1233 Biological Significance of High Expression of Serum Exosome MiR-96-5p to Suppress FoxO1 in Patients with Head and Neck Squamous Carcinoma

Objective: To explore the effect and molecular mechanism of Exos-miR-96-5p on the malignant biological behaviors of HNSCC cells such as proliferation, invasion and migration by confirming the targeted regulation of miR-96-5p in serum exosomes (Exos) of head and neck squamous cell carcinoma (HNSCC) patients on forkhead box transcription factor O subfamily 1 (FoxO1). Methods: Serum specimens of HNSCC patients (25 cases) admitted to our hospital from September 2020 to January 2022 and healthy individuals (40 cases) examined during the same period were extracted by ultracentrifugation, and miR-96-5p expression in Exos was detected by RT-PCR; after labeling Exos with PKH67 dye, they were co-cultured with HNSCC cell line-AGZY-973 to observe the effect of HNSCC cells on Exos uptake. AGZY-973 cells were divided into: Control group (normal culture with DMEM medium), HNSCC patient serum Exos group (co-culture with DMEM medium and 50μL HNSCC patient serum Exos suspension), healthy control serum Exos group (co-culture with DMEM medium and 50μL healthy control serum Exos suspension). The cell proliferation was detected by CCK-8 method; apoptosis rate was detected by flow cytometry; migration and invasion were detected by Transwell; miR-96-5p expression was detected by RT-PCR; FoxO1 and pro-apoptotic protein Bax, death mediator (Bim) and apoptotic protein p21 expression were detected by Western blot. Dual luciferase reporter assay was performed to verify the relationship between miR-96-5p and FoxO1 targeting; 36 nude mice were divided into Control group, Exos group, Exos-miR-96-5p inhibitor group, Exos-inhibitor-NC group, Exos-pcDNA FoxO1 group, Exos-pcDNA NC group, 6 in each group. The control group was inoculated with AGZY-973 cell culture, and the remaining groups were inoculated with 50 μL of Exos suspension of HNSCC patient serum, Exos-miR-96-5p inhibitor suspension, Exos-inhibitor-NC suspension, Exos-pcDNA-FoxO1 suspension, and Exos-pcDNA-NC suspension in AGZY-973 cell culture, respectively, and after 28 days, tumor volume and tumor weight were examined to verify the effect of serum Exos on miR-96-5p and FoxO1 regulation in HNSCC patients. Results: Serum Exos of HNSCC patients and healthy controls had intact membranes with a cup-shaped morphology, and both could be taken up by AGZY-973 cells. The expression of miR-96-5p in serum Exos of HNSCC patients was higher than that in serum Exos of healthy controls (P<0.05). Compared with the Control group, the serum Exos group of HNSCC patients had elevated miR-96-5p expression, decreased expression of FoxO1 and apoptotic proteins such as Bax, Bim and p21, elevated cell proliferation, migration and invasion ability, and decreased apoptosis rate (P<0.05); serum Exos of healthy controls had higher miR-96-5p expression on AGZY-973 cells miR-96-5p/FoxO1 axis and biological behavior were not significantly affected (P>0.05). MiR-96-5p had a target-regulatory relationship with FoxO1. Transfection of miR-96-5p inhibitor or pcDNA-FoxO1 in serum Exos of HNSCC patients partially reversed the pro-tumor growth effect exerted by Exos (P<0.05). Conclusion: Serum Exos of HNSCC patients can target down FoxO1 by transmitting miR-96-5p, and plays a role in promoting the development of malignant biological behaviors such as proliferation, invasion, migration and growth of HNSCC cells.

1240 Mechanism of Entinostat Inhibits Proliferation Migration and Invasion of Oral Squamous Cell Carcinoma Cells by Regulating miR-103a-3p/PIEZO1 Pathway

Objective: To observe the effect of entinoslate on the proliferation, migration and invasion of oral squamous cell carcinoma cell line WSU-HN6, and to explore the potential relationship between its mechanism and miR-103a-3p / mechanically sensitive ion channel protein (PIEZO1) pathway. Methods: WSU-HN6 cells were treated with entenolate (0.0, 0.5, 1.0, 2.0, 4.0 μmol/L) for 24, 48 and 72 hours. The optimum concentration was 2.0 μmol/L and the optimum treatment time was 48 hours. The WSU-HN6 cells were transfected by liposome method. Cell proliferation rate was detected by cell counting kit (CCK8), and cell migration and invasion were detected by Transwell test. Western blot (WB) was used to detect E-cadherin, N-cadherin and PIEZO1 protein; the expression of miR-103a-3p and PIEZO1 was detected by real-time fluorescence quantitative polymerase chain reaction (RT-qPCR);the binding ability of miR-103a-3p to PIEZO1 was detected by double luciferase reporter gene experiment. Results: Entinostat (0.5, 1.0, 2.0, 4.0 μmol/L) inhibited the proliferation of WSU-HN6 cells in a concentration and time-dependent manner. The optimal concentration was 2.0 μmol/L for 48 hours. Compared with the 0.0 μmol/L group, the amount of cell migration and invasion significantly decreased, the expression of E-cadherin significantly increased, the expression of N-cadherin significantly decreased, the expression of miR-103a-3p significantly increased, and the expression of PIEZO1 significantly decreased in the 2.0 μmol/L group (P<0.05). After inhibiting miR-103a-3p, the cell proliferation rate, migration and invasion were all significantly reduced, the expression of E-cadherin was significantly reduced, and the expression of N-cadherin was significantly increased, and the overexpression of PIEZO1 had similar functions. The double luciferase report experiment showed that miR-103a-3p targeted and negatively regulated the mRNA and protein expression of PIEZO1. Knockdown of PIEZO1 significantly enhanced the inhibitory effect of entinostat on cell proliferation, migration and invasion, and weakened the promoting effect of inhibiting miR-103a-3p on cell proliferation, migration and invasion. Conclusion: Entinostat inhibits the proliferation, migration and invasion of oral squamous cell carcinoma cells. The potential mechanism of this anticancer effect may be related to the up regulation of miR-103a-3p and the inhibition of PIEZO1.

1247 Effects of Palmatine on Proliferation Apoptosis and Cisplatin Sensitivity in BRAF/KRAS Mutant Non-Small Cell Lung Cancer Cells

Objective: To investigate the protective effect and mechanism of Palmatine on proliferation, apoptosis and cisplatin sensitivity in BRAF/KRAS mutant non-small cell lung cancer cells (NSCLC). Methods: The BRAF/KRAS mutant NSCLC cell line A549 was used as the research subject, and the effects of Palmatine on cell proliferation, apoptosis and cisplatin sensitivity were detected by thiazolyl blue method and flow cytometry, respectively. Western blotting was used to detect the expression of Bcl-2 and PARP protein. Results: Compared with the control group, the proliferation viability, Bcl-2 and PARP protein expression were significantly lower and the apoptosis rate was significantly higher in the 1 μmol/L, 5 μmol/L and 10 μmol/L groups (P<0.05). Compared with the control group, the proliferation viability, Bcl-2 and PARP protein expression were significantly reduced and the apoptosis rate was significantly increased in the palmatine and cisplatin groups (P<0.05); compared with the cisplatin group, the proliferation viability, Bcl-2 and PARP protein expression were further reduced and the apoptosis rate was further increased in the cisplatin+palmatine group (P<0.05). Conclusion: Palmatine can inhibit the proliferation of BRAF/KRAS mutant NSCLC cells, induce apoptosis, and enhance the sensitivity of cisplatin.

1251 Ramipril Reduces Blood Pressure in SHR Rats by Activating ERK1/2-Cx43 Signaling Pathway

Objective: To observe the effects of angiotensin converting enzyme inhibitor Ramipril (RMP) on blood pressure in spontaneously hypertensive rats (SHR) and explore the mechanism.Methods: Fifty male SHR rats were randomly divided into SHR group, RMP+SHR group, RMP+ gamperic acid [CBX, GAP junction 43 (Cx43) blocker] +SHR group, RMP+CBX+U0126[extra cellular signal-regulated kinase 1/2 (ERK1/2) inhibitor]+SHR, n=10/ group. control group (n=10) and SHR group were given normal saline 40mL/kg i.v. The other three groups were given RMP 10mg/kg i.v., RMP 10mg/kg+CBX 13.5mg/kg i.v., RMP 10mg/kg +CBX 13.5mg/kg+U012612mg/kg i.v., respectively. After 48 h of administration, BP-6 rat noninvasive blood pressure tester, hematoxylin eosin staining and Western blot analysis were used to observe the changes of physiological indexes of rats.Results: Compared with control group, SHR group showed hypertension symptoms (all P<0.05), and the phosphorylation level of Ser368 Cx43 (p-Cx43) and phosphorylation of ERK1/2 (p-ERk1/2) were down-regulated (all P<0.05). Compared with SHR group, the blood pressure of RMP+SHR group was decreased (all P<0.05), and the expressions of p-Cx43 and p-ERK1/2 were increased (all P<0.05). Compared with RMP+SHR group, the blood pressure of RMP+CBX+SHR group was significantly increased, and the expression of P-Cx43 was decreased (all P<0.05), while the expression of p-ERK1/2 was not significantly changed (P>0.05), while the blood pressure of RMP+U0126+SHR group was significantly increased. The expression of p-Cx43 and p-ERK1/2 was decreased (P<0.05). Compared with RMP+CBX+SHR group or RMP+U0126+SHR group, the blood pressure in RMP+CBX+U0126+SHR group was increased, and the expression of p-Cx43 and p-ERK1/2 was decreased (all P<0.05). Conclusion: This study confirmed that RMP can improve the symptoms of hypertension in SHR by activating ERK1/2 -Cx43 signaling pathway.

