1 Effects of Hyperbaric Oxygen Regulation of TLR4/NF-κB Pathway on Apoptosis in Hippocampus of Septic Rats

Objective: To explore the effect of hyperbaric oxygen (HBO) on apoptosis in hippocampus of rats with sepsis-related encephalopathy (SAE) by regulating the Toll-like receptor 4 (TLR4)/nuclear factor (NF)-κB pathway. Methods: 45 rats were randomly divided into control group, SAE group and SAE+HBO group (n=15). The SAE model was constructed by intraperitoneal injection of LPS (15 mg/kg, 3 days). Rats in SAE+HBO group received HBO intervention daily. The cranial nerve function, hippocampal neuron damage, apoptosis, and inflammatory response levels in each group were tested and compared. TLR4 and NF-κB transcription and translation levels in the hippocampus were detected. Results: The above indicators of the three groups of rats were significantly different (P<0.05). The escape latency (44.06±5.12 s), IL-1β (76.25±8.06 pg/mL), IL-6 (91.37±10.74 pg/mL), hippocampal neuron damage and apoptosis (6.17±0.78 cells/field of view), TLR4 and NF-κB mRNA and protein levels in SAE group were significantly higher than those of the control group, and the number of crossing platforms (2.94±0.35 times) was significantly lower than that of the control group (P<0.05). The escape latency (35.86±3.06 s), IL-1β (48.68±5.21 pg/mL), IL-6 (64.47±7.06 pg/mL), hippocampal neuron damage and apoptosis (3.42±0.52 cells/field of view), TLR4 and NF-κB mRNA and protein levels in the SAE+HBO group were significantly lower than those in the SAE group, and the number of crossing platforms (4.98±0.66 times) was significantly higher than in the SAE group (P<0.05). Conclusion: HBO may alleviate the damage and apoptosis of hippocampal neurons caused by SAE by inhibiting the TLR4 /NF-κB pathway, thereby protecting nerve function.

6 The Molecular Mechanism of Circ_DONSON Targeting MiR-671-5p on the Radiosensitivity of Lung Cancer Cells

Objective: To investigate the effect of circ_DONSON on the radiosensitivity of lung cancer cells and its possible mechanism. Methods: A total of 43 cases of lung cancer tissues and adjacent tissues were collected, and real-time quantitative polymerase chain reaction (RT-qPCR) was used to detect the expression of circ_DONSON and microRNA-671-5p (miR-671-5p) in the tissues. Lung cancer cells A549 were cultured in vitro and irradiated with different doses (2 Gy, 4 Gy) of X-ray, and then RT-qPCR was used to detect the expression of circ_DONSON and miR-671-5p in the cells. After A549 cells transfected with si-NC, si-circ_DONSON, miR-NC, miR-671-5p mimic, or si-circ_DONSON and miR-671-5p inhibitor were irradiated with 4 Gy X-ray, cell counting kit-8(CCK-8) method and flow cytometry were used to detect cell proliferation activity and apoptosis, respectively; the colony formation experiment was used to detect the number of colonies formed, and the western blot method was used to detect the expression of Cleaved-caspase3 protein in cells. The dual luciferase reporter gene experiment verified the regulatory relationship between circ_DONSON and miR-671-5p. Results: The expression of circ_DONSON in lung cancer tissues was 4.27±0.38, which was higher than that of adjacent tissues (0.80±0.13, P<0.05), but the expression of miR-671-5p was 0.37±0.07, which was lower than that of adjacent tissues (0.37±0.07, P<0.05). Compared with the A549 cells of control group, the expression of circ_DONSON in A549 cells irradiated with different doses (2 Gy, 4 Gy) of X-ray were increased (3.07±0.142 Gy/3.69±0.124 Gy vs 1.00±0.00, P<0.05), but the expression of miR-671-5p were decreased (0.23±0.022 Gy/0.45±0.034 Gy vs 1.00±0.00, P<0.05). Compared with 4 Gy X-ray irradiated A549 cells of control group or A549 cells transfected with si-NC, the cell proliferation activity and the number of colony formation of 4 Gy X-ray irradiated A549 cells transfected with si-circ_DONSON were decreased (0.48±0.04 vs 1.04±0.11,1.01±0.09; 41.33±1.70 vs 85.67±2.05,86.33±1.25; P<0.05), but the apoptotic rate and the expression of Cleaved-caspase3 protein were increased [(31.18±1.39)% vs (14.24±0.80)%, (14.30±0.84)%;0.90±0.06 vs 0.41±0.04, 0.40±0.05; P<0.05]. Compared with 4 Gy X-ray irradiated A549 cells of control group or A549 cells transfected with miR-NC, the cell proliferation activity and the number of colony formation of 4 Gy X-ray irradiated A549 cells transfected with miR-671-5p mimic were decreased (0.63±0.05 vs 1.01±0.11, 1.00±0.11; 52.67±2.05 vs 85.00±2.45, 84.00±1.63; P<0.05), but the apoptotic rate and the expression of Cleaved-caspase3 protein were increased [(28.22±1.47)% vs (14.42±0.88)%, (14.61±1.02)%; 0.77±0.04 vs 0.40±0.03, 0.41±0.05; P<0.05]. circ_DONSON could target miR-671-5p, and the expression of miR-671-5p in A549 cells transfected with si-circ_DONSON was significantly higher than that in cells transfected with si-NC (4.66±0.21 vs 1.00±0.02, P<0.05). Compared with 4 Gy X-ray irradiated A549 cells transfected with si-circ_DONSON, the cell proliferation activity and the number of colony formation of 4 Gy X-ray irradiated A549 cells co-transfected with si-circ_DONSON and miR-671-5p inhibitor were increased (0.91±0.06 vs 0.50±0.04; 71.00±2.83 vs 40.67±2.49; P<0.05), but the apoptotic rate and the expression of Cleaved-caspase3 protein were decreased [(20.66±0.96)% vs (31.13±1.57)%; 0.57±0.03 vs 0.90±0.07; P<0.05]. Conclusion: Silencing circ_DONSON may enhance the radiosensitivity of lung cancer cell A549 by targeting up-regulation of miR-671-5p.

13 The Effect of Ginsenoside Rg3 Regulateing Dickkopf-Related Protein 3 on the Proliferation and Apoptosis of Hepatocellular Carcinoma Cells

Objective: To explore the effect of ginsenoside Rg3 on the proliferation of liver cancer cells and its mechanism. Methods: The human liver cancer cell line Hep G2 was used as the research subject which was divided into control group (Control), ginsenoside Rg3 group (G-Rg3), ginsenoside Rg3+Dickkopf-related protein 3 irrelevant fragment group (G-Rg3+DKK3-NC), and ginsenoside Rg3+Dickkopf-related protein siRNA group (G -Rg3+DKK3-siRNA). G-Rg3 group cells were given G-Rg3 (40 mg/L) intervention for 48 h; 24h after G-Rg3+DKK3-NC group cells were transfected with DKK3-NC, G-Rg3 intervention was conducted 48 h; after G-Rg3+DKK3-siRNA group cells were transfected, G-Rg3 intervention was conducted 48 h; cells in the control group were cultured normally. MTT method was used to detect cell proliferation; flow cytometry was used to detect cell apoptosis; Western blotting was used to detect the expression of DKK3, Wnt, and β-catenin. Results: Compared with the control group, the apoptosis index of G-Rg3 group was significantly increased (P<0.01), cell proliferation was significantly decreased (P<0.01), DKK3 expression was significantly increased (P<0.01), Wnt and β-catenin expression were significantly decreased (P<0.01); compared with the G-Rg3 group, the apoptosis index in the G-Rg3+DKK3-siRNA group was significantly reduced (P<0.01), cell proliferation was significantly increased (P<0.01), DKK3 expression significantly decreased (P<0.01), and the expression of Wnt and β-catenin increased significantly (P<0.05 or P<0.01). Conclusion: G-Rg3 can inhibit the proliferation of liver cancer cells and promote apoptosis. The mechanism is related to the promotion of DKK3 expression and inhibition of Wnt/β-catenin signal transduction.

