Abstract:Objective: To explore quantitative parameters obtained by dynamic contrast-enhanced magnetic resonance imaging (DCE MRI) with gadolinium ethoxybenzyl diethylenetriamine pentaacetic acid (Gd-EOB-DTPA) in diagnosing and discriminating liver fibrosis (LF) in a rabbit model. Methods: LF group was established in 60 rabbits by the injection of 50% CCl4 oil solution, whereas 20 rabbits injected with normal saline served as the control group. 15 objects in the LF group and 5 in the control group were randomly selected at the end of the 4th, 5th, 6th, 15th week after injection of 50% CCl4 oil solution, respectively. All rabbits underwent pathological examination to determine the LF stage using the Metavir classification system. DCE MRI was performed, and quantitative parameters, including trans, Kep , Ve, Vp and Re were measured and compared among the different LF stages. Results: In all, 16, 14, 12, 10, 11 rabbits of stage F0, F1, F2, F3 and F4, respectively, were included in this study. trans (r=0.913) increased, Kep (r =-0.757) and Re (r =-0.804) decreased with LF stage progression (P<0.001), while no significant correlation was found for Ve or Vp. The AUC of trans was higher than that of other quantitative parameters, with an AUC of 0.96, 0.96, 0.96 and 0.99 for staging ≥F1、≥F2、≥F3、≥F4, respectively. Conclusion: Gd-EOB-DTPA DCE MRI parameters are closely associated with LF progression, indicating that this may be a promising method for quantitatively differentiating early-stage LF.