1256 Effects of LYN on Proliferation and Apoptosis of Gastric Cancer Cells through AKT/mTOR Signaling Pathway

Objective: To investigate the expression level of LYN in gastric cancer and the effect and mechanism of knockdown LYN expression on proliferation and apoptosis of gastric cancer cells.Methods: Immunohistochemistry was used to detect LYN expression in gastric cancer and adjacent tissues.Shrna-LYN was transfected into AGS (gastric adenocarcinoma) cells using Lipofectamine 2000, and untransfected gastric cancer cells were used as control. CCK8 and flow cytometry were used to detect the effects of LYN knockdown on AGS cell proliferation and apoptosis, respectively.Western blot analysis was used to investigate the changes in the expression levels of AKT/mTOR signaling pathway related proteins and apoptosis related proteins Bcl2, Bax, Caspase9 and Casapse3 induced by LYN knockdown, and to study its proliferation and apoptosis mechanism.Bioinformatics software was used to screen out the target gene miR-496, and western blot was used to detect the effect of transfection miR-496 on LYN expression. Rescue experiments were performed to analyze the interaction between miR-496 and LYN. Results: The sample number of LYN overexpression in gastric cancer tissues was 58 cases, which was significantly higher than that in paracancer tissues (28 cases) (P<0.05). After transfection 48 and 72h, the absorbance values of sh-LYN group were (0.0992±0.0346) and (1.0953±0.0379), which were significantly lower than those of NC group (1.4123±0.1161) and (1.6717±0.0120),(P<0.05).After transfection, the apoptosis rate of sh-LYN group (16.3567±0.5556) was significantly higher than that of NC group (P<0.05). After LYN knockdown, the expression levels of Bcl2 (0.4629 ± 0.0294), P-Akt (0.5565 ± 0.0473), P-MTOR (0.6167 ± 0.0459) and p70S6K (0.6049 ± 0.0570) decreased compared with NC control group. The expression of Bax (1.407 ± 0.0528), Caspase-9 (1.5200 ± 0.1063), caspase-3 (1.4170 ± 0.0751) increased. Transfection of miR-496 could inhibit LYN expression in AGS cells. Conclusion: Knockdown LYN expression can inhibit AKT/mTOR signaling pathway, thereby inhibiting proliferation and promoting apoptosis of gastric cancer cells, and its effect on gastric cancer cells is regulated by miR-496.

1262 Effect of Baicalin on IL-33/ST2 Signaling Pathway in Rats with Recurrent Oral Ulcers and Its Mechanism

Objective: To investigate the therapeutic effect of baicalin on model rats with recurrent oral ulcer (ROU) and its mechanism by regulating the interleukin-33/suppression of tumorigenicity 2 (IL-33/ST2) signaling pathway. Methods: Fifty rats were injected with autologous antigens to construct ROU models and randomly divided into model group, low, medium, and high dose baicalin (25, 50, 100 mg/kg) groups, and levamisole group (17.5 mg/kg); another 10 rats injected with Freund's complete adjuvant were taken as the normal group. After the completion of intragastric administration, the number, duration and interval of ulcers of rats in each group were observed and recorded; flow cytometry was used to detect the contents of CD4+ and CD8+ T in lymphocyte subsets in the peripheral blood of rats in each group; enzyme-linked immunoassay was used to detect the levels of interferon-γ (IFN-γ), IL-17, IL-2, and tumor necrosis factor-α (TNF-α) in the serum of rats in each group; reduced glutathione (GSH), superoxide dismutase (SOD), malondialdehyde (MDA) test kits were used to detect the levels of GSH, SOD and MDA in the serum of rats in each group; Western blotting was used to detect the levels of IL-33, ST2, and nuclear factor-kB (NF-kB) protein in the oral mucosa tissues of rats in each group. Results: Compared with the normal group, the number of ulcers in the model group increased, the duration increased, and the interval time prolonged, the CD4+ and CD4+/CD8+ decreased, the CD8+ increased, the contents of IFN-γ, IL-17, IL-2, TNF-α, and MDA increased, the contents of GSH and SOD reduced, the expression of IL-33, ST2, and NF-kB proteins was up-regulated (P<0.05). Compared with the model group, the number and duration of ulcers in the low, medium and high dose baicalin groups decreased, and the interval time prolonged, the CD4+ and CD4+/CD8+ increased, the CD8+, the contents of IFN-γ, IL-17, IL-2, TNF-α, and MDA decreased, the contents of GSH and SOD increased, the expression of IL-33, ST2, and NF-kB proteins was down-regulated (P<0.05), and they were dose-dependent. There was no statistically significant difference in indicators between the high-dose baicalin group and levamisole group (P>0.05). Conclusion: Baicalin has a significant therapeutic effect on ROU model rats. It may inhibit the IL-33/ST2 pathway, regulate the imbalance of T lymphocyte subsets, and improve inflammatory response and oxidative damage.

1268 Effect of Naringenin Regulation of PERK/eIF2α/ATF4/CHOP Axis on Proliferation Apoptosis Migration and Invasion of Thyroid Cancer Cells

Objective: To analyze the effects of naringin on the proliferation, apoptosis, migration and invasion of thyroid cancer cells B-CPAP by regulating protein kinase R-like ER kinase/eukaryotic translation initiation factor alpha 2/activating transcription factor 4/CCAAT/ enhancer-binding protein homologous protein (PERK/eIF2α/ATF4/CHOP) axis. Methods: B-CPAP cells were treated with 1, 5, 10, 20 μmol/L naringin, B-CPAP cells treated with 5 μmol/L PERK inhibitor GSK2606414+20 μmol/L naringin were used as the inhibitor group, and B-CPAP cells without naringin as control group. MTT method was used to detect the proliferation of B-CPAP cells in each group; flow cytometry was used to detect apoptosis of B-CPAP cells in each group; cell scratch test was used to detect the migration of B-CPAP cells in each group; Transwell experiment was used to detect the invasion of B-CPAP cells in each group; and Western blot method was used to detect the expression levels of PERK/eIF2α/ATF4/CHOP axis-related proteins, Vimentin, E-cadherin, N-cadherin, Bcl-2 related X protein(Bax), B-cell lymphoma-2(Bcl-2) in B-CPAP cells of each group. Results: Compared with the control group, the proliferation rate of B-CPAP cells [(100.00±0.00)% vs. (90.73±5.25)%, (76.52±3.19)%, (62.84±3.25)%, (48.39±2.14)%] decreased sequentially after treatment with 1, 5, 10, and 20 μmol/L naringin for 48, in a concentration-dependent manner (P<0.05); after treatment with 20 μmol/L naringin for 12 h, 24 h, and 48 h, the proliferation rate of B-CPAP cells decreased sequentially[(82.37±3.06)% vs. (71.45±2.73)% vs. (48.39±2.14)%], in a time-dependent manner (P<0.05). Compared with the control group, the apoptosis rate of B-CPAP cells [(6.37±0.44)% vs. (9.46±0.83)%, (18.59±2.16)%, (33.41±4.29)%, (50.76±5.17)%] increased with naringin concentration increasing (1, 5, 10, 20 μmol/L), the wound healing rate[(62.74±2.86)% vs. (55.26±2.49)%, (46.38±2.17)%, (39.15±1.84)%, (33.42±1.39)%] and the number of transmembrane cells of B-CPAP cells[(70.54±6.12) vs. (57.78±5.62), (42.59±4.86), (29.27±3.91), (65.62±5.83)] decreased, the levels of p-PERK/PERK, p-eIF2α/eIF2α, ATF4, CHOP, E-cadherin and Bax proteins in B-CPAP cells gradually increased, the levels of Vimentin, N-cadherin and Bcl-2 proteins in B-CPAP cells gradually reduced, in a concentration-dependent manner (P<0.05). The B-CPAP cell proliferation rate[(94.16±5.38)% vs. (48.39±2.14)%], scratch healing rate[(58.65±2.73)% vs. (33.42±1.39)%], and number of penetrating cells[(65.62±5.83)% vs. (16.49±3.04)%] in the inhibitor group were higher than those in the 20 μmol/L group (P<0.05), the apoptosis rate[(7.82±0.65)% vs. (50.76±5.17)%] and levels of p-PERK/PERK, p-eIF2α/eIF2α, ATF4, CHOP, E-cadherin and Bax proteins were lower than those in the 20 μmol/L group, the levels of Vimentin, N-cadherin and Bcl-2 proteins were higher than those in the 20 μmol/L group (P<0.05). Conclusion: Naringin can inhibit the proliferation of B-CPAP cells, promote their apoptosis, and reduce the migration and invasion abilities of B-CPAP cells by activating the PERK/eIF2α/ATF4/CHOP axis.

