353 Comparison and Evaluation of Guinea Pig Models of Cough Variant Asthma

Objective: To compare the differences in terms of airway responsiveness,airway inflammation and pathology among three cough variant asthma (CVA) guinea pig models,and to explore appropriate evaluation criteria to evaluate the advantages and disadvantages of each model. Methods: Three-week-old male Hartley guinea pigs were randomly divided into normal control group,model A group,model B group and model C group. Based on the classical asthmatic guinea pig modeling method,model A was given a reduced dose of ovalbumin during two sensitizations,model C was given cyclophosphamide prior to sensitization to reduce the sensitization state,while model B was given a combination of intra-peritoneal and intramuscular injections,the concentration of ovalbumin was doubled and sensitized once. After 2 weeks,three groups of guinea pigs were nebulized with 1% OVA. The number of capsaicin-induced coughs,changes in airway resistance,the percentage of eosinophils in alveolar lavage fluid and the pathological changes of lung tissue were observed in each group after nebulization. Results: Compared with the normal control group,the cough frequency of guinea pigs in model A,B and C groups was significantly increased,and the difference was statistically significant (P<0.05),but the cough frequency between the three model groups was not statistically significant (P>0.05). During the experimental observation,2 guinea pigs in both model A and model B had obvious wheezing cough respectively. When airway hyperresponsiveness in the guinea pigs was set at the dose of MeCh 0.04 mg/mL,the airway resistance was higher in group A than in group B than in group C (P<0.05). Compared with the normal control group,the percentage of eosinophils in the bronchoalveolar lavage fluid (BALF) of guinea pigs in the three model groups was significantly increased (P<0.05),with a significant increase in model B,but there was no significant difference between the three model groups (P=0.051). In each model group,the local mucosal epithelial injury in the bronchial wall of guinea pigs in each model group was shed off,congestion,edema and infiltration of a large number of inflammatory cells were observed in all layers of the bronchial wall. According to the pathological grading score of guinea pig lungs,the lung histopathological score of guinea pigs in the three model groups was significantly increased (P<0.05),but there was no statistically significant difference between the groups (P>0.05). Conclusion: The cough symptoms and airway hyperresponsiveness of the three models are roughly consistent with the characteristics of clinical symptoms of CVA. Amongst them,there were four guinea pigs in model A and model B with obvious wheezing. Model C's behavior and airway hyperresponsiveness were more stable,the proportion of eosinophils in BALF,and the pathological results were close to the clinical characteristics of CVA patients. Therefore,we believe that the established model C modeling method is feasible and more optimized.

358 Construction and Functional Identification of Recombinant Adeno-Associated Virus Type 9 Containing Human Carabin Gene

Objective: To construct recombinant adeno-associated virus type 9 containing human carabin gene and identify its function. Methods: PUC57-carabin and pAAV9-IRES-ZsGreen plasmids was digested by EcoRI + XhoI,then the enzymatic digestion product was ligated by T4DNA Ligase. The pAAV9-carabin-IRES-ZsGreen plasmid was screened in JM109 cells. The pAAV9-carabin-IRES-ZsGreen overexpression vector plasmid was transfected into 293AAV cells using high-efficiency transfection HET kit reagent to package rAAV9-carabin-IRES-ZsGreen. The viral titer was detected by Q-PCR. AC16 cells were infected with rAAV9-carabin-IRES-ZsGreen p,then the carabin protein expression was detected by western blot. Results: The pAAV9-carabin-IRES-ZsGreen overexpression plasmid was successfully constructed. RAAV9-carabin-IRES- ZsGreen was successfully packaged with a titer of 5.09E+12 vg/ml. The expression of carabin protein was significantly increased after AC16 cells were infected. Conclusion: RAAV9-carabin-IRES-ZsGreen has been successfully constructed and can express carabin efficiently in cardiomyocytes.

362 Effects of Imperatorin on Proliferation,Apoptosis and RhoA/ROCK1 Signaling Pathway in Human Prostate Cancer PC3 Cells

Objective: To investigate the effect of imperatorin on proliferation,apoptosis and Ras homolog gene family member A(RhoA)/Rho-associated coiled coil-containing protein kinase 1(ROCK1) signaling pathway in human prostate cancer PC3 cells. Methods: PC3 cell group,5-fluorouracil group (100μg/mL),low-dose and high-dose imperatorin group (60μg/mL,120μg/mL) were set up. Six parallel samples were set in each well of the above groups for 72 h. Cell proliferation was measured by MTT assay,and cell invasion and migration was measured by Transwell chamber system,apoptosis measured by flow cytometry,RhoA and ROCK1 mRNA and protein levels measured by qRT-PCR and Western blot. Results: Compared with PC3 cell group,the OD value,survival rate,penetration number,RhoA,ROCK1 mRNA and protein expression levels in 5-fluorouracil group,low-dose and high-dose imperatorin group were decreased,the apoptosis rate were increased(P<0.05). Compared with 5-fluorouracil group,the OD value,survival rate,penetration number,RhoA,ROCK1 mRNA and protein expression levels in low-dose and high-dose imperatorin group were increased,the apoptosis rate were decreased(P<0.05). Compared with low-dose imperatorin group,the OD value,survival rate,penetration number,RhoA,ROCK1 mRNA and protein expression levels in high-dose imperatorin group were decreased,the apoptosis rate were increased(P<0.05). Conclusion: Imperatorin can significantly inhibit the proliferation,invasion and migration,promote its apoptosis of human prostate cancer PC3 cells. The mechanism may be related to the inhibition of RhoA,Rock 1 mRNA and protein expression by imperatorin and then inhibiting the activation of RhoA/ROCK1 signaling pathway.

368 Cyasterone Inhibits IL-1β Induced Chondrocyte Apoptosis by Up-regulating MiR-204 Expression

Objective: To investigate the effects of Cyasterone on the expression of microRNA (miR)-204 and the apoptosis of chondrocytes induced by interleukin (IL)-1β. Methods: Human normal chondrocytes C28/I2 were cultured in vitro,and set up ①control group (no treatment),IL-1β group (50 mg/L IL-1β),low-dose Cyasterone group (2.5 mg/L Cyasterone + 50 mg/L IL-1β),medium dose Cyasterone group (5 mg/L Cyasterone + 50 mg/L IL-1β) and high dose Cyasterone group (10 mg/L Cyasterone + 50 mg/L IL-1β); ②normal control group (no transfection), negative control group (transfect NC-siRNA) and miR-204 siRNA group (transfect miR-204 siRNA),three groups were adminstered 50 mg/L IL-1β and 10mg/L Cyasterone. The proliferation of C28/I2 cells was detected by CCK-8 method; the apoptosis of C28/I2 cells was detected by flow cytometry; the levels of IL-6,IL-8 and tumor necrosis factor-α (TNF-α) in the supeRNAtant of C28/I2 cells were detected by enzyme-linked immunosorbent assay (ELISA); the expression of miR-204 in C28/I2 cells was detected by real-time fluorescent quantitative PCR (qRT-PCR); in addition,the protein expressions of Cytochrome C,Bcl-2-related X protein (Bax) and Caspase-3 in C28/I2 cells were detected by Western blot (WB). Results: Compared with those in control group,the cell viability and miR-204 expression level of C28/I2 cells in IL-1β group were significantly lower (P<0.05),and the apoptosis rate,the expression levels of IL-6,IL-8 and TNF-α,the protein expression levels of Cytochrome C,Bax and Caspase-3 were significantly higher (P<0.05); moreover,compared with those in the IL-1β group,with the addition of Cyasterone and the increase of its dosage,the cell viability and the expression level of miR-204 in C28/I2 cells increased gradually (P<0.05),and the apoptosis rate,the expression levels of IL-6,IL-8 and TNF-α,the protein expression levels of Cytochrome C,Bax and Caspase-3 decreased gradually (P<0.05). Compared with negative control group,the apoptosis rate of miR-204 siRNA group were significantly increased (P<0.05). Conclusion: Cyasterone may inhibit the IL-1β-induced apoptosis of chondrocytes by up-regulating the expression of miR-204.

