Abstract:Objective: To study the expression and significance of microRNA-125b (mir-125b) and placental growth factor (PIGF) in villi and serum of women with early abortion.Method: 105 cases of early spontaneous aborted women admitted to our hospital from July 2017 to October 2019 were selected as the research objects,and 105 cases of induced abortion in the same period were selected as the control group.Peripheral venous blood serum was collected and chorionic tissue was separated from all abortion tissues.Real-time quantitative PCR was used to detect the mRNA expression levels of miR-125b and PIGF in serum and villus tissue,and immunoblotting (WB) was used to detect the protein expression levels of PIGF in villus tissue.The serum PIGF level was detected by enzyme linked immunosorbent assay (ELISA),and the correlation of mir-125b and PIGF protein levels in serum and chorionic tissue was analyzed by Pearson method.The predictive value of serum mir-125b and PIGF levels in patients of spontaneous abortion in early pregnancy was analyzed by receiver operating characteristic (ROC).Result: Compared with the control group,the level of mir-125b in chorionic tissue of pregnant women in spontaneous abortion group increased significantly,and the level of PIGF mRNA and protein decreased significantly (P<0.05);The level of miR-125b in serum increased significantly and the level of PIGF decreased significantly;The difference was statistically significant (P<0.05).Pearson analysis showed that there was a negative correlation between mir-125b level and PIGF protein level in chorionic tissue,serum of spontaneous abortion pregnant women (r=-0.643,P<0.05).Serum levels of mir-125b and PIGF were 0.896 (95% CI:0.850-0.943) and 0.884 (95% CI:0.838-0.929) respectively for predicting the area under curve (AUC) of spontaneous abortion in early pregnancy.The sensitivity and specificity of mir-125b (>1.630) in predicting spontaneous abortion were 95.30% and 84.60%,respectively.The sensitivity and specificity of PIGF (<42.553 pg/mL) in predicting spontaneous abortion were 90.60% and 77.90%;the AUC,specificity and sensitivity of combined detection for predicting spontaneous abortion in early pregnancy were 0.954,92.30% and 89.60%,respectively.Conclusion: The levels of mir-125b and PIGF in chorionic tissues of women suffering early spontaneous abortion are up-regulated,and they are negatively correlated,which may have some predictive value for the occurrence of spontaneous abortion.
