Expression of MicroRNA-145 in Peripheral Blood of Children with Lower Respiratory Tract Infection of Respiratory Syncytial Virus and Its Association with Th1/Th2 Balance

Abstract:Objective: To explore the relationship between miR-145 and Th1/Th2 balance by detecting the levels of miR-145 and Th1/Th2 cytokines in peripheral blood of children with lower respiratory tract infection caused by respiratory syncytial virus (RSV). Methods: 60 children with RSV bronchiolitis and pneumonia who were admitted to our hospital from June 2018 to November 2019 were selected as the experimental group, and 60 healthy children who had health examination in our hospital during the same period were selected as the control group. Real-time quantitative PCR (qRT-PCR) was adapted to detect the expression of miR-145 in peripheral blood mononuclear cells of all subjects, Th1 subgroup and Th2 percentage were detected by double fluorescence flow cytometry, and the expression levels of Interleukin-2(IL-2), Interleukin-4(IL-4), Interferon-γ(IFN-γ) and Interleukin-5(IL-5) in plasma of all subjects were detected by enzyme-linked immunosorbent assay (ELISA). Results: The expression level of miR-145 in the experimental group was significantly higher than that in the control group (P<0.05); the percentage of Th1 cells and Th1/Th2 ratio in the experimental group were significantly lower than those in the control group (P<0.05), and there was no significant difference between the two groups (P>0.05); the expression level of IL-2 and IFN-γ in plasma of experimental group was significantly lower than that of control group, and the expression level of IL-4 and IL-5 was significantly higher than that of control group (P<0.05); the levels of miR-145 of the experimental group were negatively correlated with the levels of IL-2, IFN-γ, Th1/Th2 ratio (P<0.05), and positively correlated with the levels of IL-4 and IL-5 (P<0.05). Conclusion: The up-regulation of miR-145 expression in peripheral blood of children with RSV lower respiratory tract infection is closely related to Th1/Th2balance.