374 Effects of Compound Qishen Extract on Wound Healing and HIF-1α/VEGF/VEGFR2 Pathway in Diabetic Foot Ulcer Model Rats

Objective: To investigate the effect of compound Qishen extract on wound healing and hypoxia inducible factor-1α (HIF-1α)/vascular endothelial growth factor (VEGF)/vascular endothelial growth factor receptor 2 (VEGFR2) pathway in diabetic foot ulcer model rats. Methods: To establish diabetic foot ulcer rat model,forty-eight rats were successfully established and randomly divided into model group,metformin group (0.2g/kg),compound Qishen extract low dose group (4g/kg),compound Qishen extract high dose group (8g/kg),12 rats in each group; 12 normal rats were used to establish ulcer wound as control group. On the first day after successful modeling,metformin group,compound Qishen extract low dose group and compound Qishen extract high dose group were given 0.2g/kg metformin,4g/kg and 8g/kg compound Qishen extract respectively,and the control group and model group were intragastrically given the same volume of normal saline,once a day for 4 weeks. At the end of the administration period,the healing rate of ulcer wound was measured,the pathological changes of ulcer wound tissue in diabetic foot rats and normal rats were observed,and the levels of HIF-1α,VEGF,VEGFR2 mRNA and protein in ulcer wound tissues of diabetic foot rats and normal rats were measured. Results: In the control group,the wound tissue structure was normal,with a large number of fibroblasts and capillaries; in the model group,a large number of inflammatory cells infiltrated,and the number of fibroblasts and capillaries decreased significantly; after the intervention of metformin and compound Qishen extract,the wound tissue fibroblasts and capillaries increased,and the inflammatory cell infiltration decreased. Compared with the control group,the wound healing rate,ulcer wound tissue HIF-1α,VEGF,VEGFR2 mRNA and protein levels in the model group were decreased (P<0.05); compared with the model group,the wound healing rate,ulcer wound tissue HIF-1α,VEGF,VEGFR2 mRNA and protein levels in the metformin group and the low and high dose groups of compound Qishen extract were increased (P<0.05); the wound healing rate,ulcer wound tissue HIF-1α,VEGF,VEGFR2 mRNA and protein levels in compound Qishen extract high dose group were higher than those in compound Qishen extract low dose group (P<0.05); compared with metformin group,the above indexes of rats in the compound Qishen extract low dose group were lower (P<0.05); but there was no significant difference between the two groups (P>0.05); there was no significant difference between the compound Qishen extract high dose group and metformin group (P>0.05). Conclusion: Compound Qishen extract can promote wound healing of diabetic foot ulcer model rats,and its mechanism may be related to the promotion of HIF-1 α,VEGF,VEGFR2 mRNA and protein levels by Compound Qishen extract,and then activate HIF-1α/VEGF/VEGFR2 pathway.

379 Effects of Ovariectomy on Lipid Metabolism in the Aorta of Female Hyperlipemia Rats

Objective: To investigate the effect of ovariectomy on aortic lipid metabolism in female hyperlipidemia rats. Methods: Healthy female SD rats were randomly divided into blank group,model group 1 ; model group 2,hyperlipidemia rat model was established in model group 1,and ovariectomized+ hyperlipidemia rat model was established in model group 2. After 8 weeks,the mRNA levels of CAV1,LDLR,VLDLR,ABCA1,ApoA1 and SRB1 were detected by real-time quantitative PCR,serum total cholesterol,triglyceride,low density lipoprotein cholesterol and high density lipoprotein cholesterol of rats were determined by Elisa. Results: Compared to the blank group,the TC、TG、LDL-C contents of model 1 and model 2 increased significantly (P<0.01),the HDL-C content decreased significantly (P<0.01),the TC,TG,LDL-C content of model 2 group was significantly higher than that of model 1 group (P<0.01),The HDL-C content was significantly less than that of model 1 group (P<0.01).Compared with the blank group,the mRNA relative expression of ABCA1,ApoA1,CAV1,SRB1,VLDLR,LDLR in model group 1 and model group 2 decreased significantly (P<0.01),and the expression in model group 2 was lower than that in model group 1(P<0.01). Conclusion: Ovariectomy leads to the decrease of estrogen and affects aortic lipid metabolism in female rats by ABCA1,ApoA1,CAV1,SRB1,VLDLR,LDLR mRNA,resulting in abnormal serum lipid content in rats.

384 Transfection of STAT3 Overexpression Plasmid Mediated by Recombinant Lentivirus Promotes Differentiation of Bone Marrow Mesenchymal Stem Cells into Neural Cells in Fetal Rats with Spina Bifida Aperta

Objective: To observe the action of recombinant lentivirus mediated STAT3 overexpression plasmid transfected into bone marrow mesenchymal stem cells on neural cell trans-differentiation in spinal cord tissue of fetal rats with spina bifida aperta. Methods: The fetal rat model of spina bifida aperta was established by retinoic acid and treated by microsurgical injection of stem cells. They were divided into blank control group (12 cases),negative control group (10 cases) and experimental group (11 cases). OD value of BMSCs viability was determined by CCK-8 kit. The expression of STAT3,pSTAT3,nerve markers (GFAP,NSE,NF and nestin) and apoptosis related factors (Caspase-8 and Bcl-2) were tested by the method of real-time PCR and Western blot. Results: OD value in experimental group was the highest at 8 hours after transplantation by CCK-8 kit,and then gradually decreased,which was statistically significant compared with blank control group and negative control group (P<0.05). There was no statistical difference in OD peak time and value between blank control group and negative control group. The expression levels of pSTAT3,GFAP,NSE,NF and nestin in experimental group were remarkably higher than those in blank control group and negative control group (P<0.05),but STAT3 expression in experimental group were statistically decreased (P<0.05). The relative expression levels of Caspase-8 and Bcl-2 in experimental group were tremendously lower than those in blank control group and negative control group (P<0.05). Conclusion: Transfection of recombinant lentivirus mediated STAT3 overexpression plasmid with BMSCs can improve the efficiency of transforming into neural cells and provide new seed cells for the treatment of congenital spina bifida aperta.

390 Correlation Study of Oxygenation Index Platelet Count and Plasma PCT Level with the Severity of Sepsis

Objective: To explore the correlation between oxygenation index,platelet count (PLT) and plasma procalcitonin (PCT) level and the severity of sepsis. Methods: A retrospective analysis was performed among 126 sepsis patients admitted to the hospital between January 2018 and October 2020,and the subjects were divided into sepsis group (n=38),severe sepsis group (n= 47) and septic shock group (n=41) according to the severity. General clinical data and laboratory indicators of the 3 groups were comparatively analyzed,and the correlations among oxygenation index,PLT,PCT,the acute physiology and chronic health evaluation II (APACHEⅡ) score and the sequential organ failure assessment (SOFA) score were analyzed. Meanwhile,ROC curve was used to analyze the value of oxygenation index,PLT and PCT in evaluating death in patients with sepsis within 28 days after admission. Results: The APACHE Ⅱ score,SOFA score and the mortality rate within 28 days after admission were significantly higher in the septic shock group than in the severe sepsis group and the sepsis group (P<0.05). There were significant differences in red blood cell distribution width (RDW),white blood cell count (WBC),PLT,C-reactive protein (CRP),serum creatinine (Scr),PCT,blood lactate and oxygenation index among the 3 groups (P<0.05). Besides,the septic shock group had the highest RDW,WBC,CRP,Scr,PCT and blood lactate levels,and the lowest PLT and oxygenation index (P<0.05). The oxygenation index,and PLT was significantly negatively correlated with APACHEⅡ score and SOFA score (P<0.05),while PCT was significantly positively correlated with APACHEⅡ score and SOFA score (P<0.05). Oxygenation index and PLT in patients with sepsis were positively correlated with PLT,but negatively with PCT Correlation (P<0.05); PLT and PCT were negatively correlated (P<0.05). ROC curve analysis showed that oxygenation index,PLT and PCT could predict 28 days mortality of patients with sepsis,and the area under curve (AUC) was 0.728,0.799 and 0.762,respectively. Conclusion: Oxygenation index,PLT and PCT are related to the condition of patents with sepsis,indicating prognostic value. Besides,the expression levels of the three indexes are correlated in patients with sepsis. They can be used as auxiliary clinical indicators for diagnosis and evaluation of sepsis.

