Abstract:Objective: To culture a polyclonal antibody against the oprC protein of the outer membrane receptor of Acinetobacter baumannii (A.baumannii) and to evaluate its immune effect. Methods: The full gene sequence of oprC was queried on NCBI, and the full length of oprC was amplified using A.baumannii DNA as a template. The antibody titer was determined by enzyme-linked immunosorbent assay (ELISA), and the results showed that the potency of oprC antiserum was 1∶12800. The improvement of the survival rate, the number of bacteria in the blood, and the damage of multiple organ tissues of the mice challenged with oprC protein polyclonal antibody was detected. Results: The oprC polyclonal antibody could improve the survival rate of mice, reduce the load of bacteria (P<0.05), and relieve tissue damage to the lung, spleen, and kidney to some extent. Conclusion: This study confirmed that oprC recombinant protein can improve the survival rate of mice, reduce pathological damage, and achieve immune protection.
