177 Effects of Rho-Associated Kinase on High Glucose-Induced Murine Heart Endothelial Cell Senescence
XU Changwu, QIU Liqiang, JIANG Hong
DOI: 10.3969/j.issn.1006-6233.2018.02.001
Objective: To investigate the role of Rho-associated kinase in the high glucose-induced murine heart endothelial cells (MHECs) senescence. Methods: MHECs senescence was induced by culturing with 22 mM glucose for 72 h. MHECs in control group were treated with normal culture media and the MHECs in High glucose (HG) group were treated with 22 mM glucose. MHECs in Y27632 group were pretreated with Rho-associated kinase inhibitor Y27632 before cultured in high glucose. In addition, mannitol was added to exclude the effect of osmotic pressure. The ratio of senescent cells was identified by senescence-associated β -galactosidase (SA-βG) staining and flow cytometry was used to analyze the cell cycle. The expression of senescence associated protein p21, p-MBS(Thr853) and MBS was explored by Western blot. The telomerase activity in endothelial cells and the level of nitric oxide (NO) in culture medium assay kit were detected with respective kits. Results: Compared to the control group, treatment of high glucose could significantly increase the ratio of SA-βG positive cells (44.00%±3.63% vs. 4.17%±0.70%,P<0.01), enhance ROCK activity, increase the ratio of cells in G0/G1 stage and the expression of p21, inhibit telomerase activity of the cells and NO level in culture medium (all P<0.05). However, treatment of ROCK inhibitor Y27632 could greatly inhibit ROCK activity and effectively decrease the ratio of senescent cells (both P<0.05). Also, treatment of Y27632 could decrease the ratio of cells in G0/G1 stage and the expression of p21 and reverse the decrease of telomerase activity and NO level in cells cultured in high glucose medium (all P<0.05). Conclusion: Rho related kinase activation is involved in high glucose induced endothelial cell senescence, and inhibition of Rho related kinase can reduce the senescence of endothelial cells induced by high glucose, and provide a new direction for the prevention and treatment of diabetic angiopathy.

DOI: 10.3969/j.issn.1006-6233.2018.02.028
Objective: To analyze the distribution of all kinds of irregular antibodies causing hemolytic disease of the newborn (HDN) in Rh blood group system to provide important basis for clinical prevention, diagnosis and the treatment of Rh-HDN. Methods: The ABO and Rh blood group identification (neonatal ABO blood group identification was done only by positive test) by the tube method. Three tests of neonatal hemolysis were carried out by microcolumn gel method. The antibody screening and antibody identification were carried out by microcolumn gel method and polyamine method. The application of 2 test were compared between the two groups. Result: In 246 cases of compatible ABO blood type of the parents, there were 61 cases of irregular antibodies detected in Rh blood group system (24.60%), and the distribution of antibodies were anti-D(32 cases), anti-E(2 cases), anti-cE(4 cases), anti-C(3 cases), anti-c(2 cases) and anti-Ce(4 cases) respectively. In 273 cases of incompatible ABO blood type of the parents, there were 25 cases of irregular antibodies detected in Rh blood group system (8.93%), and the distribution of antibodies were anti-D(15 cases), anti-E(2 cases), anti-cE(4 cases), anti-C(2 cases), anti-c(1 case) and anti-Ce(1 case) respectively. The detected number of Rh antibodies in the group without ABO antibodies was higher than the number of the group with ABO antibodies. The differences was statistically significant (P<0.05). Conclusion: Anti-D is still the main cause of Rh-HDN. However, the detection rates of anti -E and anti-cE were also high in other Rh systems, especially in the samples of the same or compatible ABO blood group. We should attach importance to the RhD positive pregnant women. If the ABO blood group is the same or coincide, the screening of unexpected antibodies in Rh blood group system is of great clinical significance for the prevention and early treatment of Rh-HDN.