17 Protective Effect and Mechanism of Cannabinoid 1 Receptor on and Microglia Activation in Mice with Spinal Cord Injury

Objective: To investigate the protective effect and mechanism of cannabinoid 1 receptor (CB1R) on and microglia activation in mice with spinal cord injury. Methods: Thirty-six female C57BL/6 mice were randomly divided into control group, model group and drug group. The mouse spinal cord injury model was constructed by modified Allen's method. The behavioral evaluation was performed on the 1th, 7th, and 14th day after the operation, respectively. Western blotting was used to detect the protein expression of myelin basic protein (MBP), brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) in spinal cord tissue. Enzyme-linked immunosorbent assay was used to determine the serum levels of IL-6, IL-17 and TNF-α in mice. Immunofluorescence double staining was used to detect the expression of calcium ion binding regulator (IBA)-1 and ED-1 positive cells in spinal cord tissue. Results: Compared with the control group, the BMS score and the inclination of the oblique plate in the model group were significantly reduced, the serum levels of IL-6, IL-17 and TNF-α, the protein expression of MBP, BNDF and NGF and the number of IBA-1/ED-1 positive activated microglia in the model group was significantly increased (P<0.05). Compared with the model group, the BMS score, the inclination of the oblique plate and the protein expression of MBP, BNDF and NGF in the drug group were significantly increased, the serum levels of IL-6, IL-17 and TNF-αand the number of IBA-1/ED-1 positive activated microglia (P<0.05). Conclusion: CB1R reduces spinal cord injury by inhibiting microglia activation and inflammation.

21 The Effects of MiR-93-5p on Podocyte Damage in Diabetic Nephropathy by Targeting DUSP8 Gene

Objective: To observe the effects of miR-93-5p and dual-specificity phosphatase 8 (DUSP8) on high glucose-induced HPC cells, and explore its mechanism. Methods: HPC cells were treated with 25mM high glucose for 6, 12, and 24 hours. The optimal treatment time was 12h by the secretion of the inflammatory factors interleukin-6 (IL-6) and Tumor Necrosis Factor-alpha (TNF-α) in the cells, apoptosis rate and apoptosis-related protein split Cleaved caspase-3 (Cleaved caspase-3), Bcl-2 associated X protein (Bax), B-cell lymphoma-2(Bcl-2). The miR-con group (transfected miR-con), miR-93-5p group (transfected miR-93-5p mimics), anti-miR-con group (transfected anti-miR-con), anti-miR- 93-5p group (transfected anti-miR-93-5p mimics), high glucose (HG)+si-con group (transfected si-con), HG+si-DUSP8 group (transfected si-DUSP8), HG+miR-93-5p +pcDNA group (co-transfected miR-93-5p mimics and pcDNA), HG+miR-93-5p+ pcDNA-DUSP8 group (co-transfected miR-93-5p mimics and pcDNA-DUSP8) were all transfected to HPC cells with liposome methods . Fluorescence quantitative polymerase chain reaction (qRT-PCR), Western blotting (WB), Enzyme linked immunosorbent assay (ELISA), flow cytometry were used to detect the expression of miR-93-5p, DUSP8, cell secretion of IL-6, TNF-α , and protein expression of DUSP8, Cleaved caspase-3, Bax, Bcl-2, and cell apoptosis rate; Dual luciferase reporter experiment detects the fluorescence activity of cells. Results: Compared with the low glucose (NG) group, the expression of miR-93-5p was significantly decreased, the expression of DUSP8 was substantially increased, the levels of IL-6 and TNF-α were substantially increased, protein expression of Cleaved caspase-3, Bcl-2 were significantly decreased, Bax protein expression was significantly increased, the cell apoptosis rate was markedly increased in the HG (6, 12, 24 h) group. The optimal treatment time was 12 h. Overexpression miR-93-5p depressed the secretion of IL-6, TNF-α, apoptosis rate, and facilitated proteins of Cleaved caspase-3 and Bcl-2, and suppressed Bax protein in HPC cells induced by high glucose. MiR-93-5p inhibited the fluorescent activity of wild-type DUSP8 cells. Silencing DUSP8 had a protective effect similar to that of overexpression miR-93-5p, and overexpression DUSP8 reversed the effects of overexpression miR-93-5p on the inhibition of IL-6 and TNF- α secretion and apoptosis in injured podocytes. Conclusion: MiR-93-5p could inhibit the secretion and apoptosis of inflammatory factors in podocytes induced by high glucose, and its mechanism is related to miR-93-5p targeting DUSP8.

28 Effects of Cyclopamine on Cell Proliferation and Apoptosis in Rats with Benign Prostatic Hyperplasia

Objective: To observe the effect of Cyclopamine on cell proliferation and apoptosis in rats with benign prostatic hyperplasia. Methods: Thirty BPH rats were randomly divided into model group (BPH group) and Cyclopamine group (Cyclopamine group), and 15 rats without modeling served as normal control group (Normal group). Rats in Cyclopamine group were intraperitoneally injected with Cyclopamine, and rats in normal group and BPH group were intraperitoneally injected with the same dose of normal saline, once a day, for 7 consecutive days. After the administration, the rats were killed to obtain the prostate tissue of each group, and the indexes were determined. Results: Compared with normal group, the wet weight, volume and prostate index of BPH group and Cyclopamine group were significantly increased (P<0.05). The glandular epithelium showed papillary hyperplasia, and the number of glands increased, the glandular cavity enlarged, the stroma and blood vessels dilated with hyperemia and edema. The expression rate of apoptotic bodies was significantly decreased (P<0.05), and the proliferation rate was significantly increased (P<0.05). Compared with BPH group, the wet weight, volume and prostate index of Cyclopamine group were significantly lower [(3.18±0.28)g vs (2.03±0.22)g; (2.30±0.26)ml vs (1.45±0.27)ml; (0.78±0.23)% vs (0.46±0.17)%] (P<0.05). The histopathological changes of prostate were improved significantly. The expression rate of apoptotic bodies increased significantly [(3.17±0.68)% vs (5.25±0.77)%] (P<0.05). The proliferation rate of stromal and epithelial cells decreased significantly [OD 4d (1.35±0.09)/(1.62±0.17) vs (0.94±0.11)/(1.26±0.15); 5d (2.24±0.16)/(2.73±0.27) vs (1.74±0.17)/(2.16±0.20); cell number 4d (21.57±1.89)/(31.32±2.54) vs (17.88±1.26)/(25.44±2.39); 5d (33.66±3.16)/(44.97±4.29) vs (29.14±2.42)/(39.75±3.82)] (P<0.05). Conclusion: Cyclopamine can inhibit the proliferation of prostatic epithelial cells and stromal cells, and promote the occurrence of apoptosis.

33 The Possible Mechanism of Surgery Combined with 125I Brachytherapy for Patients with Oral Salivary Adenoid Cystic Carcinoma

Objective: To study the related growth factors and clinical efficacy of surgery combined 125I brachytherapy patients with nts with oral salivary adenoid cystic carcinoma(ACC). Methods: 86 patients with ACC from April 2016 to March 2018 were divided into observation group (41 cases) and control group (39 cases). The control group had surgery, and the observation group had surgery combined with 125I brachytherapy. The clinical efficacy were compared between two groups in the three-year follow-up. Before treatment and 6 months after treatment, serum related growth factors were compared between two groups. Results: In the three-year follow-up, the observation group remission rate and control rate were higher than those of the control group (82.93% vs 58.97%, 92.86% vs 74.36%)(χ2=5.598, 4.932, P<0.05). 6 months after therapy, observation group had serum VEGF, bFEG, MMP-9 lower than control group [(238.54±36.25 vs 266.46±32.23)pg/m,(215.42±34.25 vs 276.26±36.12)pg/mL,(493.24±24.36 vs 524.12±30.43)pg/mL](t=3.634, 7.732, 5.023, P<0.05). Conclusion: Surgery combined with 125I brachytherapy can improve clinical efficacy of ACC patients, and its mechanism may be related to reducing serum VEGF, BFEG and MMP-9.

37 Changes of Serum CP CRP and Cys-C Levels in Patients with Type 2 Diabetes and the Predictive Value for Macrovascular Disease

Objective: To explore the changes of serum C-peptide (CP), C-reactive protein (CRP) and Cystatin C (Cys-C) levels in patients with type 2 diabetes (T2DM) and their predictive value for macrovascular disease. Methods: A total of 85 patients with T2DM complicated with macrovascular disease in our hospital from April 2018 to March 2020 were selected as the study group, and 85 patients with pure T2DM were selected as the control group. The clinical data, serum CP, CRP, and Cys-C levels were compared between the two groups. Logistic regression was used to analyze the influencing factors of T2DM macrovascular disease. Pearson correlation analysis was used to explore the relationship between serum CP, CRP, and Cys-C levels and carotid intima-media thickness (IMT). The receiver operating characteristic (ROC) curve analysis was used to predict value of serum CP, CRP, and Cys-C levels for T2DM with macrovascular disease. Statistics of the incidence of major adverse cardiac events (MACE) in the two groups was recorded in the six-month follow-up. Results: In the study group, age ≥60 years, the proportion of diabetes ≥10 years, TG, LDL-C, carotid IMT serum CRP and Cys-C had higher levels than those of the control group, and the CP level was lower than that of the control group (P<0.05). Logistic regression model showed, age correction ≥60 years, diabetes course ≥10 years, serum TG, LDL-C levels, carotid IMT, serum CP, CRP, Cys-C levels were still related to T2DM complicated by macrovascular disease (P<0.05). Serum CP levels in patients with T2DM complicated with macrovascular disease are negatively correlated with carotid IMT, and serum CRP and Cys-C levels are positively correlated with carotid IMT (P<0.05). The maximum AUC of serum CP, CRP, and Cys-C in predicting T2DM with macrovascular disease is 0.948, the corresponding sensitivity is 90.59%, and the specificity is 96.47%. The incidence of MACE in the study group 21.18% was higher than that in the control group (9.41%) (P<0.05). Conclusion: Serum CP, CRP, and Cys-C are abnormally expressed in patients with T2DM, and each index is an influencing factor in the pathogenesis of macrovascular disease. Combined detection can provide an important reference for clinical prediction of macrovascular disease.

