ZHAO Wenming, ZHOU Bingjuan, LI Jinmei, et al
DOI: 10.3969/j.issn.1006-6233.2018.03.001
Objective: To investigate the relationship between the expression of -1 - alpha (HIF-1-α), glucose transporter 1 (Glut-1) and the degree of differentiation and molecular subtypes of breast cancer. Methods: Immunohistochemical method was used to detect the expression of HIF-1α and Glut-1 in 102 cases of breast cancer and 60 cases of adjacent normal tissues. The expression rates of HIF-1α and Glut-1 in breast cancer tissues with different degree of differentiation and molecular typing were compared. Results: The positive expression rates of HIF-1α and Glut-1 protein in breast cancer tissues were 62.75% and 68.63%, respectively, which were higher than those in 11.67% normal tissues. The difference was statistically significant (P < 0.05). The positive expression rates of HIF-1α and Glut-1 protein in high differentiated breast cancer tissues were 34.78% and 40.91%, respectively, which were lower than those of moderately differentiated 60%, 68.88%, and poorly differentiated 85.29% and 88.24% (P < 0.025). The differentiation of moderately differentiated breast cancer was lower than that of poorly differentiated ones (P < 0.025). There was no significant difference in positive expression rates of HIF-1α and Glut-1 protein between Luminal A type breast caner and Luminal B type breast caner (46.67% and 51.11% vs 54.55% and 63.64%) (P > 0.017). However, the rates in Luminal A and Luminal B type breast cancer were all lower than those in tissues with over-expressed HER-2 (85.71%, 92.86%) and triple negative breast cancer (90.48% and 95.24%) (P < 0.017). There was no significant difference between HER-2 overexpression type and three yin type (P > 0.017). Conclusion: The expression of HIF-1α and Glut-1 in breast cancer patients is highly positive, and the positive expression of HIF-1 alpha and Glut-1 in different degree of differentiation and molecular subtype of breast cancer is different.

357 The Relationship between the Expression Level of Tumor Tissue and Plasma miR-146a and the Clinicopathological Features and Prognosis of Patients with Intrahepatic Cholangiocarcinoma
ZOU Yong, YU Changwen
DOI: 10.3969/j.issn.1006-6233.2018.03.002
Objective: To evaluate the association of circulating and tumor tissue miR-146a expressions with the clinicopathological properties and their correlations with prognosis in surgical patients with intrahepatic cholangiocarcinoma (ICC). Methods: 66 ICC patients were enrolled in the study. ICC tissues sample and plasma sample before operation were collected. The expression of miR-146a was determined by quantitative polymerase chain reaction (qPCR). Results: MiR-146a expressions in plasma were positively associated with that in tumor tissue (R=0.342, P=0.005). Tissue miR-146a was negatively correlated with poor differentiation (R=-0.253, P=0.041), TNM stage (R=-0.316, P=0.010), as well as ECOG performance (R=-0.386, P=0.001), while plasma miR-146a was inversely associated with N stage (R=-0.251, P=0.042) and TNM stage (R=-0.282, P=0.022). Kaplan-Meier curves suggested that high expression of tissue miR-146a was correlated with prolonged overall survival (OS) (P<0.001), while plasma miR-146a didn't (P=0.061). For the purpose of evaluating the prognostic value of tumor tissue or plasma miR-146a for NSCLC patients, univariate Cox regression analysis was performed, which indicated that tissue miR-146a could predict better OS (P<0.001); ECOG high performance (P=0.036), T high stage (P=0.005), N high stage (P=0.032), M high stage (P=0.006), and TNM high stage (P=0.019) could predict worse OS. Furthermore, multivariate Cox regression analysis revealed that only tissue miR-146a high expression was an independent factor for prolonged OS (P=0.001), while T high stage was a risk factor for OS. Conclusion: The expression level of miR-146a in tumor tissue and plasma is correlated with the clinicopathological characteristics of ICC patients, and miR-146a in tumor tissue may serve as a credible prognostic marker for ICC patients.

DOI: 10.3969/j.issn.1006-6233.2018.03.003
Objective: To investigate the expression of miR-381 in tissues of esophageal cancer and its clinical significance. Methods: 108 cases of esophageal cancer admitted to our hospital were selected as the research object, and the QRT-PCR method was used to detect the expression of miR-381 in postoperative cancer tissues and adjacent normal tissues.Analyzed the relationship between the expression of miR-381 and the clinicopathological features of esophageal cancer patients. Results: The expression of miR-381 in esophageal cancer tissues was significantly lower than that in adjacent normal tissues (0.47 ± 0.22 vs 1.02 ± 0.97),and the difference was statistically significant (P <0.05). The expression level of miR-381 in esophageal cancer was closely related to the TNM staging, differentiation degree, infiltration degree and lymph node metastasis (P <0.05), and the expression level of miR-381 in esophageal cancer tissues had no significant correlation with clinicopathological features such as gender, age, tumor diameter and tumor location (P> 0.05). Conclusions: MiR-381 in esophageal cancer is low expression, and its expression level is closely related to the occurrence and development of esophageal cancer, miR-381 may be a potential biomarker for the diagnosis of esophageal cancer and a new therapeutic target.

YUAN Changhe, WEI Jie
DOI: 10.3969/j.issn.1006-6233.2018.03.004
Objective: To explore the value of shear wave elastography (SWE) combined with breast imaging reporting and data system (BI-RADS) grading in the differential diagnosis of breast masses and its relationship with pathological features. Methods: 96 patients with breast masses were examined by conventional breast ultrasonography and SWE, and the BI-RADS grading and modified SWE scoring were performed. With the diagnostic results of pathology as the golden standard, the diagnostic efficiency of BI-RADS grading and its combination with SWE in the differential diagnosis of breast masses was evaluated by the area under receiver operating characteristic (ROC) curve (AUC). The strain ratio of ultrasonic elastography of normal tissues and mass tissues was measured by SWE, and its relationship with pathological features was analyzed. Results: In the 96 patients, there were benign and malignant masses in 67 cases and 29 cases respectively diagnosed by pathology. There were statistically significant differences of BI-RADS grade and SWE score between benign and malignant masses (P < 0.05). The ROC curve showed that the AUC of BI-RADS grading and SWE combined with BI-RADS grading in the differential diagnosis of breast masses were 0.83 and 0.91 respectively. The specificity, positive predictive value, negative predictive value of SWE combined with BI-RADS grading in the diagnosis of benign and malignant breast masses were significantly higher than those of BI-RADS grading alone. In the 96 patients, the strain ratio of ultrasonic elastography was lower than 3.08 in 68 cases and more than 3.08 in 28 cases, with statistically significant differences between the two in the shape of masses, boundary, aspect ratio, echo, calcification, blood flow and other pathological features (P < 0.05). Logistic regression and multivariate analysis showed that shape of masses, boundary, aspect ratio, echo, calcification and blood flow were independent influencing factors of elasticity strain ratio larger than 3.08 (P < 0.05). Conclusion: SWE combined with BI-RADS grading can fully display the histological and pathological features of breast masses, which is helpful for the differential diagnosis of benign and malignant breast masses.
