1409 Hsa_circ_0044556 Mediates the Killing of Hepatocellular Carcinoma Cells by NK Cells via miR145/PD-L1

Objective: To investigate the role of hsa_circ_0044556 in the immune escape of hepatocellular carcinoma cells and its related mechanisms. Methods: Tumor tissues and their paraneoplastic tissues from hepatocellular carcinoma patients were collected to detect hsa_circ_0044556 and PD-L1 expression levels by qRT-PCR and correlation analysis were performed. The expression levels of hsa_circ_0044556 and PD-L1 in normal hepatic epithelial HL-02 cells and hepatocellular carcinoma SMMC-7721 and HCCLM3 cells were detected by qRT-PCR. The targeting relationship between hsa_circ_0044556 and miR145 and miR145 and PD-L1 was examined by informatics prediction and luciferase reporter assay. The si-hsa_circ_0044556, si-miR145 and si-NC were transfected into HCCLM3 cells by liposome transfection method, grouped as: si-NC group, si-hsa_circ_0044556 group, si-miR145 group and si-hsa_circ_0044556 + si-miR145 group. The expression level of PD-L1 and cell proliferation ability was compared among the groups. The sensitivity to NK cell killing was tested by NK cell toxicity assay. Results: The expression levels of hsa_circ_0044556 and PD-L1 were elevated in tumor tissues compared with paraneoplastic tissues (P<0.05), and hsa_circ_0044556 was positively correlated with PD-L1 expression (r=0.5481). In HL-02, SMMC-7721, and HCCLM3 cells, hsa_circ_0044556, and PD-L1 expression levels were sequentially increased (P<0.05). Hsa_circ_0044556 was targeted to inhibit miR145 expression; miR145 was targeted to inhibit PD-L1 expression. Compared with the si-NC group, the si-hsa_circ_0044556 group showed elevated miR145 gene expression, decreased PD-L1 protein expression, diminished cell proliferation, and increased sensitivity to NK cell killing (P<0.05); compared with the si-NC group, the si-miR145 group had elevated PD-L1 gene and protein expression, increased cell proliferation capacity and decreased sensitivity to NK cell killing (P<0.05); compared with the si-hsa_circ_0044556 group, the si-hsa_circ_0044556 + si-miR145 group had PD-L1 expression level increased, cell proliferation capacity increased, and sensitivity to NK cell killing decreased (P<0.05). Conclusion: In hepatocellular carcinoma cells, hsa_circ_0044556 mediates immune escape of tumor cells by promoting the proliferation of hepatocellular carcinoma cells through the miR145/PD-L1 signaling axis while inhibiting sensitivity to NK cell killing.

1414 Experimental Study on the Neuroprotection of Albiflorin in Mice with Neonatal Hypoxic-Ischemic Brain Injury

Objective: To observe the effects of albiflorin on brain cell apoptosis, oxidative stress, and inflammation in neonatal hypoxic-ischemic brain injury mice and to explore its potential mechanism. Methods: The hypoxic brain injury (HIE) model of neonatal mice was established. 30 mg/kg albiflorin was administered by gavage to model mice, which were set as albiflorin group. Mice in the sham operation group and model group were given distilled water by gavage. 2,3,5-triphenyl tetrazolium chloride (TTC) staining was used to measure the area of cerebral infarction, TDT mediated dUTP biotin nick end labeling (Tunel) method was used to measure brain cell apoptosis, and Western blot (WB) was used to measure protein expression of activated Caspase-3, cleaved caspase-9 phosphatidylinositol 3-kinase (PI3K), phosphorylation (p) - PI3K, serine/threonine kinase (AKT), p-AKT, mammalian target of rapamycin (mTOR) and p-mTOR in brain tissue. Enzyme-linked immunosorbent assay (ELISA) was used to detect malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH), interleukin (IL)-6, IL-18, and IL-1β in brain tissue. Intercellular cell adhesion molecule-1 (ICAM-1) was detected by immunohistochemistry. Results: Compared with the sham operation group, the cerebral infarction rate, brain cell apoptosis rate, cleaved caspase-3 and cleaved caspase-9 protein, MDA, ICAM-1, IL-6, IL-18, and IL-1β in the model group were significantly increased, and SOD, GSH, p-PI3K, p-AKT and p-mTOR decreased in brain tissue (P<0.05). The above indexes of mice in the albiflorin treatment group were significantly closer to those in the sham operation group. Conclusions: AAlbiflorin alleviates cerebral infarction in HIE young rats, inhibits brain cell apoptosis, oxidative stress, and inflammatory response, and protects against nerve injury in young rats. Its mechanism may be related to the activation of the PI3K / Akt / mTOR signal pathway.

1420 Study of the Regulatory Mechanism of the Interaction Between Numb1 and ITGB1 in Gastric Cancer Progression and Drug Resistance

Objective: To investigate the interaction between cohesive protein 1 (Numb1) and integrin β1 (ITGB1) on gastric cancer progression and drug resistance and its related mechanisms. Methods: The expression levels of Numb1 and ITGB1 in tumors and adjacent tissues of 21 patients with gastric cancer were detected by qRT-PCR, and the correlation between the expression levels of Numb1 and ITGB1 was analyzed. The overexpressed vectors (flag-Numb1, flag-ITGB1, and flag-NC) were transfected into BGC-823 cells by lentivirus, and the binding sites of Numb1 and ITGB1 were detected by the immunoprecipitation method. si-NC, si-Numb1, and si-ITGB1 were transfected into BGC-823 cells. The mRNA expression levels of Numb1 and ITGB1 were detected by qRT-PCR and the protein expression levels of Numb1 and ITGB1 were detected by Western blot. CCK8 detected cell proliferation and sensitivity to oxaliplatin, cell migration was detected by cell scratch assay, and cell invasion was detected by Transwell. Results: The expression levels of Numb1 and ITGB1 were elevated in tumor tissues compared to paraneoplastic tissues (P<0.05), and Numb1 was positively correlated with ITGB1 expression levels (r=0.6270). In cellular assays, ITGB1 protein was detected in the immunoprecipitation products of flags overexpressing flag-Numb1, and Numb1 protein was detected in the immunoprecipitation products of flags overexpressing flag-ITGB1. Compared with the si-NC group, the si-Numb1 group showed reduced levels of Numb1 and ITGB1 mRNA and protein expression (P<0.05), reduced cell proliferation (P<0.05), increased sensitivity to oxaliplatin (P<0.05), and reduced cell migration and invasion (P<0.05). Compared with the si-NC group, the si-ITGB1 group showed reduced ITGB1 mRNA and protein expression levels (P<0.05), reduced cell proliferation (P<0.05), increased sensitivity to oxaliplatin (P<0.05) and reduced cell migration and invasion (P<0.05). Conclusion: Numb1 regulates ITGB1 expression levels through interaction with ITGB1 and promotes proliferation, migration, invasion and resistance to chemotherapeutic agents in gastric cancer cells

1425 Activation of the Nrf2 Pathway by MCT2 Regulates Apoptosis and Pyroptosis in H/R Retinal Ganglion Cells

Objective: To explore the effect and mechanism of monocarboxylate transporters 2 (MCT2) on apoptosis and pyrolysis of retinal ganglion cells after hypoxia/reoxygenation (H/R). Methods: Retinal ganglion cells RGC-5 were randomly divided into control group, H/R group, MCT2 group, MCT2+ML385 group, and pcDNA3.1-MCT2 recombinant plasmid was transfected into RGC-5 cells via liposome to establish Cell H/R model, and treat cells with Nrf2 inhibitor ML385. After the cells were treated accordingly, transfection efficiency was measured by real-time fluorescence quantitative PCR and protein blotting (Western blot), cell proliferation activity was measured by CCK-8, apoptosis level was measured by flow cytometry, PI apoptosis index was measured by Hoechst/PI staining, expression levels of apoptosis-related and pyrolysis-related proteins were measured by Western blot, and expression of Nrf2 and Caspase-1 were observed by immunofluorescence staining. Results: After transfection of pcDNA3.1-MCT2 into RGC-5 cells, both intracellular MCT2 mRNA and protein relative expression were significantly upregulated (P<0.05); after H/R treatment, cell proliferation activity decreased, apoptosis rate increased, PI apoptosis index increased, intracellular Bax and Caspase-3 protein relative expression were both upregulated while Bcl-2 protein relative The relative expression of Bax and Caspase-3 proteins was upregulated while that of Bcl-2 protein was downregulated, the relative expression of GSDMD, Caspase-1, NLRP3, and IL-1β proteins were upregulated, the fluorescence intensity of Nrf2 was reduced and that of Caspase-1 was enhanced. After transfected with pcDNA3.1-MCT2 and then treated with H/R, cell proliferation activity increased, apoptosis rate decreased, PI apoptosis index decreased, relative expression of intracellular Bax and Caspase-3 proteins were down-regulated, relative expression of Bcl-2 protein was up-regulated. The relative expression of GSDMD, Caspase-1, NLRP3, and IL-1β were all down-regulated, the fluorescence intensity of Nrf2 was enhanced and that of Caspase-1 was diminished, all differences were statistically significant (P<0.05); while cells treated with transfected pcDNA3.1-MCT2 with H/R followed by Nrf2 inhibitor ML385 showed a decrease in cell proliferation activity, an increase in apoptosis rate and PI apoptosis index, intracellular Bax and Caspase-3 protein relative expression was upregulated, Bcl-2 protein relative expression was downregulated, GSDMD, Caspase-1, NLRP3, and IL-1β protein relative expression was upregulated, Nrf2 fluorescence intensity was diminished and Caspase-1 fluorescence intensity was enhanced, all differences were statistically significant (P<0.05). Conclusion: MCT2 inhibits H/R-induced apoptosis and focal death in retinal ganglion cells, and this mechanism of action may be related to the activation of the Nrf2 pathway.

