1057 Changes in the Behavioristics of IFN-α Induced Depression in Rats and in the Expression Level of 5-HTA2R in the CAl Region of Hippocampal Tissue

Objective: To investigate the mechanism of depression by examining the behavioural and hippocampal histology and its 5-HTA2R expression levels in different ways of preparing rat depression models. Methods: Thirty rats were selected and divided by random number method into normal group,interferon group and CUMS model group,10 rats each.The rat depression model was prepared by subcutaneous injection of IFN-α for 12 weeks,and the traditional CUMS rat depression model was used as a control for a comparative study.Behavioural indicators such as body weight,results of the absenteeism test,sugar-water preference test and the time of feeding inhibition in the new environment were recorded at different times of the intervention; histological changes in the hippocampus of rats were observed by HE staining and auxin staining; changes in the expression level of 5-HTA2R in rats were detected by immunohistochemistry. Results: Compared with the control group,there was a statistical difference between the interferon group and the model group in terms of rat body weight and various behavioral experiment scores (P<0.05); compared with the model group,the duration of the new environmental inhibition of feeding was significantly longer in the interferon group,and there was a statistical difference (P<0.05),but there was no statistical difference (P>0.05) in the rat body weight change,the score of the absenteeism test and the results of the sugar-water preference experiment.There was a statistically significant difference (P<0.05) when the duration of the intervention was prolonged,with a gradual decrease in body weight,scores in the sugar-water preference test and the score of the absenteeism test,and a gradual increase of the duration of inhibition of feeding in the new environment (P>0.05).Both HE staining and auxin staining showed that the hippocampal CAl region of the model group showed obvious signs of injury,and the model group showed obvious signs of injury,with reduced cell numbers,increased cell gaps,fewer cell layers,sparse,disorganized,incomplete cell structure,and atrophy,with some cells showing vacuolation,smaller nuclei,solidified,and also visible swollen,lightly stained,fragmented,or even lost nuclei,showing eosinophilic staining.The interferon group was similar to the model group; the number of 5-HTA2R immunoreactive positive neurons in the hippocampal CAl region was reduced in the interferon group compared to the normal group,with a significant difference (71.6% vs 56.3%,χ2=4.85 P<0.05),and the number of 5-HTA2R receptor immunoreactive positive neurons in the hippocampal CAl region was slightly reduced compared to the model control group,but not statistically different (52.6% vs 56.3%,χ2=0.32 P>0.05). Conclusion: The ability of interferon to reduce the level of 5-HTA2R expression in the CAl region of the hippocampus may lead to depression in rats,providing a theoretical basis for the development of strategies for the preparation of depression models.

1061 SP2509 Induces Apoptosis and Autophagy in Osteosarcoma Cells by Regulating LSD1 Expression in Combination with Cisplatin

Objective: To investigate the effect and mechanism of SP2509 combined with cisplatin on apoptosis and autophagy of osteosarcoma cells. Methods: The expression of LSD1 in osteosarcoma cells MG-63,HuO9,U-2OS and human osteoblast hFoB1.19 were detected by qRT-PCR.The IC50 values of SP2509 and cisplatin were detected by CCK8 method.MG-63 cells were divided into control group,SP2509 group (20μmoL/L),cisplatin group (10μmoL/L),SP2509(20μmoL/L) combined with cisplatin group (10μmoL/L),the level of apoptosis was detected by flow cytometry,and the expressions of LDSS1,LC3-Ⅱ,p62,caspase 3 and Bcl-2 were detected by Western blot. Results: The expression of LSD1 in osteosarcoma cells MG-63,HuO9 and U-2OS was significantly higher than that in hFoB1.19,and the expression of LSD1 was the highest in MG-63.The IG50 values of SP2509 and cisplatin were 40 and 20μmoL/L.Compared with the control group,the apoptosis level of MG-63 cells in SP2509 group and cisplatin group increased significantly (P<0.05),the expression of caspase 3 increased significantly (P<0.05),the expression of Bcl-2 decreased significantly (P<0.05),the expression of LC3-Ⅱ increased significantly (P<0.05),the expression of p62 decreased significantly (P<0.05),and the expression of LSD1 decreased significantly (P<0.05).The level of apoptosis in SP2509 combined with cisplatin group was significantly higher than that in SP2509 group and cisplatin group (P<0.05),the expression of Caspase 3 and LC3-Ⅱ was significantly higher than that in SP2509 group and cisplatin group (P<0.05),and the expression of Bcl-2,P62 and LSD1 was significantly lower than that in SP2509 group and cisplatin group (P<0.05). Conclusion: SP2509 combined with cisplatin can induce apoptosis and autophagy of osteosarcoma cells by regulating the expression of LSD1.

1066 Effect of Nckα Gene Silencing on Malignant Biological Behavior of Epithelial Ovarian Cancer Cells

Objective: To investigate the effect of Nckα gene silencing on the malignant biological behavior of epithelial ovarian cancer cells. Methods: The expression characteristics of Nckα were analyzed in normal ovarian tissue sections,benign ovarian tissue sections and ovarian cancer tissue sections.The effects of Nckα silencing on ovarian cancer cell lines SKOV3 and OVCAR-3 were detected by immunofluorescence,cell invasion,migration,cell proliferation,subcutaneous xenograft animal model,and Western blot. Results: Nckα gene silencing could reduce the invasion and migration ability of ovarian cancer cells.The absorbance of SKOV3 and OVCAR3 cells with Nckα silence was significantly lower than that of the control group,and the absorbance of SKOV3 and OVCAR3 cells with Nckα overexpression was significantly higher than that of the control group (P<0.05).The tumor volume of Nckα silenced group was lower than that of control group (P<0.05).The protein expressions of PI3K P110 α,p70S6K,AKT and P-Akt in SKOV3 cells and OVCAR3 cells silenced by Nckα were significantly lower than those in the control group.The protein expressions of PI3K P110 α,p70S6K,AKT and P-Akt in SKOV3 cells and OVCAR3 cells with Nckα overexpression were significantly higher than those in the control group (P<0.05). Conclusion: Nckα gene silencing can reduce the proliferation and migration ability of epithelial ovarian cancer cells,and delay the growth rate of subcutaneous graft tumor,which may be related to the regulation of PI3K P110 α,p70S6K,AKT and P-Akt protein expression by Nckα gene silencing.

1071 Study of the Effect of HOXC10 on the Stem Cell-Like Properties of Lung Cancer Cells Through Targeted Regulation of CST1 Expression

Objective: To investigate the effect of HOXC10 on the stem cell-like characteristics of lung cancer cells through targeted regulation of CST1 expression. Methods: TLung cancer cell line A549 was cultured under serum-free conditions to form tumour spheres.WB detected the expression of HOXC10 and stemness marker proteins CD44,CD133,Nanog,SOX2 and OCT4 respectively.ShNC and ShHOXC10 A549 cells were constructed to detect the expression of stemness marker proteins CD44,CD133,Nanog,SOX2 and OCT4,tumour sphere-forming assay to detect cell self-renewal ability and CCK8 assay to detect cell proliferation ability.ShNC and ShHOXC10 lung cancer mouse subcutaneous tumour models were constructed,tumour volume and weight were measured,and the expression of HOXC10,CST1 and stemness marker proteins CD44 and CD133 were detected in tumour tissues by immunohistochemistry.CST1 mRNA and protein expression in A549 cells of ShNC and ShHOXC10 groups were detected by qPCR and WB.ChIP-PCR and ChIP-qPCR were used to detect HOXC10 and CST1 promoter sequence binding,and dual luciferase reporter genes were used to verify the regulation of CST1 promoter activity by HOXC10.The effect of CST1 overexpression on the expression of stemness marker proteins CD44,CD133,Nanog,SOX2,OCT4 and tumour spheroid formation and cell proliferation was examined by back-complementing pcDNA3.1-CST1 in ShHOXC10 cells. Results: HOXC10 was highly expressed in the tumour spheres of A549 cells,and knockdown of HOXC10 significantly inhibited the expression of stemness protein markers CD44,CD133,Nanog,SOX2,and OCT4,inhibiting the proliferation and tumour sphere-forming ability of A549 cells.Subcutaneous tumour assays showed that knockdown of HOXC10 inhibited the tumour-forming ability of A549 cells (P<0.001) and tumour weight (P<0.0001) and volume (P<0.01) were significantly reduced. Conclusion: HOXC10 regulates the stem cell-like properties of lung cancer cells through CST1.

