Abstract:Objective: To discuss insulin-like growth factor receptor 1 (IGF-1) and its receptor (IGF-1R) expression in scar tissue and the effect of ribozyme on the expression level. Methods: 18 cases of Pathological scar tissue from operation excision (9 cases of scar keloid, 9 cases of scar hyperplasia ) were selected in Department of Burn and Plastics, Guangzhou Red Cross Hospital from 2015 May to 2016 July, meanwhile 20 cases of normal skin tissue specimens were collected. The expression level of tissue was observed by using hematoxylin eosin staining, the expression level of IGF-1 mRNA in each tissue specimen and its receptor mRNA were detected by using real-time fluorescence quantitative PCR, scar tissue fibroblasts were derived to culture cells to obtained IGF-1 and its receptor ribozyme, the cells in the logarithmic growth phase with good growth state in 2-4 generation + ribozyme were selected to culture,72h later, the expression level of IGF-1 mRNA and its receptor mRNA were detected by using real-time fluorescence quantitative PCR. Results: The differences were statistically significant in the expression level of IGF-1 mRNA in normal skin, hypertrophic scar and keloid tissue and its receptor mRNA (P <0.05), the comparison between groups showed that normal skin hypertrophic scar tissue < hypertrophic scar tissue < scar keloid tissue and the differences were statistically significant (P <0.05).Conclusion: The expression level of IGF-1 mRNA in scar tissue and its receptor mRNA are significantly higher than those in normal tissues, ribozyme has the effect on blockade of IGF-1 mRNA and its receptor mRNA expression, which may play a role in inhibiting scar formation.
