Abstract:Objective: To explore the expressions and significance of Th17, Treg, and related cytokines in peripheral blood in patients with SLE. Methods: A total of 65 patients with SLE admitted in our hospital from May 2016 to April 2017 were included in the study and divided into the activity group (n=33) and the non-activity group (n=32) according to SLEDAI. Moreover, 25 healthy individuals who came for physical examinations were served as the control group. The fasting peripheral venous blood was collected. The scattering velocity immunoturbidimetry was use to detect the serum IgA, IgG, IgM, C3, C4 levels. FCM was used to detect the expressions of Th17 and Treg. ELISA was used to detect the expressions of IL-6, IL-17, IL-23, and TGF-β. Results: IgA, IgG, and IgM in the activity group were significantly higher than those in the non-activity group and the control group, while C3 and C4 were significantly lower than those in the non-activity group and the control group. IgA and IgG in the non-activity group were significantly higher than those in the control group. Th17 and Th17/Treg in the activity group were significantly higher than those in the non-activity group and the control group, while Treg was significantly lower than that in the non-activity group and the control group. Th17 and Th17/Treg in the non-activity group were significantly higher than those in the control group, while Treg was significantly lower than that in the control group. IL-6, IL-17, and IL-23 in the activity group were significantly higher than those in the non-acitivity group and the control group, while TGF-β was significantly lower than that in the non-acitivity group and the control group. IL-6, IL-17, and IL-23 in the non-activity group were significantly higher than those in the control group. Conclusion: The expressions of peripheral blood Th17, Treg, and related cytokines in patients with SLE are abnormally expressed, and correlated with the disease activity, sugessting that Th17/Treg imbalance plays a vital role in the pathogenesis of SLE.