395 Preparation of Puerarin Combined with Tricalcium Phosphate Scaffold and Its Repairing Effect on Steroid-Induced Avascular Necrosis of the Femoral Head in Rats

Objective: To prepare puerarin/tricalcium phosphate scaffold material,observe its cell compatibility,and test its effect on bone repair in rats with steroid-induced avascular necrosis of the femoral head. Methods: Using puerarin and tricalcium phosphate as materials,the puerarin/tricalcium phosphate scaffold material was prepared according to the method of negative pressure adsorption. The surface morphology of the scaffold material was observed by scanning electron microscope,and the compressive strength and porosity were detected,as well as the effects of different concentrations of puerarin/tricalcium phosphate scaffold material on the activity and growth of bone cells MLO-Y4. 60 SD rats were randomly divided into blank control group,model group and composite scaffold group,20 rats in each group. In the model group and the composite stent group,2 mg/kg LPS was injected into the tail vein once a day for 2 days. After the last injection of LPS,20 mg/kg methylprednisolone was injected intraperitoneally,once a day for 3 days,to build a femoral head necrosis model. After modeling,rats in the composite scaffold group were implanted with puerarin/tricalcium phosphate scaffold material; after 12 weeks,the blood of rats was collected,and the content of calcium and phosphorus in serum samples was detected by an automatic biochemical analyzer. ELISA kits were used to detect the content of bone alkaline phosphatase (BALP) and osteocalcin (BGP). Micro-CT scanners were used to detect the bone mineral density (BMD) and trabecular bone volume fraction (Tb.BV/TV),trabecular bone thickness (Tb.Th) and trabecular bone number (Tb.N). Hematoxylin-eosin staining (HE) was used to observe the histopathological changes of femur in each group. Immunohistochemical staining was used to detect the expression of VEGF and Runχ2 in femoral tissue. Results: The prepared puerarin/tricalcium phosphate scaffold material has a porous network structure,the compressive strength increases,and the porosity decreases; the puerarin/tricalcium phosphate scaffold material extract can increase the activity of MLO-Y4 cells. Compared with the control group,the femoral tissue of the model group showed obvious necrosis,BMD decreased,Tb.BV/TV,Tb.Th and Tb.N decreased,serum calcium and phosphorus levels decreased,and BALP levels increased,BGP level decreased,expression of VEGF and Runχ2 in femoral tissue decreased. Compared with the model group,the bone sclerosis of rats in the composite scaffold group was significantly improved,BMD increased,and Tb.BV/TV,Tb.Th and Tb.N increased,serum calcium and phosphorus levels increased,BALP The level decreased and BGP level increased,and the expression of VEGF and Runχ2 in femoral tissue increased. Conclusion: The puerarin/tricalcium phosphate scaffold material has a good repairing effect on the necrotic femur of hormonal femoral head necrosis rats.

401 Effects of TPVB and INB on the Levels of TNF-α MMP-9 S-100β and ADP in Patients Undergoing Thoracotomy

Objective: To investigate the analgesic effect of TPVB and INB on patients undergoing thoracotomy,and the influence of different analgesic methods on the serum levels of TNF-α,MMP-9,S-100β and ADP. Methods: Prospectively,400 patients who underwent thoracotomy in our hospital from January 2018 to January 2020 were selected as study subjects,and random number table was used to divide the subjects into TPVB group and INB group,with 200 patients in each group. VAS was used to evaluate the analgesic effect of patients in both groups. Serum levels of TNF-α,MMP-9,S-100β and ADP were detected by ELISA,and perioperative complications were collected from patients in both groups. Results: VAS score was significantly different concerning factors of time,group difference,and time-group interaction (P<0.05). VAS scores 6h,12h,24h and 48h after operation in the TPVB group were significantly lower than those in the INB group (P<0.05). Serum TNF-α,MMP-9,S-100β,and ADP levels were significantly different under the influence of time,group difference,and interaction between time and group (P<0.05). The serum levels of TNF-α,MMP-9 and S-100β in the two groups were significantly higher than those before surgery 24h after surgery (P<0.05),and ADP was significantly lower than that before surgery (P<0.05). The serum levels of TNF-α,MMO-9 and S-100β in TPVB group 24h after operation were significantly lower than those in INB group (P<0.05),and ADP was significantly higher than that in INB group (P<0.05). There was no significant difference in complications and total incidence between the two groups (P>0.05). Conclusion: Both TPVB and INB have good analgesic effect,but TPVB has better analgesic effect and can reduce the influence of inflammatory reaction and nerve function.

406 Study on the Relationship Between Mitogen-Activated Protein Kinase MAP2K5-Rs4776970 Polymorphism and Abnormal Metabolism and Chronic Vascular Inflammation

Objective: To investigate the relationship between mitogen-activated protein kinase MAP2K5-rs4776970 single nucleotide polymorphism (SNP) and abnormal metabolism and chronic vascular inflammation. Methods: A total of 1224 patients were included in the physical examination. According to the revised cholesterol standard (ATPⅢ*),they were divided into the metabolic syndrome (MS) group and the non-MS group,of which 520 were in the MS group and 704 were in the non-MS group. Real-time fluorescence quantitative PCR combined with TaqMan-MGB probe for SNP detection. High-sensitivity C-reactive protein (hs-CRP) is an immunoturbidimetric method. Results: The frequency of AA (7.5% vs 4.0%) distribution of MAP2K5-rs4776970 in the MS group was significantly increased (P<0.05).As to the distribution frequency of TT type (56.2% vs 59.5%)and TA type (36.3% vs 36.5%),there was no significant difference between the two groups (P=0.954). The frequency of A allele was significantly higher in the MS group than in the non-MS group (25.7% vs. 22.2%,P=0.048). After adjusting age and gender by gender and age,Logistic regression analysis indicated that the incidence of MS had no significant correlation with MAP2K5- rs4776970 genotype (OR=0.851,95%CI:0.7~1.035,P=0.106). Correlation analysis suggested that MAP2K5-rs4776970 genotype was associated with waist circumference (WC),body mass index (BMI) and Hs-CRP (P=0.015; P=0.011; P=0.049),compared with MAP2K5-rs4776970TT and TA genotypes,WC,BMI and Hs-CRP levels in AA genotype were significantly increased (P<0.01). Conclusion: MAP2K5-rs4776970 gene polymorphism may be related to obesity and chronic inflammation of blood vessels,but it is not an independent risk factor for MS.

410 The Relationship Between Lumbar and Hip Bone Density and Thyrotropin in Postmenopausal T2DM Patients with Normal Thyroid Function

Objective: To analyze the correlation between thyroid stimulating hormone (TSH) and lumbar and hip bone density in postmenopausal type 2 diabetes mellitus (T2DM) patients with normal thyroid function. Methods: A retrospective selection of 82 menopausal women admitted to the Department of Endocrinology in our hospital from May 2018 to May 2019 were made the research subjects. The subjects' fasting venous blood was collected,and biochemical indexes,thyroid function and bone metabolism indexes were measured; dual-energy X-ray bone density meter was used to measure the bone mineral density of the lumbar spine and total hip. Based on the median of TSH,TSH<2.59μIU/mL was regarded as low TSH group,and TSH≥2.59μIU/mL was regarded as high TSH group,with 41 cases in each. The differences of various indicators between the two groups were compared and their correlation with bone mineral density was analyzed. Results: Comparison of FBG,HbA1c,TC,HDL-C and LDL-C showed no significant difference in age,T2DM course,BMI,glucose and lipid metabolism index in the two groups (P>0.05). TSH level in the low TSH group was significantly lower than that in the high TSH group. There were no significant differences in the bone metabolism indexes of calcium,phosphorus and ALP between the two groups (P>0.05),but the levels of PINP and β-CTx in the low TSH group were significantly higher than those in the high TSH group (P<0.05). Comparison of bone mineral density between the two groups showed that T values of lumbar vertebral bone density and hip bone density in the low TSH group were significantly higher than those in the high TSH group (P<0.05). Pearson analysis showed that TSH was negatively correlated with PINP,β-CTx and r was -0.294 and -0.264,respectively (P<0.05). TSH was positively correlated with lumbar bone density and hip bone density T,and r was 0.617 and 0.410,respectively (P<0.05). Conclusion: In postmenopausal T2DM women with normal thyroid function,TSH level is negatively correlated with bone metabolism indexes PINP and β-CTx,while it is positively correlated with lumbar vertebral bone density and hip bone density T value.

414 Relationship Between Serum Levels of Hcy Hs-CRP TNF-α IL-6 and the Severity of Acute Ischemic Cerebral Infarction

Objective: To investigate the severity of acute ischemic cerebral infarction with high-sensitivity C-reactive protein (hs-CRP),tumor necrosis factor-α (TNF-α),serum homocysteine (Hcy) and interleukin-6 (IL -6),and to explore the relationship between them. Methods: Patients who underwent acute ischemic cerebral infarction in our hospital from January 2015 to December 2018 were enrolled. Another 120 healthy people in our outpatient clinic were selected as the control group. According to the severity of the disease,they were divided into mild stroke (115 cases),moderate stroke (85 cases),and severe stroke (60 cases). According to the area of cerebral infarction,they were divided into small area infarction (122 cases) and medium area infarction (83 cases) and large area infarction (55 cases); the level of each inflammatory factor in serum was tested by enzyme-linked immunosorbent assay (ELISA),and the relationship between the above factors and NIHSS score was analyzed by Pearson correlation analysis. The severity of the disease and the area of cerebral infarction and serum Hcy,hs-CRP,IL-6 and TNF-α levels were statistically analyzed. Results: The levels of Hcy,Hs-CRP,TNF-α and IL-6 in the observation group were significantly higher than those in the control group (P<0.05). The levels of Hs-CRP,Hcy,TNF-α,and IL-6 in the mild infarction group were significantly lower than those in the moderate and severe groups,and the patients in the moderate group were significantly lower than the recombination (P<0.05). The levels of Hs-CRP,Hcy,TNF-α,and IL-6 in patients with small infarction were significantly lower than those in moderate infarction group and large infarction group,and those in moderate infarction group were significantly lower than those in large infarction group (P<0.05). Hs-CRP,Hcy,TNF-α,and IL-6 were used as independent variables,and NIHSS score was used as the dependent variable. Pearson correlation analysis showed that Hs-CRP,Hcy,TNF-α,IL-6 and NIHSS scores were positively correlated (P<0.05). Conclusion: There is a significant positive correlation between serum levels of Hcy,hs-CRP,IL-6 and TNF-α and acute ischemic cerebral infarction. These indicators can be used to assess the patient's condition.

