Abstract:Objective: To investigate the relationship of leucine-rich G-protein-coupled receptor 5(LGR5), human epidermal growth factor receptor 2 (Her-2), Ki-67 and clinicopathologic parameters and prognosis in colorectal cancer (CRC). Methods: Clinical data of 132 CRC patients admitted to the First People's Hospital of Fangchenggang City from January 2020 to July 2021 were retrospectively analyzed. The expressions of LGR5, Her-2 and Ki-67 in CRC tissues were detected by immunohistochemical method, and the relationship between them and clinicopathological parameters was analyzed. After 12 months of follow-up, patients were divided into good prognosis group (114 cases) and poor prognosis group (18 cases) according to the prognosis. Receiver operating characteristic curve (ROC curve) was used to analyze the prognostic values of LGR5 and Her-2 in CRC patients. Results: The positive expression rates of LGR5, Her-2, and Ki-67 in CRC tissues were significantly higher than those in adjacent normal tissues (P<0.05). The protein expressions of LGR5, Her-2, and Ki-67 were not significantly correlated with gender, age, tumor site, and differentiation degree of CRC patients (P>0.05). The expressions of LGR5 and Ki-67 were correlated with the degree of invasion, TNM stage, lymph node metastasis and distant metastasis (P<0.05). The expression of Her-2 protein was not correlated with the degree of invasion (P>0.05), but with TNM staging, lymph node metastasis and distant metastasis (P<0.05). The positive expression rates of LGR5, Her-2, and Ki-67 protein in poor prognosis group were higher than those in good prognosis group (P<0.05). ROC curve showed that the combination of LGR5 and Her-2 protein (AUC=0.804, P<0.01) had higher predictive value. Conclusion: LGR5, Her-2, and Ki-67 are highly expressed in CRC tissues, and their expression levels are closely related to the clinicopathological parameters of patients. The combined detection of LGR5 and Her-2 has high predictive value for the prognosis of patients.