1076 Expression and Significance of CXCL1/CXCR2 in Intestinal and Brain Tissues of Neonatal Rats with Necrotizing Enterocolitis

Objective: To investigate the expression of chemokine CXCL1 and its receptor CXCR2 in the intestinal tissues and brain tissues of neonatal Sprague-Dawley rats with necrotizing enterocolitis (NEC),and to explore the pathogenesis of NEC-related preterm brain injury,providing a theoretical basis for the prevention and treatment of preterm brain injury. Methods: Forty-three SPF one-day-old rats(1 died) purchased from Experimental Animal Center of North Sichuan Medical University were selected and randomly divided into the experimental group (21 rats) and the control group (21rats).NEC model of immature rats was established,that is,the experimental group was gavaged 3% Dextran Sulfate Sodium Salt (DSS) 4 times a day for 3 days.The control group was gavaged with saline in the same way.The neonatal mice (6 rats at each time point) were sacrificed by head decapitation at 3h,24h and 72h after intragastric administration.The whole brain and intestine tissues were removed and the contents of CXCL1 and CXCR2 proteins in the intestine and brain tissues were measured by Western Blot at 3h,24h and 72h after gavage,respectively.The western blot bands were analysed in grey scale using GelPro software.Three animals from each group were stained with hematoxylin-eosin at 72 h after gavage for pathological sections of brain and intestinal tissues.Pathological tissue was examined for pathological description and analysis. Results: Compared with the control group,the intestinal tissue of rats in the experimental group was swollen,gas accumulation in the intestinal lumen was obvious,and the intestinal wall was thinner.The intestinal villi were damaged,exfoliated and necrotic,the intestinal glandular body was disorganized and missing,the submucosa and muscle layer was edema and separated,the base was thinned,and inflammatory cells were infiltrated.Brain tissue cell hierarchy is not clear,periventricular white matter porous,loose,decreased glial cells.WB results showed that,compared with the control group,the intestinal CXCL1 protein content in the experimental group increased at 3h and 72h after intragastric administration,with statistical significance (F=9.476,P<0.05).There was no significant difference in the intestinal CXCR2 protein content between the control group and the experimental group at each time point after intragastric administration (F=2.012,P>0.05).Compared with the control group,the brain CXCL1 protein content in the experimental group increased at 72h after intragastric administration,and the difference was statistically significant (F=35.767,P<0.05).Compared with the control group,the expression of CXCR2 protein in the brain tissues of the experimental group increased at 3h、24h and 72h after intragastric administration,with statistical significance (F=50.083,P<0.05). Conclusion: CXCL1/CXCR2 plays an important role in NEC-associated brain injury in preterm infants.And CXCL1/CXCR2 may be involved in the pathogenesis of gut-brain axis in transmitting intestinal inflammation to developing brain injury.

1080 Protective Effect of Umbilical Cord Mesenchymal Stem Cell-Derived Exosomes Carrying TSP-1 on Retinal Degeneration in Rats with Retinal Vein Occlusion

Objective: To investigate the protective effect of human umbilical cord mesenchymal stem cell (hUCMSC)-derived exosomes carrying thrombospondin-1 (TSP-1) in a rat model of retinal vein occlusion (RVO). Methods: The hUCMSCs were randomly divided into hUCMSC group, NC-hUCMSC group and TSP-1-hUCMSC group, the NC-hUCMSC group and TSP-1-hUCMSC group were infected with the lentivirus containing TSP-1 gene sequence and the negative control lentivirus, respectively, and the cell transfection effect was determined by fluorescent inverted microscope, real-time fluorescent quantitative PCR and Western blot. The hUCMSC-derived exosomes carrying the TSP-1 gene were isolated, the morphology was observed by transmission electron microscopy, the diameter was determined by the nanoparticle tracking analysis method, and the expression of the exosomal marker proteins CD9, CD63 and Alix was detected by Western blot. The RVO model of SD rats was established by laser photodynamic method and divided into model group, hUCMSC-Exo group, TSP-1-hUCMSC-Exo group, each with 10 rats, and another 10 healthy ratswerewolf made the control group. The hUCMSC-Exo group was subconjunctivally injected with 100 μl of hUCMSC-derived exosomes, and the TSP-1-hUCMSC-Exo group was injected with 100 μL of hUCMSC-derived exosomes containing TSP-1 lentivirus infection. 1 day, 7 days, 14 days, 21 days after the establishment of the RVO model, the fundus images were observed. 21 d later, the vascular lesions were examined by fluorescein fundus angiography (FFA), and the histopathological changes of the retina were observed by HE staining, Real-time fluorescent quantitative PCR and Western blot were used to detect the mRNA and protein levels of hypoxia-inducible factor-1α (HIF-1α), vascular endothelial growth factor (VEGF) and vascular cell adhesion molecule-1 (VCAM-1) in retinal tissues. Results: The inverted fluorescence microscope showed that there was obvious green fluorescent protein expression in the cells transfected by the lentivirus, and the relative expression of TSP-1 mRNA and protein in hUCMSC in the TSP-1-hUCMSC group were significantly increased (P<0.05), indicates that the cell transfection was successful. The separated particles are spherical with a diameter of approximately 100 nm. CD9, CD63 and Alix proteins are all highly expressed, indicating that exosomes have been successfully isolated. Compared with the control group, different degrees of white occluded blood vessels and plaques appeared in the eyes of the model group on the 1st, 7th, 14th and 21th day of the model, and the model was judged to be successful; in addition, the retinal blood vessels of the model group showed tortuous and dilated veins. Uneven, fluorescence staining and exudation of blood vessel wall, retinal tissue structure was obviously damaged, and the relative expression of HIF-1α, VEGF and VCAM-1 mRNA and protein in the tissue were significantly increased (P<0.05). Compared with the model group, the hUCMSC-Exo group and TSP-1-hUCMSC-Exo group reduced the fluorescence staining of the blood vessel wall, restored the retinal structure, and the relative expression of HIF-1α, VEGF and VCAM-1 mRNA and protein decreased significantly (P<0.05). Compared with the hUCMSC-Exo group, the TSP-1-hUCMSC-Exo group did not see obvious fluorescence leakage in the blood vessels, the retina structure was clearer, and the relative expressions of HIF-1α, VEGF and VCAM-1 mRNA and protein in the tissue were significantly decreased ( P<0.05). Conclusion: HUCMSC-derived exosomes carrying TSP-1 can alleviate retinal damage in RVO rats and protect the retina. The mechanism may be related to the inhibition of the expression of HIF-1α, VEGF and VCAM-1.

1087 Efficacy of Low-Frequency Pulsed Electrical Stimulation and Neostigmine on Postpartum Urinary Retention

Objective: To explore the clinical effects of low-frequency pulse electrical stimulation combined with neostigmine on postpartum urinary retention. Methods: A total of 80 patients with postpartum urinary retention admitted from April 2016 to October 2021 were selected as the research objects, and they were divided into the control group and the observation group by simple random method, with 40 cases in each group. The control group used neostigmine intramuscular injection to induce urination and pelvic floor muscle function exercise, and the study group combined low-frequency pulse electrical stimulation on the basis of the control group. Observe the clinical efficacy of the two groups of patients, and compare the urinary status, grading of urinary retention and sexual function of the patients. Results: The total effective treatment rate in the observation group was 92.50%, which was significantly higher than 75.00% in the control group. The difference was statistically significant (P<0.05). The first urination time in the observation group was shorter than that in the control group, and the residual urine volume was less than that in the control group, with statistically significant difference between the two groups(P<0.05). The urination rate of the observation group (52.50%) was significantly higher than control group (17.50%) within 1 hour after treatment,. and the difference was statistically significant (P<0.05). After 6 weeks of treatment, there was no urinary retention in the observation group (60.00%) , which was significantly higher than in the control group (30.00%),and the difference between the groups was statistically significant (P<0.05). After 6 weeks of treatment, all sexual function scores were higher in the observation group than in the control group, and the difference between the groups was statistically significant (p<0.05) Conclusion: The combination of low-frequency pulse electrical stimulation and neostigmine in the treatment of postpartum urinary retention can help patients to speed up urination, effectively improve the treatment rate of urinary retention, help restore urodynamic function,improve the sexual quality of life, and achieve good clinical effects.

1091 Relationship Between CT Signs and Clinicopathological Characteristics of Multifocal Lung Adenocarcinoma with Ground-Glass Nodules

Objective: To analyze the relationship between CT signs and clinicopathological characteristics in patients with multifocal lung adenocarcinoma with ground-glass nodules (MLA). Methods: A total of 80 patients with MLA confirmed by pathology in the hospital between July 2018 and August 2021 were included as the research subjects. A total of 203 lesions were detected and divided into preinvasive lesion group (92 nodules), microinvasive adenocarcinoma group (69 nodules) and invasive adenocarcinoma group (42 nodules). CT examination was performed on all patients, and the CT signs and clinicopathological characteristics of the three groups were analyzed. Results: There were statistical differences in nodule morphology, lobular sign, vacuole sign, spicule sign, air bronchogram, pleural indentation, tumor-lung interface, vascular characteristics and ground-glass nodule types among preinvasive lesion group, microinvasive adenocarcinoma group and invasive adenocarcinoma group (P<0.05). The proportions of round/elliptical nodules, lobular sign, vacuole sign, spicule sign, air bronchogram, pleural indentation, clear tumor-lung interface, vascular crossing sign and some solid nodules of invasive adenocarcinoma group were higher than those of the other two groups (P<0.05), and the proportions of vacuole sign, spicule sign, pleural indentation, vascular crossing sign and some solid nodules of microinvasive adenocarcinoma group were higher than those of preinvasive lesion group (P<0.05). There were statistically significant differences in the average size of nodules and average size of solid nodules among preinvasive lesion group, microinvasive adenocarcinoma group and invasive adenocarcinoma group (P<0.05), and further pairwise comparison showed that the two indicators of invasive adenocarcinoma group were larger than those of the other two groups (P<0.05), but there were no statistical differences between preinvasive lesion group and microinvasive adenocarcinoma group (P>0.05). Conclusion: CT signs of patients with MLA are closely related to pathological characteristics, and can provide a basis for clinical diagnosis and pathological classification.