1275 Comparison of the Effects of Different Enhancers of the Mbk-2 Gene on Embryonic Development in Cryptobacterium Hidradenum

Objective: To Explore the effects of mbk-2 and its enhancers on nematode embryonic development using the mbk-2 mutant Cryptobacterium hidradenum as a model organism. Methods: Targeted silencing of mbk-2 enhancers pptr-1, pptr-2, rsa-1 and rsa-2 by RNAi technology were used to observe changes in nematode egg unhatched rate and embryonic developmental stages after silencing of mbk-2 enhancers. Results: The unhatched rate of mbk-2 mutant nematode eggs was 1.56 ± 0.25%, significantly higher than that of the N2 wild-type group (0) (P<0.05); the unhatched rates of N2 wild-type nematode eggs fed with pptr-2, rsa-1 and rsa-2 RNAi bacteria were 5.77 ± 0.24%, 4.40 ± 0.03% and 6.03 ± 1.19%, respectively, all significantly higher (P<0.05) than the normal group of N2 fed with L4440 bacteria (0). The unhatched rates of mbk-2 mutant nematode eggs fed by pptr-1, pptr-2, rsa-1 and rsa-2 RNAi bacteria were 3.78±0.14%, 75.53±2.29%, 41.79±0.76% and 51.65±8.23%, respectively, all of which were significantly higher than the unhatched rates of mbk-2 mutant nematode eggs (P<0.05), with the pptr-2 (75.53±2.29%) RNAi group having the highest unhatched rate. The unhatched rates of mbk-2 mutant nematode eggs fed by pptr-1, pptr-2, rsa-1 and rsa-2 RNAi bacteria were 3.78±0.14%, 75.53±2.29%, 41.79±0.76% and 51.65±8.23%, respectively, all of which were significantly higher than the unhatched rates of mbk-2 mutant nematode eggs (P<0.05). with the pptr-2 (75.53±2.29%) RNAi group having the highest unhatched rate. DIC images showed that the rsa-2 RNAi group was significantly abnormal compared to the mbk-2 mutant nematode at the unicellular stage of embryonic development (spindle establishment, absence of prokaryotic encounters close to the posterior of the cell and absence of prokaryotic transfer to the centre of the embryo) (P<0.05). The pptr-2 RNAi group was significantly abnormal compared to the mbk-2 mutant nematode at the four-cell stage of embryonic development (large differences in cell size, multiple nuclei and abnormal intercellular contacts) (P<0.05). Conclusion: The mbk-2 and its enhancer play an important regulatory role in nematode embryonic development.

1279 Association of Serum ICAM-1 VCAM-1 and CTRP9 Expression Levels with Coronary Vascular Calcification in Patients with Diabetic Nephropathy

Objective: To explore the association of the expression levels of serum intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1) and complement C1q tumor necrosis factor related protein 9 (CTRP9) with coronary vascular calcification in patients with diabetic kidney disease (DKD). Methods: Totally 152 patients with DKD were selected between May 2019 and October 2021, and were divided into calcification group (68 cases) and non-calcification group (84 cases) according to whether they were complicated with coronary calcification. The general data, related biochemical indicators and levels of serum ICAM-1, VCAM-1 and CTRP9 (measured by enzyme-linked immunosorbent assay) were compared between the two groups. Multivariate Logistic regression analysis was used to explore the influencing factors of coronary vascular calcification in DKD, and ROC curves were plotted to explore predictive value of serum ICAM-1, VCAM-1 and CTRP9 levels on vascular calcification in DKD. Results: The proportion of DKD clinical stage in calcification group was higher than that in non-calcification group (P<0.05), but there were no statistical differences in gender, age, body mass index, smoking history, drinking history and duration of diabetes mellitus between the two groups (P>0.05). The levels of HbA1c, LDL-C, blood calcium and mAlb were higher in calcification group than in non-calcification group (P<0.05), but the differences in the levels of TG, TC, HDL-C, blood phosphorus, BUN and SCr were not statistically significant between the two groups (P>0.05). Serum ICAM-1 and VCAM-1 levels in calcification group were significantly higher than those in non-calcification group (P<0.05), while the CTRP9 level was significantly lower than that in the non-calcification group (P<0.05). Clinical stage of DKD and high levels of HbA1c, ICAM-1 and VCAM-1 were the risk factors for coronary vascular calcification in patients with DKD (P<0.05), and high level of CTRP9 was a protective factor (P<0.05). The areas under the ROC curves of serum ICAM-1, VCAM-1 and CTRP9 on predicting vascular calcification in DKD were 0.710, 0.761 and 0.752 (P<0.05). Conclusion: The expressions of serum ICAM-1 and VCAM-1 are increased, while the expression of CTRP9 is decreased in patients with DKD complicated with coronary vascular calcification. The detection of the three levels has certain predictive value on coronary vascular calcification in DKD.

Objective: To explore the application of 1.5T magnetic resonance diffusion weighted imaging (DWI) in rectal cancer and the relationship between apparent diffusion coefficient (ADC value) and microvessel density (MVD), vascular endothelial growth factor (VEGF) and lymphatic vessel density (LVD). Methods: The clinicopathological data of 86 patients with rectal cancer admitted to our hospital from April 2018 to April 2021 were retrospectively selected. All patients were examined by DWI before treatment, and their MVD, VEGF and LVD expression levels were measured by immunohistochemistry. The value of DWI in the diagnosis of rectal cancer, and the relationship between ADC value and MVD, VEGF and LVD with Person correlation were analyzed. Results: ADC values were higher in patients with highly differentiated, T2 and N0 rectal cancers than those with low+moderately differentiated, T4 and N1~2 stages (P<0.05), but MVD, VEGF and LVD expression were lower in patients with highly differentiated, T2 and N0 rectal cancers than those with low+moderately differentiated, T4 and N1~2 stages. Person correlation showed that ADC values were negatively correlated with MVD, VEGF and LVD (r=-0.428, -0.505, -0.474, P<0.05). ROC results showed that ADC values predicted the degree of differentiation, T-stage and N-stage of rectal cancer with AUCs of 0.904, 0.839 and 0.897, respectively.Sensitivity and specificity: 83.3% and 87.8% for differentiation, 68.4% and 89.6% for T-stage and 91.9% and 85.7% for N-stage. Conclusion: The sensitivity and specificity of 1.5T MRI DWI sequences in predicting T-stage, N-stage and differentiation degree of rectal cancer are high, and the ADC values are closely related to MVD, VEGF and LVD expression, which can provide a reference basis for clinical treatment and prognosis assessment.

Objective: To study the respective effects of high phosphorus or high parathyroid hormone levels on smooth muscle cell calcification. Methods: Human thoracic aortic vascular smooth muscle cells (VSMC) were cultured in vitro for 14 days and divided into control group, high phosphorus group (2.6 mmol/L), high parathyroid hormone (parathyroid hormone 1-34, PTH1-34) group a (PTH1-34 concentration 10-11 mol/L), high parathyroid hormone (parathyroid hormone 1-34, PTH1-34) group b (PTH1-34 concentration 10-10 mol/L), and observed in respect of cellular calcium salt deposition after alizarin red staining; real-time quantitative PCR and western blotting were used to detect the gene and protein expression of bone morphogenetic protein 2 (BMP-2) and Runt-related transcription factor 2 (Runx-2). The intracellular calcium content of each group was determined by applying the intracellular calcium content kit. Resulst: Calcification of vascular smooth muscle cells in the high phosphorus group was detected by alizarin red staining, and expression of calcification proteins BMP-2 and Runx-2 was significantly increased by western blotting. Calcification of vascular smooth muscle cells cultured in the high parathyroid hormone group was not detected by alizarin red staining, and BMP-2 and Runx-2 expression in vascular smooth muscle cells in the high parathyroid hormone group was not increased by real-time quantitative PCR and western blotting compared with the control group. expression did not increase compared with the control group. The intracellular calcium content of cells in the high phosphorus group was significantly higher than that of the control group, and the intracellular calcium content of cells in the high parathyroid hormone a and b groups did not differ from that of normal control cells (P>0.05). 1295 PI-RADS v2.1 in the Diagnosis of Clinically Significant Cancer of the Prostate

