Study on the Relationship Between Mitogen-Activated Protein Kinase MAP2K5-Rs4776970 Polymorphism and Abnormal Metabolism and Chronic Vascular Inflammation
Abstract:Objective: To investigate the relationship between mitogen-activated protein kinase MAP2K5-rs4776970 single nucleotide polymorphism (SNP) and abnormal metabolism and chronic vascular inflammation. Methods: A total of 1224 patients were included in the physical examination. According to the revised cholesterol standard (ATPⅢ*),they were divided into the metabolic syndrome (MS) group and the non-MS group,of which 520 were in the MS group and 704 were in the non-MS group. Real-time fluorescence quantitative PCR combined with TaqMan-MGB probe for SNP detection. High-sensitivity C-reactive protein (hs-CRP) is an immunoturbidimetric method. Results: The frequency of AA (7.5% vs 4.0%) distribution of MAP2K5-rs4776970 in the MS group was significantly increased (P<0.05).As to the distribution frequency of TT type (56.2% vs 59.5%)and TA type (36.3% vs 36.5%),there was no significant difference between the two groups (P=0.954). The frequency of A allele was significantly higher in the MS group than in the non-MS group (25.7% vs. 22.2%,P=0.048). After adjusting age and gender by gender and age,Logistic regression analysis indicated that the incidence of MS had no significant correlation with MAP2K5- rs4776970 genotype (OR=0.851,95%CI:0.7~1.035,P=0.106). Correlation analysis suggested that MAP2K5-rs4776970 genotype was associated with waist circumference (WC),body mass index (BMI) and Hs-CRP (P=0.015; P=0.011; P=0.049),compared with MAP2K5-rs4776970TT and TA genotypes,WC,BMI and Hs-CRP levels in AA genotype were significantly increased (P<0.01). Conclusion: MAP2K5-rs4776970 gene polymorphism may be related to obesity and chronic inflammation of blood vessels,but it is not an independent risk factor for MS.