1095 Correlation Between Type 2 Diabetes Mellitus and Urinary Stone Species

Objective: To analyze the correlation between type 2 diabetes mellitus (T2DM) and urinary stone species. Methods: One hundred and sixty-five patients with urinary calculi treated admitted in our hospital from January 2019 to December 2021 were enrolled for the research and divided into T2DM group (n = 35) and non-T2DM group (n = 130) according to the occurrence of T2DM before operation. The stone components were analyzed and compared between the two groups by infrared spectroscopy, and the influencing factors of uric acid stones were analyzed by logistic multivariate regression. Results: Calcium oxalate was the most common type of stones in T2DM group and non-t2dm patients, accounting for 48.5% (17/35) and 66.2% (86/130) respectively, followed by uric acid stones, accounting for 28.6% (10/35) and 10.0% (13/130) respectively. There was significant difference in the stone components between the two groups (P < 0.05), and the proportion of uric acid stones in T2DM group was significantly higher than that in non-T2DM group, but there were no significant differences in the proportions of other three stone components between two groups. We further divided all 165 patients with urinary calculi into uric acid calculi group (n = 23) and non-uric acid calculi group (n = 142). The age, body mass index, proportion of hypertension history, proportion of T2DM history, fasting blood glucose, blood triglyceride, uric acid, albumin and serum creatinine (SCr) levels in the uric acid stone group were significantly higher than those in the non-uric acid stone group, and the urine pH in uric acid stone group was significantly lower than that in non-uric acid stone group (P < 0.05). Result of multivariate logistic regression analysis showed that T2DM, blood uric acid, SCr level and urinary pH were the independent influencing factors of uric acid stones (P < 0.05). Conclusion: The stone species of patients with urinary calculi is related to T2DM, and patients with T2DM are more prone to uric acid stones, which is worthy of clinical attention.

1099 Association of ACE and Arg16Gly Genes with Clinical Prognosis and Treatment Response in Children with Asthma

Objective: To analyze the relationship between ACE and Arg16Gly genes and clinical prognosis and treatment response of children with asthma. Methods: A total of 140 children with asthma admitted to our hospital from January 2020 to January 2021 were screened, and the genes related to asthma in oral mucosa cells were detected by DNA sequencing and fluorescent quantitative polymerase chain reaction (PCR). The clinical prognosis characteristics of children with different types of gene mutations and the response to glucocorticoid treatment were compared, and the value of non-invasive detection of asthma genes in children to guide clinical prognosis evaluation and rational drug use were analyzed. Results: The ACE I/D genotype of 140 children was DD, and the dominance ratio of D gene to I gene was 1.98. Adr2arg16Gly genotype is mainly AG, and the dominance ratio of G gene to A gene is 1.89. The disease control rate and total effective rate of ACE I/D genotype DD children are lower than those of type II and ID children. The disease control rate and total effective rate of children with ADR2ARG16Gly genotype GG are lower than those of children with AA and AG genotype. Conclusion: There are significant differences in the expression of SNP in asthma gene-related sites in children with asthma, and it is closely related to the clinical prognosis and drug treatment response of children. Children with ACE I/D genotype DD type and ADRB2 Arg16Gly genotype GG type generally have poor clinical prognosis and limited response to glucocorticoid treatment.

1103 Expression Level and Significance of High Mobility Group Proteins B1 PCT and TGF-β1 in Neonatal Pneumonia

Objective: To analyze the expression levels and significance of HMGB1,PCT and TGF-β1 in neonates with pneumonia. Methods: Eighty children with neonatal pneumonia admitted to our hospital from January 2020 to January 2021 were selected as the neonatal pneumonia group.According to the bacterial infection or not,they were divided into non-bacterial infection group and bacterial infection group,and according to the severity of the disease,they were divided into mild group (34 cases),moderate syndrome group (25 cases) and severe group (21 cases).According to the disease recovery,the patients were divided into acute stage group (50 cases) and remission stage group (30 cases).In addition,80 healthy neonates in the Department of Neonatology of our hospital were selected as healthy control group.The venous blood of all subjects was extracted,and the expression levels of HMGB1 and TGF-β1 were determined by double-antibody sandwich ELISA method,and the expression levels of PCT were detected by flying immunofluorescence detector,and their correlation was analyzed. Results: The expression levels of SERUM HMGB1,PCT and TGF-β1 in neonatal pneumonia group were higher than those in healthy control group (P<0.05).The expression levels of SERUM HMGB1,PCT and TGF-β1 in the bacterial infection group were higher than those in the non-bacterial infection group (P<0.05).The levels of serum HMGB1,PCT and TGF-β1 in severe group were higher than those in mild and moderate group (P<0.05).The expression levels of SERUM HMGB1,PCT and TGF-β1 in acute group were higher than those in recovery group (P<0.05).HMGB1 was positively correlated with PCT (r=0.261,P=0.019).HMGB1 was positively correlated with TGF-β1 (r=0.245,P=0.027).PCT was positively correlated with TGF-β1 (r=0.479,P=0.001). Conclusion: The high expression of HMGB1,PCT and TGF-β1 in neonatal pneumonia was positively correlated with bacterial infection,severity and disease outcome,and all three were involved in the occurrence and development of this disease.

1107 Changes in the Expression of MicroRNA-650 in Osteosarcoma Tissues and Its Significance

Objective: To investigate the expression characteristics of miR-650 in osteosarcoma (OS) tissue,and to analyze the relationship between miR-650 and clinicopathological parameters and prognosis. Methods: Eighty osteosarcoma tissue specimens and paraneoplastic tissue specimens (>5 cm from the tumour) were collected after surgical resection,and real-time PCR was used to detect the expression of miR-650 in tissues,according to the mean expression of miR-650 in OS group,the patients were divided into high expression group of miR-650 (miR-650≥5.03,42 cases) and low expression group of miR-650 (miR-650 < 5.03,38 cases).Information regarding the clinicopathological parameters of patients was collected to compare the differences in miR-650 expression in patients with different clinicopathological parameters.The relationship between miR-650 and prognosis of patients with osteosarcoma was analysed using Kaplan-Meier,COX risk proportion regression at regular follow-up for 3 years after surgery. Results: The relative expression level of miR-650 in osteosarcoma tissue was higher than that in paratumoral tissue (P<0.001),and miR-650 was associated with Enneking stage,lymph node metastasis and distant metastasis in OS patients (P<0.001).Kaplan-Meier survival curve analysis showed that the 3 years survival rate of OS patients in the group with high miR-650 expression was lower than that in the group with low miR-650 expression (P<0.01).Multivariate Cox risk ratio regression analysis showed that lymphatic metastasis[OR=1.545(95%:1.474~1.635)],distant metastasis[OR=1.702(95%:1.664~1.815)],miR-650≥5.03[OR=1.645(95%:1.564~1.748)] were risk factors for death in OS patients (P<0.01). Conclusion: The expression of miR-650 in OS tissues is elevated,which is closely related to the clinicopathological characteristics and prognosis of patients,and may be used as a marker and therapeutic target for the prognosis evaluation of OS.