Objective: To investigate the diagnostic role of multiparametric MRI (mpMRI) for clinically significant carcinoma of the prostate, using PI-RADS v2.1 as the diagnostic standard.Methods: A total of 143 cases of suspected prostate cancer were enrolled for mpMRI, including 72 cases of clinically significant cancer confirmed by radical pathology. The films were independently blinded by an experienced imaging physician following PI-RADS v2.1. Accuracy, sensitivity, and specificity were calculated by plotting the ROC curve and calculating the area under the curve (AUC) using puncture/radiological pathology as the gold standard. Results: The AUC of PI-RADS v2.1 score in the diagnosis of csPCa was 0.900 (95% CI: 0.847 ~ 0.952). The accuracy, sensitivity, and specificity of PI-RADS≥3 were 69.7%, 95.7%, and 51.4%, respectively. The accuracy of PI-RADS≥4 (excluding PI-RADS≥3+1 in the peripheral zone [PZ]) was 98.6% (69/70). Conclusion: PI-RADS v2.1 ad high sensitivity in the diagnosis of csPCa, and DCE increased the sensitivity in the detection of PZ cancer. PI-RADS≥4 (excluding PI-RADS≥3+1 in the PZ) indicated that the probability of clinically significant cancer was as high as 9%, providing urologists with strong evidence support for subsequent management.

1298 Analysis of the Effect of Whole Brain Radiotherapy on the Recent Neurocognitive Function and Prognosis of Brain Metastases from Lung Cancer

Objective: To explore the effect of whole brain radiotherapy (WBRT) on recent neurocognitive function and prognosis of patients with lung cancer brain metastasis, and to analyze the safety of WBRT in the treatment of lung cancer brain metastasis.Methods: A retrospective study was conducted with a selection of 91 patients with lung cancer brain metastases treated in our hospital from January 2017 to October 2021. All patients were treated with WBRT. The recent neurocognitive function of patients before and after treatment was compared. A 4-month outpatient or telephone follow-up observation was conducted to record the survival and death status of patients. They were divided into survival group and death group. The factors affecting the death of patients were analyzed by Cox regression.Results: There were significant differences in neurocognitive function before and after treatment. After treatment, the speed of language memory, visual memory, attention, and information processing was significantly better (P<0.05). After 91 patients were treated with WBRT, 45 patients died within 4 months after discharge through follow-up investigation. As of the end of follow-up, 46 patients were in a state of survival, and the case fatality rate was 49.45% 4 months after discharge. Compared with the dead patients, the proportion of squamous cell carcinoma and adenocarcinoma was larger, the proportion of small cell carcinoma was smaller, and the proportion of extracranial metastasis, deep metastasis, and uncontrolled primary tumor was less in the surviving patients (P<0.05). Cox regression analysis showed that histological diagnosis, extracranial metastasis, depth of metastatic tumor, and control of primary tumor were significantly correlated with the prognosis of patients with brain metastasis of lung cancer after treatment (P<0.05). Conclusion: WBRT treatment improves the neurocognitive function of patients with brain metastasis from lung cancer. Histological diagnosis, extracranial metastasis, depth of metastasis, and control of primary tumor are the risk factors affecting the prognosis of patients.

Objective: To explore the efficacy and safety of GnRH-a combined with ursamine in the treatment of adenomyosis by hysteroscopic endometrial resection.Methods: 107 patients with adenomyosis treated in our hospital from March 2016 to March 2021 were selected and randomly divided into control group (53 cases, subcutaneous injection of 3.75mg GnRH-a, once / 4 weeks, a total of 3 months) and observation group (54 cases, subcutaneous injection of 3.75mg GnRH-a, once / 4 weeks + ursamine, 1 tablet/time, stopped for 7 days after 21 days, a total of 3 months). The pain score, uterine volume, menstrual volume, and ovarian function were counted before operation and 3 months after treatment. The clinical efficacy after 3 months of treatment and adverse reactions during treatment were counted. The recurrence within 12 months was counted. Results After 3 months of treatment, the levels of vas, uterine volume, PBAC, FSH, and LH in the observation group were lower than those in the control group (P<0.05). The curative effect of the observation group was better than that of the control group (P<0.05). The total incidence and recurrence rate of adverse reactions in the observation group were lower than those in the control group (P<0.05). Conclusion: The follow-up treatment of adenomyosis with GnRH-a combined with ursamine after hysteroscopic endometrial resection is effective, which effectively alleviates the degree of dysmenorrhea, and restores ovarian function, reduces the incidence of postoperative adverse reactions, and reduces the recurrence rate to a certain extent.

1307 Analysis of the Relationship Between Serum NSE S100B and MBP Levels and the Severity of the Disease in Patients with Glioma

Objective: To explore the relationship between expression levels of serum neuron-specific enolase (NSE), S100B, and myelin basic protein (MBP) and disease severity in patients with glioma and the postoperative changes, to provide a reference for finding biological markers of glioma. Methods: 98 cases of patients with glioma were enrolled in the study group, and another 98 healthy examiners were used in the control group. Serum NSE, S100B, and MBP levels were compared before and after operation in the two groups, and their relationship with pathological grading and Karnofsky performance score (KPS) was analyzed. Results: The levels of serum NSE, S100B, and MBP in the study group were significantly higher than those in the control group (P<0.05). The levels of serum NSE, S100B, and MBP in the grade III group were significantly higher than those in the grade I-II group (P<0.05), and the levels of S100B and MBP in the grade IV group were significantly higher than those in grade III group and grade I-II group (P<0.05). The levels of serum NSE, S100B, and MBP in glioma patients were negatively correlated with preoperative KPS score (P<0.05). The levels of serum NSE, S100B, and MBP were increased first and then decreased before the operation, and at 1, 3, and 7 d after the operation (P<0.05), and the levels of serum NSE, S100B, and MBP at 1 and 3 d after operation were higher than those before operation (P<0.05), and the levels of serum NSE and S100B at 7 d after operation were lower than those before operation (P<0.05), and serum MBP level was higher than that before operation (P<0.05). Conclusion: The levels of serum NSE, S100B, and MBP in patients with glioma were significantly increased, and they were closely related to pathological grading, brain tissue damage, and therapeutic effect.

1311 Changes in Peripheral Blood Th1/Th2 Cytokine Levels and Clinical Significance in Patients with Bacterial Pneumonia

Objective: To explore the changes and clinical significance of levels of peripheral blood helper T cell 1/helper T cell 2 (Th1/Th2) cytokines in patients with bacterial pneumonia. Methods: 123 patients with bacterial pneumonia who were treated in the hospital between January 2018 and January 2021 were selected as the research subjects. CURB-65 scoring system was used to assess the severity of the disease, and the patients were divided into a moderate-to-severe group (CURB-65 score ≥2 points, 72 cases) and a mild group (CURB-65 score of 0-1 point, 51 cases). The baseline data, Th1/Th2 and Th1/Th2 secretory cytokines [interferon-γ (IFN-γ), interleukin-4 (IL-4)] were compared between the two groups, and the correlation between levels of Th1/Th2, IFN-γ and IL-4 and CURB-65 score in patients with bacterial pneumonia, and receiver operating characteristic (ROC) curve was plotted to analyze the diagnostic efficiency of the above indicators on moderate-to-severe bacterial pneumonia. Results: There were no significant differences in gender, age, smoking history, comorbidities, and other baseline data between the two groups (P>0.05). The white blood cell count (WBC) and procalcitonin (PCT) levels in the moderate-to-severe group were significantly higher than those in the mild group (P<0.05). The levels of Th1/Th2 and IFN-γ were significantly lower in the moderate-to-severe group than those in the mild group (P<0.05) while the level of IL-4 was significantly higher than that in the mild group (P<0.05). Spearman correlation analysis showed that the CURB-65 score of patients with bacterial pneumonia was significantly negatively correlated with Th1/Th2 and IFN-γ (P<0.05), and was significantly positively correlated with IL-4 (P<0.05). ROC curve analysis showed that the AUC values of the above indicators in diagnosing moderate-to-severe bacterial pneumonia were 0.904, 0.886, and 0.818, and the cut-off values were 2.42, 9.34 pg/ml, and 10.98 pg/ml, and the AUC of the combination of the three indicators was 0.985, with high diagnostic efficiency (P<0.05). Conclusion: The levels of Th1/Th2 and its cytokines in peripheral blood of patients with bacterial pneumonia are related to the severity of the disease. The above indicators have a reference value for the evaluation and treatment of the disease.