1431 Effect of miR-21-3p Targeting VEGFA to Regulate PI3K/AKT Signaling Pathway on Renal Injury in Preeclamptic Rats

Objective: To explore the effect of microRNA-21-3p (miR-21-3p) on renal injury in preeclampsia (PE) by targeting vascular endothelial growth factor A (VEGFA) to regulate the phosphatidylinositol 3-kinase/protein kinase B (PI3K/AKT) signaling pathway Amelioration of renal injury in rats. Methods: Sixty pregnant mice were divided into normal group, model group, mimics NC group,miR-21-3p mimics group,and miR-21-3p inhibitor group randomly. Except for the normal group, PE models were established for the remaining four groups. After successful modeling, the mimics NC group was injected with the miRNA-NC empty plasmid adenoviral vector; the miR-21-3p mimics group was injected with the adenoviral vector carrying the miR-21-3p gene; and the miR-21-3p inhibitor group was injected with the adenoviral vector carrying the miR-21-3p gene expression inhibitor, all for 7 consecutive days. At the end of the experiment, blood was collected from arteries, and the levels of SCr, BUN, IL-6, and TNF-α in serum and urinary protein in urine were measured; sections were prepared by HE staining and the renal histopathological changes were observed by light microscopy; the relative expression levels of miR-21-3p and VEGFA mRNA were detected by RT-PCR; VEGFA, PI3K, p-PI3K, AKT, p-AKT protein expression levels by Western Blot. A dual luciferase reporter gene assay was performed to verify the targeting relationship between miR-21-3p and VEGFA. Results: Compared with the normal group, the renal histopathological changes in the model group were significant, with significantly higher levels of urinary protein, SCr, BUN, TNF-α, and IL-6 (P<0.05), decreased expression of miR-21-3p in renal tissues (P<0.05), VEGFA mRNA expression as well as VEGFA, p-PI3K/PI3K and p-AKT/AKT protein expression levels were significantly increased (P<0.05). There was no significant difference in the changes of the above indicators between the mimics NC group and the model group (P>0.05). Compared with the mimics NC group, urinary protein, SCr, BUN, TNF-α, and IL-6 levels were significantly decreased in the miR-21-3p mimics group (P<0.05), miR-21-3p expression was increased in renal tissues (P<0.05), and VEGFA mRNA expression as well as VEGFA, p-PI3K/PI3K, p-AKT/ AKT protein expression levels were significantly decreased (P<0.05); SCr, BUN, urinary protein, IL-6, TNF-α levels were significantly increased in the miR-21-3p inhibitor group (P<0.05), miR-21-3p expression was decreased in renal tissues (P<0.05), VEGFA mRNA expression as well as VEGFA, p-PI3K /PI3K, p-AKT/AKT protein expression levels were significantly increased (P<0.05). Validation of the dual luciferase reporter gene assay showed that VEGFA was the target gene of miR-21-3p. Conclusion: miR-21-3p may regulate the PI3K/AKT signaling pathway by targeting VEGFA to ameliorate renal injury in PE rats.

1438 The Expression Levels of STIL and PRR11 in Cervical Cancer Tissues and Their Correlation with Lymph Node Tissue Metastasis

Objective: To analyze the expression levels of SCL/TALl interrupting locus (STIL) and proline-rich protein 11 (PRR11) in cervical cancer tissues and their correlation with lymph node metastasis. Methods: A total of 143 patients with cervical cancer admitted to the hospital were enrolled as the research subjects between January 2020 and January 2022. During surgery, cervical cancer tissues and adjacent normal para-carcinoma tissues were collected. The levels of STIL and PRR11 in cancer tissues and para-carcinoma tissues were detected by Western Blot. The mRNA levels of STIL and PRR11 were detected by quantitative real-time polymerase chain reaction (qRT-PCR). The levels of STIL and PRR11 in patients with different FIGO staging, and between patients with or without lymph node metastasis were compared. The diagnostic value of STIL and PRR11 for lymph node metastasis was analyzed by receiver operating characteristic (ROC) curves, which were represented by the area under the curve (AUC). The correlation between STIL and PRR11 was analyzed by the Pearson method. Results: The levels of STIL and PRR11, and relative expression levels of their genes in cervical cancer tissues were significantly higher than those in para-carcinoma tissues (P<0.05). The higher the FIGO staging, the higher the levels of STIL and PRR11 (P<0.05). The levels of STIL and PRR11 in patients with lymph node metastasis were significantly higher than those without metastasis (P<0.05). AUC values of STIL and PRR11 in the diagnosis of lymph node metastasis were 0.733 and 0.729, respectively (P<0.05). Pearson correlation analysis showed that there was a significant correlation between STIL and PRR11 in cancer tissues (P<0.05). Conclusion: The expressions of STIL and PRR11 are up-regulated in cervical cancer tissues. And their increase indicates the increased risk of lymph node metastasis. STIL and PRR11 can assess lymph node metastasis in cervical cancer.

1442 Study on the Effect of Different Treatment Modalities for Type II Caesarean Scar Pregnancy on Ovarian Function Indicators and Recurrence of Caesarean Scar Pregnancy

Objective: To investigate the effect of different treatment methods on ovarian function indexes and recurrence of CSP in type Ⅱ cesarean scar pregnancy (CSP). Methods: A total of 210 patients with type II CSP admitted to our hospital from February 2018 to January 2021 were selected as the research subjects,and they were divided into 3 groups (70 cases in each) by random number table method.Injection of methotrexate (MTX) combined with hysterectomy (group A),hysteroscopic hysterectomy combined with Foley's balloon compression therapy (group B),and laparoscopy combined with hysteroscopy (group C).The operation time,intraoperative blood loss,hospital stay,menstrual recovery time,and endometrial thickness recovery time were compared among the three groups.All patients were followed up for 12 months,and the hormone indexes estradiol (E2) and follicle-stimulating hormone (FSH) levels,luteinizing hormone (LH),and antral follicle numbers in different time periods after surgery were compared in the three groups,and the incidence of CSP during re-pregnancy was compared among the 3 groups. Results: Intraoperative blood loss,hospital stay,menstrual recovery time,and endometrial thickness recovery time in group B were less than those in groups A and C (P<0.05),and the operation time in groups A and B were shorter than those in group C (P<0.05).There were no significant changes in the levels of hormone indicators E2,FSH,LH and the number of antral follicles in the menstrual cycle in different time periods after surgery in the three groups (P>0.05).Within 12 months of follow-up,the incidence of CSP in group B was significantly lower than that in groups A and C (P<0.05). Conclusion: MTX combined with evacuation,hysteroscopic evacuation combined with Foley's balloon compression therapy and laparoscopy combined with hysteroscopy in the treatment of type Ⅱ CSP will not affect the ovarian function of patients,and in the three treatment methods,the uterine cavity is not affected.Endoscopic hysterectomy combined with Foley's balloon compression therapy can reduce the chance of recurrence of CSP.

1447 Preparation of Polyclonal Antibody Against oprC Protein of Acinetobacter Baumannii and Its Immune Effect

Objective: To culture a polyclonal antibody against the oprC protein of the outer membrane receptor of Acinetobacter baumannii (A.baumannii) and to evaluate its immune effect. Methods: The full gene sequence of oprC was queried on NCBI, and the full length of oprC was amplified using A.baumannii DNA as a template. The antibody titer was determined by enzyme-linked immunosorbent assay (ELISA), and the results showed that the potency of oprC antiserum was 1∶12800. The improvement of the survival rate, the number of bacteria in the blood, and the damage of multiple organ tissues of the mice challenged with oprC protein polyclonal antibody was detected. Results: The oprC polyclonal antibody could improve the survival rate of mice, reduce the load of bacteria (P<0.05), and relieve tissue damage to the lung, spleen, and kidney to some extent. Conclusion: This study confirmed that oprC recombinant protein can improve the survival rate of mice, reduce pathological damage, and achieve immune protection.