43 Relationship Between Expression Level of Serum Calcium-Binding Protein Complex S100A8/A9 and Unstablecarotid Artery Plaques in Elderly Patients with Hypertension

Objective: To explore the relationship between expression level of serum calcium-binding protein complex S100A8/A9 and unstablecarotid artery plaques in elderly patients with hypertension. Methods: The clinical data of 152 elderly patients with hypertension admitted to the hospital between April 2018 and April 2021 were retrospectively analyzed. The patients were grouped according to whether the carotid artery plaque was stable or not, including stable group (n=61) and unstable group (n=91). The levels of serum S100A8/A9, blood uric acid (UA), fasting plasma glucose (FPG) and blood lipids [triacylglycerol (TG), high-density lipoprotein (HDL), low-density lipoprotein (LDL)] and intima-media thickness (IMT) of common carotid artery and internal carotid artery were compared between the two groups. Pearson correlation analysis was used to explore the relationship between serum S100A8/A9 and other clinical indicators in patients with different plaque properties. Results: There was no statistical difference in TG level between the two groups (P>0.05). The levels of serum S100A8/A9, UA, FPG, HDL and LDL and IMT values of common carotid artery and internal carotid artery in unstable group were higher than those in stable group (all P<0.05). According to Pearson correlation analysis, there was no statistical significance in the correlation between serum S100A8/A9 and levels of FPG and TG in patients with different plaque properties, and there was also no statistical significance in the correlation between serum S100A8/A9 and LDL and internal carotid artery IMT of patients in stable group (all P>0.05). Serum S100A8/A9 of patients in unstable group was positively correlated with UA, HDL, LDL, common carotid artery IMT and internal carotid artery IMT, and serum S100A8/A9 in stable group was positively correlated with UA, HDL and common carotid artery IMT (all P<0.05). Conclusion: The formation of unstable carotid artery plaques in elderly patients with hypertension may be related to the levels of serum S100A8/A9, UA, FPG, HDL and LDL.

47 Relationship of Serum PA ChE and TBA Levels with Cirrhotic Portal Hypertension Complicated by Upper Gastrointestinal Bleeding

Objective: To investigate the relationship of serum prealbumin (PA), cholinesterase (ChE) and total bile acid (TBA) levels with cirrhotic portal hypertension complicated by upper gastrointestinal bleeding. Methods: A total of 273 patients with cirrhotic portal hypertension and upper gastrointestinal bleeding (cirrhosis bleeding group) were selected between January 2019 and April 2021, including 58 patients with grade A liver function, 119 patients with grade B liver function, and 96 patients with grade C liver function. Meanwhile, 82 patients with upper gastrointestinal bleeding caused by gastroduodenal ulcer and without history of liver cirrhosis were selected as the non-cirrhosis bleeding group, and 65 healthy volunteers without history of liver cirrhosis were selected as the healthy control group. Serum PA, ChE and TBA levels in the 3 groups were compared. The correlation of serum PA, ChE and TBA levels with liver function classification was analyzed. Results: Serum PA and ChE levels in the cirrhosis bleeding group were lower than those in the non-cirrhosis bleeding group and the healthy control group (P<0.05), while serum TBA levels were higher than those in the non-cirrhosis bleeding group and the healthy control group (P<0.05). Serum PA and ChE levels of patients with grade C liver function were lower than those of patients with grade B and grade A (P<0.05), while serum TBA levels were higher than those of patients with grade B and grade A (P<0.05). Serum PA and ChE levels were negatively correlated with the grade of liver function (rs=-0.553, rs=-0.439, P<0.05), while serum TBA level was positively correlated with the grade of liver function (rs=0.628, P<0.05). Conclusion: Patients with cirrhotic portal hypertension and upper gastrointestinal bleeding have low serum PA and ChE levels, and high serum TBA level. Serum PA, ChE and TBA are closely related to liver function classification of the patients.

51 Clinical Correlation Between the Expression of Dual Specificity Phosphatases Gene Family and Hepatocellular Carcinoma Development

Objective: To analyze characteristics of dual specificity phosphatases (DUSPs) gene family expression in hepatocellular carcinoma (HCC) and their correlation with severity of clinical prognosis and immune cells infiltration. Methods: Characteristics of DUSPs mRNA expression and its correlation with severity of clinical prognosis and immune cell infiltration were analyzed in public database: Oncomine、GEPIA、Kaplan-Meier plotter,TCGA,TIMER. 19 paired HCC tissues were collected from our hospital to analyze DUSPs protein expression characteristics and their correlation with the pathological grade and clinical stage. Gene enrichment analysis (GSEA) was used to predict the possible mechanism of DUSPs involved in HCC progression. Results: Public database analysis results showed that compared with adjacent tissues, DUSP12 mRNA was highly expressed in HCC tumour tissues (P<0.05), and high expression of DUSP12 mRNA was associated with decreased overall survival rate (P< 0.001), increased pathological grade (P<0.001), increased elevated immune cells infiltration (P<0.001) and elevated expression of immune checkpoint related molecules (P<0.001). Immunohistochemical staining results of paired HCC postoperative tumour and adjacent tissues collected in our hospital showed that compared with adjacent tissues, DUSP12 protein was highly expressed in HCC tumour tissues (P<0.001), and further increased with the increase of pathological grade (P<0.05) and clinical stage (P<0.05). GSEA predicted that DUSP12 was involved in HCC progression by regulating cell cycle, RNA splicing, bile acid synthesis and retinol metabolism. Conclusion: DUSP12 expression is significantly up-regulated in HCC tumour tissues, and HCC patients with high expression of DUSP12 have decreased overall survival rate, increased pathological grade, elevated immune cell infiltration and expression of immune checkpoint related molecules. DUSP12 can be used as an important biomarker to predict the pathological grade and prognosis of HCC patients.

57 Level and Significance of Serum STOX1 and Soluble Fms-Like Tyrosine Kinase-1 in Pregnant Women with Preeclampsia

Objective: To study the level and significance of Serum STOX1 and soluble Fms-like tyrosine kinase-1 (sFlt-1) in pregnant women with preeclampsia. Methods: Ninety pre-eclampsia pregnant women admitted to our hospital from February 2018 to February 2020 were selected as the research subjects. They were divided into 45 cases in mild group and 45 cases in severe group according to the condition, and 40 healthy pregnant women in the same period were selected as control group. Heart rate, blood pressure [systolic blood pressure (SBP) and diastolic blood pressure (DBP)], mean arterial pressure (MAP), ratio of umbilical artery peak systolic to peak diastolic peak (S/D), 24h proteinuria, platelet count, alanine aminotransferase (ALT), aspartate aminotransferase (AST), blood creatinine (Scr), placental growth factor (PLGF), STOX1 and sFlt-1 were compared among the three groups. Pearson correlation was used to analyze the relationship between serum STOX1 and sFlt-1 in preeclamptic women. Results: The heart rate, SBP, DBP, MAP, S/D, 24h proteinuria, ALT, AST, Scr, STOX1, sFlt-1 of the control group were significantly lower than those of the mild and severe groups. The heart rate, SBP, DBP, MAP, S/D, 24h proteinuria, ALT, AST, Scr, STOX1, sFlt-1 were significantly lower than the severe group; the platelet count and PLGF of the control group were significantly higher than those of the mild and severe groups. The platelet count and PLGF of the mild group were significantly higher than the severe group, the differences were statistically significant (P<0.05). By Pearson's correlation analysis, STOX1 was positively correlated with heart rate, SBP, DBP, MAP, S/D, and 24h proteinuria (P<0.05), negatively correlated with PLGF (P<0.05), and not correlated with platelet count, ALT, AST or Scr (P>0.05). sFlt-1 was positively correlated with heart rate, SBP, DBP, MAP, S/D, and 24h proteinuria (P<0.05), negatively correlated with PLGF (P<0.05), and not correlated with platelet count, ALT, AST, or Scr (P>0.05). Conclusion: The serum STOX1, sFlt-1 levels of preeclampsia pregnant women increase with the progression of the disease. Clinically, the changes of SBP, DBP, MAP, 24h proteinuria, and PLGF can be combined to predict and control the condition of pregnant women to prolong the gestational week Termination of pregnancy.

