Abstract:Objectives: To investigate the effect and mechanism of allicin on the proliferation of rat vascular smooth muscle cells. Methods: Rat thoracic aortic vascular smooth muscle cells (VSMCs) were cultured in vitro, and 20 ng/mL platelet-derived growth factor BB (PDGF-BB) was used to induce proliferation of VSMCs. The experiment subject was divided into 4 groups: control group (group A), PDGF-BB stimulation group (group B), PDGF-BB + allicin treatment group (group C), PDGF-BB + allicin + lipopolysaccharide treatment group (group D). The proliferation of each group was detected by CCK-8 assay and flow cytometry. Western blotting was used to detect the expression levels of total nuclear factor kappa B p65 (NF-κB p65) and phosphorylated NF-κB p65 (P-p65) in each group. The contents of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in the cell culture supernatants of each group were detected by ELISA. Results: The results of cell proliferation assay showed that compared with group A, the proliferation activity of VSMCs in group B was significantly enhanced. Compared with group B, the proliferation activity of VSMCs in group C was inhibited. Compared with group C, the proliferation activity of VSMCs in group D was enhanced (P<0.05). Western Blot results showed that compared with group A, the expression level of P-p65 protein in group B was significantly increased. Compared with group B, the expression level of P-p65 protein in group C was decreased. Compared with group C, the expression level of P-p65 protein in group D was higher than that in group C (P<0.05). In addition, the total of NF-κB p65 protein levels in each group showed no significant difference (P>0.05).The results of ELISA showed that compared with group A, the levels of TNF-α and IL-1β in group B were increased significantly. Compared with group B, the levels of TNF-α and IL-1β in group C were lower. Compared with group C, the levels of TNF-α and IL-1β in group D were increased (P<0.05). Conclusions:Allicin can inhibit the proliferation of VSMCs induced by PDGF-BB, and the mechanism is related to the inhibition of NF-κB pathway and the reduction of inflammatory response.
