Abstract:Objective: To evaluate the effect of platelet derived growth factor receptor alpha(PDGFRA) on the proliferation and invasion of lung cancer cells. Methods: The mRNA expression level of PDGFRA in eight lung cancer cell lines and normal lung cell HBE was detected by PT-PCR. Two groups of lung cancer lines(A549) from the high expression level of PDGFRA were selected, then stable knock down and over-expression PDGFRA cell lines were constructed through lentiviral transfection. Furthermore, the effect of PDGFRA on cell proliferation and invasion in these cells were evaluated by MTS, scratch test, Transwell and western-blot test. Results: Over-expression of PDGFRA showed that cell proliferation and invasion significantly increased, meanwhile, E-candherin and Vimentin also increased, scratch test and transwell revealed that PDGFRA significantly improved the cell invasion. Whereas, knock down of PDGFRA decreased the cell proliferation, the expression of E-candherin and Vimentin decreased and the cell invasion also significantly also decreased compared with the control group. Conclusion: PDGFRA can promote the cell proliferation and contribute to the cell invasion in lung cancer cells.