61 Efficacy of Double Filtration Plasmapheresis Combined with Immunosuppressant in the Treatment of Severe Lupus Nephritis and Its Influence on Levels of Peripheral Blood Alb BUN and WBC

Objective: To observe the efficacy of double filtration plasmapheresis (DFPP) combined with immunosuppressant in the treatment of severe lupus nephritis (LN) and its influence on the levels of peripheral blood urinary Albumin (Alb), blood urea nitrogen (BUN) and white blood cell (WBC). Methods: Patients with severe lupus nephritis who were treated in the hospital between January 2019 and December 2020 were retrospectively analyzed, and they were divided into observation group (n=67) and routine group (n=75) according to the treatment regimens. The routine group was treated with immunosuppressant and hormone, and the observation group was administered double filtration plasmapheresis on this basis. The clinical efficacy, liver-kidney function indicators, immune indicators and occurrence of adverse reactions were compared between the two groups after 6 months of treatment. Results: After treatment, the total remission rate of observation group was significantly higher than that of routine group (93.33% vs 79.10%) (P<0.05). After 6 months of treatment, the levels of Alb and serum total bilirubin (TBIL) of the two groups of patients were significantly enhanced compared to before treatment (P<0.05) while the levels of BUN, serum creatinine (Scr) and urine protein were significantly decreased compared to before treatment (P<0.05), and the differences between the groups were statistically significant after treatment (P<0.05). There was no statistical significance in the difference of urine protein before and after treatment between the two groups (P>0.05), and there were statistical significance in the differences of Alb, BUN, Scr and TBIL (P<0.05). After 6 months of treatment, the positive rate of anti-DsDNA antibody and levels of WBC and vascular cell adhesion molecule-1 (VCAM-1) in the two groups were significantly lower than those before treatment (P<0.05) while the level of complement C3 was significantly higher than that before treatment (P<0.05), and there were statistically significant differences between the two groups after treatment (P<0.05). There was no statistical significance in the difference of positive rate of DsDNA antibody before and after treatment between the two groups(P>0.05), but there were statistical significance in the differences of WBC level, urine VCAM-1 level and complement C3 level (P<0.05). During treatment, the incidence rate of adverse reactions was 18.67% in observation group and was 14.93% in routine group respectively (P>0.05). Conclusion: Immunosuppressant combined with DFPP has a better effect in the treatment of severe lupus nephritis. It can effectively improve the liver-kidney function, regulate the immune level and accelerate the recovery of disease, and it has safety of medication and is recommended for clinical application.

66 The Diagnostic Value of Serum Amyloid A, Retinol Binding Protein and Urine β2 Microglobulin in Early Renal Injury During Pregnancy Hypertension

Objective: To analyze the diagnostic value of serum amyloid protein A (SAA), retinol binding protein (RBP) and urine beta 2 microglobulin (β2-MG) in early renal injury during pregnancy hypertension disease (HDP). Methods: 120 HDP patients with gestational age of 20~40 weeks in our hospital from March 2019 to March 2021 were selected and divided into mild group (32 cases), moderate group (53 cases) and severe group (35 cases) according to the severity of the disease. In addition, 50 normal pregnant women hospitalized in the same period were selected as the control group. 3 ml of fasting elbow venous blood and 5 ml of midstream urine were collected in the morning. 24 h urine was collected after bladder emptying. The levels of SAA, RBP and urine were compared among the four groups. Receiver operating characteristic curve (ROC) was used to analyze the diagnostic value of each index for early renal injury of HDP. Results: Serum SAA, RBP and urine β2-MG averages increased gradually in the control group, mild group, moderate group, and severe group, and the differences between any two groups were statistically significant (P<0.05). There was a positive correlation between serum SAA, RBP, or urine β2-MG and 24h urinary protein level (r=0.751、0.878、0.668,P<0.05). The combined detection of serum SAA, RBP and urine β2-MG had an ROC curve area (AUC) of 0.918, a sensitivity of 0.895 and a specificity of 0.933 to predict HDP. Conclusion: Serum SAA, RBP and urine β2-MG levels can reflect the degree of kidney damage in HDP patients to a certain extent. Joint testing can improve the sensitivity and specificity of early kidney damage diagnosis in HDP patients, and it has great value for the diagnosis and assessment of the early kidney damage in HDP patients.

71 Effect of Continuous Bedside Hemofiltration on Serum Lipoprotein (a) Cholinesterase Activity and Cardiac Function Indexes in Patients with Acute Organophosphorus Pesticide Poisoning

Objective: To explore the effect of continuous bedside hemofiltration on serum lipoprotein (a) [Lp (a)], cholinesterase (ChE) activity and cardiac function indexes in patients with acute organophosphorus pesticide poisoning (AOPP). Methods: A total of 160 patients with AOPP admitted to the hospital from May 2016 to May 2020 were selected and randomly divided into observation group (n=86) and control group (n=74). The control group received conventional symptomatic treatment, and on this basis, the observation group received continuous bedside hemofiltration. The clinical effects, changes in serum Lp(a) level, ChE activity and cardiac function indexes at different time points were compared between the two groups. Results: The coma duration, mechanical ventilation time/hospital stay and serum ChE activity recovery time of the observation group were shorter than those of the control group. The dosage of atropine was lower than that in the control group, and the mortality rate was lower than that in the control group (P<0.05). After treatment, serum Lp(a), ChE activity and cardiac function indexes in the two groups changed over time to varying degrees. 24h, 48h and 72h after treatment, the observation group had lower serum Lp(a) levels and higher left ventricular ejection fraction (LVEF) than the control group (P<0.05). 48h and 72h after treatment, serum ChE activity in the observation group was higher than that in the control group, and the left ventricular end systolic diameter (LVIDs) and B-type natriuretic peptide (BNP) levels were lower than those in the control group (P<0.05). 24h after treatment, there were no significant differences in serum ChE activity, LVIDs and BNP levels between the two groups (P>0.05). Conclusion: Continuous bedside hemofiltration is effective in the treatment of AOPP. It can effectively improve the patients' serum Lp(a) level and ChE activity, reduce myocardial damage and promote cardiac function recovery.

76 Renal Protective Effect of Sufentanil Preconditioning on Patients After Off-Pump Coronary Artery Bypass Grafting

Objective: To study the renal protective effect of sufentanil preconditioning on patients after off-pump coronary artery bypass grafting (OPCAPG). Methods: Patients who received OPCABG in our hospital from January 2018 to December 2020 were randomly divided into low-dose group, medium-dose group and high-dose group. 0.5μg/kg, 1.0μg/kg and 1.5μg/kg sufentanil was given 5 minutes before the recanalization of anterior descending artery. The differences of surgery conditional, renal function, inflammatory cytokines and oxidative stress indexes were compared among the three groups. Results: There was no difference in operation time, tracheal extubation time, ICU stay and postoperative hospital stay among the three groups (P>0.05); 24 hours after operation, Scr, BUN and UACR levels in the low-dose group were significantly higher than those before operation (P<0.05); Scr, BUN and UACR levels in the medium dose group and high dose group were not different from those before operation (P>0.05); TNF-α, IL-1β, IL-6, MDA and 8-OHdG levels in the three groups were significantly higher than those before operation (P<0.05); SOD, GSH levels were significantly lower than those before operation(P<0.05) and TNF-α, IL-1β, IL-6, MDA and 8-OHdG levels of middle dose group and high dose group were lower than those in low dose group, while SOD and GSH levels were higher than those in low dose group (P<0.05). Conclusion: 1.0μg/kg and 1.5μg/kg sufentanil pretreatment has protective effect on renal function after OPCAPG. Inhibition of inflammatory response and oxidative stress is the related molecular mechanism.