1316 Analysis of Short-Term and Long-Term Effect of Modified Self-Traction Post-Dissection Esophageal-Jejunostomy after Laparoscopic Total Gastrectomy in Patients with Gastric Cancer

Objective: To investigate the short-term and long-term clinical effect of modified self traction transection esophagojejunostomy in patients with gastric cancer after laparoscopic total gastrectomy.Methods: 96 patients with gastric cancer from January 2020 to February 2021 were randomly divided into 48 patients in the traditional group and 48 patients in the improved group. Patients in the traditional group were treated with traditional Overlap, and patients in the control group were treated with modified self traction and transection of esophagojejunostomy. The intraoperative and postoperative recovery of the two groups were recorded. The nutritional status and quality of life of the two groups were compared before and 1 year after the operation. The postoperative complications of the two groups were recorded. Result: The operation time and esophagojejunostomy time in the traditional group were significantly higher than those in the improved group (P<0.05), but there was no significant difference in intraoperative bleeding, lymph node dissection, first postoperative anal exhaust time, and hospital stay between the two groups (P>0.05); there was no significant difference in preoperative nutritional level between the two groups (P>0.05). The body weight, albumin, and prognostic nutritional index of the two groups one year after the operation were significantly higher than those before the operation. There was no significant difference in body weight, albumin, prognostic nutritional index, and the increase difference of each index between the two groups one year after operation (P>0.05). One year after the operation, the quality of life of the two groups was significantly better than that before the operation. At the same time, there was no significant difference between the two groups in the score one year after the operation, the difference in score increase, and the incidence of postoperative complications (P>0.05).Conclusion: Modified self-traction transected esophagojejunostomy is a safe and reliable anastomosis method, which significantly shortens the operation time, improves the postoperative nutritional status and quality of life of patients, and the short-term and long-term curative effect is satisfactory.

1321 Differential Diagnosis of Idiopathic Right Ventricular PVC or VT with Arrhythmogenic Right Ventricular Cardiomyopathy Concomitant PVC or VT by Hoffmayer ECG Integral

Objective: To explore the clinical significance of the Hoffmayer ECG integral method in differential idiopathic right ventricular premature beat (PVC) or ventricular tachycardia (VT) with arrhythmogenic right ventricular cardiomyopathy (ARVC) PVC or VT in early stage. Method: 40 cases of heart disease patients admitted to Chengde Central Hospital from Nov. 2017 to Dec. 2020 were collected. Among the patients, 30 cases had right ventricular outflow tract PVC or VT, and 10 cases had ARVC sinus rhythm concomitant PVC or VT. The electrocardiogram of 40 patients in sinus rhythm with premature ventricular beats or ventricular tachycardia was analyzed by two ECG physicians according to the Hoffmayer ECG score method. The Hoffmeyer ECG scores of the 40 patients were analyzed using the total score method, then the Hoffmayer ECG scores of the 40 patients were analyzed using the individual score method, then compared to the clinical diagnosis, and finally, the negative predictive value, positive predictive value, specificity and sensitivity of the diagnosis were calculated for this analysis. Result: Statistically significant differences were found between the ARVC with premature or ventricular tachycardia group and the idiopathic premature or ventricular tachycardia group in terms of the proportion of total Hoffmayer points ≥5 and inversion of TV1-V3 (3 points) in the sinus rhythm chest leads with premature or ventricular tachycardia. The correct diagnosis of right ventricular idiopathic ventricular premature beats or ventricular tachycardia and ARVC patients with ventricular premature beats or ventricular tachycardia was 80.0% in sensitivity and 90.3% in specificity with a positive predictive value of 80.0% and a negative predictive value of 81.3% in 30 and 10 patients, respectively, using a Hoffmayer ECG score of ≥5. The negative predictive value, sensitivity, positive predictive value, and specificity of the single criterion of Tv1-v3 inversion in the thoracic leads of sinus rhythm for the diagnosis of ARVC patients with ventricular premature or ventricular tachycardia were 93.1%, 80.0%, 72.7%, and 90.0% respectively, with statistically significant differences between the two groups.Conclusion: The Hoffmayer ECG integration method ≥5 is effective in identifying patients with ARVC with right ventricular outflow tract ventricular premature beats or ventricular tachycardia or idiopathic right ventricular outflow tract ventricular premature beats or ventricular tachycardia. This method is specific, sensitive, rapid, simple, and has high clinical value.

1325 Significance of CYP2C19 Gene Polymorphism in Guiding Antiplatelet Drug Selection Therapy

Objective: To investigate the role of CYP2C19 gene polymorphism in guiding the use of ticagrelor after percutaneous transluminal angioplasty (PTA) in patients with acute ischemic stroke (AIS). Methods: A total of 525 patients with AIS who underwent PTA from January 2019 to March 2021 were enrolled as the research subjects. According to the results of CYP2C19 genotyping, patients were divided into three groups. 185 patients (35.24%) in group A (rapid metabolic type); 259 patients (49.34%) in group B (intermediate metabolic type); 81 patients (15.43%) in group C (poor metabolic type). Group B and C were further subdivided into doubled-clopidogrel group B1 and C1, ticagrelor group B2 and C2 randomly. Stroke, death from any cause, recurrent ischemic stroke, disability, bleeding and dyspnea within 3 months after operation were followed up to evaluate the effectiveness and safety. Results: After PTA, the platelet aggregation rate in group B1 was significantly higher than that in group B2 (P<0.05); the platelet aggregation rate in group C1 after PTA was significantly higher than that in group C2 (P<0.05). Secondary outcomes and disability events happened much more frequently in group B1 than in group B2 (P<0.05), while total bleeding events, minimal bleeding events and dyspnea occurred much more frequently in group B2 than in group B1 (P<0.05). The incidences of primary outcomes, secondary outcomes and disability in group C1 were obviously increased than incidences of those in group C2 (P<0.05). Conclusion: For patients with AIS undergoing PTA in CYP2C19 intermediate metabolic type with low risk of bleeding, the use of tigretol is recommended instead of dual-dose clopidogrel, and for patients with AIS undergoing PTA in CYP2C19 slow metabolic type, the direct use of tigretol is recommended as a clinically important way to reduce the disability rate of acute ischaemic stroke, the risk of recurrent cerebrovascular disease after surgery and to improve the prognosis.

1331 Analysis of the Medium and Long-Term Efficacy of Percutaneous Minimally Invasive Locking Plates for Mid-Clavicle Fractures

Objective: To analyze the medium and long-term effecacy of percutaneous minimally invasive locking plates for mid-clavicular fractures by comparing the indexes after different surgical methods. Methods: A total of 84 patients with mid-clavicular fractures who were treated in our hospital from January 2019 to January 2021 were selected as the research subjects, and randomly divided into the observation group and the control group according to the random number table method, 42 cases in each group. Patients in the control group were treated with incisional repositioning locking plate, while the observation group was treated with percutaneous minimally invasive locking plate. The general data, improvement of shoulder joint function 1 month after surgery, the constant score before and after surgery, as well as the improvement of quality of life and patient satisfaction were compared between the two groups. Results: One month after the operation, it was found that the excellent and good rate of shoulder joint improvement in the observation group (85.71%) was significantly higher than that in the control group (69.05%), and the difference was statistically significant (P<0.05). The constant scores of the two groups of patients were significantly improved after operation, and the constant scores of the observation group at 1 year after operation were higher than those of the control group, and the difference between the two groups of patients at 1 year after operation was statistically significant (P<0.05); one year after surgery, the quality of life assessment indexes such as physiological function, physical pain, mental health and social function improved significantly in both groups, and the improvement was more obvious in the observation group than in the control group, with a statistically significant difference (P<0.05); the overall satisfaction rate of patients in the observation group (97.62%) was significantly higher than that of the control group (78.57%), with a statistically significant difference (P<0.05) Conclusion: Percutaneous minimally invasive locking plate for the treatment of middle clavicle fractures is less traumatic for patients. Compared with the traditional open reduction and locking plate, the indicators are better than traditional incisional repositioning locking plate at 1 month and 1 year after operation, and the patient satisfaction is better. It is clinically replicable.

Objective: To analyze the application value of dual-energy CT imaging in the evaluation of postoperative effect of hepatic arterial chemoembolization for primary hepatocellular carcinoma (HCC). Methods: Imaging data of 40 patients with HCC treated by hepatic artery embolization chemotherapy admitted from January 2019 to January 2020 were retrospectively analyzed, with conventional CT images and CT dual-energy iodograms obtained by dual-energy CT scan examination 4 weeks after surgery. Using the results of DSA as the gold standard, the diagnostic efficacy of two CT examination methods for residual lesions after liver cancer surgery was compared. the iodine concentration values (IC) and standard iodine concentration values (NIC) of CT dual-energy iodograms of patients' postoperative residual lesions and normal liver tissue were measured to determine their residual lesion activity. Results: Using DSA findings as the gold standard, the sensitivity, specificity and accuracy of dual-energy CT in detecting residual lesions after HCC were higher than those of conventional CT, and the difference was statistically significant (P<0.05). The ROC curve showed that the AUC of dual-energy CT for the diagnosis of residual lesions after HCC was 0.861, which was greater than that of conventional CT at 0.658. The IC and NIC values of residual active lesions after HCC were higher than those of normal liver tissue, and the difference was statistically significant (P<0.05). Conclusion: Dual-energy CT imaging has significant clinical value in the detection of residual lesions after hepatic arterial chemoembolization for liver cancer, and its IC value and NIC values have a certain potential in judging the degree of lesion activity.

