Abstract:Objective: To investigate the effect and mechanism of tetramethylpyrazine on cerebral ischemia-reperfusion injury in rats. Methods: A rat model of cerebral ischemia-reperfusion injury was established by suture-occluded method. The effects of tetramethylpyrazine on the infarct size, brain water content, neurological symptoms and pathological morphology of brain, as well as the expression of IL-6, TNF-α and MCP-1 in serum and brain tissue of rats were evaluated. The protein expression levels of VEGF, PI3K, P-AKT and AKT in rat brain were detected by Western Blot. Results: Tetramethylpyrazine has protective effects on cerebral ischemia-reperfusion injury, which can alleviate cerebral infarct size , brain water content, neurological symptoms and pathological morphology (P<0.05). In addition, it can down-regulate the expression of pro-inflammatory cytokines IL-6, TNF-α and MCP-1 and increase the expression of VEGF, PI3K and P-AKT/AKT in brain tissue (P<0.05). Conclusion: Tetramethylpyrazine pretreatment has a protective effect on cerebral ischemia-reperfusion, which may be related to the activation of VEGF-mediated PI3K-AKT signaling pathway.