81 Comparison of the Sensitivity and Accuracy of Ultrasound-Guided Fine Needle Aspiration Cytology for Different Thyroid Nodules

Objective: To compare the sensitivity and accuracy of ultrasound-guided fine-needle aspiration cytology with different sizes of thyroid nodules. Methods: A retrospective analysis method was used to collect the clinical data of patients with thyroid nodules found in the outpatient department of our hospital from April 2018 to April 2021. All patients underwent high-resolution ultrasound, US-FNAC and histopathological examination. According to the maximum diameter of the nodules, 136 nodules finally included in the study were divided into four groups. Group A: the maximum diameter was 4~5mm.Group B: 6~10mm.Group C: 11~20mm. Group D:>20mm. The Bethesda classification of patients with different diameters of thyroid nodules and its relationship with pathological diagnosis results were compared and the diagnostic efficacy of US-FNAC for different diameter thyroid nodules was analyzed. Results: In the Bethesda classification results of US-FNAC cytopathology, there were 1 in category I, 59 in category II, 2 in category III, 2 in category IV, 38 in category V, and 37 in category VI. The malignant rates were 0.00 %, 30.56%, 0.00 %, 100.00 %, 97.37 %, and 100.00 %, respectively, according to the pathological results after operation. The undiagnosed rate of US-FNAC in group A was the highest (4.76 %), but there was no significant difference in the undiagnosed rate between groups (P>0.05). The frequency difference of positive and negative US-FNAC results in ABCD groups was statistically significant (P20 mm nodules had the highest diagnostic sensitivity and accuracy, 100.0 % and 98.3 %, respectively). The diagnostic sensitivity and accuracy of 4~5 mm nodules were low, 92.9% and 90.1 %, respectively. Conclusion: US-FNAC has high value in diagnosing thyroid nodules of different sizes. It has high diagnostic accuracy for thyroid nodules of 11~20mm and >20mm. Patients with thyroid nodules of 4~5mm should be actively followed up. It can be combined with hematological examination or molecular marker detection in clinical diagnosis to avoid missed diagnosis.

86 The Effect of Aerobic Exercise Combined with Core Muscle Training on Cardiopulmonary Function and Muscle Strength of Lower Limbs in Patients with Spinal Cord Injury

Objective: To explore the effects of aerobic exercise combined with core muscle training on the cardiopulmonary function and muscle strength of the lower extremities in patients with spinal cord injury (SCI). Method: A retrospective analysis was conducted with the clinical data of 88 patients with incomplete spinal cord injury (ISCI) admitted to our hospital from January 2018 to December 2020. According to different rehabilitation training methods, they were divided into study group and control group. Among them, 41 patients who underwent conventional rehabilitation measures were the control group, and 47 patients who had aerobic exercise and core muscle training based on conventional rehabilitation measures were the study group. ECG monitors were used to monitor and compare the resting heart rate (RHR), systolic blood pressure (SBP), diastolic blood pressure (DBP), oxygen saturation (SaO2), and maximum oxygen uptake (VO2 Max) before and after rehabilitation training interventions. Pulmonary function tester was used to compare the vital capacity (VC), forced vital capacity (FVC), maximum voluntary ventilation (MVV) before and after rehabilitation training intervention. Forced expiratory volume in 1s (FEV1) accounted for the percentage of the predicted value. Manual muscle testing (MMT) was performed to evaluate and compare the quadriceps and hamstring muscle strength of the two groups of patients. The ASIA Lower Extremity Motor Function Assessment Scale (ASIA-LEMS) was used to evaluate and compare the lower extremity motor function of the two groups of patients. Results: Comparison of cardiac function index levels after intervention in the study group was significantly lower than the control group, SBP, DBP, SaO2, VO2 Max levels and the level difference before and after intervention was significantly higher than the control group (P<0.05); lung function index levels were compared after intervention. The levels of VC%, FVC%, MVV%, FEV1% and the level difference before and after the intervention in the study group were significantly higher than those in the control group (P<0.05); quadriceps MMT, hamstring Muscle MMT, ASIA-LEMS scores of the study group after intervention of comparison of muscle strength and motor function of the lower limbs significantly higher than those of the control group (P<0.05). Conclusion: Compared with conventional rehabilitation training, aerobic exercise combined with core muscle training can further improve the cardiopulmonary function of patients with SCI, strengthen the muscle strength of their lower extremities, and help patients recover their standing and walking abilities, and ultimately improve the quality of life of patients.

91 Analysis of the Effect of Laparoscopic Assisted Lam on Patients with Uterine Subserosal Myoma with Diameter>10 cm

Objective: To analyze the effect of laparoscopic assisted small incision hysteromyomectomy (LAM) in patients with subserosal myoma with diameter with diameter>10cm. Methods: A total of 80 patients with subserosal fibroids with diameter>10cm in our hospital from November 2017 to November 2020 were selected and divided into routine group (39 cases) and observation group (41 cases) with random number table method. The conventional group was treated with traditional open procedure, and the observation group was treated with LAM assisted laparoscopy. The capillary diameter, blood perfusion and blood flow pattern integral of the 2 groups before and 1 and 7 days after surgery were detected. The operative time, intraoperative blood loss, postoperative exhaust time and length of hospital stay of the 2 groups were observed and recorded. The surgical treatment effect and postoperative complications were statistically analyzed. Results: The capillary diameter and blood perfusion of the two groups were lower than those of the two groups at 1 and 7 days after the operation, and the blood flow status and VAS scores were higher than those of the two groups before the operation. The difference was statistically significant (P<0.05); The capillary diameter and blood perfusion of the group were higher than those of the routine group at 1 and 7 days after the operation, and the blood flow status and VAS score were lower than those of the routine group at 1 and 7 days after the operation. The difference was statistically significant (P<0.05). The observation group's postoperative exhaust time, hospitalization length, intraoperative blood loss, postoperative pain disappearance time, and fibroids volume were lower than those of the conventional group, and the operation time and total effective rate of treatment were higher than those of the conventional group. The difference was statistically significant (P<0.05) Conclusion: Laparoscopic assisted LAM has a good therapeutic effect in the treatment of patients with subserosal myoma with diameter>10cm, which effectively alleviates the pain of patients and improves the local microcirculation function and surgical related indicators of patients.

97 Clinical Retrospective Study of Laparoscopic NOSES Ⅳ Operation for Colorectal Cancer

Objective: To compare the clinical effect of the Laparoscopic radical resection of colorectal cancer with incisionless anastomosis and traditional laparoscopic surgery to explore the feasibility and safety of Laparoscopic radical resection of colorectal cancer with incisionless anastomosis. Methods: A total of 120 patients with colorectal cancer admitted from January 2014 to January 2021 were selected as the research objects and divided into test group and control group. Among them, 60 cases of colorectal cancer underwent the laparoscopic radical resection of colorectal cancer with incisionless anastomosis(test group), and 60 cases underwent traditional Laparoscopic radical resection of colorectal cancer(control group). By comparing the basic conditions before operation, operation length, intraoperative blood loss, the number of lymph node dissection, postoperative hospital stay, eating and defecation conditions and postoperative complications and other clinical indicators. Results: The sleep quality score(6.26±1.16), pain score(2.95±0.79), and length of stay(11.55±3.79d)in the test group were better than those in the control group (P<0.05). Postoperative pathological findings in both groups showed no cut margin-positive cases. There were no statistical significance on the recovery time of gastrointestinal function, intraoperative blood loss, intraoperative lymph node dissection, and postoperative complications in the two groups(P>0.05).Conclusion: Laparoscopic radical resection of colorectal cancer without incision anastomosis has the advantages of less trauma, faster recovery after surgery, and low pressure on the patient's heart. It is worthy of promotion.

102 Effect of Absolute Ethanol Interventional Embolization Under Total Cerebral Angiography and Minimally Invasive Craniotomy on Neurological Function of Children with Cerebral Arteriovenous Malformation

Objective: To explore the effects of absolute ethanol interventional embolization under total cerebral angiography and minimally invasive craniotomy on neurological function of children with cerebral arteriovenous malformation (bAVM). Methods: A prospective study was conducted on 62 children with bAVM in our hospital from December 2018 to December 2020. The patients were divided into embolization group (n=31) and craniotomy group (n=31) with the random number table. The craniotomy group was treated with minimally invasive craniotomy, and the embolization group was treated with absolute ethanol intervention embolization under total cerebral angiography. The two groups were compared on cure rate, complications, NIHSS score, serum neurological function indexes [brain-derived neurotrophic factor (BDNF), EF hand-calcineurin (S100B protein), neuron-specific enolase (NSE)], matrix metalloproteinase-9 (MMP-9), tissue matrix metalloproteinase inhibitor-1 (TIMP-1) levels before operation, 7 days after operation, and 1 month after operation. Prognosis 6 months after surgery was compared as well. Results: Re-examination of DSA 1 month after operation showed that both groups of malformations disappeared completely, and the cure rate was 100%. There was no statistically significant difference in the incidence of complications between the two groups (P>0.05). The NIHSS score of the embolization group was lower than that of the craniotomy group 7 days after the operation (P<0.05), there was no statistically significant difference in NIHSS scores between the two groups at 1 month after operation (P>0.05). The level of serum BDNF in the embolization group was higher than that in the craniotomy group at 7 days postoperatively, and the levels of serum S100B protein, NSE, MMP-9 and TIMP-1 were lower than those in the preoperative and craniotomy groups (P<0.05). There were no significant difference in the levels of serum BDNF, S100B protein, NSE, MMP-9 and TIMP-1 between the two groups at 1 month after operation (P>0.05); there was no statistically significant difference in the prognosis of the two groups (P>0.05). Conclusion: Interventional embolization with absolute ethanol under total cerebral angiography for the treatment of bAVM in children can achieve comparable results in minimally invasive craniotomy, and the security is acceptable, Interventional embolization with absolute ethanol can improve the local pathological environment early and avoid brain tissue damage caused by craniotomy, which is conducive to early recovery of nerve function.

