Abstract:Objective: To investigate the effect and mechanism of injection of raltitrexed on cancer cells in gastric cancer patients. Methods: 84 nude mice were purchased to establish the human gastric cancer cell MGC-803 subcutaneous tumormodel,after successful modeling ,micewere randomly divided into 3 groups, the conventional group received intraperitoneal injection of saline, the raltitrexed group were given intraperitoneal injection of raltitrexed, the 5-Fu group were given intraperitoneal injection of 5-Fu, nude miceweight, tumor volume, tumorweight ,inhibition rate,apoptosis rate, cycle distribution of cancer cellsafter intervention were observed. Results: Nude miceweight, tumor volume ,tumor weight of the conventional groupwere significantly higher than those of the raltitrexed group and the 5-Fu group, the difference had statistical significance (P<0.05);The raltitrexed group'snude mice weight was higher than that of the 5-Fu group, the difference had no statistical significance (P>0.05), tumor volume, tumor weight of the raltitrexed group were lower than those of the 5-Fu group, the difference had statistical significance(P<0.05);The nude mice cancer cells S phase distribution of the conventional group was significantly higher than that of the raltitrexed group and the 5-Fu group, the difference had statistical significance(P<0.05); The S phase distribution of the raltitrexed group and the 5-Fu group had no significant difference, the difference hadno statistical significance (P>0.05);There was no significant difference between the raltitrexedgroup and the 5-Fu group in G0/G1 , the difference hadno statistical significance (P>0.05);G0/G1 phase distribution of the raltitrexed group and the 5-Fu group was lower than that of the conventional group, the difference had statistical significance (P<0.05); G2/M phrase distribution among the three groups has no obvious difference, the difference hadno statistical significance(P>0.05);The conventional group's cancer cell inhibition rate, apoptosis rate were significantly lower than those of the raltitrexed group and the 5-Fu group, the difference had statistical significance(P<0.05); While those of the raltitrexed group were significantly higher than those of the 5-Fu group, the difference had statistical significance (P<0.05). Conclusion: Adoptingraltitrexed for gastric cancer patients has similaranti-tumor effect with 5-Fu, which can effectively inhibit the growth of human gastric cancer cell MGC-803 xenograft tumor in nude mice and induce apoptosis of cancer cells.