1452 Relationship between Levels of Serum PCT and hs-CRP and Degree of Myocardial Damage in Acute Stage of Pediatric Pneumonia

Objective: To analyze the relationship between levels of serum PCT and hs-CRP and the degree of myocardial damage in children with acute stage of pneumonia. Methods: 137 children with acute stage of pediatric pneumonia were analyzed. Among them, 82 cases with myocardial damage were enrolled in the myocardial damage group and 55 cases without the occurrence of myocardial damage were included in the non-myocardial damage group. According to the degree of myocardial damage, the children were divided into 33 cases in the mild group, 28 cases in the moderate group, and 21 cases in the severe group. The baseline data and levels of serum myocardial damage markers [creatine kinase isoenzyme (CK-MB), cardiac troponin I (cTnI)], serum PCT, and hs-CRP were analyzed in the myocardial damage group, and non-myocardial damage group, and the changes in levels of serum PCT and hs-CRP in mild, moderate and severe groups were compared. Pearson correlation analysis was used to analyze the correlation between levels of serum PCT and hs-CRP and CK-MB and cTnI in children. The receiver operating characteristic (ROC) curve was adopted to analyze the diagnostic value of serum PCT, hs-CRP, and the combination of the two in predicting the occurrence of myocardial damage in children with acute stage of pneumonia. Results: There were no statistical differences in gender, age, and time from onset to admission between the two groups (P>0.05). The levels of serum CK-MB and cTnI in the myocardial damage group were significantly higher than those in the non-myocardial damage group (P<0.05). The levels of serum PCT and hs-CRP were significantly higher in the myocardial damage group than those in the non-myocardial damage group (P<0.05). The differences in serum PCT and hs-CRP levels among the three groups of children with different myocardial damage degrees were statistically significant, among which the levels were significantly lower in the mild group than those in the moderate group and severe group (P<0.05) and were significantly lower in the moderate group than those in the severe group (P<0.05). Serum PCT and hs-CRP levels in children with acute stage of pneumonia complicated with myocardial damage were positively correlated with serum CK-MB levels (r=0.621, 0.778, all P<0.001), and were positively correlated with serum cTnI level (r=0.501, 0.6668, all P<0.001). Logistic regression analysis revealed that serum PCT and hs-CRP levels were correlated with myocardial damage in children at the acute stage (P<0.05). ROC analysis showed that the AUC values of serum PCT, hs-CRP, and their combination were 0.858, 0.914, and 0.955 respectively in the diagnosis of occurrence of myocardial damage in children with acute stage of pneumonia (all P<0.05), and the sensitivities were 85.37%, 90.24%, and 93.90% and the specificities were 72.73%, 83.64%, and 70.91% respectively. Conclusion: The high expressions of serum PCT and hs-CRP may indicate myocardial damage in children with acute stage of pneumonia, and they can be used for clinical auxiliary diagnosis.

1457 Relationship between LncRNA PVT1 and ABCG2 mRNA Expression and Pathological Characteristics in Breast Cancer Tissues and Analysis of KM Curve

Objective: To explore the relationship between long-chain non-coding RNA plasmacytoma ectopic variant gene 1 (LncRNA PVT1), adenosine triphosphate binding transporter G2 (ABCG2) mRNA expression and pathological characteristics in breast cancer tissues, and analysis of KM curve. Methods: The case data of 80 breast cancer patients who underwent surgical treatment in our hospital between January 2017 and July 2018 were selected for retrospective study.During the operation, breast cancer tissues and adjacent tissues (2cm from the lesion) were collected to compare the expression of LncRNA PVT1 and ABCG2 mRNA in the cancer tissues and adjacent tissues.The correlation between LncRNA PVT1, ABCG2 mRNA expression and pathological characteristics in cancer tissues were analyzed.The expression of LncRNA PVT1 and ABCG2 mRNA in cancer tissues of patients with and without recurrence were compared.COX regression was used to analyze the correlation between LncRNA PVT1 and ABCG2 mRNA expressions and recurrence in cancer tissues.The disease-free survival of patients with LncRNA PVT1 and ABCG2 mRNA expression in different cancer tissues was compared. Results: The expression of LncRNA PVT1 (0.076±0.011) and ABCG2 mRNA (3.27±0.45) in cancer tissues was higher than that in adjacent tissues (0.003±0.001),(0.82±0.26)(t=59.114,42.165,P<0.05); the expression of LncRNA PVT1 and ABCG2 mRNA in cancer tissues was related to clinical stage, pathological grade, and lymph node metastasis (P<0.05); the expression of LncRNA PVT1 (0.084±0.010) and ABCG2 mRNA (3.75±0.59) in cancer tissues of relapsed patients was higher than that of non-relapsed patients (0.074±0.009),(3.13±0.47)(t=4.048,4.644,P<0.05); COX regression analysis showed that the high expression of LncRNA PVT1 and ABCG2 mRNA in cancer tissues were independent risk factors for postoperative recurrence (P<0.05); Kaplan-Meier survival analysis showed that patients with high expression of LncRNA PVT1 and ABCG2 mRNA in cancer tissues had a lower 3-year disease-free survival rate than those with low expression (P<0.05). Conclusion: The abnormally high expression of LncRNA PVT1 and ABCG2 mRNA in the cancer tissues of breast cancer patients is associated with pathological characteristics and disease-free survival, and is expected to become a new therapeutic target.

Objective: To observe the effects of miR-216a-3p and miR-128-3p on the phenotypic expression of OSCC cells,and explore their potential mechanisms. Methods: The expression of miR-216a-3p,miR-128-3p and G protein signal regulator 17 (RGS17) in cancer tissues,adjacent tissues,human normal oral keratinocytes (hNOK) and OSCC cancer cells (SCC-9,SCC-25,HN4) were detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction (RT-qPCR); mimic NC group (transfected mimic),mimic-miR-216a-3p group (transfected miR-216a-3p mimic),mimic-miR-128-3p group (transfected miR-128-3p mimic),si-NC group (transfected si-NC),si-RGS17 group (transfected si-RGS17),mimic-miR-216a-3p+pcDNA group (co-transfected miR-216a-3p mimic and pcDNA),and mimic-miR-216a-3p+pcDNA-RGS17 group(co-transfected miR-216a-3p mimic and pcDNA-RGS17) were transfected into SCC-9 cells.5-bromo-2-deoxyuracil (EdU) staining and clonogenic assay were used to detect cell proliferation; Transwell test was used to detect the ability of cell migration and invasion; Annexin V / propidium iodide (PI) staining was used to detect the ability of apoptosis; Double luciferase report assay and RNA sedimentation assay were used to verify the targeting relationship; The RGS17 protein was detected by Western blot (WB). Results: Compared with adjacent tissues or hNOK cells,the expression of miR-216a-3p and miR-128-3p in OSCC tissues (stage Ⅰ,Ⅱ,Ⅲ) and cancer cells (SCC-9,SCC-25,HN4) was significantly decreased,and the expression of RGS17 was significantly increased (P<0.05).Compared with the mimic-NC group,the positive rate of EdU,the rate of clone formation,the number of migrating and invasive cells in the mimic-miR-216a-3p and mimic-miR-128-3p groups were significantly lower,and the apoptosis rate was significantly higher (P<0.05).The cells in si-RGS17 group had similar changes to those in mimic-miR-216a-3p and mimic-miR-128-3p cells.miR-216a-3p and miR-128-3p jointly targeted and negatively regulated RGS17.Overexpression of RGS17 significantly attenuated the proliferation,migration-invasion inhibition and apoptosis-promoting effects of miR-216a-3p and miR-128-3p on SCC-9 cells. Conclusion: MiR-216a-3p and miR-128-3p could inhibit the proliferation,migration and invasion of OSCC cells and promote apoptosis,which may be related to targeting RGS17.

1469 Effects of EMG Biofeedback Combined with Task-Oriented Circuit Therapy on Walking Impairment in Stroke Patients with Hemiplegia

Objective: To explore the effects of EMG biofeedback combined with task-oriented circuit therapy for walking impairment in stroke patients with hemiplegia. Methods: A total of 102 stroke patients with hemiplegia in our hospital from May 2019 to March 2022 were randomly divided into experiment group and control group,21 cases in each group.EMG biofeedback was used in control group while EMG biofeedback combined with task-oriented circuit therapy was used in experiment group.The walking ability,leg myodynamia and life quality were recorded and compared. Results: After treatment,the MAS score,FAC score,FMA score and active dorsal expansion ROM of ankle joint in experiment group were significantly higher than those in control group (P<0.05).the EMG value of tibialis anterior muscle in experiment group was significantly higher than that in control group (109.92±18.36 vs 89.41±10.35,P<0.05),while the EMG value of gastrocnemius muscle in experiment group was significantly lower than that in control group(8.39±2.03 vs 13.04±1.87,P<0.05).The experiment group also had significantly higher SF-36 scale scores and Barthel Index scale scores than the control group (P<0.05). Conclusion: The EMG biofeedback combined with task-oriented circuit therapy in stroke patients with hemiplegia can improve myodynamia of shank,enhance walking ability and life quality.