1112 Serum KL-6 Level and Its Relationship with T Lymphocyte Subsets in RA Patients with Interstitial Lung Disease

Objective: To investigate the serum salivary liquefied carbohydrate antigen-6 (KL-6) level and its relationship with T lymphocyte subsets in patients with rheumatoid arthritis (RA) with interstitial lung disease (ILD). Methods: Seventy-five patients with RA with ILD in our hospital from September 2020 to September 2021 were selected as the study group,and 75 patients with RA alone in the same period were selected as the control group according to the 1:1 matching case-control principle.The general data of the two groups were compared,the serum KL-6 level was detected by enzyme-linked immunosorbent assay,the T lymphocyte subsets (CD3+,CD4+,CD8+) were detected by flow cytometry,and RA activity was assessed using the Disease activity score 28 (DSA 28),and the patients with different degrees of pulmonary ventilation disturbance and pulmonary diffusion dysfunction in the study group were compared.Serum KL-6 level,and the correlation between serum KL-6 level and T lymphocyte subsets,RA activity,degree of pulmonary ventilation disturbance,and degree of pulmonary diffusion dysfunction in the study group were analyzed.All patients in the study group received symptomatic treatment,and the curative effect was evaluated after 6 months of treatment.The serum KL-6 levels before treatment,1 month,3 months,and 6 months of treatment in patients with different curative effects were compared,and the value of serum KL-6 in predicting curative effect was evaluated by receiver operating characteristic (ROC) curve. Results: The serum KL-6 level and DSA 28 score of the study group were higher than those of the control group (819.65±85.34 vs 418.93±52.67,t=34.605,P<0.001;3.46±0.85 vs 2.59±0.77,t=6.569,P<0.001),and the levels of CD3+,CD4+ and CD8+ were lower than those of the control group (41.69±5.39 vs 44.86±5.72,t=3.493,P =0.001;17.82±3.24 vs 23.69±3.86,t=10.087,P<0.001;18.05±2.81 vs 25.73±3.42,t=15.026,P<0.001).The serum KL-6 level in the moderate patients was higher than that in the mild patients (P<0.05); the serum KL-6 level in the study group was negatively correlated with the levels of CD4+ and CD8+ (r=-0.526,P<0.001,r=-0.713,P<0.001),and with the DSA 28 score,the degree of pulmonary ventilation disorder,and the degree of pulmonary diffusion dysfunction were positively correlated (r=0.429,P<0.001,r=0.802,P<0.001,r=0.851,P<0.001); the serum KL-6 levels in patients with poor efficacy before treatment 1 month,3 months and 6 months of treatment were higher than those in patients with good efficacy (P<0.05); the area under the curve (AUC) of serum KL-6 level before treatment for predicting poor efficacy in patients with RA and ILD was 0.838,and the predictive sensitivity and specificity were 75.02% and 88.14%,respectively. Conclusion: The level of serum KL-6 in RA patients with ILD is significantly increased,and its level is closely related to the abnormality of T lymphocyte subsets,and it can assist in clinical evaluation of patients' condition and predict curative effect,and has high application value.

1118 Analysis of the Efficacy and Toxic Effects of Different Combination Modalities of Radiotherapy for Non-Dmall Cell Lung Cancer Patients and The Impact on Prognostic Factors

Objective: To investigate the efficacy,toxic effect and prognostic factors of different combination modalities of radiotherapy and chemotherapy in patients with non-small cell lung cancer. Methods: The 200 patients with pathologically confirmed non-small cell carcinoma were divided into 3 groups using different treatment modes,including 78 patients in the chemotherapy-only group,68 patients in the sequential chemoradiotherapy group and 54 patients in the synchronous chemoradiotherapy group,and the clinical efficacy,toxic effects and prognostic factors of the three groups were analysed. Results: The short-term total effective rates (complete remission + partial remission) of chemotherapy alone group,sequential chemoradiotherapy group,and synchronous chemoradiotherapy group were 39.74% (31/78),73.53% (50/68),81.48% (44/54),respectively. The comparison between sequential chemoradiotherapy group and the chemotherapy alone group,and the comparison between the synchronous chemoradiotherapy group and the chemotherapy alone group,had statistical significance (P<0.05); the mortality rates of the chemotherapy alone group,the sequential chemoradiotherapy group,and the synchronous chemoradiotherapy group were67.95%(53/78),67.65%(46/68),53.70%(29/54),respectively.Median survival times averaged 12 months (95% confidence interval 6.6-17.2),10 months (95% confidence interval 7.0-14),25 months (95% confidence interval 23.1-28.6).The median survival time in the synchronous chemoradiotherapy group was higher than that in the sequential chemoradiotherapy and chemotherapy alone groups,as seen by statistical analysis (P<0.05).There was a difference between the simultaneous chemoradiotherapy group and the chemotherapy alone group by statistical analysis (P<0.05),and the overall survival rates at 1,2 and 3 years for the three groups were different by statistical analysis (P<0.05).The main side effects in the three groups were bone marrow suppression.The rates of grade Ⅲ and Ⅳ leukopenia in the chemotherapy alone,sequential chemoradiotherapy and synchronous chemoradiotherapy groups were 8.97% (7/78),1.47% (1/68) and 12.96% (7/54) respectively,and there was a statistical difference between the sequential chemoradiotherapy group and the chemotherapy alone and synchronous chemoradiotherapy groups (P<0.05).The decrease in grade Ⅲ and Ⅳ haemoglobin in the chemotherapy alone group,sequential chemomradiotherapy group and simultaneous chemoradiotherapy group was 1.28% (1/78),0 (0/68) and 0 (0/54),and there was a difference in the chemotherapy alone group compared with the sequential chemoradiotherapy group by statistical analysis (P<0.05).Grade Ⅲ and Ⅳ gastrointestinal reactions were 8.97% (7/54),1.47% (1/68) and 0 (0/54) in the chemotherapy alone group,sequential chemoradiotherapy group and synchronous chemoradiotherapy group,respectively,and were statistically different in the sequential chemoradiotherapy group compared with the chemotherapy alone group and the sequential chemoradiotherapy group compared with the synchronous chemoradiotherapy group (P<0.05).In the sequential chemoradiotherapy group and synchronous chemoradiotherapy group,7.35% (5/68) and 5.56% (3/54) of grade Ⅱ~Ⅲ radiation lung injury were found respectively,with no difference by statistical analysis (P>0.05).The percentage of grade Ⅱ~Ⅲ radiation esophagitis in the sequential chemoradiotherapy and synchronous chemoradiotherapy groups was 5.88% (4/68) and 11.11% (6/54),respectively,with no difference by statistical analysis (P>0.05).After univariate analysis,age,gender,PS score,clinical stage,cumulative organ count and pre-treatment haemoglobin level (P<0.2) were all multifactorial for small cell carcinoma of the lung.Screening by the ENTER method showed a=0.05 age,PS score and clinical stage were prognostic influencing factors for non-small cell carcinoma. Conclusion: The main factors affecting non-small cell lung cancer are age,PS score,and clinical stage.The clinical stage is directly related to the survival rate of patients.The earlier the stage,the more obvious the median survival time and survival rate; the prognosis of the patient is also crucial and should be given special attention in clinical work.

1124 A Controlled Study of the Effects of Cryo-Balloon Catheter Ablation on the Coagulation System Inflammatory Response and Prognosis of Patients with Atrial Fibrillation

Objective: To analyze the effects of cryo-balloon catheter ablation on the coagulation system,inflammation and prognosis of patients with atrial fibrillation. Methods: A total of 93 patients with atrial fibrillation in our hospital from Jul. 2017 to Nov.2021 were selected and divided into cryo group (n=47) and RF group (n=46) according to different treatment plans.Radiofrequency ablation was performed in the RF group,and cryo-balloon catheter ablation was performed in the cryo group.The two groups were compared in terms of operative time,ablation time,success rate of pulmonary vein isolation (PVI),complications and preoperative,1 d and 3 d postoperative levels of coagulation parameters [activated partial thromboplastin time (APTT),fibrinogen (FIB),D-dimer (D-D)],inflammatory parameters [interleukin-6 (IL-6),supersensitive-C-reactive protein (hs-CRP),white blood cell count (WBC) WBC)] levels, 6 month and 12 months cardiac function parameters preoperative and postoperative [left ventricular ejection fraction (LVEF),left ventricular end-systolic diameter (LVESD),left ventricular end-diastolic diameter (LVEDD)],and 6-month and 12-month postoperative recurrence rates. Results: The difference between the two groups was not statistically significant when comparing the success rate of PVI (P>0.05); the operative time and ablation time in the cryo group were shorter than those in the RF group (P<0.05); compared with the preoperative period,the APTT level decreased in both groups at 1 d and 3 d postoperatively,but was higher in the cryo group than in the RF group; the plasma FIB,D-D,WBC and serum IL-6 and hs-CRP levels increased,but were lower in the cryo group than in the RF group (P<0.05).The incidence of complications was 6.38% in the cryo group compared with 13.04% in the RF group,and the difference was not statistically significant (P>0.05); the LVEF levels increased and the LVESD and LVEDD levels decreased at 6 and 12 months postoperatively compared with the preoperative levels in both groups,and the cryo group was better than the RF group (P<0.05); the recurrence rate at 12 months postoperatively was 19.15% in the cryo group compared with 39.5% in the RF group.The recurrence rate of cryo group at 12 months after surgery was 19.15% lower than that of the RF group,which was 39.13% (P<0.05). Conclusion: The cryo-balloon catheter ablation for the treatment of atrial fibrillation has the advantages of convenient operation,good postoperative cardiac function improvement,low recurrence rate,etc.,and has little impact on the body's coagulation system and mild inflammatory response.