418 Diagnostic Value of Serum CA125 and Human Epidiymis Protein 4 for Epithelial Ovarian Cancer and Its Relationship with Clinical Pathological Characteristics and Prognosis

Objective: To analyze the diagnostic value of serum carbohydrate antigen 125 (CA125) and human epidiymis protein 4 (HE4) for epithelial ovarian cancer and its relationship with clinical pathological characteristics and prognosis. Methods: The clinical data of 96 patients with epithelial ovarian cancer (malignant group) and 90 patients with benign ovarian disease (benign group) admitted in our hospital from April 2014 to April 2019 were retrospectively analyzed. Serum HE4 and CA125 levels were compared,and the results of pathological diagnosis were used as the gold standard to compare the sensitivity,specificity and accuracy of serum HE4 and CA125 levels in the diagnosis of epithelial ovarian cancer and benign ovarian disease. The clinicopathological characteristics and short-term prognosis of all patients with epithelial ovarian cancer were recorded. The expressions of serum HE4 and CA125 in patients with different pathological features were compared,and logistic regression model was used to analyze the independent risk factors affecting the prognosis of patients with epithelial ovarian cancer. Results: The levels of HE4 and CA125 in the malignant group were higher than those in the benign group (P<0.05). The clinical efficacy of serum HE4 in the diagnosis of epithelial ovarian cancer is higher than that of serum CA125 (P<0.05). The sensitivity and accuracy of the combined diagnosis of serum HE4 and CA125 were 96.88% and 94.09%,respectively,which were higher than those of any single serum detection (P<0.05),and there was no difference in the specificity between the combined diagnosis and single serum HE4 diagnosis (P>0.05).The positive expression rates of serum HE4 and CA125 in patients with FIGO stage Ⅲ + Ⅳ and low differentiation are higher than those in patients with FIGO stage Ⅰ + Ⅱ and medium and high differentiation (P<0.05). The patients who are in FIGO stage Ⅲ + Ⅳ,and have low degree of differentiation,CA125> 500U / ml,positive serum HE4 and lymphatic metastasis have low survival rates (P<0.05). FIGO stage Ⅲ + Ⅳ,lymphatic metastasis,low degree of differentiation,CA125> 500U / mL,and positive HE4 are independent risk factors affecting the prognosis of patients with epithelial ovarian cancer (P<0.05). Conclusion: ① Serum HE4 and CA125 are highly expressed in patients with epithelial ovarian cancer,which can be used as an early screening method in the high population,but the clinical value of combined diagnosis is better than that of single serum detection. ②The expression levels of serum HE4 and CA125 are closely related to the clinicopathological characteristics and prognosis of patients. FIGO stage Ⅲ+Ⅳ,lymphatic metastasis,low degree of differentiation,CA125> 500U / mL,positive HE4 are independent risk factors that affect the prognosis of patients with epithelial ovarian cancer,and can be used as an effective indicator to predict the prognosis of patients.

425 An Investigation of the Relationship Between Preoperative and Postoperative Corneal Astigmatism and Visual Acuity Recovery in Patients with High Myopia and Cataract

Objective: To investigate the corneal astigmatism in patients with high myopia and cataract before and after operation,and to analyze the relationship between corneal astigmatism and postoperative visual acuity recovery. Methods: A total of 90 cataract patients (90 eyes) who underwent phacoemulsification in our hospital from December 2017 to July 2020 were retrospectively analyzed. According to whether they had complicated high myopia,they were divided into non high myopia group (44 cases,44 eyes) and high myopia group (46 cases,46 eyes). The corneal curvature in all directions,corneal curvature,corneal astigmatism on anterior and posterior surfaces and visual acuity were measured before and one week after operation. Results: Compared with before operation,the surface curvature above the nose and the level of astigmatism on the anterior and posterior surfaces of the high myopia group were significantly increased (P<0.05),and the type of astigmatism on the anterior and posterior surfaces was significantly different (P<0.05). The corrected visual acuity was improved in both groups one week after operation,compared with those in the high myopia group; the postoperative corrected visual acuity of the non-high myopia group was better (P<0.05). Conclusion: The postoperative visual acuity of the high myopia with cataract patients is worse than that of the non-high myopia with cataract patients,and the postoperative posterior supranasal corneal curvature is larger. Therefore,to reduce the damage of corneal posterior surface,more attention should be paid to the careful operation of such patients and skillfully control the ultrasonic energy when making the corneal lateral incision.

429 The Effect of Percutaneous Intervertebral Foraminal Endoscopy and Disc Endoscopic Nucleus Excision in the Treatment of Lumbar Disc Herniation and Analysis of Influencing Factors for Recurrence

Objective: To explore the efficacy of percutaneous foraminal endoscopy and disc endoscopic nucleus pulposus in the treatment of lumbar disc herniation (LDH) and the factors affecting postoperative recurrence. Methods: 120 patients with LDH admitted to our hospital from March 2017 to March 2019 were selected as the research objects,and they were divided into groups A and B by simple random grouping method,with 60 cases in each group. Patients in group A underwent percutaneous transforaminal endoscopy,and patients in group B underwent endoscopic discectomy. The perioperative indicators of patients were recorded and compared,the visual analogue score (VAS) and the changes of the lumbar Oswestry dysfunction index (ODI) during the one year follow-up period were compared,and the modified MacNab standard was used to evaluate the efficacy of the two groups. At the last follow-up interview,patients were divided into recurrence group (17 cases) and non-recurrence group (103) according to their recurrence status. Single factor analysis and multivariate logistic regression were used to analyze the influencing factors of LDH patients' recurrence. Results: The operation time of group A was longer than that of group B,and the amount of intraoperative blood loss,incision length,and hospital stay of group A were shorter than those of group B. The difference was statistically significant (P<0.05). The patients' VAS score and ODI index showed a significant downward trend over time (P<0.05).The clinical efficacy rate of patients in group A was higher than that in group B,but the difference was not statistically significant (P>0.05). Univariate analysis showed that age,course of disease,type of disease,grade of degeneration,operative segment,intraoperative fibrous annulus damage,intraoperative incomplete removal of nucleus pulposus,etc. may be factors influencing postoperative recurrence of LDH patients (P<0.05),And gender,BMI,history of smoking,history of drinking,comorbidities,operation method,operation time,prominent position,etc. were not related to postoperative recurrence of LDH patients (P>0.05). Multivariate logistic regression analysis showed that age <60 years,course of disease <3 years,and extracted LDH were independent protective factors for postoperative recurrence (P<0.05),L4~L5,L5~S1 surgical segment,intraoperative fibroblast damage,intraoperative incomplete removal of nucleus pulposus were independent risk factors for postoperative recurrence (P<0.05),and the degree of degeneration was not related to postoperative recurrence (P>0.05). Conclusion: Both percutaneous foraminal endoscopy and discectomy nucleus pulposus can effectively treat lumbar disc herniation. In patients with recurrence after surgery,age,disease course,disease type,surgical segment,intraoperative annulus fibrosus damage,intraoperative incomplete removal of nucleus pulposus are the main influencing factors. Patients with the above risk factors need to be actively intervened to reduce the recurrence rate after surgery.