1473 Characterization of Hepatitis B Virus/S-Gene Mutation and Its Association with Immune Escape of Hepatitis B Vaccine

Objective: To study the characteristics of hepatitis B virus (HBV) S-gene mutation and its association with immune escape of hepatitis B vaccine. Methods: 1,973 inpatients in the hospital between March 2021 and March 2022 were selected as samples for a cross-sectional study.All patients completed hepatitis B (HepB) vaccination according to the standard procedures of 0,1 month and 6 months.HBV serum markers and HBV S-gene mutation were detected. Results: There were 47 HBsAg positive samples among 1973 patients (2.38%),the rate of HBsAg positivity was higher in those aged >18 years than in those aged 1-18 years,and the rate of HBsAg positivity was lower in those with HBV-negative parents than in those with HBV-positive mothers,all with statistically significant differences (P<0.05).Compared with the standard sequence,162 nucleotide mutation sites and 98 amino acid mutation sites were detected in 47 HBsAg-positive samples,among which the mutation sites in the a determinant (aa124-147) mainly included I/T126A/N/I/S,Q129H/R,M133L,K141E,T143S,D144A/EH and G145R/A,mutation sites in the a determinant upstream (aa99-123) were mainly Q101R/H,L110I,G112K,S113T/G,S117G and P120Q/R/L,and mutation sites in the a determinant downstream (aa148-169) mainly included Y161F/C,E164D,F161Y and R160K.Among 47 HBsAg-positive samples,30 samples were found to have amino acid mutation with the detection of 63.83%,among which 19 had amino acid polarity change (according for 63.33%) and 11 occurred in the a determinant (aa124-147),including 5 mutation sites of T126I,Q129R,T143S,D144A and G145A. Conclusion: HBV S-gene mutation is closely associated to the immune escape of HepB vaccine.The mutation sites of T126I,Q129R,T143S,D144A and G145A in the a determinant (aa124-147) can cause changes in the polarity and hydrophilicity of HBsAg,resulting in the decrease of antigenicity and the occurrence of immune escape.

1479 Changes of Serum IGF-1,IGFBP-3 and Hcy Levels in Children with Congenital Hypothyroidism and Their Relationship with Physical Development

Objective: To investigate the changes in serum insulin growth factor-1 (IGF-1),insulin-like growth factor binding protein-3 (IGFBP-3) and homocysteine (Hcy) levels in children with congenital hypothyroidism (CH) and their relationship with physical development. Methods: One hundred and ten children with CH who received replacement therapy at our hospital (observation group) and 59 normal newborns at the same time (healthy control group) were selected as study subjects.Both groups were followed up for 1 year.The differences in levels of serum IGF-1,IGFBP-3 and Hcy and physical development indicators [height,body mass,height standard deviation score (HtSDS),weight standard deviation score (WTSDS)] were compared between observation group and healthy control group at enrollment and after 1 year of follow-up.Pearson correlation analysis was used to analyze the correlation between levels of serum IGF-1,IGFBP-3 and Hcy and physical development indicators of children patients with CH after 1 year of follow-up. Results: There were no significant differences between the two groups in terms of gender,age,delivery methods and maternal birth age (all P>0.05),and the TSH level of observation group was significantly higher than that of healthy control group (P<0.05) while the levels of FT3 and FT4 were significantly lower than those in control group (P<0.05).The levels of IGF-1 and IGFBP-3 in observation group and healthy control group after 1 year of follow-up were higher than those at enrollment (P<0.05) while the level of Hcy was significantly lower than that at enrollment (P<0.05).At enrollment and after 1 year of follow-up,the levels of serum IGF-1 and IGFBP-3 in observation group were lower than those in healthy control group (P<0.05) while the level of serum Hcy was higher (P<0.05).At enrollment,there were no statistically significant differences in the height,body mass,HtSDS and WTSDS between the two groups (P>0.05).The height and body mass in observation group and healthy control group after 1 year of follow-up were higher than those at enrollment (P<0.05),and the height and body weight of healthy control group were higher or heavier than those of observation group (P<0.05).According to Pearson correlation analysis,levels of serum IGF-1,IGFBP-3 and Hcy in children with CH were not correlated with HtSDS and WTSDS (P>0.05),but levels of IGF-1 and IGFBP-3 were positively correlated with height and body mass (all P<0.05),and Hcy level was negatively correlated with height and body mass (all P<0.05). Conclusion: The levels of serum IGF-1,IGFBP-3 and Hcy in children with CH after replacement therapy have been significantly improved,and they have a significant correlation with physical development.

1484 Relationship between Serum miR-483-5p and miR-487b Expression and Inflammatory Factors Disease and Prognosis in Patients with Acute Lung Injury Induced by Sepsis

Objective: To investigate the relationship between miR-483-5p and miR-487b and inflammatory factors, disease condition, and prognosis in patients with acute lung injury (ALI) induced by sepsis. Methods: A total of 133 sepsis patients admitted to the ICU of our hospital from September 2018 to December 2021 were selected, including 51 cases complicated with ALI (ALI group) and 82 cases without ALI (NALI group). According to oxygenation index [oxygen partial pressure/inhaled oxygen concentration], ALI group was divided into mild group (201 ~ 300mmHg) (16 cases), moderate group (101 ~ 200mmHg) (20 cases) and severe group (≤100mmHg) (15 cases). According to the 28-day survival status, the ALI group was divided into the survival group (28 cases) and the death group (23 cases). Another 43 volunteers were selected as the control group. The serum expressions of mir-483-5p and mir-487b and the levels of inflammatory factors in different groups were compared. Pearson analyzed the correlation between the expressions of miR-483-5p and miR-487b and the levels of inflammatory factors in patients with sepsis complicated with ALI. Receiver operating characteristic curve (ROC) analysis of the prognostic value of miR-483-5p and miR-487b in patients with sepsis-induced ALI. Results: The expression of miR-483-5P, the levels of interleukin-6 (IL-6), tumor necrosis factor -α (TNF-α), and interleukin-33 (IL-33) on the baseline in the ALI group were higher than those in NALI group and control group (P<0.05), and the expression of miR-487b on the baseline was lower than those in NALI group and control group (P<0.05). The expression of miR-483-5p, IL-6, TNF-α, and IL-33 on the baseline, 3d hospitalization, and 7d hospitalization in the severe group were higher than those in the moderate and mild groups (P<0.05), and the expression of miR-487b was lower than those in the moderate and mild groups (P<0.05). Serum miR-483-5p expression, IL-6, TNF-α, IL-33 levels on the baseline, 3d hospitalization, and 7d hospitalization in the death group were higher than those in the survival group (P<0.05), and miR-487b expression was lower than those in the survival group (P<0.05). The expression of miR-483-5P was positively correlated with the levels of IL-6, TNF-α, and IL-33 on the baseline, 3d hospitalization, and 7d hospitalization in patients with sepsis (P<0.05). The expression of miR-487b was negatively correlated with the levels of IL-6, TNF-α, and IL-33 on the baseline, 3d hospitalization, and 7d hospitalization (P<0.05). The area under the curve predicted by the combination of miR-483-5p and miR-487b on 7d hospitalization was 0.964, which was higher than those predicted by miR-483-5p and miR-487b alone (z=2.612, 3.141,P<0.05). Conclusion: The expression of miR-483-5p is up-regulated and miR-487b is down-regulated in serum of ALI patients induced by sepsis, which is related to excessive inflammatory response, aggravation of the disease, and poor prognosis.

Objective: To investigate the levels and clinical significance of retinol-binding protein 4 (RBP4) and N-terminal probrain natriuretic peptide (NT-proBNP) in patients with chronic heart failure. Methods: A total of 89 cases in the observation group were patients diagnosed with chronic heart failure in our hospital from May 2018 to August 2019,and a total of 75 cases in the control group were volunteers who underwent physical examination in our hospital during the same period.According to NYHA classification,the subjects in the observation group were divided into 3 groups:NYHA II (n=23),III (n=34),and IV (n=32).All subjects were required to draw fasting venous blood in the morning to measure and record fasting RBP4 and NT-proBNP. Results: The levels of RBP4 and NT-proBNP in the observation group were significantly higher than those in the healthy control group,and the levels of serum RBP4 and NT-proBNP gradually increased with the severity of heart failure. Conclusion: RBP4 and NT-proBNP are closely related to chronic heart failure,and they are of great significance to the early diagnosis and prognosis of heart failure.

