Abstract：Objective: To identify miRNAs regulated by Helicobacter pylori infection and their mechanism of action in gastric cancer. Methods: Expression levels of miR-125a-5p and EVA1A were assessed using qRT-PCR in Hp-infected patient tissues.The biological functions of the miR-125a-5p/EVA1A axis in gastric cancer cell proliferation,migration and invasion were investigated using cell functional experiments after Hp infection.The binding relationship between miR-125a-5p and EVA1A was confirmed through a dual luciferase assay. Results: miR-125a-5p was down-regulated in Hp-infected patient tissues and Hp-infected gastric cancer cells.Functional experiments revealed that the overexpression of miR-125a-5p could inhibit gastric cancer cell proliferation and invasion.The targeting of miR-125a-5p down-regulated the expression of EVA1A,and overexpressing EVA1A could reverse the effects of miR-125a-5p overexpression on gastric cancer phenotype. Conclusion: Hp-induced miR-125a-5p,which targets EVA1A,inhibits the proliferation and invasion of gastric cancer.MiR-125a-5p and EVA1A may represent potential therapeutic targets for gastric cancer.