436 An Analysis of Risk Factors Related to the Depth of Invasion in Esophageal Carcinoma

Objective: To study the related risk factors inherent in the invasive depth of esophageal cancer,and provide a more accurate method to judge the invasive depth of esophageal cancer. Methods: A total of 174 patients (174 lesions) diagnosed with esophageal cancer via gastroscopy and pathological diagnosis in the Second Affiliated Hospital of Zhengzhou University from June 2014 to June 2020,including 102 males (58.6%),72 females (41.4%),were retrospectively analyzed. The age,gender,tumor location,tumor size,preoperative PLR,NLR,LMR and postoperative pathological diagnosis were observed and recorded. Results: High PLR was significantly correlated with the depth of invasion (> SM1 vs ≤SM1:OR=1.017,95% CI=1.006 ~ 1.028,P = 0.002); tumor length was significantly correlated with the depth of invasion (> SM1 vs ≤SM1:OR=1.581,95% CI=1.234 ~ 2.024,P<0.001),where AUC was 0.74 and 0.796. The sensitivity was 0.495 and 0.722,and the specificity respectively was 0.883 and 0.714. The AUC,sensitivity and specificity of PLR combined with tumor length was 0.812,0.866 and 0.61. Conclusion: PLR and tumor length are accurate in their diagnostic capabilities for the depth of the invasion of esophageal cancer,and their combination can further improve accuracy and provide a more reliable basis for treatment options.

440 Comparison of Clinical Effects of Two Surgical Methods for Patients with Osteoporotic Thoracolumbar Vertebral Compression Fractures

Objective: To observe effects of two different treatment methods (percutaneous puncture and kyphoplasty) for patients with osteoporotic thoracolumbar compression fracture (Osteoporotic thoracolumbar compression fracture,OTCF). Methods: The selected study period was from January 2016 to October 2019. The 84 patients treated in this hospital who had OTCF were divided into observation group (n=42) and control group (n=42) by simple random grouping. For patients in the observation group,the treatment was percutaneous kyphoplasty,while for patients in the control group,percutaneous vertebroplasty was adopted. The overall efficiency,Barthel index and VAS pain score,the recovery of the anterior edge of the vertebral body,the amount of bone cement injection,and the leakage rate were compared. Results: The observation group's effective rate was (95.2%) higher than that of the control group(78.6%) (P<0.05). There was no significant difference between the two groups of patients in the Barthel index and VAS pain score before and half a year after the operation (P>0.05). The observation group had more bone cement injections,and the observation group had significantly lower bone cement leakage rate (P<0.05). The height of the anterior edge of the vertebral body and the Cobb s angle between the two groups of patients were compared. There was an interaction between groups and time (P<0.001),and the observation group had greater changes (P<0.001). The functional exercise time and functional recovery time of patients in the observation group were significantly shorter (P<0.05). Conclusion: In the treatment of OTCF patients,percutaneous kyphoplasty has certain advantages,especially in the effect of reduction.

443 Effects of Total Knee Replacement and Artificial Femoral Head Replacement on Femoral Neck Fracture and Deep Venous Thrombosis in Lower Limbs

Objective: To investigate the use of artificial femoral head replacement and total hip replacement in patients with femoral neck fractures,and to compare the intraoperative conditions,postoperative curative effects,and postoperative complications. Methods: A total of 92 patients with femoral neck fractures admitted to our hospital from October 2017 to October 2018 were selected and randomly divided into observation group and control group,with 46 cases in each. The control group underwent artificial femoral head replacement treatment. Patients in the observation group were treated with total hip arthroplasty. The clinical effects of the two groups were observed. The relevant indexes (intraoperative blood loss,operation time and drainage volume) of the two groups were compared. The blood D-dimer,FIB,HCT and whole blood viscosity levels,postoperative complications (deep vein thrombosis of the lower extremities,prosthesis loosening,infection,etc.) were compared. Results: The total effective rate of the observation group was 95.65%,and that of the control group was 73.91%. The effective rate of the observation group was higher (Z=2.936,P<0.05). The operation time,intraoperative blood loss and drainage volume of patients in the observation group increased compared with those in the control group (P<0.05). 2 weeks after treatment,the blood D-dimer,FIB,HCT,and whole blood viscosity levels of the two groups were significantly reduced (P<0.05). And the blood D-dimer,FIB,HCT and whole blood viscosity levels of the observation group were lower than those of the control group (P<0.05). The incidence of complications in the observation group was significantly lower than that in the control group (P<0.05). Conclusion: Total hip arthroplasty for patients with femoral neck fractures has a better clinical effect than artificial femoral head replacement,with fewer postoperative complications,but more bleeding and operation time. It is worthy of further clinical promotion.

447 Analysis of Incidence and Risk Factors of Adverse HT and Prognosis in Patients with Acute Cerebral Infarction After Mechanical Thrombolysis

Objective: To analyze the occurrence and risk factors of hemorrhagic transformation (HT) and poor outcome in patients with acute cerebral infarction after mechanical thrombectomy. Methods: The clinical data of 80 patients with acute cerebral infarction admitted to our hospital from January 2019 to January 2020 were retrospectively analyzed. The modified Rankin Scale (mRS) was used to evaluate the clinical outcome of the patients 90 days after the onset of the disease. According to the occurrence of HT,they were divided into HT group and non-HT group; according to mRS score,they were divided into good outcome group and poor outcome group. Multivariate logistic regression analysis was used to analyze the independent risk factors that affect patients' poor outcome and HT. Results: The gender,history of diabetes,history of atrial fibrillation,fasting blood glucose,time from puncture to recanalization,number of thrombectomy,mTICI grade after thrombectomy were compared between the HT group and the non-HT group with statistically significant differences (P<0.05). Using the above factors as independent variables and HT as the dependent variable for multivariate logistic regression analysis,the results showed that atrial fibrillation,the time from puncture to recanalization,the number of thrombectomy times,and the mTICI grade after thrombectomy were the independent risk factors for HT after embolization (P<0.05) that led to mechanical retrieval in patients with acute cerebral infarction. Patients in the good outcome group and the poor outcome group have history of diabetes,atrial fibrillation,baseline National Institutes of Health Stroke Scale (NIHSS) score,middle cerebral artery occlusion vessel,vertebrobasilar artery occlusion vessel,anterior circulation stroke,HT and brain The comparison of parenchymal hematoma was statistically significant (P<0.05). Multivariate logistic regression analysis with the above factors as independent variables and poor outcome as the dependent variable showed that diabetes,baseline NIHSS score,and cerebral parenchymal hematoma were all independent factors that caused poor outcomes in patients with acute cerebral infarction after mechanical thrombectomy (P<0.05). Conclusion: Complicated brain parenchymal hematoma,high baseline NIHSS score,and diabetes are independent risk factors for adverse outcomes in patients with acute cerebral infarction after mechanical thrombectomy,while atrial fibrillation is an independent risk factor for HT.

Objective: To study the effect of modified transumbilical single port laparoscopic technique in the treatment of tubal ectopic pregnancy. Methods: A total of 120 patients with tubal ectopic pregnancy admitted to our hospital from January 2018 to December 2019 were selected as the research objects. According to the random number remainder grouping method,they were divided into the conventional laparoscopy group (40 cases),the transumbilical single port laparoscopy group (40 cases),and the modified transumbilical single port laparoscopic group (40 cases). Three groups were respectively treated with conventional three-port laparoscopic surgery,transumbilical single port laparoscopic surgery,and modified transumbilical single port laparoscopic technology,and the perioperative indicators (operating length,intraoperative blood loss,postoperative blood HCG return to normal time,hospitalization time),gastrointestinal function recovery indicators (exhaust time,defecation time),laboratory indicators (white blood cell count,C-reactive protein,D-dimer),incision aesthetic satisfaction,and complication rate were compared in the three groups. Results: Under comparison,the values of the transumbilical single port laparoscopic group and the modified transumbilical single port laparoscopic group were similar,and the difference was not statistically significant (P>0.05). However,compared with the conventional laparoscopic group,the difference was statistically significant (P<0.05). The three groups of incision aesthetic satisfaction and complication rate were compared,and the difference was statistically significant (P<0.05). Conclusion: The improved transumbilical single port laparoscopic technique in the treatment of tubal ectopic pregnancy has a better application effect and a lower incidence of complications. It is worthy of promotion and application in future work.

457 Changes and Clinical Significance of NLR, ELR and PLR in Early-onset Neonatal Sepsis

Objective: To observe the clinical significance and the changes on the neutrophil-to-lymphocyte, the eosinophil-to-lymphocyte, the platelet-to-lymphocyte with Neonatal septicemia. Methods: From April 2017 to May 2020, 58 newborns with early-onset sepsis in our hospital were selected as sepsis group, and 40 non infected newborns in the same period were selected as control group. The levels of NLR, ELR and PLR were compared between the two groups. The changes and clinical significance of NLR, ELR and PLR in early-onset neonatal sepsis were evaluated. Results: The levels of NLR and ELR in sepsis group were significantly higher than those in control group (P<0.05). ROC curve analysis showed that area under the curve NLR > ELR > PLR, NLR cutoff value was 1.987, sensitivity was 78.87%, specificity was 88.11%, area under the curve was 0.752, ELR cutoff value was 0.117, sensitivity was 70.81%, specificity was 77.25%, area under the curve was 0.689. Conclusion: The diagnostic efficiency of NLR is higher than that of ELR, which can assist in the diagnosis of early-onset neonatal sepsis, and has a certain reference value for the diagnosis and evaluation of children's diseases.

