimicrobial therapy should be administered according to the drug sensitivity results, and combined evaluation is beneficial to the prognosis of patients.

147 Correlation of Serum Hcy, HbA1c and APN with Cognitive Impairment in Patients with Acute Cerebral Infarction

Objective: To analyze the correlation between serum homocysteine (Hcy), glycosylated hemoglobin (HbA1c) and serum adiponectin (APN) and cognitive impairment in patients with acute cerebral infarction (ACI). Methods: 102 patients with acute cerebral infarction treated in the hospital between July 2018 and February 2022 were selected as observation subjects, and the above patients were divided into normal cognition group (n=57, MMSE score ≥ 27 points) and cognitive impairment group (n=45, MMSE score<27 points) according to the Mini-Mental State Scale (MMSE). The general data of the two groups of patients were compared, and the laboratory indicators such as Hcy, HbA1c and APN were measured. The risk factors of cognitive impairment in patients with ACI were analyzed. According to MMSE, the patients with cognitive impairment were classified into mild group (17 cases), moderate group (15 cases) and severe group (13 cases). The changes in the above serum indicators in different groups of patients were compared, and their correlation with cognitive impairment and severity was analyzed. Results: There were no statistically significant differences in the course of disease, years of education, gender, drinking history, location of cerebral infarction, triglyceride (TG) and total cholesterol (TC) between normal cognition group and cognitive impairment group (P>0.05). The age in cognitive impairment group was older than that in normal cognition group, and the proportions of patients with hypertension, coronary heart disease and diabetes mellitus were higher than those in normal cognition group, and the levels of low-density lipoprotein cholesterol (LDL-C), Hcy and HbAlc were higher compared to normal cognition group (P<0.05). Hypertension (OR=1.442, 95%CI: 1.131~1.839), coronary heart disease (OR=1.342, 95%CI: 1.069~1.684), diabetes mellitus (OR=1.493, 95%CI: 1.027~2.171) and elevated levels of Hcy (OR=1.525, 95%CI: 1.053~2.209) and HbAlc (OR=1.650, 95%CI: 1.100~2.476) were risk factors for cognitive impairment in patients with ACI, and elevated APN level (OR=0.659, 95%CI: 0.458~0.949) was a protective factor for cognitive impairment in patients with ACI (P<0.05). The levels of Hcy and HbAlc in moderate group were higher than those in mild group while the APN was lower than that in mild group (P<0.05). The levels of Hcy and HbAlc were higher in severe group than those in mild and moderate groups while the APN was lower than that in mild and moderate groups (P<0.05). Hcy and HbAlc were negatively correlated with MMSE score (r=-0.569, -0.451, P<0.05), and APN was positively correlated with MMSE score (r=0.447, P<0.05). Conclusion: Elevated levels of Hcy and HbAlc are risk factors for cognitive impairment in patients with ACI, and high APN is a protective factor for cognitive impairment. The levels of Hcy and HbAlc are increased with the aggravation of cognitive impairment and are negatively correlated with cognitive function score, while APN is decreased with the aggravation of cognitive impairment and is positively correlated with cognitive function score. These indicators are helpful to guide the evaluation of cognitive impairment in patients with ACI.

153 Effect of Dispelling Pathogenic Factors and Purging Solid Needling on Facial Muscle Recovery in Patients with Facial Neuritis