1130 Serum LncRNA CASC15 LncRNA MALAT1 Expression and Clinical Significance in Hemodialysis Patients with End-Stage Renal Disease

Objective: To investigate the expression and clinical significance of serum LncRNA CASC15 and LncRNA MALAT1 in hemodialysis patients with end-stage renal disease. Methods: Patients with end-stage renal disease who underwent regular dialysis treatment in our hospital from January 2019 to December 2020 were selected as the research group,and 62 patients with CKD stage Ⅲ-Ⅳ who started dialysis treatment for the first time in our hospital were randomly selected according to the ratio of 1:1 as the control group.Blood samples were tested for creatinine (SCr),uric acid (UA),eGFR,total cholesterol (TC),triglycerides (TG),urea (BUN),haemoglobin,C-reactive protein (CRP),calcium and phosphorus by real-time fluorescence quantitative polymerase chain reaction (RT-PCR) based on SYBR Green method to amplify the target sequences.Patients in the study group were followed up for the occurrence of clinical endpoint times within 1 year of starting dialysis. Results: The expressions of serum LncRNA CASC15 and LncRNA MALAT1 in the study group were significantly higher than those in the control group (P>0.05).); LncRNA MALAT1 high expression of SCr,UA,BUN was higher than LncRNA MALAT1 low expression group,eGFR and hemoglobin were lower than low expression group,the difference was statistically significant (P<0.05); uremic patients with different LncRNA CASC15 expression levels developed cardiovascular disease.There were statistically significant differences in events and all-cause mortality (P<0.05),and differences in the expression levels of different LncRNA CASC15 and LncRNA MALAT1 for giving up treatment (all-cause death) were statistically significant (P<0.05).There was no significant difference in the incidence of cerebrovascular accident,serious infection and gastrointestinal bleeding among the expression level groups (P>0.05). Conclusion: LncRNA CASC15 and LncRNA MALAT1 are significantly up-regulated in the serum of end-stage renal disease hemodialysis patients,which are closely related to the clinical parameters of patients and the time of dialysis clinical endpoints,and are expected to be therapeutic targets for in-depth research.

1135 Analysis of the Effect of Different Auxiliary Intubation on Difficult Intubati on Endoscopic Retrograde Cholangiopancreatography and the Influencing Factors of Postoperative Acute Pancreatitis

Objective: To study the effect of different auxiliary intubation on the intubation effect of difficult intubation endoscopic retrograde cholangiopancreatography and the influencing factors of postoperative acute pancreatitis. Methods: Ninety-four patients admitted to our hospital from October 2018 to December 2021 were selected for retrospective analysis and grouped according to the different auxiliary intubation methods of the patients,namely the guidewire group (36 patients,double guidewire intubation method),needle group (30 patients,needle knife nipple pre-incision method) and trans-pancreatic group (trans-pancreatic duct nipple pre-incision method).The intubation success,intubation time and postoperative clinical effect of blood amylase were observed in the three groups.The adverse events such as acute pancreatitis,multiple intubation and balloon dilation were compared among the three groups,and the influencing factors of acute pancreatitis were analyzed and studied. Results: Treatment effect:The differences in successful intubation (55.6%,93.3% and 71.4%,respectively,χ2=11.688,P=0.003) and postoperative blood amylase (526.35±213.54 U/L,186.35±53.54 U/L and 289.45±136.68 U/L,respectively,F=42.197,in the guide wire,needle and trans-pancreatic groups.P<0.001) was statistically significant; the difference in intubation time was not statistically significant (P>0.05).Adverse event profile:The differences in the incidence of acute pancreatitis (8.3%,26.7%,10.7%),multiple intubations (16.7%,33.3%,17.9%) and balloon dilation (19.4%,26.7%,21.4%) were not statistically significant (P>0.05) between the three groups of patients.The differences in univariate screening for acute pancreatitis were statistically significant (P<0.05) for multiple intubations (OR=4.714,95% CI:1.426 to 15.588),balloon dilation (OR=4.714,95% CI:1.426 to 15.588),prophylactic medication (OR=0.030,95% CI:0.007 to 0.132).Binary logisitc multifactorial regression yielded independent risk factors affecting the development of acute pancreatitis,mainly prophylactic medication (OR=0.001,95% CI:0.000-0.043),age (OR=1.191,95% CI:0.990-1.433),time to intubation (OR=0.308,95% CI:0.116-0.820),blood amylase 0.820),blood amylase (OR=1.168,95%CI:1.041~1.311). Conclusion: The success rate of intubation was best with needle knife nipple preincision,followed by trans-pancreatic duct nipple preincision and bottom with double guidewire intubation; independent risk factors affecting acute pancreatitis after ERCP were multiple intubations,balloon dilation,prophylactic medication,age,time of intubation and blood amylase.

1141 Effects of Aryl Hydrocarbon Receptor AhR on Myocardial Cell Injury and Pyroptosis in CVB3-Induced Viral Myocarditis Mice

Objective: To explore the effect of inhibiting aryl hydrocarbon receptor (AhR) on myocardial injury and cell pyroptosis in mice with coxsackievirus 3 (CVB3)-induced viral myocarditis (VMC). Methods: Totally 60 BALB/c mice were randomly divided into control group (Control), AhR inhibitor group (AhR inhibitor), model group (VMC), and VMC+AhR inhibitor group (VMC+AhR inhibitor), 15 mice in each group. Mice in VMC group and VMC+AhR inhibitor group were intraperitoneally injected with CVB3 to establish a myocarditis model. Mice in AhR inhibitor group and VMC+AhR inhibitor group were intraperitoneally injected with AhR inhibitor CH223191 (10 mg/kg). After 10 days, the cardiac ultrasound indexes of each group were detected by the high-resolution Vevo2100 small animal ultrasound imaging system; ELISA method was used to detect serum cardiac troponin l (cTnl), tumor necrosis factor-α (tumor necrosis factor-α, TNF-α), interleukin-1β (interleukin-1β, IL-1β) and IL-18; Hematoxylin-Eosin (HE) staining was used to observe the pathological changes of myocardial tissue; TUNEL staining to detect the apoptosis of myocardial tissue cells; immunohistochemical staining to determine the positive expression levels of NOD-like receptor pyrin domain containing 3 (NLRP3) and Gasdermin-D (GSDMD) in myocardial tissue; real-time fluorescent quantitative PCR and Western blot to detect expression levels on mRNA and protein of cysteinyl aspartate specific proteinase (caspase-1) , apoptosis-associated speck-like protein containing CARD (ASC), NLRP3 and GSDMD in myocardial tissue. Results: Compared with the control group, FS and EF decreased, LVIDd and LVAWd increased, the levels of cTnl, TNF-α, IL-1β and IL-18 in serum increased, the cell arrangement in myocardial tissue was disordered, accompanied by a large number of inflammatory cell infiltration, the increase of TUNEL positive rate in myocardial tissue, the NLRP3 positive expression rate and GSDMD positive expression rate. The relative expression of caspase-1, ASC, NLRP3 and GSDMD mRNA and protein were all upregulated, and the differences were statistically significant (P<0.05); Compared with the VMC group, FS and EF were elevated, LVIDd and LVAWd were reduced, serum levels of cTnl, TNF-α, IL-1β and IL-18 were decreased, myocardial histopathological damage was significantly reduced, TUNEL positive rate in myocardial tissue was decreased, meanwhile, NLRP3 positive expression rate and GSDMD positive expression rate. The relevant expression of caspase-1, ASC, NLRP3 and GSDMD mRNA and protein were downregulated, and the differences were statistically significant (P<0.05). Conclusion: Inhibition of AhR played a protective role against CVB3-induced viral myocarditis in mice anda was able to reduce myocardial tissue damage and inhibit cardiomyocyte scorching.

1148 Correlation Analysis Between Bone Mineral Density and Serum Vitamin D IGF -1 and IGFBP-3 in Children with Dwarfism

Objective: To analyze the correlation between bone mineral density and serum vitamin D,insulin-like growth factor-1(IGF-1) and insulin growth factor binding protein-3(IGFBP-3) in children with short stature. Methods: One hundred and forty children with dwarfism admitted to our hospital from March 2016 to March 2021 were selected and included in the Idiopathic short stature (ISS) group and Growth hormone deficiency (GHD) group according to their growth hormone (GH) levels after 2 drug provocation tests,and 40 healthy children with physical examination during the same period were selected and included in the control group.The bone mineral density and serum vitamin D,IGF-1 and IGFBP-3 levels of children in each group were detected,and the correlation between bone mineral density and serum indexes of children with short stature was calculated by Pearson correlation analysis. Results: The bone mineral density of children in the control group,ISS group and GHD group decreased in turn,and the difference between the groups was statistically significant (P<0.05).Children's serum 25- hydroxyvitamin D,IGF-1 and IGFBP-3 in the control group,ISS group and GHD group decreased in turn,and the difference between the groups was statistically significant (P<0.05).Pearson correlation analysis showed that the bone mineral density of ISS and GHD children was positively correlated with serum 25- hydroxyvitamin D,IGF-1 and IGFBP-3 (r=0.627,0.660,0.625,P<0.05). Conclusion: Children with short stature caused by different causes have different degrees of decrease in bone density,and the decrease of bone density in children is closely related to the decrease of serum 25-hydroxy-vitamin D,IGF-1,IGFBP-3.