Methods: Retrospective analysis was performed on the clinical data of 137 patients who received laparoscopic anterior resection for rectal cancer and 132 women who received open anterior resection for low rectal cancer in our hospital from January 2010 to October 2020,and they were divided into the laparoscopic group and the open group. The laparotomy group received conventional anterior resection for rectal cancer,while the laparoscopy group received laparoscopic assisted anterior resection for rectal cancer. The differences of perioperative clinical indicators (operative time,intraoperative blood loss,length of proximal bowel resection,length of distal bowel resection,postoperative first exhaust time,postoperative first out of bed time,and hospital stay length) were compared between the two groups. The occurrence of RVF was analyzed and the patients were accordingly divided into RVF group and non RVF group,whose basic information was compared and the difference was applied to the Logistic model and quantified with RVF as the dependent variable (Y=1),with menopause,tumor distance from anal edge,Duke 's stage,consistent way,the terminal ileum preventive colostomy as independent variables. Risk factors of RVF were clarified. Results: There were no statistically significant differences between the two groups in operation time,length of proximal bowel resection and length of distal bowel resection (P>0.05). Intraoperative blood loss,postoperative exhaust time,postoperative out of bed time and hospital stay length in the laparoscopic group were all lower than those in the open group,and the differences were statistically significant (P<0.05). After statistical analysis,RVF occurred to 13 patients in the laparoscopic group during hospitalization,with an incidence of 9.49%. Patients with RVF were set as RVF group (n=13),and those without RVF were set as the non-RVF group (n=124). There was no significant difference in BMI,living area,pathological type,smoking and alcohol consumption between the two groups (P>0.05). The proportion of patients in RVF group ≥60 years old,menopause,tumor distance from the anal edge ≤6cm,Duke's stage C,double anastomosis and no preventive terminal ileostomy was significantly higher than that in the non-RVF group,and the difference was statistically significant (P<0.05). Multivariate Logistic regression analysis confirmed that ≥60 years of age,menopause,tumor distance from the anal edge ≤6cm,Duke's stage C,double anastomosis,and non-prophylactic terminal ileostomy were all risk factors for RVF after laparoscopic anterior rectal resection for patients with low rectal cancer (P<0.05). Conclusion: Compared with open treatment,laparoscopic anterior rectal resection for low rectal cancer can reduce intraoperative bleeding,and patients recover quickly after surgery. Multivariate Logistic regression analysis confirmed that RVF after laparoscopic anterior rectal cancer resection was affected by many factors,which should be paid close attention to by clinicians.

467 Effect of Left Subclavian Artery Bypass Technique on Neurological Complications After Aortic Dissection Endovascular Exclusion

Objective: To investigate the effect of left subclavian artery bypass technique on neurological complications after aortic dissection endovascular exclusion. Methods: The clinical data of 43 patients with Stanford type B aortic dissection (TBAD) admitted to our hospital from June 2016 to June 2018 were retrospectively analyzed. The patients were divided into group A (n=23) and group B (n=20) according to the different surgical methods; group A received left subclavian artery bypass technology combined with aortic dissection endovascular exclusion,and group B received chimney stent implantation combined with aortic dissection endovascular exclusion. The operation conditions,postoperative complications during hospitalization,and death rate within 2 years of follow-up were compared between the two groups. Results: There was no significant difference in age,gender,medical history and dissection stage between the two groups (P>0.05). The operation time of group A was shorter than that of group B (P<0.05),and the amount of intraoperative blood loss was less than that of group B (P<0.05),while there was no significant difference in the rate of intraoperative leakage and success rate between group A and group B (P>0.05). The incidences of internal leakage and spinal cord ischemia in group A were lower than those in group B (P<0.05),while the incidences of cerebral infarction and left upper limb ischemia in group A were not significantly different from those in group B (P>0.05). After 2 years of follow-up,up to June 2020,there were 2 deaths in group A and 5 deaths in group B,there was no significant difference in mortality between group A and group B (χ2= 1.062,P=0.303). Conclusion: Compared with chimney stent implantation combined with aortic dissection endovascular exclusion,the implementation of left subclavian artery bypass technology combined with aortic dissection endovascular exclusion can significantly shorten the operation time,reduce intraoperative bleeding,reduce the incidences of leakage and spinal cord ischemia during the hospitalization in patients with TBAD. There was no significant difference in intraoperative leakage rate,success rate of operation,incidence of cerebral infarction,incidence of left upper limb ischemia,mortality and so on between the two groups.

471 Clinical Effects of Minimally Invasive Intracranial Hematoma Aspiration and Drainage Combined with Rt-PA in the Treatment of Spontaneous Intra-Cerebral Hemorrhage and the Influence on Neurological Function and Complications

Objective: To investigate the clinical effects of minimally invasive intracranial hematoma aspiration and drainage combined with recombinant tissue-type plasminogen activator (rt-PA) in the treatment of spontaneous intracerebral hemorrhage and the influence on neurological function and complications. Methods: This study retrospectively reviewed 146 patients with spontaneous intracerebral hemorrhage treated in the hospital between May 2016 and May 2019. Patients treated with conventional craniotomy were included in the control group (72 cases) and those treated with minimally invasive intracranial hematoma aspiration and drainage combined with rt-PA were included in the observation group (74 cases). The two groups were compared in terms of operation-related indexes,clinical effects,neurological function and complications. Results: The observation group had significantly shorter operation time and hospital stay than the control group (P<0.05). The comparison of hematoma volume between the two groups before operation,at 1 d and 7 d after operation showed statistically significant differences (P<0.05). The hematoma volume of the two groups decreased at different time points after operation,and the observation group had larger hematoma volume than the control group at 1 day after operation (P<0.05),but no significant difference was observed in hematoma volume between the two groups at 7 days after operation (P<0.05). No patient died during the follow-up period. The excellent and good rate of GOS was 67.57% in the observation group,and 50.00% in the control group (P>0.05). Before treatment,no significant differences were found between the two groups in NIHSS scores and SSS scores (P>0.05). After treatment,the above scores decreased in both groups,and the observation group had significantly lower scores than the control group. In addition,the difference values before and after treatment were higher in the observation group than in the control group (P<0.05). The rebleeding rate and blood-brain barrier index were significantly lower in the observation group than in the control group (P<0.05). There was no significant difference in the incidence of intracranial infection between the two groups (P>0.05). Conclusion: Minimally invasive intracranial hematoma aspiration and drainage combined with rt-PA for treating spontaneous intracerebral hemorrhage has characteristics of less surgical trauma,fast postoperative recovery,and good clinical effects. It is conductive the recovery of cranial nerve function and reduction of damage to the cranial nerve,with a low incidence of complications.

476 Expression Levels and Clinical Significance of Neutrophil Surface Differentiation Antigens CD67 and CD24 in Paroxysmal Nocturnal Hemoglobinuria

Objective: To analyze the expression levels and clinical significance of neutrophil surface differentiation antigens CD67 and CD24 in paroxysmal nocturnal hemoglobinuria (PNH). Methods: The clinical data of 32 patients with PNH (PNH group) in our hospital were retrospectively analyzed,and the clinical data of 28 patients with aplastic anemia (AA) (AA group) and 32 healthy subjects with physical examination (control group) admitted to the hospital during the same period were collected. The differences in the levels of neutrophil surface differentiation antigens CD67,CD24,CD55 and CD59 were compared among the three groups. The correlation among the expression levels of the above indicators was evaluated in PNH group,and receiver operating characteristic curves (ROC curves) were used to evaluate the diagnostic value of neutrophil surface differentiation antigens on PNH. Results: There were no statistically significant differences in gender,age,body mass index,smoking history and drinking history among the three groups (P>0.05). Comparison of expression levels of neutrophil surface CD67,CD24,CD55 and CD59 in the three groups showed that PNH grouP<AA grouP<control group (P<0.05). Pearson correlation analysis showed that expression levels of neutrophil surface CD67,CD24,CD55 and CD59 were significantly positively correlated with each other in PNH group (P<0.05). After ROC curve analysis,it is found that the expression levels of neutrophil surface CD67,CD24,CD55 and CD59 had high diagnostic value on PNH (AUC=0.961,0.967,0.972,0.995,P<0.05),and their cut-off values were 81.88%,83.64%,84.42% and 81.37% respectively,and the combination of the four had the highest diagnostic value (AUC=1.000,P<0.05). Conclusion: The expression levels of neutrophil surface differentiation antigens CD67 and CD24 are severely reduced in PNH,and they are consistent with the changes of expression levels of CD55 and CD59,and the combined detection of the four indicators is of high value in diagnosing PNH,and can guide clinical diagnosis and treatment.

