Abstract:Objective: To observe its effect on leukemia cell apoptosis by using Ad-GINS2 infection. Methods: NB4 cells and K562 cells were infected by Ad-GINS2 and using fluorescence microscope observe the infection effect of NB4 cells and K562 cells; Apoptosis and cell cycle distribution of NB4 cells and K562 cells were detected by FCM;Bax and bcl-2 protein were detected by Western blotting test. Results: Fluorescence were observed by fluorescence microscope after 72h; Apoptosis rate was decreased significantly using FCM technology compared with the untreated group and no-load group(P<0.05);Results of cell cycle distribution showed that S, phase rate were increased and G1 phase rate were decreased in NB4 cells compared with the untreated group and no-load group(P<0.05);S G2 phase rate was increased and G1 phase rate were decreased in K562 cells compared with the untreated group and no-load group(P<0.05); Bax protein was decreased and bcl-2 protein was increased detected by Western blotting test. Conclusion: The recombinant adenovirus vector of GINS2 gene successfully infect NB4 and K562 cells and inhibit cell apoptosis, and then found that apoptosis may be related to the process of cell cycle, which lays a foundation for further clarifying the role of GINS2 gene white in leukemia.