1340 Expression and Clinical Significance of OPN TIMP-2 and STAT3 Proteins in Patients with Endometriosis

Objective: To analyze the expression and clincal significance of osteopontin (OPN), tissue inhibitor of matrix metalloproteinase 2 (TIMP-2) and signal transducer and activator of transcription 3 (STAT3) proteins in endometriosis (Endometriosis, EMS) patients.Methods: Clinical data of 117 patients with endometriosis admitted to our hospital from January 2019 to January 2020 were selected [EMS group, according to the American Fertility Society scoring standard (r-AFS) staging, 53 cases in stages I-II and 64 cases in stages III-IV; menstrual cycle: 66 cases in the proliferative phase and 51 cases in the secretory phase], and another 117 cases admitted to the same period with uterine 103 non-EMS patients with fibroids or infertility treated surgically (control group) were selected. The expression of OPN, TIMP-2 and STAT3 in different populations, different r-AFS stages and different menstrual cycles were compared, and the correlation between OPN, TIMP-2 and STAT3 and r-AFS stages was analyzed.Results: The expression of OPN and STAT3 in EMS group was significantly higher than that in control group, while the level of TIMP-2 was significantly lower than that in control group (P<0.05); the expression of OPN and STAT3 in stage III-IV was significantly higher than that in stage I-II, while the level of TIMP-2 was significantly lower than that in stage I-II (P<0.05); the expression of OPN and STAT3 in proliferative stage was significantly higher than that in secretory stage, while the TIMP-2 levels were significantly lower than those in the secretory phase (P<0.05); serum OPN and STAT3 were positively correlated with r-AFS stage, and TIMP-2 was negatively correlated with r-AFS stage in EMS patients (P<0.05). Conclusion: OPN and STAT3 expression were increased and TIMP-2 expression was down-regulated in EMS patients, which were closely related to the occurrence and progression of the disease, and monitoring their expression could provide a reference for disease development prediction and regression.

Objective: To explore the clinical significance of serum microRNA-17 (miR-17) and miR-25 expression in patients with preeclampsia (PE) and its influence on pregnancy outcome.Methods: Seventy-four cases of PE patients in our hospital from February 2018 to August 2020 were selected as the PE group, and 74 cases of normal pregnant women without pregnancy comorbidities and complications were selected as the healthy group during the same period. The general data and serum miR-17 and miR-25 levels of the two groups were compared, and the diagnostic value of serum miR-17 and miR-25 levels in relation to the occurrence of PE level on PE was analyzed, and the correlation between serum miR-17 and miR-25 levels and clinical characteristics and pregnancy outcome of PE patients was analyzed. Results: The diastolic blood pressure, systolic blood pressure, and serum miR-17 levels in the PE group were higher than those in the healthy group, and week of delivery, neonatal birth weight, and serum miR-25 levels were lower than in the healthy group (P<0.05); logistic regression analysis showed that serum miR-17 and miR-25 levels were correlated with PE (P<0.05);receiver operating characteristic (ROC) curve shows that the AUC for the combined diagnosis of PE with miR-17 and miR-25 was the largest at 0.918; there were significant differences in diastolic blood pressure, systolic blood pressure, gestational week of delivery, and neonatal birth weight among patients with PE with different serum miR-17 and miR-25 levels (P<0.05); miR-17 and miR-25 expression levels were associated with clinical characteristics (diastolic blood pressure, systolic blood pressure) and pregnancy outcome (gestational week of delivery, neonatal birth weight (P<0.05).Conclusion: Serum miR-17 and miR-25 of PE patients are expressed pathologically, and they are related to the severity of the disease and pregnancy outcome.

1349 Clinical Efficacy and Safety of Uurethroplasty with Transverse Pedicled Prepuce Island Flap for Hypospadias with Penile Head

Objective: To analyze the clinical value of urethroplasty with transverse cutting pedicled prepuce island flap (Duckett operation) for hypospadias with penile head. Methods: The clinical data of 129 cases of hypospadias with small penis head treated in our hospital from March 2018 to March 2021 were retrospectively analyzed. According to the therapeutic methods, they were divided into Duckett group (n=67) and Snodgrass group (n=62). The operation-related indexes, the incidence of postoperative complications, the treatment effect and family satisfaction rate of the two groups were compared. Results: Compared with snodgrass group, the operation time and blood loss in Duckett group were higher, and the difference was statistically significant (P<0.05). The incidence of postoperative urinary fistula in Duckett group was lower than that in Snodgrass group (P<0.05). The cure rate of Duckett group was higher than that of Snodgrass group 6 months after operation, and the difference was statistically significant (P<0.05). Six months after operation, the satisfaction rate of guardians in Duckett group was 86.57%, which was higher than that in Snodgrass group (67.74%, P<0.05). Conclusion: Although Duckett's operation in the treatment of hypospadias with small penis head is more complicated and the intraoperative trauma is slightly higher than that of Snodgrass's operation, the incidence of postoperative complications in children is lower, and the cure rate and satisfaction rate of guardians are higher.

1353 Bcl-2 and MACCI Protein Expression in Cervical Epithelial Tissue Before and After Chemotherapy in Patients with Cervical Cancer and the Predictive Efficacy of ROC

Objective: To explore the expression of B lymphocyte tumor-2 gene (Bcl-2) and colon cancer metastasis related gene 1 protein (MACCI) in cervical epithelial tissue before and after chemotherapy in patients with cervical cancer. Methods: The retrospective analysis of 89 cervical intraepithelial neoplasia (cervical neoplasia group) and 121 cervical cancer patients (cervical cancer group) were collected between March 2018 and November 2020, all of whom underwent chemotherapy intervention, and MACCI and Bcl-2 expression were detected using the immunohistochemical SP method, and after comparing the gene protein expression of the two groups, the Bcl-2 and MACCI gene expression of cervical cancer patients before and after chemotherapy was then compared, and the predictive efficacy of Bcl-2 and MACCI was analysed by ROC curve. Results: The MACCI expression (7.68 ± 1.58)% and Bcl-2 expression (0.29 ± 0.04)% were higher in cervical cancer patients than in the cervical neoplasia group (P<0.05), and ROC curve analysis showed that MACCI and Bcl-2 predicted cervical cancer with AUC values of 0.924 and 0.968, respectively. meanwhile, MACCI expression (5.98 (±1.44)% and Bcl-2 expression (0.21±0.03)% were lower than those before chemotherapy (P<0.05), and the efficacy was positively correlated with MACCI and Bcl-2 by Spearman method analysis; ROC curve analysis showed that the AUC values of MACCI and Bcl-2 for diagnosing cervical cancer were 0.924 and 0.968, respectively; the AUC values of MACCI and Bcl-2. Conclusion: Bcl-2 and MACCI are more effective in the assessment of cervical cancer incidence and regression.

Objective: To investigate the value of ambulatory blood pressure combined with quantitative 24-hour urine protein monitoring in pre-eclampsia and its pregnancy outcome. Methods: From January 2019 to December 2020, 78 cases of pregnant women with pre-eclampsia were selected for the study. The patients were divided into 32 cases in the observation group with adverse pregnancy outcomes and 46 cases in the control group without adverse pregnancy outcomes according to whether the mothers and infants had adverse pregnancy outcomes. 24-hour ambulatory blood pressure monitoring and quantitative urine protein analysis were performed respectively. Results: The dSBP, dDBP, nSBP and 24hDBP were all higher in the observation group than in the control group (P<0.05); the final birth weight (FBW) of the children in the observation group was significantly lower than that of the control group (t=6.261, P<0.001); the 24-hour urine protein quantification was significantly higher in the observation group than in the control group (t=2.986, P<0.001); all patients in the observation and control groups had significantly higher SBP blood pressure load values and DBP blood pressure load values at night than during the day (P<0.001). Conclusion: Patients with pre-eclampsia have a high incidence of abnormal circadian rhythms, characterized by non-arrythmic circadian rhythms and elevated blood pressure at night, and ambulatory blood pressure and proteinuria monitoring can indicate adverse pregnancy outcomes.

