Abstract:Objective: To evaluate the value of metagenomic next generation sequencing (mNGS) in the diagnosis of tuberculosis. Methods: 4 suspected tuberculosis patients were collected, and their alveolar lavage fluid (BALF) or pleural effusion were detected by mNGS. Chinese and English literatures from Wanfang, CNKI and PubMed databases were searched and analyzed. Results: 2 of the 4 patients with pleural effusion mNGS indicated Mycobacterium tuberculosis (MTB) and MTB complex, clinically diagnosed as tuberculous pleurisy. BALF acid-fast staining was positive in 2 patients, and the result of BALF mNGS indicated MTB complex group, which confirmed the diagnosis of pulmonary tuberculosis. A total of 154 literatures in Chinese and English were searched to provide mNGS, traditional culture, and GeneXpert MTB/RIF (Xpert) detection data as inclusion criteria. Finally, a total of 453 patients diagnosed with MTB infection were included. Conclusion: mNGS is more sensitive to the diagnosis of MTB infection than Xpert and traditional culture. Combined application can significantly improve the sensitivity of tuberculosis detection.