480 The Relationship Between FA,VR of Cerebellar Dentate Nucleus and the Condition of Neonatal Pathological Jaundice by DTI Technique and Its Value in Predicting ABE

Objective: To analyze the relation between the severity of the disease and magnetic resonance diffusion tensor imaging (DTI) technology in determining fractional anisotropy (FA) and volume reconstructio (VR) of the cerebellar dentate nucleus in children with neonatal pathological jaundice and to analyze the value of predicting the occurrence of acute bilirubin encephalopathy (ABE). Methods: A total of 85 children with simple neonatal pathological jaundice in our hospital from June 2018 to June 2020 were selected as the jaundice group,and 73 children with neonatal pathological jaundice secondary to ABE during the same period were selected as the ABE group. All have been tested by DTI technology. The general information,disease severity,neonatal behavioral neurological assessment (NBNA) score,laboratory indicators [total bile acid (TBA),total bilirubin (TSB),direct bilirubin (DBil),alanine aminotransferase (ALT),γ-glutamyltransferase (GGT)] levels,FA and VR in the cerebellar dentate nucleus were compared between the two groups. The relationship between FA and VR of cerebellar dentate nucleus and the severity of pathological jaundice,NBNA score,laboratory indicators and the occurrence of ABE were analyzed,and the predictive value of FA and VR of cerebellar dentate nucleus on the occurrence of ABE was evaluated. Results: There was a statistically significant difference in the degree of illness and NBNA score between the two groups (P<0.05). The TSB,DBil,TBA and VR of the cerebellar dentate nucleus in the ABE group were higher than those in the jaundice group,and the FA of the cerebellar dentate nucleus was shorter than that in the jaundice group (P<0.05). The FA of the cerebellar dentate nucleus in children with neonatal pathological jaundice was positively correlated with the NBNA score,and negatively correlated with the severity of the disease; TSB,DBil,and TBA. The VA of the cerebellar dentate nucleus was negatively correlated with the NBNA score,and positively correlated with the severity of the disease,TSB,DBil,and TBA (P<0.05). The severity of illness,NBNA score,TSB,cerebellar dentate nucleus FA,VR and ABE were significantly correlated (P<0.05). The combination of FA and VR in the cerebellar dentate nucleus predicted the highest AUC in ABE,which was 0.810,and the sensitivity and specificity were 71.23% and 78.82%,respectively. Conclusion: The FA and VR of the cerebellar dentate nucleus are significantly correlated with the severity of neonatal pathological jaundice. Early detection of FA and VR of the cerebellar dentate nucleus by DTI can provide effective auxiliary means for clinical evaluation of the severity of the disease and predicting the occurrence of ABE.

486 Expression and Clinical Significance of LncRNA MNX-AS1 in Non-Small Cell Lung Cancer

Objective: To investigate the expression and clinical significance of long non-coding RNA (lncRNA) MNX1-AS1 in non-small cell lung cancer (NSCLC). Methods: The primary lesions and adjacent tissues of 180 patients with NSCLC from Jun.2015 to Jun.2017,diagnosed by histopathology and surgically removed,were collected. Real time quantitative PCR (RT-qPCR) was used to detect the expression of MNX1-AS1. The relationship between the expression of MNX1-AS1 and the clinicopathological characteristics of NSCLC patients was analyzed. Kaplan-Meier method and Cox regression analysis were used to analyze the prognosis of patients with different expression levels of MNX1-AS1. Results: The relative expression of MNX1-AS1 in NSCLC tissues was significantly higher than that in adjacent tissues (P<0.001). The expression of MNX1-AS1 was significantly correlated with tumor differentiation,TNM stage and lymph node metastasis in NSCLC patients. The progression free survival,overall survival and 5-year overall survival rate of patients with high expression of MNX1-AS1 were lower than those of patients with low expression of MNX1-AS1 (P<0.05). Multivariate Cox regression analysis showed that MNX1-AS1 level was an independent prognostic factor in NSCLC patients. Conclusion: MNX1-AS1 was highly expressed in NSCLC and was closely related to its pathological characteristics and prognosis. It might be a new biomarker for early diagnosis and prognosis evaluation of NSCLC patients.

490 Clinical Efficacy and Safety of Intravitreal Injection of Compaccip in the Treatment of Choroid Neovascularization Secondary to Pathological Myopia

Objective: To explore clinical efficacy and safety of intravitreal injection of Compaccip in the treatment of choroid neovascularization secondary to pathological myopia. Methods: A total of 100 CNV patients from July 2017 to July 2019 were included and divided into research group(n=50)which received conventional western medicine treatment,and control group (n=50),which had intravitreal injection of Compaccip. Before and 3 months after treatment the best corrected visual acuity (BCVA) was applied to evaluate vision of patients,and also conducted were the observation of CNV area before and after treatment,the analysis of changes in intraocular pressure (IOP) and calculation of the thickness of macular central (CMT),and the measurement and comparison of the levels of vascular endothelial growth factor (VEGF) and C-reactive protein of the two groups. Results: 3 months after treatment,BCVA of patients in both groups was increased compared with that before treatment,and the difference was statistically significant (P<0.05),but there was no statistically significant difference (P>0.05) between the two groups. 3 months after treatment,the visual acuity of both groups increased,but there was no statistically significant difference between the two groups (P>0.05).Compared with those before treatment,the CNV area,IOP,CMT,VEGF and CRP in the two groups decreased significantly (P<0.05),but there was no statistically significant difference between the two groups (P>0.05).Compared within the two groups,there was no statistically significant difference in complications and recurrence rates (P>0.05). Conclusion: The effect of intravitreal injection of Conbercept in the treatment of pathological myopia CNV is equivalent to that of Ranibizumab. It can effectively reduce inflammatory indicators,with fewer complications and recurrences,and high safety.

494 Diagnostic Value of Combined Detection of Serum TK1,CEA,CA72-4 and CA242 for Gastric Cancer

Objective: To investigate the diagnostic efficiency of combined detection of serum thymidine kinase (TK1),carcinoembryonic antigen (CEA),carbohydrate antigen (CA) 72-4 and CA242 for gastric cancer. Methods: 120 patients with gastric cancer who were admitted to the hospital from February 2018 to February 2019 were included in the gastric cancer group. Another 100 patients with gastric diseases who underwent gastroscopy in the hospital for stomach discomfort and 50 healthy adults receiving physical examination during the same period were included in the benign gastric disease group and the control group. Levels of serum TK1,CEA,CA72-4 and CA242 were determined. The indicators were compared between the groups and the diagnostic efficiency of each index was analyzed by ROC curve. Results: The levels of serum TK1,CEA,CA72-4 and CA242 in the gastric cancer group were significantly higher than those in the benign gastric disease group and the control group (P<0.05),without significant difference between the benign gastric disease group and the control group (P>0.05). In the gastric cancer group,serum indexes in patients with advanced gastric cancer were significantly higher than those in patients with early gastric cancer (P<0.05). The areas under the curves of TK1,CEA,CA72-4 and CA242 alone for diagnosis of gastric cancer were 0.781,0.764,0.769 and 0.719,respectively. The areas under the curves of different tumor markers for diagnosis of gastric cancer were larger than that of single diagnosis. The area under the curve of combined diagnosis of the four indexes and TK1,CEA,CA242 combination was larger. Conclusion: The elevation of serum TK1,CEA,CA72-4 and CA242 is witnessed in patients with gastric cancer. The combined diagnosis of these indicators can improve the diagnostic efficiency for gastric cancer.