1494 Identification of Relationship between Helicobacter Pylori Infection and Serum SERPIN E1 Protein Level in Adults

Objective: To identify the relationship between Helicobacter pylori (Hp)infection and serum SERPIN E1 protein level in adults and offer a theoretical basis for investigating the mechanisms by which Hp induces gastric cancer. Methods: A total of 417 subjects were allocated to the Hp-negative group(n=192) or the Hp-positive group(n=225). All participants had fully detailed records of biochemical data and results of serum SERPIN E1 testing. Results: Serum SERPIN E1 level was significantly different (P<0.01) between the Hp-negative group and Hp-positive group: 5.38(4.56~6.18)(ng/ml))versus 8.20(6.91~9.34) (ng/ml). All the participants were divided into 4 groups by quartile values of serum SERPIN E1 and a linear correlation between the prevalence of Hp infection and quartiles of serum SERPIN E1 levels was observed(P<0.001). Based on the receiver operating characteristic curve (ROC) for predicting Hp infection in participants, the serum level of SERPIN E1 with optimal sensitivity (84.4%) and specificity (83.3%) was about 6.49ng/ml and the area under the ROC curve was 0.906. Conclusion: Serum level of SERPINE1 is closely associated with Hp infection in adults. SERPIN E1 may play an important role in the pathogenesis of Hp-induced gastric cancer.

1498 Efficacy of Probiotic-Assisted Estramiprazole-Based Quadruple Regimen in the Treatment of Hp-Positive Gastric Ulcer and Analysis of the Effect on Intestinal Flora

Objective: To investigate the efficacy of a probiotic-assisted quadruple regimen based on esomeprazole in the treatment of Helicobacter pylori (H. pylori, Hp) positive gastric ulcer and its effect on the intestinal flora. Methods: A total of 123 patients with Hp-positive gastric ulcers who were admitted to our hospital from May 2019 to May 2021 were selected for research, and they were divided into a study group (N=63) and a control group (N=60) by a simple random method using a random number table. The group was treated with a quadruple regimen based on esomeprazole, and the research group was given probiotics (Saccharomyces boulardii) on this basis. Both groups were treated for 14 days, and the Hp eradication rate, ulcer surface healing, intestinal flora, and adverse effects between the two groups were compared. Results: The rate of ulcer surface healing and Hp eradication were higher in the study group than in the control group, with statistically significant differences (P<0.05). The levels of Clostridium perfringens, Bifidobacterium, and Lactobacillus were lower in the control group than before treatment (P<0.05), while the levels of Enterobacter, Yeast, and Enterococcus were higher (P<0.05) and the B/E values were lower (P<0.05). The levels of Bifidobacterium and Lactobacillus in the study group were higher than before treatment (P<0.05), the levels of Clostridium perfringens were lower than before treatment (P<0.05), and the levels of Enterobacter, Enterococcus and Yeast were not significantly different from before treatment (P>0.05). Conclusion: The probiotic-assisted estramiprazole-based quadruple regimen for the treatment of patients with H. pylori Hp-positive gastric ulcers significantly improves clearance rates, promotes healing of the ulcer surface, and has a positive effect on promoting a balanced intestinal flora environment, making it suitable for promotion.

1502 Study on the Current Status of Malnutrition Risk and Factors Affecting Patients with Type 2 Diabetes Mellitus Combined with Macroangiopathy

Objective: To explore the current situation and influencing factors of malnutrition risk in patients with type 2 diabetes mellitus complicated with macroangiopathy, to provide a basis for formulating malnutrition prevention and intervention measures. Methods: 554 patients with type 2 diabetes admitted to the Department of Endocrinology of our hospital from July 2020 to June 2021 were selected. After admission, whether they were complicated with the macrovascular disease was evaluated. At the same time, the nutritional risk of all patients was evaluated by NSR2002. Among the patients with the macrovascular disease, patients with NSR ≥ 3 were included in the risk group and patients with NSR<3 were included in the control group. The related data of the two groups were compared, and the related factors that may cause the risk of malnutrition were analyzed by logistic multifactor analysis. Results: Among 554 patients with type 2 diabetes in this study, 447 patients were complicated with macroangiopathy, and the detection rate of macroangiopathy was 80.7%; according to the evaluation of NSR2002, 138 of all patients had malnutrition risk, and the detection rate of malnutrition risk was 24.9%. Among 447 diabetes patients with the macrovascular disease, 122 patients at risk of malnutrition were included in the risk group, while the other 325 patients without malnutrition risk were included in the control group. There were significant differences in body mass index, blood urea nitrogen level, albumin level, prealbumin level, triglyceride level, and glycosylated hemoglobin level between the two groups. At the same time, the hospitalization time of patients in the risk group was significantly longer than that of patients in the control group; the difference was statistically significant (P<0.05). After the analysis of the logistic multivariate regression analysis model, it was found that low body mass index, low albumin, low triglyceride, and high glycosylated hemoglobin were the risk factors affecting the risk of malnutrition in patients, and the difference was statistically significant (P<0.05). Conclusion: Type 2 diabetes combined with macroangiopathy has a high detection rate and more patients are at risk of malnutrition. Active clinical interventions to identify associated risk factors are valuable in improving patient prognosis.

1506 Effects of Caffeine Citrate Combined with Mild Hypothermia on the Levels of S100B and MMP-9 in Premature Infants with Brain Injury

Objective: To observe the effect of caffeine citrate combined with mild hypothermia on the levels of S100B and MMP-9 in premature infants with brain injury. Methods: A total of 60 premature infants with brain injury who were treated in the hospital from May 2018 to May 2021 were selected and randomly divided into 2 groups. 30 children in the control group were given basic treatment and caffeine citrate and 30 children in the experimental group were given basic treatment, caffeine citrate combined with sublimation. The levels of S100B, matrix metalloproteinase-9 (MMP-9), neurofilament protein H phosphorylated isoform (pNF-H), glial fibrillary acidic protein (GFAP), tumor necrosis factor-α (TNF-α), Toll-like receptor-4 (TLR-4), interleukin-1β (IL-1β), superoxide dismutase (SOD), lipid peroxide (P-LPO) and reactive oxygen species (ROS) were measured before and after treatment. The levels of lipid peroxide (P-LPO) and reactive oxygen species (ROS) were recorded, and the time to recovery of primitive reflexes, time to recovery of muscle tone, time to recovery of consciousness, time to voluntary suckling, and days of hospitalization were recorded in both groups. Results: The level of TNF-α, TLR-4, and IL-1β were lower in the experimental group than in the control group (P<0.05), the level of SOD was higher in the experimental group than in the control group (P<0.05), the level of P-LPO and ROS were lower in the experimental group than in the control group (P<0.05), and the level of S100B, MMP-9, pNF-H, and GFAP water was lower in the experimental group than in the control group (P<0.05). The time to recovery of primitive reflexes, time to recovery of muscle tone, time to recovery of voluntary suckling, time to recovery of consciousness, and days of hospitalization were shorter in the test group than in the control group (P<0.05), and the NB-NA elevation score was higher in the test group than in the control group (P<0.05). Conclusion: Compared with the basic treatment and caffeine citrate treatment, the combination of caffeine citrate and subhypothermia treatment for preterm infants with brain injury can reduce inflammatory factors in the body, inhibit the inflammatory response, improve patients' oxidative stress levels, reduce oxygen-free radical damage, reduce S100B, MMP-9, pNF-H, and GFAP levels, enhance patients' neurological function and promote their recovery.

1511 Clinical Value of NLR MAGE and PNI on Predicting Intra-Abdominal Infection after Anatomical Hepatectomy

Objective: To explore the clinical value of the neutrophil-to-lymphocyte ratio (NLR), mean amplitude of glycemic excursions (MAGE), and prognostic nutritional index (PNI) in predicting the intra-abdominal infection after anatomical hepatectomy. Methods: 192 patients who underwent anatomical hepatectomy in the hospital were selected between October 2018 and October 2021, including 43 patients with postoperative intra-abdominal infection (infected group) and 149 patients without intra-abdominal infection (non-infected group). All patients received routine blood examination and blood glucose monitoring within 3 d before surgery. The NLR, MAGE, and PNI were calculated and the differences between groups were analyzed. The receiver operating characteristic curve (ROC curve) was used to evaluate the value of the three indicators for predicting the postoperative intra-abdominal infection. Results: There were statistical differences between the infection group and the non-infected group in terms of the presence of diabetes mellitus, surgical time, and indwelling time of the intra-abdominal drainage tube (P<0.05). NLR, MAGE, and PNI in the infected group before surgery were significantly higher than those in the non-infected group (P<0.05). The areas under the ROC curves of NLR and MAGEPNI for predicting intra-abdominal infection after anatomical hepatectomy were 0.825 (95%CI:0.763~0.876), 0.957 (95%CI:0.918~0.981) and 0.914 (95%CI:0.865~0.949) respectively, with good predictive efficiency. Logistics regression analysis showed that the presence of diabetes mellitus, surgical time≥ 5h, drainage tube indwelling time≥3d, NLR>2.35, MAGE>0.83, and PNI>43.79 were the risk factors for intra-abdominal infection after anatomical hepatectomy (P<0.05). Conclusion: Preoperative NLR, MAGE, and PNI have certain predictive value on postoperative intra-abdominal infection in patients with anatomical hepatectomy, and can provide a reference for clinical prevention and treatment.