Objective: To explore the effect of hyperlipidemia on the sensitivity of regular uric acid lowering therapy in patients with primary gout. Methods: The clinical data of 120 gout patients treated with regular uric acid lowering therapy in the Department of Rheumatology and immunology of the tirsa affiliated hospital of Hainan Medical College from April 2020 to June 2021 were analyzed retrospectively. According to the treatment sensitivity, they were divided into sensitive group (78 cases) and drug-resistant group (42 cases). Gender, age, course of disease, family history of gout, baseline value of serum uric acid and blood lipid indexes triglyceride (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C) other relevant data were collected and compared between the two groups. The incidence of hyperlipidemia in the two groups was counted, The internal relationship between hyperlipidemia and the sensitivity of regular uric acid lowering therapy was analyzed. Results: The course of disease, combined with gout stone, frequency of gout attack, history of hypertension, baseline value of serum uric acid and serum creatinine (SCR) level in the drug-resistant group were significantly higher than those in the sensitive group (P< 0.05).The incidence of serum TG (1.68 ± 0.40) mmol / L and hyperlipidemia in the drug-resistant group was 38.10% higher than that in the sensitive group (1.54 ± 0.31) mmol / L and 12.82%, and HDL-C (1.06 ± 0.27) mmol / l was lower than that in the sensitive group (1.21 ± 0.23) mmol / L, the difference was statistically significant (P< 0.05). Multivariate logistic regression analysis showed that the course of disease (OR= 1.308, 95% CI:), higher baseline value of serum uric acid (OR= 2.017, 95% CI: 1.426 ~ 2.608), gout stone (OR = 1.208, 95% CI: 1.031 ~ 1.385), history of hypertension (OR= 1.250, 95% CI: 1.071 ~ 1.429), and hyperlipidemia (OR= 1.529, 95% CI: 1.138 ~ 1.920) were independent influencing factors of poor sensitivity of regular uric acid lowering therapy in patients with gout (P< 0.05).Conclusion: There are many influencing factors of uric acid lowering drug treatment sensitivity in patients with primary gout. Hyperlipidemia is an independent risk factor with poor treatment sensitivity, which deserves more attention in clinic.

112 Clinical Observation of Visual Quality After Femtosecond Laser Assisted Cataract Surgery Combined with TECNIS Symfony Intraocular Lens Implantation

Objective: To study the visual quality after femtosecond laser-assisted cataract surgery (FLACS) combined with TECNIS Symfony intraocular lens (IOL) implantation. Methods: A total of 148 patients (148 eyes) with cataract who underwent FLACS combined with IOL implantation in the hospital between August 2018 and October 2020 were selected and divided into study group and control group by simple random grouping method with 74 patients in each group. Patients in the study group were treated with TECNIS Symfony IOL, while patients in the control group were treated with traditional spherical monofocal IOL. Changes in visual acuity, contrast sensitivity (CS), wavefront aberration, corneal refractive power and astigmatism before and after treatment were compared between the two groups, and visual quality after surgery was evaluated. Results: There was no significant difference in uncorrected distant visual acuity, corrected visual acuity, CS from different angles, total low order aberrations and satisfaction with distant vision between the two groups (P>0.05). However, the uncorrected middle and near visual acuity, satisfaction with near vision and middle vision, spectacles-independent rate while reading and VF-14 score of the study group were significantly higher than those of the control group. The overall aberrations and overall high-order aberrations were significantly lower than those of the control group (P<0.05). After surgery, overall refractive power of the two groups was significantly reduced, and the overall astigmatism was significantly increased (P<0.05), but the comparison between the two groups before and after the operation showed no statistical significance (P>0.05). Conclusion: FLACS combined with TECNIS Symfony IOL implantation can better meet the vision needs of patients with cataract and provide them with high visual quality.

117 Application Analysis of Nasal High Flow Oxygen Humidification Therapy in Patients with Chronic Obstructive Pulmonary Disease and Respiratory Failure

Objective: To analyze the application effect of nasal high flow oxygen humidification therapy (hfnc) in patients with chronic obstructive pulmonary disease (COPD) and respiratory failure. Methods: The 158 patients with COPD respiratory failure who were treated in our hospital from January 2019 to January 2021 were selected as the research subjects, and they were simply randomly divided into group A and group B, with 79 cases in each group. Group A was treated with low flow oxygen inhalation through nose, and group B was treated with HFNC. The oxygen metabolism, respiration, blood gas analysis and lung function of the two groups before and after treatment were compared, and the clinical efficacy and adverse reactions of the two groups after treatment were compared. Results: There was no significant difference in oxygen metabolism, respiration, blood gas analysis, and lung function between the two groups before treatment (P>0.05); after treatment, compared with before treatment, the levels of VO2, respiratory frequency, peak airway resistance, airway resistance, PaO2 were decreased, the levels of CaO2, DO2, SaO2, PaCO2, FEV1, FVC, PEER were increased in the two groups, and the changes in the indexes of group B were higher, and the difference was significant (P<0.05). The clinical total effective rates of group A and group B were 78.48% and 94.93% respectively, and the difference was significant (P< 0.05). The incidences of epistaxis, dry nose and dry mouth in group A were higher than those in group B (P< 0.05). Conclusion: Compared with traditional nasal low flow oxygen inhalation therapy, HFNC can effectively improve oxygen metabolism, respiration, blood gas and lung function, improve clinical efficacy and reduce adverse reactions in the treatment of COPD with respiratory failure.

122 Effect of Platelet/Lymphocyte Ratio on Cognitive Dysfunction in Patients with Cerebrovascular Disease

Objective: To analyze the effect of platelet/lymphocyte ratio (PLR) on cognitive dysfunction in patients with cerebrovascular disease. Methods: 158 patients with cerebral small vessel disease in our hospital from January 2018 to January 2021 were retrospectively analyzed, according to whether the patients had cognitive impairment, and they were divided into a disorder group (n=67) and a control group (n =91). The general data and clinical data of the two groups were compared. Logistic multivariate analysis of the risk factors of cognitive dysfunction was conducted in patients with cerebral small vessel disease. Results: The levels of IMT, FBG, TC, TG, LDL, Hcy, hs-CRP and PLR in the disorder group were higher than those in the control group. The incidence of hyperhomocysteinemia, number of LI lesions, WMLS score and lacunar cerebrovascular disease in the disorder group were higher than those in the control group. Plasma HO-1 and albumin levels in the disorder group were lower than those in the control group, and the difference was statistically significant (P< 0.05). Logistic multivariate analysis showed that primary school education or below, number of LI lesions ≥5, WMLS score ≥10, IMT≥2mm, Hcy≥15μmol/L, PLR≥150, and plasma HO-1≤0.6μg/L were risk factors for cognitive dysfunction in patients with cerebral vascular disease (P< 0.05). PLR was positively correlated with the number of LI lesions, WMLS score, IMT and Hcy (P< 0.05), and negatively correlated with education level and plasma HO-1 (P< 0.05) in cerebrovascular disease patients. Conclusion: Education level, number of Li lesions, WMLs score, IMT, Hcy, PLR and plasma HO-1 levels were risk factors of cognitive impairment in patients with cerebral small vessel disease.

