Abstract:Objective: To investigate the effect of apoptosis of somatic proteins from Trichinella spiralis muscle larva on small cell lung cancer H446 cells and correlation of C-kit expression. Methods: The H446 cells were divided into groups, somatic proteins of Trichinella spiralis muscle larva as experiment group, cis-platinum(DDP) as positive control group and untreated cells as negative control group. The effects of somatic protein and DDP on the proliferation activity of H446 cells were detected by MTT colorimetry; the cell apoptosis rate of H446 cells were induced by somatic proteins and DDP by flow cytometry(FCM) respectively; the transcriptional level of C-kit mRNA and protein expression of C-kit protein were detected by Real time PCR and Western Blot method respectively. Results: The results of MTT detection showed that the proliferation of H446 cells decreased with the increase of the concentration of insect body protein and the prolongation of the time of H446 cells, and the insect body protein had a significant inhibitory effect on H446 cells, showing a dose time effect. Flow cytometry showed that somatic proteins and DDP could induce apoptosis of H446 cells. Real time PCR and Western Blot showed that the transcriptional level of C-kit mRNA and protein expression of C-kit protein were decreased of somatic proteins group and DDP group respectively, compared with negative control group. Conclusion: The somatic protein of Trichinella spiralis muscle larvae can inhibit the growth of H446 cells and induce apoptosis of H446 cells. C-kit gene may be one of the molecular targets for the treatment of small cell lung cancer.