Objective: To analyze the clinical effect of dispelling pathogenic factors and purging solid needling on the recovery of facial muscle function in patients with facial neuritis. Methods: Seventy patients with facial neuritis admitted to our hospital from May 2019 to November 2021 were selected and divided by random number table method into 35 cases in the treatment group, which were treated with the dispelling pathogenic factors and purging solid needling, and 35 cases in the conventional group, which were treated with conventional traditional acupuncture. The clinical efficacy of the two groups was evaluated and compared by analyzing the H-B facial nerve function classification before and after treatment, and the changes in the Chinese medicine symptom and sign score, facial muscle strength and electromyography before and after treatment in the two groups were evaluated. Results: After treatment, the TCM symptom and sign scores (angle of mouth deviation, nasolabial fold depth, shrugging nose, cheek puffing and air leakage, food residue) decreased in both groups compared with the pre-treatment scores in the two groups ( P<0.05), while the decrease in the above scores in the treatment group was greater than that in the conventional group ( P<0.05); the facial muscle strength increased in both groups after treatment compared with the pre-treatment scores in the two groups ( P<0.05), while the increase in the treatment group was greater than that in the conventional group ( P<0.05); the latency of motor nerve conduction in the orbicularis oris and nasal muscles was reduced in both groups after treatment compared with that before treatment in the two groups ( P< 0.05), while the reduction in the treatment group was greater than that in the conventional group (P<0.05); the wave amplitude of nerve conduction in the orbicularis oris and nasal muscles was prolonged in both groups after treatment compared with that before treatment in the two groups ( P< 0.05), while the prolongation in the treatment group was greater than that in the conventional group (P<0.05). Conclusion: Dispelling pathogenic factors and purging solid needling method is more conducive to the improvement of facial muscle strength in patients with facial neuritis and promote functional recovery.

158 Efficacy and Safety of Bosentan Combined with Treprostinil in the Treatment of Pulmonary Arterial Hypertension

Objective: To investigate the efficacy and safety of macitentan combined with tadalafil in the treatment of pulmonary arterial hypertension. Methods: 80 patients with pulmonary arterial hypertension were selected as the research subjects of this prospective study. The case collection time was from March 2021 to March 2022. All included cases were screened according to the inclusion and exclusion criteria. The 80 patients were divided into a control group and an experimental group of 40 patients each, and the grouping method was a simple randomized grouping method. The control group was treated with oral tadalafil tablets, and the observation group was treated with macitentan tablets on top of the control group. Both groups were treated for 12 consecutive weeks. The echocardiographic indexes such as pulmonary artery flat pressure (MPAP), mean systemic arterial pressure (MSAP), systemic circulation resistance (SVR) and pulmonary circulation resistance (PVR) were compared between the two groups before and after treatment, and the six-minute walk distance test (6MWD) and the Borg dyspnoea score were compared, and serum vascular endothelial factor-1 (ET) and C-reactive protein (CRP) levels were compared between the two groups of patients. The safety of the two groups of patients during treatment was compared. Results: MPAP, MSAP, SVR and PVR were lower in both the control and test groups at the end of treatment compared to the pre-treatment period, and the difference in MPAP, MSAP, SVR and PVR was more significant in the test group compared to the control group before and after treatment (P<0.05). After treatment, MPAP, MSAP, SVR, PVR in the control group and the test group were lower than those before treatment, and MPAP, MSAP, SVR, PVR in the test group were more significant than those before treatment (P<0.05). After treatment, Borg scores of the control group and the test group decreased compared with those before treatment, and Borg scores of the test group were more significant than those of the control group before and after treatment (P<0.05). After treatment, 6MWD of the control group and the test group increased compared with those before treatment, and 6MWD of the test group was more significant than those of the control group before and after treatment (P<0.05). After treatment, the levels of serum ET-1 and CRP in the control group and the test group were lower than those before treatment, and the levels of serum ET-1 and CRP in the test group were significantly lower than those in the control group (P<0.05). There were no adverse events in both groups during the treatment. Conclusion: Macitentan combined with tadalafil in the treatment of pulmonary hypertension can significantly reduce hemodynamic indexes, improve respiratory symptoms, reduce inflammatory response, and the treatment is safe.

