Abstract:Objective: To investigate the effect of procaine on proliferation and apoptosis of cervical cancer cells and its mechanism. Methods: SiHa cells were treated with procaine with final concentrations of 0.0 mm / L, 0.5 mm / L, 1 mm / L, and 2 mm / L. pcDNA and pcDNA trail were transfected into untreated SiHa cells respectively, which were recorded as pcDNA group and pcDNA trail group. After Si NC and Si trail were transfected into SiHa cells, they were treated with procaine with 2 mm / L, which were recorded as procaine 2 mm / L + Si NC group and procaine 2 mm / L + Si trail group, respectively The liposome method was used. MTT method was used to detect cell proliferation; flow cytometry was used to detect apoptosis; QRT PCR was used to detect trail mRNA expression; Western blot was used to detect protein expression. Results: Procaine can inhibit the expression of CyclinD1, Bcl-2, p21, Caspase-8 and Bax, inhibit the proliferation of SiHa, promote apoptosis and promote the expression of trail. Overexpression of trail inhibited the proliferation of SiHa and promoted apoptosis. Inhibition of TRAIL expression reversed the inhibition of proliferation and apoptosis of cervical cancer cell line SiHa by procaine. Conclusion: Procaine can inhibit the proliferation of SiHa and promote the apoptosis of cervical cancer cells, which may be related to the regulation of trail. It will provide new ideas and targets for the treatment of cervical cancer.