1516 Quantitative Assessment of the Risk of Goiter-Derived Airway Compression Based on Ultrasound Volumetric Imaging

Objective: To evaluate whether thyroid ultrasound volume imaging can quantitatively evaluate the risk of airway compression caused by goiter. Methods: The data of 84 subjects who underwent thyroid ultrasound volume imaging and airway (neck) CT imaging from January 2017 to December 2020 were collected retrospectively. The render surface mode was adopted in the three-dimensional ultrasonic image analysis. The thin layer imaging assisted multiplanar reconstruction (MPR), curved surface reconstruction (CPR), and volume reconstruction (VR) were used for airway CT imaging. Results: There was a significant difference in thyroid TV between the airway stenosis group and the non-stenosis group (t= 13.363,P<0.001), and it was significantly correlated with the degree of airway stenosis (r= 0.549,P<0.05). The differential efficacy of thyroid volume for airway compression was 0.679 (95%CI: 0.599-0.753,P<0.001), and the differential efficacy for tracheal stenosis was 0.658, 0.679, 0.702 and 0.787, respectively. The cut-off values were 19.69ml, 20.98ml, 23.85ml, and 29.37ml. Conclusion: Thyroid ultrasound imaging can accurately and quantitatively evaluate the risk of airway compression, which can be used as a reliable theoretical basis for the prevention and treatment of airway compression.

1519 Effect of Ultrasound-Guided Supraclavicular Complex Subclavian Brachial Plexus Nerve Block on Diaphragmatic Function in Elderly Patients

Objective: To observe the effect of ultrasound-guided supraclavicular with infraclavicular brachial plexus block on diaphragmatic function in the elderly. Methods: A total of sixty patients, scheduled for the right upper limb fracture, aged 65-79 years, falling into ASA physical status Ⅱ or Ⅲ, were selected and Simple randomly divided into two groups. Group S (ultrasound-guided supraclavicular brachial plexus block) received ultrasound-guided supraclavicular brachial plexus block with 0.33% ropivacaine 30ml; Group C (ultrasound-guided supraclavicular with infraclavicular brachial plexus block) received ultrasound-guided supraclavicular brachial plexus block with 0.33% ropivacaine 10ml, then received infraclavicular brachial plexus block with 0.33% ropivacaine 20ml. The hemi-diaphragmatic movement was measured by M-mode ultrasonography under a quiet and deep breathing test before the block procedure and 30min after the block. The sensory block of the main nerve roots of the brachial plexus, the onset time and duration of motor block were recorded and the occurrence of adverse reactions was observed. Results: The onset time of sensory block and maintaining time of sensory block or motor block had no statistical difference between the two groups. Compared with Group C, the onset time of motor block in Group S was significantly shorter (P<0.05). There was a statistically significant difference in the degree of diaphragm paralysis between the two groups under quiet (P<0.05)and deep(P<0.05) breathing tests 30min after the block。Conclusion:The combination of ultrasound-guided supraclavicular brachial plexus block with infraclavicular brachial plexus block can provide good anesthesia, decrease ipsilateral diaphragmatic paralysis, and is a reasonable clinical use for the right upper limb fracture in the elderly.

1524 Logistic Regression Analysis of Risk Factors of Osteoporosis in Patients with Diabetic Kidney Disease

Objective: To analyze the related risk factors of osteoporosis in patients with diabetic kidney disease. Methods: 243 patients with DKD treated in the hospital between September 2018 and September 2021 were selected. According to the examination results of the X-ray bone mineral density meter, the patients were divided into the osteoporosis group (OP group, 119 cases) and the non-osteoporosis group (NOP group, 124 cases). The general data of the two groups of patients were compared, and multivariate Logistic analysis was used to analyze the related factors affecting the occurrence of OP in patients with DKD. ROC curve was plotted to analyze the diagnostic value of Klotho and FGF-23 on the occurrence of OP in patients with DKD. Results: There were statistically significant differences between the OP group and the NOP group in terms of gender, GFR, BMI, Klotho, and FGF-23 (P<0.05). Multivariate Logistic analysis showed that gender and FGF-23 were risk factors that affected the occurrence of OP in patients with DKD, and Klotho was a protective factor affecting the occurrence of OP in patients with DKD (P<0.05). ROC curve analysis results showed that when the cut-off values of Klotho and FGF-23 were 333.33ng/L and 347.46ng/L, the areas under the curves of Klotho and FGF-23 in diagnosing OP in patients with DKD were 0.700 (95% CI: 0.635~0.765), 0.659 (95% CI: 0.591~0.728), and the sensitivities and specificities were 59.7% and 71.8%, 56.3% and 72.6% respectively. Conclusion: Female and high FGF-23 levels are risk factors to promote OP in patients with DKD, and a high Klotho level is a protective factor to inhibit OP in patients with DKD. Klotho and FGF-23 have good diagnostic value on OP in patients with DKD.

1529 Cosmetic Results of Glass Fiber Post and Resin Core in Restoration of Maxillary Anterior Teeth and Analysis of Dental Plaque Index and Stability

Objective: To investigate the cosmetic results of glass fiber post and resin core in the restoration of maxillary anterior teeth, and analyze the dental plaque index and stability. Methods: A total of 226 patients who underwent a restoration of maxillary anterior teeth in the hospital from May 2017 to June 2019 were selected, and randomly divided into control group and study group, with 113 cases in each. The control group received restoration with metal post and core, while the study group received restoration with glass fiber post and resin core. Cosmetic results, periodontal indicators, masticatory function, and fracture resistance were compared between the two groups. Results: The pink esthetic scores (PES) and white esthetic scores (WES) of the two groups increased over time, with an interaction effect between groups (P<0.05). The above scores showed no significant difference at 3 months after restoration (P>0.05). The study group had higher PES scores and WES scores than the control group at 12 months after restoration (P<0.05). Gingival index, bleeding index, plaque index, and probing depth in the study group were lower than those in the control group at 3 months after restoration. The differences before and after restoration were significantly greater than those of the control group (P<0.05). The excellent and good rate of masticatory function in the study group at 12 months after restoration (95.58%) was higher than that in the control group (87.61%) (P<0.05). The incidence rates of post fracture, post loosening, and falling off in the study group during 2 years of follow-up (1.77% and 3.54%) were lower than those in the control group (7.96% and 11.50%) (P<0.05). Conclusion: The application of glass fiber post and resin core in the restoration of maxillary anterior teeth can improve the cosmetic results and periodontal index, reduce dental plaque index, and improve stability.

1534 Analysis of the Short- and Long-Term Efficacy of Laparoscopic Transabdominal Preperitoneal Hernia Repair in Adults with Recurrent Inguinal Hernia

Objective: To analyze the short-term and long-term curative efficacy of transabdominal preperitoneal (TAPP) for recurrent inguinal hernia in adults. Methods: This study is a retrospective clinical study, and ninety-eight adult patients with recurrent inguinal hernia treated in our hospital from January 2019 to January 2020 were enrolled in the research and divided into study group (TAPP, n = 60) and control group (open tension-free hernia repair, n = 38) according to the operation method. The perioperative data, hospitalization length, hospitalization expense and postoperative recurrence of the two groups were compared. Results: All patients successfully completed the operation. The amount of intraoperative bleeding, postoperative eating time, postoperative first exhaust time, first time out of bed and hospital stay in the observation group were significantly lower than those in the control group, and the hospitalization cost was significantly higher than that in the control group (P<0.05). There was one case of recurrence in the observation group and the control group respectively. There was no significant difference in the incidence of postoperative complications and recurrence between the two groups (P>0.05). Conclusion: Compared with traditional open tension-free hernioplasty, TAPP has a better short-term effect on recurrent inguinal hernia in adults, with less operative trauma, faster postoperative recovery. Their surgical safety and long-term efficacy is equivalent, and the postoperative recurrence rates were both lower.

1537 Analysis of the Influencing Factors of Enamel Demineralization and the Effect of Fluoride Intervention in Adolescents after Orthodontic Treatment

Objective: To explore the influencing factors of enamel demineralization and the effect of fluoride intervention in adolescents after orthodontic treatment. Methods: The clinical data of 89 patients who received orthodontic treatment in our hospital from March 2020 to March 2021 were retrospectively analyzed. According to whether enamel demineralization occurred after treatment, they were divided into a demineralization group (43 cases) and a non-demineralization group. (46 cases), the general data of the two groups of patients were compared, and the multivariate Logistic regression analysis was used to analyze the risk factors affecting the occurrence of enamel demineralization in patients. According to the different treatment methods of 43 demineralized patients, they were divided into groups A and B. The 23 patients in group A were treated with fluoride, and the 20 patients in group B were treated with routine oral hygiene protection measures. The clinical efficacy and pre-orthodontic treatment were compared between the two groups, as well as 12 months after orthodontic treatment enamel demineralization index (Enamel Demineralization Index, EDI), plaque index (Plaque index, PLI), bleeding index (bleeding index, BI). Results: The results of multivariate Logistic regression analysis showed that the frequency of tooth brushing, the use of fluoride toothpaste, the intake of sweets, and the consumption of carbonated beverages were independent risk factors for enamel demineralization after orthodontic treatment, P<0.05; The total effective rate of group A was 95.65% (22/23), which was significantly higher than that of group B, 70.00% (14/20), P<0.05; 12 months after orthodontic treatment, the EDI of the two groups of patients was 0.22±0.04, PLI (0.22±0.04), BI (1.47±0.35) levels were higher than those before orthodontic treatment, and the levels in group A were lower than those in group B (0.35±0.06, 2.13±0.45, 2.60±0.47), P<0.05. Conclusion: Demineralization of tooth enamel often occurs after orthodontic treatment in adolescents, which is related to the frequency of brushing, the use of fluoride toothpaste, the intake of sweets, and the consumption of carbonated beverages. Fluoride treatment can significantly improve enamel demineralization, which is worthy of clinical reference.