Objective: To study the effectiveness of extracorporeal magnetic wave pelvic floor therapy combined with pelvic floor muscle training on postpartum pelvic floor dysfunction in different delivery modes. Methods: A total of 120 patients with pelvic floor dysfunctional disorders who were reviewed at 6-8 weeks postpartum in our hospital from January 2020 to January 2022,including 60 vaginal deliveries and 60 cesarean deliveries,were selected and randomly divided into control group (vaginal delivery control group A and cesarean delivery control group B) and experimental group (vaginal delivery experimental group A and cesarean delivery experimental group B).The control group was given pelvic floor muscle training and the experimental group was given pelvic floor muscle training + extracorporeal magnetic wave pelvic floor therapy.Before and after treatment,each group was compared in terms of the clinical efficacy,pelvic floor electromyography Glazer assessment,Pelvic Floor Dysfunction Questionnaire (PFDI-20),Pelvic Floor Impact Questionnaire (PFIQ-7) and clinical complications. Results: After treatment,there was no significant difference between the clinical efficacy of experimental group A and experimental group B; the total effective rate of both experimental group A and experimental group B was significantly higher than that of control group A and control group B.The difference was significant (χ2=14.471,P<0.05).Before treatment,there was no significant difference in the comparison of the maximum value of fast muscle contraction,the mean value of 10s slow muscle contraction and the mean value of 60s slow muscle contraction between the groups; after treatment,the maximum value of fast muscle contraction,the mean value of 10s slow muscle contraction and the mean value of 60s slow muscle contraction were significantly higher in each group,and the difference was higher in experimental group A and experimental group B than in control group A and control group B.The difference was significant (t=3.629,4.983,4.991,3.774,5.199,4.669,P<0.05),and the increase in the mean 10s slow muscle contraction and the mean 60s slow muscle contraction was higher in the experimental group A than in the experimental group B.Before treatment,the pre and post resting muscle tension was higher in control group B and experimental group B than in control group A and experimental group A.After treatment,the pre and post resting pelvic floor muscle tension was significantly lower in experimental group B,while the remaining groups did not change significantly; before treatment,there was no significant difference in the comparison of PFIQ-7 and PFDI-20 scores between the groups; after treatment,the PFIQ-7 and PFDI-20 scores were significantly lower in all groups,and experimental group A and experimental group B had lower scores than control group A and control group B respectively,with significant differences (P<0.05).Before treatment,there was no significant difference in the total incidence of complications such as stress urinary incontinence between the groups; after treatment,the total incidence of complications such as stress urinary incontinence was significantly lower in both experimental group A and experimental group B than in control group A and control group B.The difference was significant (χ2=9.629,11.193,P<0.05). Conclusion: The combination of extracorporeal magnetic pelvic floor therapy and pelvic floor muscle training achieved significant results in treating pelvic floor dysfunction in women with different modes of delivery,all of which were better than pelvic floor muscle training alone,and the recovery of pelvic floor dysfunction was better in women who delivered vaginally than in those who delivered by caesarean delivery.

1157 Effect of Acupoint Acupuncture Combined with Core Stabilization Training on Balance Function and Quality of Life of Post-Stroke Hemiplegic Patients

Objective: To explore the effect of acupuncture point combined with core stabilization training on quality of life and balance function of post-stroke hemiplegia patients. Methods: From February 2020 to February 2021,151 patients with post-stroke hemiplegia were collected and divided into a control group (n=75) and an observation group (n=76) according to a simple randomized grouping method.The patients in the control group were treated with acupuncture using the "Cure Shen and Tone Marrow" acupuncture point group,while the patients in the observation group received core stabilisation training on top of the control group.The National Institutes of Health Stroke Scale (NIHSS) scores,Barthel scores,Functional Movement Status (FMA) scores,Breg Balance Scale (BBS) scores,10m maximum walking speed (10MWS) and quality of life were compared between the two groups. Results: The difference in NIHSS by time was statistically significant (F time=21.241,P<0.05),there was a difference in NIHSS scores between the two groups (Fbetween groups=6.775,P<0.05),and the interaction between group and time on the effect of NIHSS scores was statistically significant (Finteraction=15.244,P<0.05),both groups showed a decreasing trend,with a greater decrease in the observation group.After treatment,Barthel and FMA scores increased significantly in both groups compared to the pre-treatment scores,and the increase in the observation group was more significant than that in the control group.The 10MWS and BBS scores increased more significantly in the observation group than in the control group.The SF-36 scores increased significantly in both groups compared to the pre-treatment scores,and the increase in the SF-36 scores was more significant in the observation group compared to the control group (P<0.05). Conclusion: Acupoint acupuncture combined with core stabilization training is beneficial to the recovery of nerve function in patients with post-stroke hemiplegia,and can improve their daily living ability,motor function,balance function and quality of life.

1162 Clinical Efficacy of COOK Balloon versus Prostaglandin Preparation for Full-Term Induction of Labour in Pregnant Women with Gestational Hypertension

Objective: To compare the clinical efficacy of cook balloon and prostaglandin preparation for full-term induction of labor in pregnant women with gestational hypertension. Methods: A total of 80 full-term pregnant women with hypertension from August 2019 to January 2021 were randomly divided into experimental group and control group.40 pregnant women in the control group were induced by prostaglandin and 40 pregnant women in the study group were induced by COOK balloon.The delivery status,complications during induction,postpartum hemorrhage and neonatal outcome of pregnant hypertensive pregnant women in the two groups were compared. Results: Delivery status:The number of vaginal deliveries,cervical maturity Bishop score,cervical maturation efficiency,oxytocin use and manual rupture of membranes were significantly higher in the experimental group than in the control group using misoprostol given vaginally,with a statistically significant difference (P<0.05).Complications during induced labor:No amniotic fluid embolism occurred during induction of labour in either group,and the incidence of complications during induction of labour was significantly lower in the experimental group than in the control group (22.50% vs.47.50),with a statistically significant difference (χ2=5.495,P=0.019).Postpartum hemorrhage and neonatal outcome:The amount of postpartum hemorrhage in the experimental group treated with cook balloon was significantly lower than that in the control group treated with misoprostol through vagina,the difference was statistically significant (t=6.973,P<0.05).There was no statistically significant difference between the two groups in terms of Apgar score,body mass,number of admissions to the nursing unit,number of cases on oxygen and complication rate (P>0.05). Conclusion: The COOK cervical dilatation balloon is superior to misoprostol given vaginally for the relief of labour,reduction of complications during induction of labour and postpartum haemorrhage during full-term induction of labour in pregnant women with gestational hypertension and is worthy of clinical promotion.

1166 A Study on the Distribution of Pathogenic Bacteria and Risk Factors for Multidrug Resistant Bacterial Infections in ICU Ventilator-Associated Pneumonia

Objective: To investigate the distribution of pathogenic bacteria and risk factors of multidrug-resistant bacteria infection in patients with ventilator-associated pneumonia in ICU. Methods: Clinical data of 150 patients with ventilator-associated pneumonia admitted to our hospital from January 2018 to December 2020 were retrospectively collected, and the distribution of pathogenic bacteria in patients was analyzed. Patients were divided into the drug-resistant bacteria group (93 cases) and the non-drug-resistant bacteria group (57 cases) according to whether multidrug-resistant bacteria infection occurred. The single factor of multidrug-resistant bacteria infection in patients with ventilator-associated pneumonia ICU was analyzed, and the risk factors of multiple drug-resistant bacteria infection in patients with ventilator-associated pneumonia in ICU were analyzed by multivariate Logistic regression. Results: A total of 240 strains of pathogenic bacteria were isolated from 150 patients with ventilator-associated pneumonia in ICU, mainly gram-negative bacteria, accounting for 84.17%, among which the top three pathogens were Klebsiella pneumoniae, Acinetobacter baumannii and Pseudomonas aeruginosa (25.42%, 20.42%, 12.50%). A total of 93 cases of multidrug-resistant bacteria infection occurred in 150 patients with ventilator-associated pneumonia in ICU, and the infection rate was 62.00%. Univariate analysis showed that the proportion of patients with diabetes mellitus, disturbance of consciousness, dysphagia, late type of stroke associated pneumonia, the treatment of the 3rd and 4th generation cephalosporins as well as complex preparations containing enzyme inhibitors and carbapenems in the past 90 days, time of ventilator ventilation ≥7 d, ICU stay ≥7 d in the drug-resistant bacteria group was higher than that in the non-drug-resistant bacteria group (P<0.05). Multivariate Logistic regression analysis showed that the disturbance of consciousness, late type of stroke associated pneumonia, the treatment of the 3rd and 4th generation cephalosporins as well as complex preparations containing enzyme inhibitors and carbapenems in the past 90 days, time of ventilator ventilation ≥7 d, ICU stay ≥7 d were all independent risk factors for multidrug-resistant bacteria infection in patients with ventilator-associated pneumonia in ICU (OR=4.644, 3.152, 4.289, 2.421, 17.167, P<0.05). Conclusion: The main pathogen of ventilator-associated pneumonia in ICU was Klebsiella pneumoniae, Acinetobacter baumannii and Pseudomonas aeruginosa, patients with ventilator-associated pneumonia in ICU correlation independent risk factors for infection multi-resistant bacteria including the occurrence of disturbance of consciousness, late onset of ventilator-associated pneumonia, indent gastric tube, time of ventilator ventilation ≥7 d, and ICU stay ≥7 d. Clinically, targeted intervention or treatment could be carried out for patients with the above characteristics to reduce the risk of multidrug-resistant bacteria infection in patients with ventilator-associated pneumonia in ICU.

