Abstract:Objective: To investigate the influences of genistein (GS) on the migration and invasion of non-small cell lung cancer (NSCLC) by inhibiting osteopontin (OPN)/focal adhesion kinase (FAK) signaling pathway. Methods: The expression levels of OPN and FAK mRNA in four NSCLC cell lines (H596,H520,A549,H1703) and normal human lung epithelial cells (BEAS-2B) were detected by qRT-PCR; taking A549 cells as the research target,the control group,the GS low concentration (GS-L) group (10μmoL/L),the GS medium concentration (GS-M) group (20μmoL/L),and the GS high concentration (GS- H) group (40μmoL/L),recombinant osteopontin (rOPN) group (15ng/mL),and GS-H+rOPN group (40μmoL/L GS+15ng/mL rOPN) were set.The proliferation rate and apoptosis rate of the cells in the above groups were detected by MTT method and flow cytometry respectively; scratch test and Transwell test were applied to detect changes in cell migration and invasion; Western blot and qRT-PCR were applied to detect the expression of OPN/FAK signaling pathway related genes and proteins. Results: Compared with BEAS-2B cells,the expression levels of OPN and FAK mRNA in A549 cells changed most obviously (P<0.05).Compared with the control group,the proliferation rate,migration rate,invasion and migration number,expression of OPN/FAK signaling pathway-related gene and protein,and apoptosis rate of A549 cells in GS-L,GS-M,and GS-H groups decreased obviously,and the apoptosis rate increased obviously (P<0.05); the proliferation rate,migration rate,invasion and migration number,expression of OPN/FAK signaling pathway-related gene and protein,and apoptosis rate of A549 cells in rOPN group increased obviously,and the apoptosis rate decreased obviously (P<0.05).Compared with the GS-H group,the proliferation rate,migration rate,invasion and migration number,expression of OPN/FAK signaling pathway-related gene and protein,and apoptosis rate of A549 cells in GS-H+rOPN group increased obviously,and the apoptosis rate decreased obviously (P<0.05).Compared with the rOPN group,the proliferation rate,migration rate,invasion and migration number,expression of OPN/FAK signaling pathway-related gene and protein,and apoptosis rate of A549 cells in GS-H+rOPN group decreased obviously,and the apoptosis rate increased obviously (P<0.05). Conclusion: GS can inhibit the migration,invasion and proliferation of NSCLC-A549 cells and promote their apoptosis,which may be related to the inhibition of OPN/FAK signaling pathway.
