Abstract:Objective: To observe the growth and apoptosis rate of recombinant human endostatin combined with paclitaxel in breast cancer cell line SK-BR-3. Methods: The growth of cells was observed under light microscope. The apoptosis rate of breast cancer cell SK-BR-3 was detected by flow cytometry. The expression of apoptosis related protein Bcl-2/Bax was observed by Realtime PCR method. The apoptosis inducing effects of different administration protocols on breast cancer cell line SK-BR-3 were compared. Results: Apoptosis of Endostar 1 g/ml after 48h was 11.6% (P<0.05), Bcl-2 expression, and the expression of Bax was not changed. The rate of apoptosis was 12.4% (P<0.05) after paclitaxel was 1 g/ml, and the expression of Bcl-2 was down regulated, while the expression of Bax was increased by 24h. Paclitaxel and sequential administration of Endostar respectively, the apoptosis rates were 16.50% and 14.18% (P<0.05), and can down regulate the expression of Bcl-2, increase the expression of Bax. The apoptosis rate of SK-BR-3 cells was 22.74%, which was significantly higher than that of other administration group (P<0.05), and the expression of Bcl-2 was down regulated and the expression of Bax was elevated at the same time when the two groups were combined and administered at the same time. Conclusion: Both endostar and paclitaxel can inhibit the growth of breast cancer cell line SK-BR-3 and induce apoptosis. The two drugs simultaneously produced the best effect.