1361 Value of Chemokine Receptor 4 in the Assessment of Postoperative Recurrent Metastasis in Patients with Colorectal Cancer

Objective: To investigate the value of chemokine receptor 4 (CCR4) in assessing postoperative recurrence and metastasis of colorectal cancer patients, and to establish a Cox survival risk score model. Methods: According to the expression of CCR4 detected by postoperative immunohistochemistry, the patients with colorectal cancer were divided into CCR4 positive group and CCR4 negative group. The general clinicopathological data of patients with CCR4 positive and CCR4 negative groups were compared. Taking postoperative tumor recurrence and metastasis as the dependent variable, multivariate Cox regression analysis was used to obtain the independent predictors of 5-year tumor recurrence and metastasis after operation, Kaplan Meier survival analysis compared the 5-year disease-free survival rate of patients between CCR4 positive group and CCR4 negative groups. Based on the independent predictors obtained by multivariate analysis, a Cox survival risk score model for predicting tumor recurrence and metastasis of colorectal cancer patients was established. Results: There were significant differences in the degree of differentiation, TNM / T stage and lymph node metastasis between the two groups (P<0.05). Multivariate Cox regression analysis showed that the degree of differentiation, TNM / T stage, lymph node metastasis and CCR4 were independent predictors of 5-year tumor recurrence and metastasis of patients after operation (P<0.05). Kaplan Meier survival analysis showed that 1-year,3-year and 5 year disease-free survival rates of the patients in CCR4 negative group were significantly higher than those of patients in CCR4 positive group (72.3%, 54.2%, 38.6% vs 54.5%, 31.9%, 24.5%, P<0.05). A Cox survival risk score model for predicting the 5-year risk of tumor recurrence and metastasis of patients with colorectal cancer was constructed based on the 4 independent predictors of differentiation, TNM / T stage, lymph node metastasis and CCR4. The best cutoff value of the risk score was -0.08, the patients were divided into high-risk group and low-risk group with the cutoff value. The Cox survival risk score model showed that the higher the Cox survival risk score, the higher the risk of postoperative tumor recurrence and metastasis of patients. Conclusion: The Cox survival risk score model combined with CCR4 can be used to predict the risk of 5-year tumor recurrence and metastasis of colorectal cancer patients after operation.

1366 Epidemiological Characteristics of Mycoplasma Pneumoniae Chlamydia Pneumoniae and Legionella Pneumophila in Children with Recurrent Respiratory Tract Infections

Objective: To analyze the epidemiological characteristics of mycoplasma pneumonia (MP), chlamydia pneumonia (CP) and legionella pneumophila (LP) in children with recurrent respiratory tract infections (RRTIs). Methods: A total of 186 children with RRTIs admitted to the hospital between September 2017 and September 2021 were enrolled in the study. The whole blood was collected to detect IgM antibodies of serum MP, CP and LP. And the screening results were analyzed. Results: The positive rate of serum MP was higher than that of CP (31.72% vs 19.89%) (P<0.017, Bonferroni correction). The positive rate of CP was higher than that of LP (19.89% vs 8.06%) (P<0.017, Bonferroni correction). There was no significant difference in positive rates of serum MP, CP and LP among children of different genders (P>0.05). The positive rates of serum MP, CP and LP in RRTIs children aged 3-6 years were 51.32%, 34.21% and 17.11%, higher than those aged 0-2 years (13.64%, 9.09%, 2.27%) and 7-12 years (21.21%, 10.61%, 1.52%) (P<0.017, Bonferroni correction). The positive rates of serum MP, CP and LP in winter were 58.54%, 41.46% and 24.39%, higher than those in spring (25.49%, 13.73%, 3.92%), summer (17.95%, 12.82%, 2.56%) and autumn (27.27%, 14.55%, 3.64%) (P<0.0083, Bonferroni correction). Conclusion: Among children with RRTIs, MP, CP and LP had the highest rate of positivity, followed by CP infection, and LP was the lowest. There is no significant difference in the three infections among children of different genders. However, there are differences in their susceptibility among children at different age groups. There are seasonal differences in the prevalence of pathogens.

Objective: To analyze the effects of medium frequency electrical stimulation combined with rehabilitation training on lower limb muscle strength, urinary retention and gait parameters of patients with thoracolumbar incomplete spinal cord injury. Methods: A total of 103 patients with thoracolumbar incomplete spinal cord injury admitted to the hospital between January 2018 and September 2021 were selected as the research subjects. They were divided into combined group (given medium frequency electrical stimulation combined with rehabilitation training, n=52) and conventional group (given conventional rehabilitation training treatment, n=51) according to the simple randomization method. The patients were continuously treated for 12 weeks. The lower limb muscle strength (muscle strengths of quadriceps femoris and tibialis anterior muscle) before and after treatment and improvement status of urinary retention were compared between the two groups of patients. Gait test was conducted to compare the changes in gait parameters such as step length, step frequency and step speed in the two groups before and after treatment. Functional Independence Measure (FIM) was used to compare the independent living ability before and after treatment in the two groups. Results: Generalized estimating equation results showed that muscle strength in both groups was improved, and the improvement of muscle strength in combined group was better than that in conventional group (P<0.05).The clinical effective rate of urinary retention treatment of 94.23% in combined group was higher than 80.39% in conventional group, and the efficacy grading was better in combined group than that in conventional group (P<0.05). There were no significant differences between the two groups of patients in terms of step frequency, step length and step speed before treatment (P>0.05). After treatment, the differences of the above gait parameters before and after treatment in combined group were higher compared with those in conventional group (P<0.05). Before treatment, the differences in scores of dimensions of FIM were not significant between the two groups (P>0.05). The differences of scores of FIM dimensions before and after treatment were higher in combined group compared to conventional group (P<0.05). Conclusion: Medium frequency electrical stimulation combined with rehabilitation training can improve the lower limb muscle strength, help improve the symptoms of urinary retention and gait, and enhance the independent living ability of patients with thoracolumbar incomplete spinal cord injury.

1375 Effect of Group B Haemolytic Streptococci on Maternal Coagulation Vaginal Microecology and Pregnancy Outcome

Objective: To investigate the effect of group B hemolytic streptococcus (GBS) on maternal blood coagulation, vaginal microecology and pregnancy outcome. Methods: A total of 210 pregnant women with GBS infection who were admitted to our hospital from January 2020 to February 2022 were selected as the research subjects, and 210 pregnant women with normal obstetric examination during the same period were selected as the control group. The coagulation function, vaginal microecology and pregnancy outcomes of the two groups of pregnant women were compared. and the association of GBS with coagulation function, vaginal microecology and pregnancy outcome was analyzed. Results: Patients in the study group had significantly lower AP, APTT and INR levels than the control group and significantly higher Fbg levels than the control group (P<0.05).The difference between the two groups was not statistically significant (P>0.05) when comparing positive Trichomonas vaginalis, but the rate of vaginal micro-ecological disorders, positive fungi, positive bacterial vaginosis, abnormal pH, and abnormal cleanliness was higher in the study group than in the control group, which was statistically significant (P<0.05). The incidence of caesarean section, premature rupture of membranes, puerperal infection, intrauterine infection, prematurity, pathological jaundice, neonatal infection and neonatal asphyxia was significantly higher in the study group than in the control group (P<0.05). Conclusion: Pregnant women with Group B haemolytic streptococcal (GBS) infection during pregnancy are at risk of blood hypercoagulation, vaginal microecological disorders and adverse pregnancy outcomes. Maintaining stable vaginal microecology, focusing on coagulation testing and early management can help to reduce the risk of adverse pregnancy outcomes.

Objective: To analyze the correlation between fetal growth restriction and abnormal umbilical artery blood flow and the related factors affecting fetal growth. Methods: A total of 120 pregnant women with fetal growth restriction treated in our hospital from January 2019 to January 2021 were selected as the observation group and 100 normal pregnant women as the control group. The ratio of systolic peak to diastolic peak of umbilical artery blood flow (S/D), blood resistance index (RI) and blood pulsation index (PI) of the two groups were observed, various factors leading to fetal growth restriction were analyzed, and statistically significant factors were included in multivariate logistic regression analysis to analyze the relevant independent risk factors of fetal growth restriction. Results: The indexes of S/D, PI and RI in the observation group were significantly higher than those in the control group (P<0.05); the results of univariate and multivariate logistic regression analysis showed that anemia, placental abnormality and umbilical cord abnormality were independent risk factors for fetal growth restriction. Conclusion: Pregnant women with fetal growth restriction in the abdomen will have obvious abnormalities in umbilical artery blood flow, and their blood flow resistance will also increase significantly. Anemia, placental abnormality, umbilical cord abnormality and fetal membrane abnormality are independent risk factors leading to fetal growth restriction. More clinical attention should be paid to this type of condition in pregnant women.