127 Study on the Relationship Between Childhood Obesity and Adult Hypertension and Left Ventricular Hypertrophy

Objective: To explore the relationship between childhood obesity and adult hypertension(HP) and left ventricular hypertrophy(LVH). Methods: A total of 450 hypertensive adults and 350 non-hypertensive adults who were examined in the second Affiliated Hospital of Shenyang Medical College from 2018 to 2019 were selected as research subjects. Patients' age range between 19 and 48, whose general data, laboratory examination index and Doppler echocardiography were completed. Retrospective survey was conducted toward selected subjects with at least 6 physical examination reports of them from childhood (6-18 years old) to adulthood. BMIs of childhood age brackets and adulthood were recorded and the trajectory curve equation of childhood BMI was constructed with the mixed effect model. The area under the curve (AUC) was recorded as AUCBMI, which represents the average level of BMI for individuals throughout childhood. Results: Spearman correlation analysis showed that childhood obesity was significantly correlated with HP and LVH in adults(r=0.545,r=0.478). Non-conditional logistic analysis showed excluding age, gender and other possible confounding factors, childhood obesity could increase the risk of HP and LVH in adults (P<0.05).The risk of adult HP and LVH was the highest when childhood obesity was extended to adult obesity (P<0.05).Conclusion: Childhood obesity is significantly related to HP and LVH in adults, so the window of opportunity for prevention and treatment of cardiovascular chronic diseases related to obesity should be moved from adulthood to childhood. It is advocated to identify high-risk children with obesity early, control risk factors from childhood, achieve prevention and treatment window "move down" , in order to reduce the risk of cardiovascular disease in adults. The quantification of the regulatory effect of childhood obesity on adult blood pressure and left ventricular remodeling will promote the development of new prevention and intervention strategies.

131 Comparative Study of Dental Pulp Stem Cells Intravenous Injection and Combined Core Decompression in the Treatment of Early Steroid-Associated Femoral Head Necrosis in Rabbits

Objective: To study the therapeutic effect of intravenous application of dental pulp stem cells on early steroid-associated femoral head necrosis in rabbits, and to provide experimental data support for clinical application. Methods: Forty seven healthy adult Japanese white rabbits were randomly divided into 4 groups. There were 5 rats in group A (normal control group), 14 rats in group B (model group), 14 rats in group C (intravenous administration group) and 14 rats in group D (stem cell transplantation group). Models of femoral head necrosis were built among group A, B, and C before relative treatment was conducted to every of the three groups. After 12 weeks of modeling, the X-ray, micro CT, tissue sections and immunohistochemical indexes were collected and statistically analyzed. Results: There were significant differences between group C and group D in bone volume fraction (BV/TV), trabecular thickness (Tb.Th), trabecular number (TB.N), structural model index (SMI), bone mineral density (BMD), void lacuna rate, trabecular area ratio and VEGF (P<0.05). The indicators of stem cell transplantation group(group D) were better than those of intravenous administration group(group C). Conclusion: Both intravenous injection of dental pulp stem cells and intratunnel injection of dental pulp stem cells after core decompression are effective in the treatment of steroid induced femoral head necrosis in rabbits, and the effect of stem cell transplantation is better than that of intravenous injection.

Objective: To explore the clinical efficacy of endoscopic retrograde cholangiopancreatography (ERCP) + lithotomy in removing extrahepatic bile duct stones, and then laparoscopic cholecystectomy for bile duct stones. Methods: A total of 98 patients with extrahepatic bile duct stones removed by traditional surgery and endoscopic retrograde cholangiopancreatography (ERCP) + choledocholithotomy in the first people's Hospital of Yunnan Province from January 2019 to January 2020 were collected and divided into two groups, 49 cases for each operation. The experimental group was treated by endoscopic retrograde cholangiopancreatography (ERCP) + choledocholithotomy. The control group underwent the traditional surgery. The clinical efficacy, operation, postoperative pain and complications of the two groups were compared. Results: The complete stone clearance rate of the experimental group was higher than that of the control group (χ2=9.910, P<0.05), and the proportion of analgesics used in the experimental group (χ2=7.127, P<0.05), the postoperative ventilation stay of the experimental group were significantly less than those of the control group (t=14.239, P<0.05). There difference of VAS score between the two groups before operation was not significant, but the average VAS score of the two groups decreased significantly after operation (P<0.05), and the VAS score of the experimental group (t=6.341, 10.759, P<0.05) was lower than that of the control group(χ2=6.078, P<0.05). Conclusion: Non-emergency operation patients take ERCP + lithotomy, and then laparoscopic cholecystectomy. This method has significant curative effect, good prognosis and less adverse reactions.

141 Correlation Between Serum IL-15, AAG Levels and Prognosis of Patients with Pulmonary Tuberculosis After Chemotherapy

Objective: To observe the correlation between serum interleukin-15 (IL-15), α1-acid glycoprotein (AAG) and prognosis of patients with pulmonary tuberculosis after chemotherapy, to guide the clinical rational evaluation of prognosis and intervention of patients with ulmonary tuberculosis after chemotherapy. Methods: A total of 221 patients with pulmonary tuberculosis admitted to our hospital from February 2017 to February 2019 were selected as the research subjects, and the baseline data of patients were collected by self-made questionnaire of our hospital. All patients received chemotherapy, and after 2 months of treatment, the levels of cytokines [interferon-γ (IFN-γ), tumor necrosis factor-α (TNF-α), macrophage migration inhibitory factor (MIF) and IL-15] and AAG were detected. The status of sputum smear having turned negative was recorded after 8 months of treatment. The prognosis of the patients was evaluated by chest X-ray examination. The patients were divided into poor prognosis group (n=69) and good prognosis group (n=152). The baseline data, cytokine levels and AAG levels of the two groups were compared, and the relationship between serum AGG level and each indicator and prognosis of patients was analyzed. Results: After 8 months of treatment, 152 of 221 patients with pulmonary tuberculosis had a good prognosis and 69 had a poor prognosis. After comparing the baseline data, cytokine levels and AAG levels between the good prognosis group and the poor prognosis group, the indicators with statistically significant differences were taken as independent variables, and the prognosis of patients was a dependent variable. Multiple regression model was established, and the results showed that the over expression of IFN-γ, TNF-α, MIF, IL-15 and down-regulation of AAG were all risk factors of poor prognosis in patients with pulmonary tuberculosis treated with chemotherapy for 2 months (OR>1, P<0.05). The receiver operating curve (ROC) was plotted and revealed that IFN-γ, TNF-α, MIF, IL-15 and AAG levels predicted the area under the curve (AUC) of the risk of poor prognosis were >0.71, all had certain predictive value. The highest AUC was predicted by AAG. Conclusion: The risk of poor prognosis in patients with pulmonary tuberculosis chemotherapy is high, which is related to the abnormal expression of IFN-γ, TNF-α, MIF, IL-15 and AAG after 2 months of treatment, of which the expression level of IL-15 and AAG has higher predictive value on the prognosis of patients with pulmonary tuberculosis after chemotherapy. The test results during treatment can guide the prognosis risk prediction and reasonable adjustment of treatment plan.

145 Effect of P Wave Terminal Potential in Lead V1 of Electrocardiogram on Stroke in Patients with Coronary Heart Disease

Objective: To analyze the effect of P-wave terminal potential (PTFV1) on stroke in patients with coronary heart disease (CHD). Methods: Using retrospective analysis, the clinical data of 122 patients with coronary heart disease admitted to our hospital from January 2018 to January 2021 were analyzed. All patients received 12-lead electrocardiogram and cardiac magnetic resonance examination. According to the level of PTFV1, 122 patients were divided into groups, including group A (>-0.03mm·s, n=42), group B (P wave upright, calculated as 0, n=40), and group C (≤-0.03mm·s, n=40). The general information, LA and LV structure and function parameters, pulmonary aortic trunk diameter and right atrium/ventricular structure, serum-related index levels, coronary heart disease and stroke events following 3-month follow-up were compared with the general data of each group of patients, and Logistic Factor analysis related factors of stroke in patients with coronary heart disease. Results: Compared with group C, the LA Vmin index level of group A and group B decreased, and the LAEF level increased, the difference was statistically significant (P<0.05). Compared with group C, the four-chamber short diameter of the right atrium apex, the inner diameter of the right ventricular short-axis outflow tract, and the four-chamber long diameter of the right ventricular apex in group A and group B decreased with statistical significance (P<0.05). Compared with group C, serum IMD and Salusin-α levels of group A and group B increased, and serum CysC, CST, and NT-proBNP levels decreased. The difference was statistically significant (P<0.05). After 8 months of follow-up, the incidence of coronary heart disease events in group C was 10%, and the incidence of stroke events was 5%. Compared with group C, the incidence of coronary heart disease events and stroke events in groups A and B were lower. PTFV1, the four-chamber short diameter of the right atrium apex, the inner diameter of the right ventricular short-axis outflow tract, and the four-chamber long diameter of the right ventricular apex were risk factors for stroke in patients with coronary heart disease (P<0.05). Conclusion: PTFV1≤-0.03mm·s is the main risk factor for stroke in patients with coronary heart disease, and it can help predict the occurrence of stroke in patients with coronary heart disease.