1172 The Effect of Presacral-Retro-Psoas Space Block-Selective Nerve Root Block on the Dosage of Anesthetics in General Anesthesia for Below-Knee Osteoporotic Fractures and Its Clinical Significance

Objective: To investigate the effect of presacral-retro-psoas space block-selective nerve root block on the dosage of anesthetics during general anesthesia in patients with osteoporotic fractures below the knee and its clinical significance. Methods: From January 2020 to June 2021,70 patients with osteoporotic fractures below the knee who were treated in the Department of Orthopaedic Surgery of our hospital and received surgical treatment were selected as subjects,and divided into general anesthesia group according to different anesthesia methods (n=37,Simple general anesthesia) and combined group (n=33,presacral-retro-psoas space block-selective nerve root block assisted by general anesthesia).The fluctuations of hemodynamic indexes [heart rate (HR),systolic blood pressure (SBP),diastolic blood pressure (DBP),mean arterial pressure (MAP)] in the combined group before and after nerve block were observed; the pain at each time point after the operation was compared between the two groups Differences in visual analogue scale (VAS) score,perioperative anesthetic dosage,postoperative recovery and other indicators. Results: There were no significant differences in HR,SBP,DBP and MAP fluctuation at 5min,10min and 20min after nerve block in combined group compared with before the block (P>0.05).The dosage of remifentanil in the combined group was (0.77±0.03) mg,less than that in the general anesthesia group (0.90±0.05) mg (P<0.05),and the number of relief analgesia of sufentanil in the combined group was (0.84±0.14) times,less than that in the general anesthesia group (1.30±0.27) times (P<0.05).There were significant differences in VAS scores at all time points in the rest and exercise states in the combined group (P<0.05),there were significant differences in VAS scores at all time points in the rest state in the general anesthesia group (P<0.05),and there was no significant difference in VAS scores at all time points in the exercise state (P>0.05).VAS scores in the combined group at 6h,12h and 24h after surgery were lower than those in the general anesthesia group (P<0.05),and VAS scores in the combined group at 12h after surgery at exercise were lower than those in the general anesthesia group (P<0.05).There was no significant difference in the time of anal exhaust between the two groups (P>0.05),and the time of passive exercise in bed in the combined group was (3.36±0.32) h,which was earlier than that in the general anesthesia group (4.02±0.33) h (P<0.05). Conclusion: The presacral-retro-psoas major space block-selective nerve root block combined with general anesthesia has stable hemodynamics in patients with below-knee osteoporotic fracture surgery,which can effectively reduce the amount of perioperative anaesthetic drugs and improve the analgesic effect,which is of specific clinical significance in promoting the postoperative recovery of patients.

1177 Analysis of the Effectiveness of Autogenous Fiber Blood Clot Transplantation Combined with Arthroscopic FasT-Fix Repair surgery in Patients with Meniscal Tears

Objective: To analyze the therapeutic effect of autogenous fiber blood clot transplantation combined with arthroscopic FasT-Fix for meniscus tear. Methods: The clinical data of 145 patients with meniscus tear admitted to our hospital from February 2019 to February 2021 were retrospectively analyzed and divided into observation group (n = 74) and control group (n = 71) according to surgical methods.The control group underwent arthroscopic FasT-Fix surgery,and the observation group underwent autogenous fiber blood clot transplantation combined with arthroscopic FasT-Fix surgery.The knee function (IKDC),pain (VAS),Lysholm and knee motion (Tegner) scores,prognosis and incidence of adverse reactions were compared between two groups before and after treatment. Results: Comparison of the difference between the two groups before and after treatment showed that the increase of Lysholm and Tegner scores in the observation group was greater than that in the control group (P<0.05),the decrease in the VAS score in the observation group was greater than that in the control group (P<0.05),and the increase in IKDC was higher than that in the control group (P<0.05).The incidence of adverse reactions in observation group (2.70%) was significantly lower than that in control group (11.27%),and the difference was statistically significant (P<0.05).Meniscus healing status of observation group was better than control group (P<0.05). Conclusion: Compared with arthroscopic FasT-Fix repair surgery alone,the combined treatment with autologous fibrillary blood clot transplantation was more beneficial to improve the Lysholm,Tegner and IKDC scores of meniscus tears,reduce the VAS scores and the incidence of adverse reactions,and promote meniscus healing.

1182 Effectiveness of 3D Printed Guides in the Restoration of Dentition Defects and the Effect on the Inflammatory Response of the Gingival Sulcus and Quality of Life

Objective: To investigate the application effect of 3D printed guide in dentition defect restoration and its effect on gingival sulcus inflammatory reaction and quality of life. Methods: A total of 86 patients with dentition defects (86 teeth) from October 2019 to October 2020 in our hospital were selected and grouped by random number table method,43 cases in each (43 teeth in each).The control group had traditional implant guides and the study group had 3D printed guides.The implant survival rate,implant cusp and neck deviation values,and complications of the 2 groups were counted,and the masticatory efficiency,occlusal force,gingival sulcus fluid inflammatory factors [interleukin-6 (IL-6),interleukin-8 (IL-8),tumor necrosis factor-α(TNF-α)],and oral health impact scale (OHIP-14) were compared before and after the surgery. Results: The deviation values of apex,neck mesio-distal,buccal-lingual,and vertical in the study group were lower than those in the control group (P<0.05); At 12 months postoperative follow-up,the survival rate of the implants in the study group was 100.00% compared to 75.00% in the control group (P<0.05);Masticatory efficiency,occlusal force and IL-6,IL-8 and INF-α levels were higher in both groups at 6 and 12 months postoperative than preoperative (P<0.05).There was no significant difference in the incidence of complications between the two groups (P>0.05); (5) OHIP-14 scores were higher than those before operation (P<0.05). Conclusion: The application of 3D printed guide plate in patients with dentition defect repair helps reduce the deviation of implant tip and neck,improve implant survival rate,improve quality of life,and has less effect on gingival crevicular fluid inflammatory reaction.

1188 Effectiveness of 3D Printed Individualized Navigation Templates in Lumbar Screw Implantation and the Effect on Patient Lumbar Spine Function and Stress Hormones

Objective: To explore the application effectiveness of 3D printed individualised navigation template in lumbar screw implantation and its effect on patients' lumbar spine function and stress hormones. Methods: A total of 122 patients with lumbar disc herniation and lumbar spinal stenosis in our hospital from January 2019 to January 2021 were selected.The random number table method was used to randomly divide 122 patients into two groups,with 61 cases in each group.The control group underwent lumbar spine screw implantation,and the 3D printed individualized navigation template assisted the lower lumbar spine screw implantation in the study group.The perioperative indicators,surgical outcomes and complications in the 2 groups were observed,and the stress hormones [catalase (CAT),malondialdehyde (MDA),reactive oxygen species (ROS)],spinal cord function [nerve conduction velocity (NCV),latency (DL)] and Oswestry Dysfunction Index Questionnaire (ODI) and visual analogue scoring method (VAS) scores before and after surgery were compared. Results: The study group had a shorter nail placement time than the control group,fewer X-ray machine fluoroscopies than the control group and a lower overall complication rate than the control group (P<0.05); The rate of surgical excellence in the study group was 86.89%,higher than that in the control group (72.13%,P<0.05);The differences in MDA,CAT and ROS between groups and time and the interaction between group and time were statistically significant (P<0.05); the effect of MDA,CAT and ROS levels of the study group was smaller than that of the control group (P<0.05); There is no statistically significant differences in NCV,DL between groups,at different times and the interaction of group and time (P>0.05); ( 5) The differences in ODI and VAS between groups and time and the interaction between group and time were statistically significant (P>0.05),and all showed a decreasing trend over time,with a greater decrease in the study group than in the control group (P<0.05). Conclusion The application of 3D printed individualized navigation template in lumbar screw implantation reduces the nail placement time,decreases complications,increases the good rate of nail placement,improves lumbar spine function,and has less impact on oxidative stress.

