Abstract:Objective: To investigate the expression of miR-191-5p in the plasma of patients with IgA nephropathy and its correlation with renal interstitial fibrosis (RIF). Methods: 80 patients with IgA nephropathy (IgA nephropathy group) admitted to our hospital from February 2017 to February 2022 were divided into stage Ⅰ~Ⅱ group (42 cases), stage Ⅲ group (38 cases) according to CKD stages. Patients were divided into the T0 group (n = 20), T1 group (n = 39), and T2 group (n = 21) according to Oxford parting, and 77 healthy volunteers were selected as the control group. Plasma miR-191-5p expression was detected by Quantitative real-time polymerase chain reaction (qRT-PCR), renal function [creatinine (Scr), urea nitrogen (BUN)] were detected by the automatic biochemical analyzer, and RIF indexes [transforming growth factor TGF -β1 and TGF-α]were detected by enzyme-linked immunosorbent assay. Pearson analysis was conducted to understand the correlation between miR-191-5p expression and renal function and RIF index and RIF area. Results: The levels of serum miR-191-5P in the IgA nephropathy group were lower than those in the control group (P<0.05), and the levels of Scr, BUN, TGF -β1, and TGF-α were higher than those in the control group (P<0.05). The expression of miR-191-5p in the stage Ⅲ group was lower than that in the stageⅠ~Ⅱ group (P<0.05), Scr, BUN, TGF-β1, and the area of RIF was higher than that in stageⅠ~Ⅱ group (P<0.05). The expression of miR-191-5p in the T2 group was lower than that in the T1 group and T0 group (P<0.05), Scr, BUN, TGF-β1, and the area of RIF was higher than that in the T1 group and T0 group (P<0.05). Plasma miR-191-5p expression was negatively correlated with RIF area, Scr, BUN, TGF-β1, and TGF-α in patients with IgA nephropathy (r=-0.509, -0.379, -0.311, -0.475, -0.486,P<0.05). Conclusion: The expression of miR-191-5p in plasma is decreased in patients with IgA nephropathy and is related to RIF and renal function decline.