1541 Analysis of the Relationship and Predictive Value between the Type of Alveolar Bone Defect and the Postoperative Outcome of GBR

Objective: To explore the relationship between the type of alveolar bone defect and the postoperative effect of guided bone regeneration (GBR). Methods: A total of 82 patients who received implants in the maxillary anterior region and underwent GBR during the same period from October 2019 to October 2021 in the Department of Stomatology of Dongguan People's Hospital were retrospectively selected as the research subjects. The bone graft resorption rate was measured by cone beam CT (CBCT) in all patients after implantation. The three-dimensional structure reconstruction of bone defect was carried out according to the patient's CBCT data, the relationship between Pearson-related bone defect morphological variables and bone graft resorption rate, and the ROC curve was used to analyze the predictive value of primary bone defect morphological variables on bone graft resorption rate. Results: Six months after GBR, the average absorption rate of 82 patients was (46.8±10.52)%. After reconstruction of the initial bone defect by CBCT, the calculation showed that the mMD was (5.39±1.58) mm, the mCA was (5.24±1.08) mm, the mBL was (5.06±1.25) mm, the rmCA/mMD was (1.03±0.29), and the rmCA was (5.06±1.25) mm. /mBL was (1.07±0.27), and rmBL/mMD was (1.00±0.33). Pearson correlation analysis showed that mCA and mBL were negatively correlated with bone graft resorption rate (r=-0.408, -0.496, P<0.05); rmCA/mMD, rmCA/mBL were positively correlated with bone graft resorption rate (r=0.636, 0.418, P<0.05). Univariate and multivariate results showed that mBL and rmCA/mMD were the influencing factors of bone graft resorption rate (P<0.05). In addition, the ROC curve showed that the AUCs of mBL and rmCA/mMD for predicting bone graft resorption rate ≥45.1% were 0.836 and 0.812, respectively, and the 95% CIs were 0.743-0.921 and 0.722-0.902, respectively. Conclusion: The initial bone defect morphology was closely related to the effect of bone grafting after GBR, and the morphological characteristic variables of the bone defect had better value in predicting the postoperative bone graft resorption rate.

1546 The Effect of Digital and Alloy Splint Impressions on the Marginal Fit of Implant Crown and Bridge Restorations

Objective: To compare the effect of digital impression and alloy splint impression on the edge of implant crown and bridge restoration. Methods: 120 patients with crown and bridge implantation from January 2019 to January 2021 in our hospital were prospectively selected as the subjects, and randomly divided into observation group (alloy splint impression) and control group (digital impression), with 60 cases in each group. The two groups were observed for 3 months, and the periodontal indexes of the two groups before and after treatment were compared. Patients were followed up for the incidence of defective restorations and the degree of closeness of the edges of the restorations and surveyed on satisfaction. Results: There was no significant difference in periodontal indexes between the two groups before treatment (P<0.05); the plaque index, periodontal pocket depth, gingival index and bleeding index in the observation group were lower than those in the control group after treatment, and the differences were statistically significant (P<0.05); the ratio of prostheses that were too long or too short and early contact in the observation group (3.3%, 5.0%) was significantly lower than that of the control group (13.1%, 16.7%), the difference was statistically significant (P<0.05); there was no statistically significant difference in the incidence of overhang and over-loose or over-tightening (P>0.05); the observation group had lower edge consistency at A1, A2, A3, A4 and A5 points than the control group, and the difference was statistically significant (P<0.05); the patient satisfaction in the observation group (95.0%) was significantly higher than that in the control group (80.0%), and the difference was statistically significant (P<0.05). Conclusion: Compared with digital impressions, alloy splint impressions can improve the tightness of the edge of implant crown and bridge restorations, which improves the patient's periodontal indicators, reduces the occurrence of too long or too short restorations, and early contact, etc, and patient satisfaction is high, which is worthwhile for application.

Objective: To screen the quantitative parameters of ontrast-enhanced ultrasound (CEUS) and shear wave elastography (SWE) in diagnosing peripheral neuropathy (PN) in type 2 diabetes mellitus (T2DM) based on Logistic regression model. Methods: Fifty patients with T2DM complicated with PN treated in our hospital from January 2019 to June 2021 were selected as PN group, 50 patients with T2DM without PN were selected as non-PN group, and 50 healthy adults were selected as control group. The conventional two-dimensional ultrasound examination [transverse diameter, anterior posterior diameter and cross-sectional area], SWE examination [elastic maximum value (Emax), elastic minimum value (Emin) and elastic mean value (Emean)], and CEUS examination [time to peak (TTP), intensity maximum value (Imax) and area under curve (AUC)] were compared among the three groups. Multivariate logistic regression analysis was performed to screen the ultrasonic examination parameters related to PN. Results: The transverse diameter, anterior posterior diameter, cross-sectional area, Emax, Emin, Emean and TTP of PN group were significantly higher than those in non-PN group and control group, the Imax and AUC of PN group were significantly lower than those in non-PN group and control group, the transverse diameter, anterior posterior diameter, cross-sectional area, Emax, Emin, Emean and TTP of non-PN group were significantly higher than those of control group, and the Imax and AUC of non-PN group were significantly lower than those of control group (P<0.017). Multivariate logistic regression analysis showed that Emean, TTP and Imax were the independent related factors of PN (P<0.05). Conclusion: CEUS parameters (including TTP, IMAX) and SWE parameters (mainly Emean) of tibial nerve have important diagnostic value for PN in T2DM patients, which is worthy of clinical application.

1554 Analysis of Risk Factors for the Development of Otitis Media with Effusion Following Endoscopic Surgery for Chronic Rhinosinusitis

Objective: To investigate the risk factors of otitis media with effusion (OME) following endoscopic surgery for chronic rhinosinusitis (CRS). Methods: The clinical data of 218 patients with CRS endoscopic surgery admitted to the hospital from February 2016 to February 2020 were reviewed. The incidence of OME in patients with CRS after endoscopic surgery was calculated. The risk factors that may influence OME in patients with CRS after endoscopic surgery were summarized and the ratio of possible risk factors in patients with and without CRS was compared. Logistic regression analysis was used to investigate the risk factors for OME in CRS patients after endoscopic surgery. Results: In this group, there were 16 patients with postoperative OME, with an incidence of 7.34%. The patients with a history of allergic rhinitis, recurrent respiratory tract infection, poor mastoid gasification, nasal polyp, and nasal decongestant used for a long time before surgery composition ratio were all higher than those without (P<0.05). The proportion of patients with regular postoperative surgical cavity cleaning, postoperative intranasal glucocorticoids, postoperative antibiotics, and mucus thinning agents was lower than that of those without (P<0.05). Logistic regression analysis showed that a history of allergic rhinitis, recurrent respiratory infections, poor mastoid gasification, nasal polyps, and nasal decongestant used for a long time before surgery were independent risk factors for OME after CRS endoscopic surgery (P<0.05). Postoperative regular intraoperative cavity cleaning, postoperative intranasal glucocorticoids, postoperative antibiotics, and mucus thinning agents were all protective factors (P<0.05). Conclusion: CRS patients are complicated with OME endoscopic surgical management risk, and the history of allergic rhinitis repeated respiratory infection, mastoid gasification, nasal polyps, nasal decongestant used for a long time before surgery are the risk factors, and postoperative regular cavity cleaning, postoperative application of intranasal corticosteroids, postoperative application of antibiotics, postoperative mucus thinning agent were the protection factors, to strengthen the prevention and control.