Objective: To explore the effects of different doses of ticagrelor combined with atorvastatin calcium tablets on hemorheology, cardiac function and serum matrix metalloprotein-9 (MMP- 9), the effect of insulin-like growth factors-1 (IGF-1). Methods: A total of 117 patients with coronary heart disease UAP who were treated in our hospital from November 2020 to November 2021 were prospectively selected as the research subjects, and were divided into groups A and B by simple random coin tossing method, and finally 61 patients in group A and 56 patients in group B were included. The patients in group B were given 90mg ticagrelor + atorvastatin calcium tablets on the basis of conventional treatment, and the patients in group A were given 45mg ticagrelor + atorvastatin calcium tablets on the basis of conventional treatment , the two groups of patients were treated continuously for 1 month. The rheology (whole blood viscosity (WBV), plasma viscosity (PV)), cardiac function (left ventricular ejection fractions (LVEF), left ventricular end diastolic diameter (LVEDD) and serum MMP-9, LVEDD) were compared between the two groups before and after 1 month of treatment(LVEF), left ventricular end diastolic diameter (LVEDD), interventricular septum thickness (IVST)) and serum MMP-9 and IGF-1 levels. Results: The results of repeated measures variance showed that the comparison between the WBV and PV levels at different points of treatment in both groups was statistically significant with time factor as the source, between-subjects effect with group as the source and within-subjects effect with time and group interaction as the source, P<0.05; after 1 month of treatment, the WBV and PV levels decreased in both groups and were lower in group A than in group B, P<0.05; The results of repeated measurement variance showed that the comparison of LVEF, LVEDD, and IVST levels at different points of treatment in both groups was statistically significant with time factor as the source, between-subjects effect with group as the source and within-subjects effect with time and group interaction as the source, P<0.05; after 1 month of treatment, LVEF levels increased in both groups and were higher in group A than in group B, LVEDD and IVST levels decreased and were lower in group A than in group B, P<0.05. Methods: A total of 218 patients with chronic HBV infection-related liver cirrhosis treated in the hospital between October 2019 and October 2021 were selected, and they were divided into combined group (given Fuzheng Qudu decoction combined with entecavir antiviral therapy, n=109) and control group (treated with entecavir antiviral therapy, n=109) according to the simple random grouping method. The clinical efficacy and scores of TCM syndromes, HBV-DNA load and positive rate and negative conversion rate of hepatitis B surface antigen (HBsAg) and hepatitis B virus e antigen (HBeAg) of the two groups were compared after 12 weeks of treatment. The levels of liver function indicators [alanine aminotransferase (ALT), serum albumin (ALB), total bilirubin (TBIL), aspartate aminotransferase (AST)] before and after treatment and levels of serum inflammatory factors [tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), interleukin-8 (IL-8)] were compared between the two groups. Results: The decreases of scores of TCM syndromes before and after treatment were higher in combined group compared to control group (P<0.05), and the clinical effective rate of 92.66% was higher than 83.49% in control group (P<0.05). The decrease of HBV-DNA load before and after treatment in combined group was higher compared with that in control group (P<0.05), and the negative conversion rates of HBV-DNA, HBsAg and HBeAg were higher compared with those in control group (P<0.05). The levels of ALT, TBIL and AST and inflammatory indicators TNF-α, IL-6 and IL-8 in combined group after treatment were lower than those in control group while the level of ALB was higher than that in control group (P<0.05), and the decreases of the above indicators in combined group before and after treatment were higher than those in control group (P<0.05). Conclusion: Fuzheng Qudu decoction combined with antiviral therapy has a significant clinical efficacy in the treatment of chronic HBV-related liver cirrhosis, and it can inhibit the body’s inflammatory response, enhance the body’s immune response, inhibit the HBV-DNA replication, reduce the viral load and improve the liver function.

1393 Nucleus Zizhu Capsule Combined with Ceftriaxone Sodium in the Treatment of Pregnancy Urinary Tract Infection and Its Influence on Vaginal Microecology and Pregnancy Outcome

Objective: To explore the effect of Nucleus Zizhu Capsule combined with ceftriaxone sodium in the treatment of pregnancy urinary tract infection and its influence on vaginal microecology and pregnancy outcome. Methods: A total of 84 pregnant urinary tract infection patients admitted to our hospital from January 2019 to January 2021 were selected as the research subjects, and they were divided into an observation group and a control group according to the simple randomization, with 42 cases in each group. The control group was treated with ceftriaxone sodium, and the observation group was treated with Nudehua Zizhu Capsules combined with ceftriaxone sodium. The curative effect, symptom scores, levels of inflammatory factors [procalcitonin (PCT), C-reactive protein (CRP), heme oxidase 1 (HO-1), interleukin-6 (IL-6)], vaginal micro-ecological conditions [density of flora, diversity of flora, pH value], pregnancy outcome before and after treatment, incidence of adverse reactions were compared. Results: The total effective rate of the observation group was higher than that of the control group, and the incidence of adverse pregnancy outcomes was lower than that of the control group (P<0.05). After treatment, symptom score, serum PCT, CRP, HO-1, and IL-6 levels in the observation group were lower than those in the control group (P<0.05). After treatment, the density, diversity and pH value of the observation group were better than those of the control group (P<0.05). There was no statistically significant difference in the incidence of adverse reactions between the two groups (P>0.05). Conclusion: Nucleus Zizhu Capsules combined with ceftriaxone sodium has a significant effect on the treatment of pregnancy urinary tract infections.It can effectively promote the improvement of symptoms and vaginal microecology, reduce body inflammation, and reduce the incidence of adverse pregnancy outcomes while ensuring safety.

1399 Effect of Different Doses of Parecoxib Sodium on Immune Indexes, Serum S100β NSE TNF-α and IL-1β Levels in Patients Undergoing Surgery for Rectal Cancer

Objective: To investigate the Effect of different doses of parecoxib sodium on immune indexes, serum S100β, NSE, TNF-α and IL-1β levels in patients undergoing surgery for rectal cancer. Methods: A total of 80 patients who underwent rectal cancer surgery in our hospital from March 2018 to June 2021 were included in the study. They were simply randomly divided into high-dose, low-dose and control groups, with 30 cases, 30 cases and 20 cases respectively. The control group was treated with sodium chloride (0.9%) 20 minutes before anesthesia , the high-dose group was given 30 mg parecoxib sodium, and the low-dose group was given 10 mg parecoxib sodium β , NSE , TNF- α And IL-1 β. Results: The number of analgesic pump presses was significantly less in the high-dose group than in the low-dose and control groups, and significantly less in the low-dose than in the control group, with statistically significant differences (P<0.05); the differences were statistically significant (P<0.05) for the comparison of intersubject effects (F time point, F interaction) and between-subject effects (F between groups) for S100β, TNF-α, IL-1β, and NSE only for the F time point comparison was statistically significant (P<0.05), while the between-group differences and group-time-interaction effects were not statistically significant (P>0.05). The differences were statistically significant (P<0.05) for the comparison of CD3+ and CD4+ within-subject effects (F time point, F interaction) and between-subject effects (F between groups), and statistically significant (P<0.05) for CD8+ only for the comparison of F time point, while neither the between-group differences nor the interaction between groups and time were statistically significant (P>0.05). Conclusion: Parecoxib sodium significantly improved immune markers, serum S100β, NSE, TNF-α and IL-1β levels in patients undergoing surgery for rectal cancer, with better results when 30mg was used.

1404 Effect of Banxia XiexinA Decoction Combined with Jinghua Weikang Capsule on CA724 and PG1 Levels in Patients with Chronic Atrophic Gastritis

Objective: To analyze the effect of Banxia Xiexin Decoction combined with Jinghua Weikang capsule on CA724 and PG1 levels in patients with chronic atrophic gastritis. Methods: A total of 100 patients with chronic atrophic gastritis admitted to our hospital from October 2019 to October 2021 were selected as the research objects, and they were divided into control group and research group, 50 cases in each. Control group was given Jinghua Weikang capsule, study group was given Banxia Xiexin Decoction combined with Jinghua Weikang capsule. The levels of CA724, PG1, inflammatory factors and gastric mucosa function related indicators in the two groups were detected, and the TCM symptom scores, adverse reactions and clinical efficacy were compared between the two groups. Results: Comparison of dry mouth and bitter taste, epigastric fullness, epigastric fullness, nausea and vomiting, CA724, PG1, IL-6, IL-1β, TNF-α, PGE2, GSH-px between the two groups before and after treatment. Dry mouth, stomach fullness, epigastric fullness, nausea and vomiting scores, CA724, IL-6, IL-1β, TNF-α levels decreased more than the control group, and PG1, PGE2, GSH-px levels increased more than the control group group (P<0.05). The adverse effects were slightly higher than the control group (P>0.05), and the treatment response rate was higher than the control group (P<0.05). Conclusion: After combined treatment with Banxia Xiexin Decoction and Jinghua Weikang Capsules, the levels of CA724 and inflammatory factors in patients with chronic atrophic gastritis decreased, the level of PG1 increased, and the gastric mucosal function improved.