1194 Incidence and influencing factors of postprandial hypotension in patients with OSAHS

Objective: To understand the incidence and factors affecting postprandial hypotension in OSAHS patients and to provide a basis for the prevention and treatment of PPH in OSAHS patients through measuring incidence of postprandial hypotension (PPH) in patients with and without obstructive sleep apnea-hypopnea syndrome (OSAHS). Method: Ninety-eight patients with OSAHS diagnosed by PSG were divided into mild,moderate and severe groups according to AHI.There were thirty-two cases in mild group,thirty-three cases in moderate group and thirty-three cases in severe group,and another thirty-three cases in control group exclud OSAHS.The general data of subjects were recorded,the differences between four groups,the incidence of PPH and the influencing factors of PPH were compared. Results: There was no significant difference in the incidence of total PPH among the four groups; the incidence of breakfast PPH in mild and moderate OSAHS group was not significantly different from the control group (P>0.05); the incidence of breakfast PPH in severe OSAHS group was statistically different from that in mild OSAHS and control group (P<0.05); there was no significant difference in the incidence of breakfast PPH between moderate and severe OSAHS group (P>0.05). AHI,hypertension and diabetes were risk factors for PPH. Conclusion: The incidence of PPH increased with the severity of hypoxia.Hypertension,diabetes and hypoxia may be risk factors for PPH.

1199 Effects of Ultra-Pulse CO2 Fractional Laser on Skin Characteristics Scores and Skin Barrier Function in Patients with Photoaging

Objective: To investigate the effects of ultra-pulse CO2 fractional laser on skin characteristics scores and skin barrier function in patients with photoaging. Methods: A total of 183 patients with skin photoaging admitted to the hospital between July 2018 and July 2020 were selected as the research subjects.The patients were divided into ultra-pulse group (n=102) and Er:YAG laser group (n=81) according to the non-randomized and concurrently controlled method.The patients in the ultra-pulse group received ultra-pulse CO2 fractional laser treatment,and the Er:YAG laser group was given Er:YAG fractional laser treatment.After 6 months of treatment,the skin characteristics scores (VISIA skin image analysis),skin barrier function [transepidermal water loss (TEwL),skin surface pH value,water content of stratum corneum,skin grease content] and level of improvement [Global Aesthetic Improvement Scale (GAIS),Global Score for Photoaging (GSP)] were compared between the two groups before treatment and at 3 months after treatment.The occurrence of adverse reactions during 6 months of treatment and satisfaction self-rating score at 3 months after treatment were compared between the two groups. Results: Before treatment,there were no statistical differences in skin characteristics scores,skin barrier function and improvement level between the two groups.The differences of scores of skin spots,texture,pores,red matter,purple matter and ultraviolet spots,TEwL,skin surface pH value,water content of stratum corneum,skin grease content,GAIS score and GSP score before and after treatment were significantly higher in ultra-pulse group than those in Er:YAG laser group (all P<0.05).The total satisfaction of patients in ultra-pulse group was significantly higher compared to Er:YAG laser group (P<0.05).Within 6 months of treatment,the differences in the occurrence of adverse reactions (pain,erythema,scar hypertrophy,hyperpigmentation) were not statistically significant between the two groups (all P>0.05). Conclusion: Ultra-pulse CO2 fractional laser has a definite efficacy on skin photoaging,and it can effectively improve the skin status and repair the skin barrier,and has high clinical application value.

Objective: To observe the efficacy and effect on renal function of azitromycin combinde with Huaiqihuang granule for the treatment of primary nephrotic syndrome with mycoplasma pneumoniae infection. Methods: Fifty-six children with recurrent PNS and Mycoplasma pneumoniae infection were randomly divided into 2 groups,28 children in the control group and 28 children in the observation group.The control group was treated with azithromycin and the observation group was treated with azithromycin combined with Sophora citrifolium granules.The data of clinical efficacy,albumin,total cholesterol(TC),urea nitrogen(BUN),creatinine(Cr)levels were observed,and the changes in the indicators of the two groups of children were analyzed and compared.Then negative time of urinary protei,time of edema disappearance and drug reactions between the two groups were observed. Results: After treatment,the total effective rates of the treatment group and control group were 92.86% and 75.00%,respectively; the difference was statistically significant (P<0.05).The plasma albumin was (36.35±8.02) g/L and 31.08±3.79) g/L,total cholesterol was (3.40±1.50) mmoL/L and (4.41±1.17) mmoL/L,urea nitrogen was (2.92±1.48) mmoL/L and (3.78±1.16) mmoL/L in the observation and control groups,respectively.The creatinine was (46.34±19.30) μmoL/L/L and (63.55±24.44) μmoL/L/L respectively,with significant differences in the biochemical parameters between the 2 groups (all P<0.05).After treatment,the time for urine protein to turn negative was (11.75±7.10) d and (15.89±5.26) d respectively in the observation group and control group,and the time for swelling to disappear was (9.32±4.07) d and (13.36±4.54) d respectively,and the difference was significant in both groups (both P<0.05).The overall adverse drug reaction rates were 17.85% (5/28 cases) and 42.86% (12/28 cases) in the observation and control groups,respectively,with statistically significant differences between the 2 groups (P<0.05). Conclusion: Azitromycin combinde with Huaiqihuang granule is effective in children with PNS combined with mycoplasma pneumoniae infection,and can significantly improve the renal function and clinical symptoms of children with a good safety profile.

1209 Effect of Microecological Preparations Supplemented with Alpha-Interferon on the Immune Function and Prognosis of Children with Asthma Combined with Mycoplasma Pneumoniae Infection

Objective: To investigate the effect of microecological preparations supplemented with α-interferon on the immune function and prognosis of children with asthma complicated by Mycoplasma pneumoniae infection. Methods: A total of 124 children with asthma complicated by Mycoplasma pneumoniae infection who were admitted to our hospital from January 2020 to August 2021 were selected for study.They were randomly divided into the study group and the control group with 62 cases in each group.Both groups were given conventional treatment such as cough suppression,wheezing and sedation with azithromycin upon admission.The control group received α-interferon atomization inhalation therapy on the basis of this,and the study group was given probiotics adjuvant therapy on the basis of the control group.After 5 days of treatment,the pulmonary function indexes,peripheral blood CD25,serum interleukin-5 (IL-5),and total immunoglobulin E (IgE) concentrations were compared between the two groups before and after treatment,and the coughing and wheezing conditions were compared between the two groups. Results: After treatment,FEV1,FVC,FEV1/FVC in the two groups were higher than those before treatment (P<0.05),CD25,IL-5 and IgE levels in the two groups were lower than those before treatment (P<0.05),and the research group The difference before and after treatment was higher than that in the control group (P<0.05).TThe cough symptom score,mean length of stay and number of wheezing episodes ≥2 during follow-up were all lower in the study group than in the control group,and the cough disappeared earlier than in the control group (P<0.05). Conclusion: The microecological preparation supplemented with α-interferon in the treatment of children with asthma complicated by Mycoplasma pneumoniae infection has an ideal effect,and has significant advantages in improving the symptoms and prognosis of children,and has a good promotion prospect.

1213 Comparison of the Efficacy of GLP1 Receptor Agonist and SGLT2 Inhibitor in Respective Combination with Metformin in Treatment of Type 2 Diabetes

Objective: To compare the effecacy of glucagon-like peptide-1 (GLP1) receptor agonist (liraglutide) and sodium-glucose co-transporters 2 (SGLT2) Efficacy of inhibitors (dapagliflozin) combined with metformin in the treatment of type 2 diabetes,respectively. Methods: From January 2021 to December 2021,120 diabetic patients admitted to our hospital were randomly divided into 3 groups,with 40 cases in each group,as control group (metformin),treatment group A (metformin + liraglutide) ),treatment group B (metformin + dapagliflozin).Automatic biochemical analyzer and enzyme-linked immunosorbent assay (ELISA) were used to detect the blood glucose indexes of patients before and after treatment:fasting blood glucose (FPG),blood glucose 2 hours after meal (2hPBG),glycated hemoglobin (glycosylated hemoglobin,HbA1c),insulin resistance index (insulin resistance index,HOMA-IR) levels,blood lipid indicators triglyceride (triglyceride,TG),total cholesterol (total cholesterol,TC),high-density lipoprotein cholesterol (high density lipoprotein cholesterol,HDL-C),low density lipoprotein cholesterol (low density lipoprotein cholesterol,LDL-C) levels. Results: Compared with before treatment,the levels of FPG,2hPBG,HBA1c and HOMA-IR significantly decreased after treatment (P<0.05),and the TG,TC and LDL-C significantly decreased (P<0.05),HDL-C significantly increased in the three groups of patients after treatment (P<0.05).Compared with the control group,the levels of serum FPG,2hPBG,HBA1c,HOMA-IR and TG significantly decreased in group A and group B after treatment (P<0.05),and TC,LDL-C and HDL-C had no significant difference in group A and group B after treatment (P>0.05).Compared with group A,the levels of FPG and 2hPBG significantly decreased (P>0.05),while the levels of HBA1c,HOMA-IR,TG,TC and LDL-C decreased,with no significant difference in group B (P>0.05).The adverse reaction rates of the three groups were 27.5%,27.5% and 25%,respectively. Conclusion: The SGLT2 inhibitor combined with metformin in reducing blood sugar and lipid-lowering in type 2 diabetes was superior to GLP1 receptor agonist combined with metformin.
