Abstract:Objective: To detect the expression of serum long non-coding RNA 00324 (Linc00324) and T helper cell 17(Th17) cytokine in patients with atopic dermatitis and to explore its role and relationship in the pathogenesis of atopic dermatitis.Methods: 80 patients with atopic dermatitis confirmed by pathology in our hospital from October 2017 to November 2019 were selected as the study objects,which were divided into 26 patients with mild atopic dermatitis,24 patients with moderate atopic dermatitis,30 patients with severe atopic dermatitis,and 35 healthy people were selected as the control group.The expression of Linc00324 in serum was detected by real-time fluorescent quantitative PCR (qRT PCR).The levels of interleukin-4(IL-4) and interleukin-10(IL-10) in Th17 cells were detected by enzyme-linked immunosorbent assay (ELISA),and Pearson method was used to analyze the correlations between serum Linc00324 and the expression levels of Th17 cytokines IL-4,IL-10 in patients with atopic dermatitis.Results: The levels of serum Linc00324,Th17 cytokines IL-4 and IL-10 in patients with atopic dermatitis were significantly higher than those in the control group (P<0.05),the levels of serum Linc00324,Th17 cytokines IL-4 and IL-10 in severe group and moderate group were significantly higher than those in mild group (P<0.05),and the levels of serum Linc00324,Th17 cytokines IL-4 and IL-10 in severe group were significantly higher than those in moderate group (P<0.05).The expression of Linc00324 in serum of patients with atopic dermatitis was positively correlated with the levels of Th17 cytokines IL-4 and IL-10 (r = 0.516,r = 0.498,P<0.05).Conclusion: Linc00324 is highly expressed in the serum of patients with atopic dermatitis,the levels of Th17 cytokines IL-4 and IL-10 are significantly higher in patients with atopic dermatitis,which are closely related to the occurrence and development of atopic dermatitis.It is suggested that Linc00324 can be used as a potential biological index for the early diagnosis and assessment of atopic dermatitis with important clinical significance.
