Abstract:Objective: To investigate the effects of different concentrations of injectable platelet rich fibrin (iPRF) on the migration, activation, proliferation and tumorigenicity of melanocytes. Methods: Control group, experimental group 1 and experimental group 2 were configured with no iPRF, iPRF concentration of 1/3, iPRF concentration of 1/2, respectively. The melanocyte migration, cell proliferation, melanin concentration and tumorigenicity were compared between groups. Results: Comparison of the three culture systems, the migration of melanocytes was the most obvious in experimental group 2, followed by experimental group 1, and the least in control group. There was no statistical difference in the proliferation of melanocytes between groups (P>0.05). The amount of melanin was the highest in the experimental group 1, and there was no statistical difference between the control group and the experimental group 2 (P>0.05). In the three culture systems, the three groups showed a dose-dependent downregulation of colony formation rate of melanocyte, but the number of clones in each group was less than 50, and the clone plate test was negative. Conclusion: Different culture systems have different migration and activation effects on melanocytes. The migration and activation effect of melanocyte showed a downward trend with the increase of iPRF concentration. iPRF may promote the production of melanin, but not the higher the concentration, the stronger the effect. iPRF did not promote the proliferation of melanocytes and had no tumorigenic effect.