162 Analysis of the Efficacy of Lrh-BNP Combined with Resulvastatin Calcium in the Treatment of Heart Failure in Dilated Cardiomyopathy

Objective: To explore the efficacy of combination of lyophilized recombinant human brain natriuretic peptide (lrhBNP) and rosuvastatin calcium in treating heart failure (HF) in dilated cardiomyopathy and its influence on cardiac function and levels of serum soluble suppression of tumorigenicity-2 (sST2), recombinant human galectin-3 (Galectin-3) ) and N-pro-brain natriuretic peptide (NT-pro BNP), so as to provide a treatment for this disease. Methods: One hundred and twenty-two patients with dilated cardiomyopathy complicated with HF who were treated in the Third People' s Hospital of Chengdu between October 2018 and October 2021 were enrolled. Random numbers were generated in Microsoft excel using the formula f(x)=rand() and then randomised using the sorting function to produce a randomised summary table, dividing the patients into an observation group (n=61) and a control group (n=61). Both groups were treated with standard basic therapy, with the control group treated with Rosuvastatin calcium in addition to the standard basic therapy and the observation group treated with lrhBNP on top of the control group. The difference in efficacy between the two groups after 7d of treatment was compared, and the differences in cardiac function [left ventricular ejection fraction (LVEF), left ventricular end-diastolic diameter (LVEDD), stroke volume (SV)] and serum markers (sST2, Galectin-3, NT-pro BNP) were evaluated before treatment and after 7d of treatment. The incidence rates of adverse reactions were analyzed within 7d of treatment. Results: After 7 d of treatment, there was a difference in the overall efficacy between both groups, and the total effective rate of the observation group at 95.08% after treatment was higher than the control group at 83.61% (P<0.05). The LVEF and SV in the two groups were increased after 7 d of treatment compared with those before treatment while the LVEDD, sST2, Galectin-3 and NT-pro BNP were reduced compared to before treatment, and the differences of the above six indicators before and after treatment in observation group were higher compared with those in control group (P<0.05). The incidence rates of adverse reactions within 7 d of treatment showed no statistical differences between the two groups (P>0.05), and there were no serious adverse events in the two groups. Conclusion: LrhBNPcombined with rosuvastatin calcium has a significant efficacy in the treatment of HF in dilated cardiomyopathy, and it has a certain regulatory effect on myocardial remodeling and fibrinolytic system function, with good safety.

167 Effect of Metoprolol and Ezetimibe Combined in the Treatment of Coronary Heart Disease in the Elderly and Its Influence on RAAS

Objective: To evaluate the efficacy of metoprolol and ezetimibe in the treatment of coronary heart disease in the elderly and their influence on renin-angiotensin-aldosterone system (RAAS). Methods: A total of 187 elderly patients with coronary heart disease who were admitted to our hospital from June 2019 to June 2021 were included in the combination group (n=93) and control group (n=94) by stratified random method. Both groups were treated with conventional medication for coronary heart disease and metoprolol, while the combination group was additionally treated with ezetimibe for three months. The changes of cardiac function, myocardial remodeling index and blood lipid level in the two groups were compared before and after treatment. The RAAS function was evaluated according to the changes of renin activity (PRA), angiotensin II (Ang II) and aldosterone (ALD) levels in patients. Results: After treatment, the cardiac output (CO), left ventricular ejection fraction (LVEF) and peak early diastolic velocity/peak late diastolic velocity (E/A) of the two groups were all higher than those before treatment, and the increase of the combined group was more significant than that of the control group, with statistical significance (P<0.05). After treatment, the left ventricular end diastolic diameter (LVEDD) and left ventricular end systolic diameter (LVESD) of the two groups decreased compared with those before treatment, and the decrease of the combined group was more significant than that of the control group, with statistical significance (P<0.05). After treatment, the triglyceride (TG), total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) of the two groups decreased compared with those before treatment, and the decrease of the combined group was more significant than that of the control group, with statistical significance (P<0.05). After treatment, the PRA, AngⅡ ⅱ and ALD of the two groups were lower than those before treatment, and the decrease of the combined group was more significant than that of the control group, with statistical significance (P<0.05). There were no obvious adverse reactions in both groups during treatment. After treatment, the total effective rate of the combined group was 93.55%, which was higher than 82.98% of the control group (P<0.05). Conclusion: The combination of metoprolol and ezetimibe can further regulate blood lipid levels in elderly patients with coronary heart disease, achieve the purpose of improving cardiac function, inhibiting myocardial remodeling and regulating RAAS function, and help to improve the prognosis of patients.