1560 Comparison of the Value of High-Resolution MRI and MDCT in the Diagnosis of Rectal Mesenteric Lymph Node Metastases

Objective: To study the value of high-resolution MRI and MDCT in the diagnosis of rectal mesorectal lymph node metastasis. Methods: From March 2018 to December 2021, 147 patients with rectal cancer diagnosed by pathological biopsy in our hospital were selected for a retrospective study, all of whom were diagnosed by high-resolution MRI and MDCT. The parameters of MDCT and MRI were analyzed. The innovation of this study is to compare the diagnostic efficiency of MDCT and MRI with the receiver characteristic curve (ROC curve) and pictures, to improve the diagnostic efficiency, and to provide a visual comparison of the diagnostic efficiency, which provides practical value for the diagnosis of mesorectal lymph node metastasis. The receiver characteristic curve (ROC curve) and pictures were used to compare the diagnostic performance of MDCT and MRI. The consistency between MDCTT staging and pathological diagnosis staging, MRIT staging, and pathological diagnosis staging were analyzed. Results: The SUV (2.25±0.76), the ratio of short to long diameter (0.86±0.14), the number of lymph nodes (7.88±1.45), and the size of lymph nodes (0.67±0.12) cm in the metastasis group were higher than those in the non-metastatic group [1.26 ±0.34, 0.74±0.13, (3.56±1.57), (0.51±0.13) cm]; the difference was statistically significant (P<0.05). The ADC (0.81±0.11) 10-3mm2/s in the metastatic group was lower than the non-metastasis group (1.02±0.16) 10-3mm2/s, which was statistically significant (P<0.05), while the periintestinal fat mixed signal sign [76 (87.4 %)], serous surface infiltration [77 (88.5%)], and venous wrapping sign [77 (88.5%)] were higher than those in the non-metastatic group [respectively 2 (3.3%), 2 (3.3%), 11 ( 18.3%)]; the difference was statistically significant (P<0.05). The AUC of MDCT and MRI were 0.887 and 0.877, respectively, and the difference was not statistically significant (Z=-1.000, P=0.317). The sensitivity was 0.839 and 0.850, respectively, and the difference was not statistically significant (χ2=2.000, P=0.157). The sexes were 0.983 and 0.839, respectively, and the difference was not statistically significant (χ2=2.000, P=0.157). The Kappa values of MDCT and MRI were 0.873 and 0.863, respectively, and the consistency of pathological diagnosis and staging showed strong consistency. Conclusion: Both high-resolution MRI and MDCT can diagnose rectal cancer mesorectal lymph node metastasis, which have high diagnostic efficiency and are suitable for clinical use.

1564 Correlation between Connexin 45 and Clinicopathological Features and Prognosis of Colorectal Cancer Patients

Objective: To analyze the correlation between connexin 45 (Cx45) and clinicopathological features and prognosis of colorectal cancer patients. Methods: 97 patients with colorectal cancer admitted to our hospital from January 2016 to January 2019 were selected, and the expression levels of Cx45 in their focus tissues, adjacent tissues, and metastatic lymph nodes were detected by tissue microarray technology. The expression differences of Cx45 in focus tissues of patients with different clinicopathological features and different prognoses were compared, and the relationship between the expression characteristics of Cx45 in focus tissues and clinical pathological features and prognosis of patients were analyzed. Results: A total of 97 lesions, 97 adjacent tissues and 86 metastatic lymph node tissues were obtained. The high expression rate of Cx45 in lesions and metastatic lymph nodes was higher than that in adjacent tissues, and the low expression rate was lower than that in adjacent tissues, with statistical significance (P<0.05). The expression of Cx45 in rectal cancer patients with different lymph node metastasis, different degree of differentiation, and different TNM staging rhythm was statistically significant (P<0.05). The overall survival time (OS) and disease-free survival time (PFS) of patients with high expression of Cx45 were shorter than those with low expression of Cx45, and the difference was statistically significant (P<0.05). Cox regression model showed that lymph node metastasis, low differentiation degree, TNM stage Ⅲ ~ Ⅳ, and high expression of Cx45 in lesion tissues were independent risk factors for OS in patients with colorectal cancer (P<0.05). Conclusion: The expression of Cx45 in colorectal cancer patients with different lymph node metastasis status, differentiation degree, and TNM stage are significantly different. Besides the clinicopathological features, the high expression of Cx45 in colorectal cancer patients is also directly related to the shortening of OS.

1569 Effect of Celecoxib on Stress Response and DVT in Patients Undergoing Knee Arthroplasty

Objective: To study the effect of oral celecoxib on stress response and DVT after knee joint replacement. Methods: From January 2016 to December 2020, 134 patients undergoing knee joint replacement in our hospital were taken as the research subject. All patients were randomly divided into an observation group (n=67) and a control group (n=67) according to the principle of stratified sampling. Patients in the control group were given routine intravenous patient-controlled analgesia combined with heparin sodium injection immediately after the operation, while those in the observation group were given oral celecoxib based on the control group. After 7 consecutive days of treatment, the contents of CA, Cor, and ACTH were measured by ELISA, and the stress response before and after treatment was compared. The coagulation function of fibrinogen and D-dimer was measured by C2000-A automatic blood coagulation analyzer produced by Pulisen Company. Adverse events, such as ecchymosis, digestive tract reaction, fatigue, and DVT occurrence probability were compared among all patients during treatment. Results: Before treatment, there was no significant difference in serum CA, Cor, ACTH levels, serum fibrinogen, and D- dimer between the two groups (P>0.05). After treatment, the levels of CA, Cor, ACTH, and the contents of serum fibrinogen and D-dimer in the two groups were significantly higher than those before treatment. Compared with the control group, the levels of CA, Cor, ACTH, and the contents of serum fibrinogen and D-dimer in the observation group were significantly lower than those in the control group (P<0.05). There was no significant difference in the incidence of adverse reactions in the observation group during treatment (P>0.05). The incidence of DVT in the observation group was 1.49%, which was significantly lower than that in the control group (10.45%), and the difference was statistically significant (P<0.05). Conclusion: Oral celecoxib treatment for patients after knee joint replacement can effectively reduce the stress reaction caused by operation and the risk of DVT, with good safety performance and remarkable treatment effect, which is worthy of clinical application.

1573 Analysis of Effect of Jintiange Capsule Combined with Calcitriol on Patients with Osteoporotic Vertebral Compression Fracture

Objective: To analyze the effect of Jintiange capsule combined with calcitriol in patients with osteoporotic vertebral compression fracture (OVCF). Methods: Totally 150 patients with OVCF admitted to our hospital from January 2017 to June 2021 were randomly divided into the combined group and the control group with 75 cases in each group. Patients in both groups received percutaneous vertebroplasty (PVP), the control group was given calcitriol orally after the operation, and the combined group was given Jintiange capsule orally for 6 months. The changes in the visual analog scale of pain (VAS), Oswestry dysfunction index (ODI), the height of the anterior edge of the vertebral body, Cobb angle of injured vertebral body, bone mineral density, and bone metabolism index were compared between the two groups. Results: Six months after treatment, the VAS score, ODI index, Cobb angle, and RANKL of the two groups decreased compared with those before the operation, while the height of the vertebral front, the bone mineral density of L1~L4, the bone mineral density of femoral neck and OPG increased. The VAS score, ODI index, Cobb angle, RANKL, the height of the anterior edge of the vertebral body, the bone mineral density of L1~L4, the bone mineral density of the femoral neck, and OPG in the group were all higher than those in the control group at 6 months after combined treatment, and the difference was statistically significant (P<0.05). Conclusion: Based on oestriol, the combination of Jintiange capsules in the postoperative adjuvant treatment of OVCF patients with PVP not only helps to alleviate patients' clinical symptoms and promotes the recovery of vertebral height but also improves patients' bone density and bone metabolism, achieving both symptomatic and primary treatment.

1577 Effectiveness of Extract of Ginkgo Biloba Leaves Injection Combined with Doperidonezide in the Treatment of Vascular Dementia in the Elderly and Its Effect on Cognitive Function and Oxidative Stress Response

Objective: To explore the effect of extract of ginkgo biloba leaves injection combined with donepezil on senile vascular dementia and its influence on cognitive function and oxidative stress. Methods: The data of 90 aged patients who had vascular dementia treatment in our hospital from May 2019 to December 2021 were retrospectively analyzed. All patients were divided into two groups, the observation group (n=45) and the control group (n=45), according to different treatment methods. People in the control group were administered donepezil hydrochloride tablets, while the people in the observation group were administered extract of ginkgo biloba leaves injection combined with donepezil hydrochloride tablets. All patients were treated continuously for 2 months. The therapeutic effect, cognitive function, self-care ability, and indexes related to oxidative stress were compared before and after treatment. Results: The total effective rate of treatment in the observation group was significantly higher than that in the control group (P<0.05). After treatment, the scores on the MMES scale and Barthel index in the two groups were significantly higher than those before treatment. After treatment, the contents of serum homocysteine (Hcy), interferon-γ (IFN-γ), and malondialdehyde (MDA) in both groups were significantly lower than those before treatment, while the contents of superoxide dismutase (SOD) in both groups were significantly higher than those before treatment. Conclusion: Extract of ginkgo biloba leaves injection combined with doxepinezi can improve the cognitive function of elderly patients with vascular dementia, improve self-care ability, and reduce the content of oxidative stress reaction index. The therapeutic effect is remarkable, and it is worthy of clinical application.