151 Clinical Study of Immune Reconstruction After Allogenic Hematopoietic Stem Cell Transplantation in Children with Aplastic Anemia

Objective: To study the immunological reconstruction of allogeneic hematopoietic stem cell transplantation (allo-HSCT) in children with aplastic anemia (AA) and the relationship with the acute graft-versus-host disease (aGVHD). Methods: From December 2017 to December 2019, 34 patients with AA underwent allo HSCT in the blood stem cell transplantation center of children's Hospital of the First Affiliated Hospital of Zhengzhou University. The lymphocyte subsets of 34 AA children in 1st, 3rd, 6th and 12th month after transplantation were analyzed retrospectively, and 15 healthy children were taken as reference. At the same time, the proportion of lymphocyte subsets between the group without aGVHD and the group with aGVHD was compared. Results: The proportion of CD3+ cells and CD8+ cells in the 1st, 3rd, 6th and 12th month after hematopoietic stem cell transplantation were higher than those in the healthy control group (P<0.05), showing a gradual downward trend. The percentage of CD4+ cells and the ratio of CD4+/CD8+ in the 1st, 3rd, 6th and 12th month after transplantation were lower than those in the healthy control group (P<0.05). The proportion of B cells in the 1st, 3rd, 6th month after transplantation was lower than that in the healthy control group, the difference was statistically significant (P<0.05), showing a gradual upward trend, and there was no significant difference between the two groups at the 12th month (P>0.05). The percentage of NK cells in the 1st month after transplantation was significantly higher than that in the healthy control group (P<0.05), but there was no significant difference between the two groups (P>0.05). The proportion of CD3+ cells and CD8+ cells in the non aGVHD group was lower than that in the aGVHD group in the 1st, 3rd months, and the proportion of NK cells was higher (P<0.05). The ratio of CD4+ cells, CD4+/CD8+ and B cells between the two groups had no significant difference in the 1st, 3rd month (P>0.05). Conclusion: CD3+ cells, CD8+ cells and NK cells are the main immune reconstitution after hematopoietic stem cell transplantation in children with aplastic anemia. AGVHD is easy to occur in children with higher proportion of CD8+ cells and low proportion of NK cells.

155 Effect of Delay Time and Scanning Time on the Quality of Direct Lower Extremity Venography by 320-Row Spiral CT

Objective: To evaluate the image quality of direct lower extremity venography by 320-row spiral CT under different CT delay time and scanning time, optimize CT machine scanning plan and obtain the best imaging parameters. Methods: 60 patients with suspected inferior vena cava or lower extremity venous diseases were performed direct CT venography of lower extremity with Toshiba 320-row spiral CT. Under the same liquid volume (200ml), contrast medium concentration (1∶7~1∶9) and injection rate of 3.0ml/s, the delay time and scanning time of group A were 50s and 10~20s respectively. In group B, the delay time was 30s and the scanning time was 20~30s. In group C, the delay time was 20 s and the scanning time was 30 ~ 40 s. All the images were sent to the post-processing workstation for VR and MIP reconstruction. The image quality of lower extremity venography was evaluated according to four grades and analyzed statistically. Results: The image quality of each experimental group was not the same. There was no significant difference between group B and group C, and the image quality was good or excellent. There was significant difference between group A and groups B and C, and the image quality was mainly good and fair. Conclusion: Under the condition of the same liquid volume (200ml), contrast medium concentration (1∶7 ~ 1∶9) and injection rate of 3.0ml/s, 320-Row spiral CT direct lower extremity venography can be used to obtain images satisfying clinical diagnostic needs by using delay time 50s and scanning time 20~30s.

159 Application of Oxycodone Hydrochloride in Sedation and Analgesia After Laparoscopic Myomectomy and Its Influence on Pain Mediators

Objective: To explore the application effect of oxycodone hydrochloride(OH) in sedation and analgesia after laparoscopic myomectomy, and analyze its influence on pain mediators. Methods: 134 patients who underwent laparoscopic myomectomy in the hospital were selected as the research subjects between May 2019 and May 2021, and were randomly divided into OH1 group (n=45), OH2 Group (n=46) and control group (n=43). All patients were given patient-controlled intravenous analgesia (PCIA) after operation. The OH1 group received PCIA with 20mg of OH and 8mg of ondansetron, the OH2 group received PCIA with 30mg of OH and 8mg of ondansetron, and the control group received PCIA with 50mg of morphine and 8mg of ondansetron. Pain degree, sedative degree, and the levels of pain mediators were compared between the groups. The rescue analgesia rate within 48h after operation, the pressing times of PCA and the occurrence of adverse reactions were recorded. Results: The OH1 group and OH2 group had significantly lower VAS scores and Ramsay scores than the control group at 12h and 24h after operation (P<0.05). The levels of PGE2 and NGF in the OH1 group and the OH2 group at 6h, 12h, and 24h after operation were lower than those in the control group (P<0.05). The rescue analgesia rates in the OH1 group and the OH2 group within 48h after operation were lower than those in the control group, and the pressing times of PCA was less than that in the control group (P<0.05). The incidences of nausea and vomiting, dizziness and drowsiness in the OH2 group during analgesia were higher than those in the OH1 group and the control group (P<0.05). Conclusion: Compared with morphine, OH can achieve better sedative and analgesic effects after laparoscopic myomectomy. In addition, low-dose OH has a lower incidence of adverse reactions and is safer.

Objective: To explore the effects of different administration methods of oxycodone on the level of pain-causing substances and bladder spasm pain in patients after transurethral resection of prostate (TURP). Methods: Researchers retrospectively analyzed the clinical data of 116 TURP patients admitted to our hospital from March 2020 to March 2021, and divided them into groups A, B, and C according to their different administration methods. 45 patients in group A adopting patient self-control intravenous analgesia (patient controlled intravenous analgesia, PCIA) were continuously pumped. 37 patients in group B received a single PCIA administration, and 34 patients in group C did not take PCIA analgesia. The levels of postoperative pain-causing substances (plasma substance P, 5-hydroxytryptamine (5-HT)), the number of bladder cramps at 48 hours, the visual analogue scale (VAS), and the occurrence of adverse reactions were compared among different groups of patients. Results: VAS scores, 48h bladder spasms and total duration of patients in groups A and B were significantly lower than those of group C. There were statistically significant differences in VAS scores, 48h bladder spasms and total duration between the three groups, P<0.05; The levels of substance P and 5-HT in groups A and B were significantly lower than those in group C. The differences in the levels of substance P and 5-HT in the three groups were statistically significant, P<0.05; Group A (35.56%), B (32.43) %) and C (20.59%) had no statistically significant difference in the total incidence of adverse reactions between patients, P>0.05. Conclusion: Both continuous pump injection and single administration of oxycodone PCIA can achieve analgesia and relieve bladder spasm after TURP, and there is no serious adverse reaction, which is worthy of clinical reference.

168 Effect of Coenzyme Q10 Combined with Sodium Fructose Diphosphate or Sodium Creatine Phosphate on Myocardial Enzyme Spectrum and Immune Function in Children with Viral Myocarditis

Objective: To explore the effect of coenzyme Q10 combined with fructose sodium diphosphate or sodium phosphate in the treatment of myocardial spectrum and immune function of viral myocarditis. Methods: A total of 270 children with viral myocarditis admitted from November 2019 to November 2020 were selected as the subjects and divided into control group (142 cases, administered coenzyme Q10 fructose sodium diphosphate and sodium fructose diphosphate) and study group (128 cases, administered coenzyme Q10 fructose sodium diphosphate and sodium creatine phosphate). The therapeutic efficacy, myocardial enzyme spectrum (serum CK-MB、CPK、cTnI、HBDH、LDH) and immune function (CD3+、CD4+、CD4+/CD8+) were compared between the two groups. Results: The total effective rate of 1, 2 and 3 weeks treatment in the study group was higher than that in the control group, and the difference was statistically significant (Wald χ2 group =7.412, Wald χ2 time point =290.769, P group =0.006, P time point<0.001). Before treatment, there was no significant difference in the values of CK-MB, CPK, cTnI, HBDH, LDH, LVEF, FS, CI, CD3+, CD4+, CD4+/CD8+ between the two groups (P>0.05); After treatment, the levels of CK-MB, CPK, cTnI, HBDH and LDH in the study group were lower than those in the control group (P<0.05), LVEF, FS, CI, CD3+, CD4+, CD4+/CD8+ were higher than those in control group (P<0.05); The difference of CK-MB, CPK, cTnI, HBDH, LDH, LVEF, FS, CI, CD3+, CD4+, CD4+/CD8+ between the two groups before and after treatment was statistically significant (P<0.05).Conclusion: The effect of coenzyme Q10 combined with sodium phosphate in the treatment of viral myocarditis in children is better than that of coenzyme Q10 combined with sodium fructose diphosphate, which can effectively improve the myocardial enzyme spectrum and improve the immunity of children.