499 The Predictive Value of CA125 CA199 HE4 and ROMA Index on the Prognosis of Ovarian Cancer

Objective: To investigate carbohydrate antigen 125 (CA125),carbohydrate antigen 199 (CA199),human epididymisprotein 4 (HE4) and risk of ovarian malignancy algorith (ROMA) index for the prognosis of ovarian cancer. Methods: A retrospective analysis of 70 cases of ovarian cancer treated in our hospital from March 2017 to March 2019 was conducted,depending on the patient's survival,39 surviving patients were classified as the survival group,31 patients who died were classified as death group. The CA125,CA199,HE4 and ROMA indexes of the two groups of patients was recorded,and logistic regression analysis was applied to the impact of the above indexes on the prognosis of ovarian cancer,ROC curve analysis was applied to the predictive value of the above indexes on the prognosis of the disease. Results: The CA125,CA199,HE4 and ROMA indexes of the death group were significantly higher than those of the survival group,and there were statistical differences between the groups (P<0.05). Multivariate logistic regression analysis showed that CA125,CA199,HE4 and ROMA index were independent factors influencing the prognosis of ovarian cancer (P<0.05). ROC curve analysis showed that the area under the curve (Area Under Curve,AUC) of the CA125、CA199、HE4、ROMA index and four combinations for the prognosis of ovarian cancer was 0.758,0.874,0.800,0.833 and 0.887,respectively. The CA199 and four combinations had better predictive value for prognosis. Conclusion: CA125、CA199、HE4 and ROMA indices are independent prognostic risk factors for ovarian cancer. The level of the above indexes can predict the prognosis of the disease,the prediction value of the four combinations is the best,and the clinical can be used to predict the prognosis of the patients through the four combined tests.

503 Effect of Alteplase on Serum Oxidative Stress Vascular Endothelial Function and Cerebral Nerve Function in Patients with ACI

Objective: To investigate the effect of alteplase on serum oxidative stress,vascular endothelial function and brain nerve function in patients with acute cerebral infarction (ACI). Methods: Between September 2018 and September 2020,160 patients with ACI were selected and randomly divided into 2 groups. The control group (n=80) underwent conventional treatment with aspirin and clopidogrel,while the observation group (n=80) underwent alteplase on the basis of the treatment for the control group. The 2 groups were compared with respect to treatment effects,serum oxidative stress,vascular endothelial function and cerebral nerve function indexes as well as adverse reactions. Results: The total response rate in the observation group was 95.00%,higher than 83.75% in the control group (P<0.05). After treatment,the levels of malondialdehyde (MDA) and endothelin (ET-1) and the National Institutes of Health Stroke Scale (NIHSS) were reduced,while the levels of superoxide dismutase (SOD) and vascular endothelial growth factor (VEGF) and mRS scores were increased in the two groups (P<0.05). In addition,the difference values of MDA,ET-1,SOD and VEGF levels,NIHSS and mRS scores before and after treatment were greater in the observation group than in the control group (P<0.05). There was no significant difference in the incidence of adverse reactions between the 2 groups (8.75% vs 6.25%) (P>0.05). Conclusion: In sum,alteplase can reduce oxidative stress and vascular endothelial damage,and improve cerebral nerve function in patients with ACI.

508 Effects of PCSK9 Inhibitor-Evolocumab on Blood Lipid Profiles and Vascular Endothelial Function in Patients with Acute Coronary Syndrome

Objective: To explore the effects of proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitor-evolocumab on blood lipid profiles and vascular endothelial function in patients with acute coronary syndrome (ACS). Methods: A total of 160 ACS patients in the hospital from June 2019 to June 2020 were enrolled and divided into observation group and control group by random number table method,80 cases in each group. The control group was given oral atorvastatin,while observation group was given subcutaneous injection of evolocumab on basis of control group. The levels of blood lipid profiles,vascular endothelial function,inflammatory factors and adverse reactions were compared between the two groups. Results: The difference values of cholesterol (TC),triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) before and after treatment in observation group were greater than those in control group (P<0.05),and difference values of serum endothelin 1 (ET-1) and nitric oxide (NO) before and after treatment were greater than those in control group (P<0.05). The difference values of serum inflammatory factors such as tumor necrosis factor α (TNF-α),interleukin 6 (IL-6) and high-sensitivity C-reactive protein (hs-CRP) before and after treatment in observation group were greater than those in control group (P<0.05). There was no significant difference in the incidence of adverse reactions between the two groups (P>0.05). Conclusion: PCSK9 inhibitor-evolocumab can effectively regulate blood lipid level of ACS patients,improve vascular endothelial function and inhibit inflammation reactions,with high medication safety.

512 Comparison of Efficacy of Teguio and Apatinib in the Treatment of Advanced Refractory Ovarian Cancer and the Effect of Drugs on HE4 CA125 and CA199

Objective: To observe the clinical efficacy of apatinib and tigio for patients with advanced refractory ovarian cancer (OC),and to explore the effect of the above drugs on human epididymal secretory protein 4(HE4),carbohydrate antigen 125(CA125) and carbohydrate antigen 199(CA199). Methods: Retrospective analysis was conducted of 88 patients with refractory OC from April 2018 to September 2019(TNM Ⅲ-Ⅳ),who were divided into observation group (treated with apatinib mesylate) and control group (treated with tigio),with 44 cases in each. The changes of HE4,CA125 and CA199 were compared between the two groups. The total effective rates,disease control rates and adverse reactions were compared between the two groups. Results: After treatment,the total effective rate of observation group was 50.00%,which was significantly higher than that of control group (P<0.05). The serum HE4,CA125,CA199 levels in the observation group decreased significantly compared with those in the control group (all P<0.05). The rates of non-hematological and hematological adverse reactions were 75.00% and 77.27% in the control group after treatment; there was a significant increase in 52.27% and 36.36% of the observation group (all P<0.05). Conclusion: Apatinib treatment for advanced refractory OC patients is ideal,with less adverse reactions and better disease control. It is considered to be related to the obvious inhibition of apatinib HE4、CA125、CA199 level,which has clinical value.

516 Effects of Sodium Valproate Combined with Levetiracetam or Lamotrigine on Electroencephalography in the Treatment of Epilepsy

Objective: To investigate the effects of sodium valproate combined with levetiracetam or Lamotrigine in the treatment of epilepsy on eeg. Methods: A total of 384 epileptic patients admitted to our hospital from July 2014 to March 2020 were selected. According to the random number table,the patients were randomly divided into control group,study group 1 and study Group 2,each of 128 cases. The control group was treated with sodium valproate,the study group 1 was treated with sodium valproate combined with levetiracetam,and the study group 2 was treated with sodium valproate combined with lamotrigine. The differences of attack control rate,eeg efficacy and cognitive function among the three groups were observed. Results: In terms of seizure control,its rates of study group 1 and 2 after treatment were significantly higher than that of control group,and the differences between groups and time points were statistically significant (P<0.05).The total effective rate of study group 1 and study group 2 was significantly higher than that of control group (P<0.05). Before treatment,there were no statistically significant differences in P300 incubation period and MoCA scores among the three groups (P>0.05); after treatment,the P300 incubation period and MoCA scores of study group 1 and study group 2 were higher than those of control group,and the differences were statistically significant (P<0.05). Conclusion: Valproate combined with levetiracetam or lamotrigine in the treatment of epilepsy has a good effect in effectively controlling seizures,reducing electrical discharge,improving eeg background,improving cognitive function. The combination drug should be selected reasonably according to the specific situation of the patients.

521 Intrathecal Continuous Infusion of Different Doses of Dexmedetomidine Combined with Sufentanil in Labor Analgesia

Objective: To investigate the clinical study of continuous intrathecal infusion of different doses of dexmedetomidine combined with sufentanil in labor analgesia. Methods: 120 full-term pregnant women admitted to our hospital from May 2018 to October 2019 were randomly divided into compatibility group (1:3) and control group A. The experimental group was divided into B1,B2 and B3 subgroups with 30 cases in each. The NRS scores of contractile pain in each group at different time points (before treatment,15min after treatment,1 hour after treatment and full-time opening of cervical orifice) were compared. The baseline fetal heart sounds of each group were compared at each time point (before treatment,15min after treatment,1 hour after treatment and full-time opening of cervical orifice). The second stage of labor was counted in each group. Apgar scores of newborns in each group (at birth,1min after birth and 5 min after birth) were compared. The complications of each group were compared. Results: The NRS scores of contractile pain in group B3 were lower than those in group A,group B1 and group B2,the difference was statistically significant(P<0.05),at 15min after treatment,1h after treatment and full-time opening of the public portal. There was no significant difference in baseline fetal heart sounds in each group at each time point (before treatment,15min after treatment,1h after treatment and full-time opening of cervical orifice) (P>0.05). The second stage of labor in group B3 was shorter than that in group A,group B1 and group B2,the difference was statistically significant(P<0.05). At birth,1 min after birth and 5min after birth,there was no significant difference in neonatal Apgar score (P>0.05). The proportion of uterine agony in group B3 was lower than that in group A,group B1 and group B2,the difference was statistically significant(P<0.05). Conclusion: During labor analgesia,a relative dose (0.04μg/kg) of dexmedetomidine combined with sufentanil can effectively relieve the pain of patients without affecting the fetal heart sound and neonatal asphyxia ratio,and has less complications and high safety.
