Abstract:Objective: To investigate the expression and significance of matrix metalloproteinase MMP-9, MMP-13 and their inhibitors TIMP-1, TIMP-3 in spinal tuberculosis intervertebral disc tissue. Methods: 48 cases of patients with spinal tuberculosis treated by operation from January 2012 to January 2016 in our hospital were selected as TB group, another 48 cases with disc herniation treated by operation were selected as ID group, and patients with traumatic spinal fractures treated in our hospital in the same period were selected as the control group. Then the positive rate and the level of expression in the three groups were detected by immunohistochemistry assay and real-time quantitative PCR (RT-PCR) technique. Results: The positive rate of MMP-9 was 93.75%, MMP-13 positive rate was 89.58%, TIMP-1 positive rate was 87.50%, TIMP-3 positive rate was 91.67% in TB group, which were all significantly higher than those in ID group (70.83%, 66.67%, 68.75%, 64.58%) and the control group (20.83%, 16.67%, 18.75%, 10.42%) (X2=56.541,54.665,49.886 and 66.544,respectively, P<0.05); The expression levels of MMP-9, MMP-13, TIMP-1, TIMP-3TB in intervertebral disc tissue group were significantly higher than those in ID group and the control group (F=12.451,37.423,17.483 and 9.167,respectively, P<0.05), the ratio of MMP-9 / TIMP-1 and MMP-13 / TIMP-3 all increased. Conclusion: MMP-9, MMP-13, TIMP-1, TIMP-3 are highly expressed in spinal tuberculosis disc tissue, which result in the imbalance of MMP-9 / TIMP-1, MMP-13 / TIMP-3, indicating that the matrix metal proteases (MMPs) and matrix metalloproteinase inhibitors (TIMPs) may participate in the damages of spinal tuberculosis intervertebral disc.
